The Ethnobotanical Investigation around National Parks in the Northern Area of Province Gyeongsangbuk-do, Korea

  • We investigated traditional knowledge about ethnobotanical uses around national parks in the northern areas of Province Gyeongsangbuk-do, with the focus on the Sobeaksan National Park area. Interviews were carried out to 138 residents at 42 places from 7 counties and/or cities, and verified species and usage information were categorized by taxonomic groups, usage, and used parts. The ethnobotanical species of the regions consisted of a total of 277 taxa: 244 species, 3 subspecies, 28 varieties, and 2 forma in 213 genera of 79 families. The significant native plants included one Endangered Species (Paeonia obovata Max.) and four endemic species (Salix hallaisanensis H. Lev., Aconitum pseudolaeve Nakai, Paulownia coreana Uyeki, and Cirsium setidens (Dunn) Nakai, Sasa coreana Nakai). About 17 Approved Species for Delivering Overseas designated by Korea Ministry of Environment have been also traditionally valuable in the regions. The main usage of the plants were edible (185 taxa) and medicinal (175 taxa). Leaves were most commonly used parts (109 taxa), followed by stems (73 taxa), fruits (69 taxa), roots (67 taxa), and wholes (57 taxa). The traditional usage of plants