It is an important issue to develop quality assessing system for biofuel for the purpose of accelerating the mass production of biofuel. It is particularly challenging to conduct testing method in the mass production of bioethanol while meeting quality specifications such as ASTM (American Society for Testing & Materials) D4806-10. In order to address this challenge, this paper proposes on-line spectroscopic quality assesment system based on Near Infrared spectrum and Partial Least Squares method in Chemometrics. As a result of testing a number of preprocessing methods and Partial Least Squares, it was found out that Savitzky-Golay method showed the best performance in terms of spectrum correction, noise reduction, and model maintenance. The proposed system allows us to assess multiple quality components continuously using spectroscopic facilities with the cheap cost. Since the value of R2 is more than 0.99, it is possible to replace the laboratory analysis.
To construct useful yeast strain for bioethanol production, we improved yeast harboring various phenotypes by using yeast protoplast fusion method. In this study, S. cerevisiae BYK-F11 strain which have ethanol tolerance, thermotolerance and ${\beta}-glucanase$ activity and P. $stipitis{\Delta}ura$ strain which has xylose metabolism pathway were fused by genome shuffling. P. $stipitis{\Delta}ura$ strain was constructed for protoplast fusion by URA3 gene disruption, resulting in uracil auxotroph. By protoplast fusion, several fused cells were selected and BYKPS-F8 strain (fused cell) showing both karyotypes from two parent strains (S. cerevisiae BYK-F11 and P. $stipitis{\Delta}ura$ strain) among 22 fused cells was finally selected. Sequentially, various phenotypes such as ${\beta}-glucanase$ activity, xylose utility, ethanol tolerance, thermotolerance and ethanol productivity were analyzed. The BYKPS-F8 strain obtained ${\beta}-glucanase$ activity from BYK-F11 strain and 1.2 fold increased xylose utility from P. $stipitis{\Delta}ura$ strain. Also, the BYKPS-F8 strain showed thermotolerance at $40^{\circ}C$ and increased ethanol tolerance in medium containing 8% ethanol. In this fused cell, 7.5 g/l ethanol from 20 g/l xylose was produced and the multiple phenotypes were stably remained during long term cultivation (260 hr). It was proved that novel biological system (yeast strains) is easily and efficiently bred by protoplast fusion among yeasts having different genus.
Bacillus licheniformis was grown in minimal nutrient medium containing 1% (w/v) of distillers dried grain with soluble (DDGS), palm kernel meal (PKM), wheat bran (WB) or copra meal (CM), and the enzyme activity of endoglucanase, ${\beta}$-glucosidase, xylanase and reducing sugars was measured to investigate a possibility of using cost-effective agricultural residues in producing cellulolytic and hemicellulolytic enzymes. The CM gave the highest endoglucanase activity of 0.68 units/mL among added substrates at 48 h. CM yielded the highest titres of 0.58 units/ml of ${\beta}$-glucosidase, compared to 0.33, 0.23, and 0.16 units/mL by PKM, WB, and DDGS, respectively, at 72 h. Xylanase production was the highest (0.34 units/mL) when CM was added. The supernatant from fermentation of CM had the highest reducing sugars than other additional substrates at all intervals (0.10, 0.12, 0.10, and 0.11 mg/mL respectively). It is concluded that Bacillus licheniformis is capable of producing multiple cellulo- and hemicellololytic enzymes for bioethanol production using cost-effective agricultural residues, especially CM, as a sole nutrient source.
On-road source emissions are major air pollutants and have been associated with serious health effects in Seoul metropolis. Thus, it is of fundamental importance to have an accurate assessment of vehicle emissions in order to implement an effective air quality management policy. As a result, there is a need to overview vehicle emission characteristics of air pollutants. This article discusses vehicle exhaust sampling and chemical analysis, emission characteristics of air pollutants, and emission regulations from passenger cars. The vehicle exhaust sampling and chemical analysis methods were described in particulate matter and gaseous compounds. In this article, chassis dynamometer, measurement instrumentation for nano-particulate matter and carbon compounds analysis device were described. For the gasoline and diesel vehicles, the effective parameters of emissions were average vehicle speed, vehicle mileage and model year. The particle number emissions for diesel nano-particles were sensitive to the sampling conditions. Also, the particle number emissions with a diesel particle filter (DPF) largely reduced rather than those without it. This article also describes different emission characteristics of air pollutants according to biodiesel or bioethanol mixing ratio. The Korean emission standards for passenger cars were compared with those of the US and EU. Finally, the objective is to give an overview of relevant background information on emission characteristics of air pollutants from passenger cars in Korea.

In bioethanol from acid hydrolysis process, neutralization of acid hydrolyzate is essential step, which resulted in dissolved cations in glucose solution. Impact of cations to Saccharomyces cerevisiae in glucose solution was investigated focused on ethanol fermentation. Both potassium and sodium cations decreased the ethanol fermentation and glucose to ethanol conversion as potassium or sodium cations. In sodium cation, more than 1.13 N sodium cation in glucose solution led to ethanol production less than theoretical yield with severe inhibition. In 1.13 N sodium cation concentration, ethanol fermentation was slowed down to reach the maximum ethanol concentration with 48 h fermentation compared with 24 h fermentation in control (no sodium cation in glucose solution). In case of potassium cation, three different levels of potassium led to silimar ethanol concentration even though slight slow down of ethanol fermentation with increasing potassium cation concentration at 12 h fermentation. Sodium cation showed more inhibition than potassium cation as ethanol concentration and glucose consumption by Saccharomyces cerevisiae.

Bioethanol derived from lignocellulosic biomass has the potential to replace gasoline. Cellulose is naturally recalcitrant to enzymatic attack, and it also surrounded by the matrix of xylan and lignin, which enhances the recalcitrance. Therefore, lignocellulosic materials must be pretreated to make the cellulose easily degraded into sugars and further fermented to ethanol. In this work, hydrothermal pretreatment on corn stover at $195^{\circ}C$ for 15 min with and without lower concentration of formic acid was compared in terms of sugar recoveries and ethanol fermentation. For pretreatment with formic acid, the overall glucan recovery was 89% and pretreatment without formic acid yielded the recovery of 94%. Compared with glucan, xylan was more sensitive to the pretreatment condition. The lowest xylan recovery of 55% was obtained after pretreatment with formic acid and the highest of 75% found following pretreatment without formic acid. Toxicity tests of liquor parts showed that there were no inhibitions found for both pretreatment conditions. After simultaneous saccharification and fermentation (SSF) of the pretreated corn stover with Baker's yeast, the highest ethanol yield of 76.5% of the theoretical was observed from corn stover pretreated at $195^{\circ}C$ for 15 min with formic acid.
A xylanolytic and cellulolytic anaerobic bacterium strain CtC72 was isolated from cattle rumen liquor. Phylogenetic analysis of 16S rRNA gene sequences revealed that strain CtC72 shared only 97.78% homology with its nearest phylogenetic affiliate Actinomyces ruminicola, showing its novelty. The strain could grow on medium containing xylan, carboxymethyl cellulose and avicel producing $CO_2$, acetate, and ethanol as major fermentation products. The whole genome analysis of the strain CtC72 exhibited a broad range of carbohydrate-active enzymes required for the breakdown and utilization of lignocellulosic biomass. Genes related to the production of ethanol and stress tolerance were also detected. Further there were several unique genes in CtC72 for chitin degradation, pectin utilization, sugar utilization, and stress response in comparison with Actinomyces ruminicola. The results show that the strain CtC72, a putative novel bacterium can be used for lignocellulosic biomass based biotechnological applications.

One-step sulfuric acid saccharification of the red alga Pterocladiella capillacea was optimized, and various detoxification methods (neutralization, overliming, and electrodialysis) of the acid hydrolysate were evaluated for fermentation with the thermotolerant yeast Kluyveromyces marxianus. A proximate composition analysis indicated that P. capillacea was rich in carbohydrates. A significant galactose recovery of $81.1{\pm}5%$ was also achieved under the conditions of a 12% (w/v) biomass load, 5% (v/v) sulfuric acid, $121^{\circ}C$, and hydrolysis for 30 min. Among the various detoxification methods, electrodialysis was identified as the most suitable for fermentable sugar recovery and organic acid removal (100% reduction of formic and levulinic acids), even though it failed to reduce the amount of the inhibitor 5-HMF. As a result, K. marxianus fermentation with the electrodialyzed acid hydrolysate of P. capillacea resulted in the best ethanol levels and fermentation efficiency.

바이오에탄올 생산공정은 당 (Sugar)을 기반으로 하는 공정과 합성가스를 이용하는 공정으로 분류할 수 있다. 이 가운데 합성가스를 이용하는 공정은 촉매를 이용한 화학적 공정과 혐기성 발효에 의한 생물학적 공정의 두 가지로 나뉜다. Clostridium ljungdahlii는 일산화탄소와 수소가 주요 성분으로 구성되는 합성가스를 이용하여 에탄올과 아세트산을 생산할 수 있는 균주 중의 하나로 알려져 있다. 합성가스 발효공정에서 pH는 미생물의 증식과 에탄올 등의 생산에 아주 중요한 요인 중의 하나이다. 본 연구에서는 pH 조건이 미생물의 생장과 에탄올 생산성에 미치는 영향을 조사하였다. C. ljungdahlii 배양은 엄격한 혐기성 조건에서 100 ml의 serum bottle과 pH 제어가 가능한 반응기를 이용한 실험결과, 회분식 배양 조건에서는 미생물의 생장과 에탄올 생산을 위한 최적 초기 pH는 7.0로 나타났다. 미생물 농도는 0.57 g/L, 에탄올 농도 0.91 g/L로 나타났다. pH 4.5 이하에서는 미생물의 생장이 멈추는 것으로 나타났다. pH 제어가 가능한 생물반응기에서는 pH 6.0 일때 에탄올 생산량이 pH 7.0 일때 보다 높게 나타났다. 일정 수준의 미생물 농도를 유지한 조건에서 합성가스를 기포식으로 주입하고 pH 5.9에서 5.4까지 제어하였을 때 미생물량과 에탄올 농도가 증가하였다. 60 시간이 지난 후에 미생물의 농도는 0.498 g/L, 에탄올은 1.056 g/L까지 이르렀다.
Certain microalgae have been known to use light and various carbon sources to produce carbohydrates, mainly in the form of starch. This is one of the pertinent feedstocks replacing agricultural products for the production of bioethanol by yeast. This study focuses upon dilute acid hydrothermal pretreatments at low cost and high efficiency to compete with current methods, and employs Chlamydomonas reinhardtii UTEX 90 as the feedstock. With dry cells of 5%(w/v), the algal biomass was pretreated with sulfuric acid(1-5%) under temperatures from 100 to $120^{\circ}C$, from 15 to 120 min. As a result, the glucose release from the biomass was maximum at 58%(w/w) after pretreatment with 3% sulfuric acid at $110^{\circ}C$ for 30 min. This method enabled not only starch, but also the hydrolysis of other oligosaccharides in the algal cell in high efficiency. Arrhenius-type of model equation enabled extrapolation of some yields of glucose beyond this range. The pretreated slurry was fermented by yeast, Saccharomyces cerevisiae S288C, resulting in an ethanol yield of 29.2% from algal biomass. This study suggests that the pretreated algal biomass is a suitable feedstock for ethanol production and can have a positive impact on large-scale applied systems.
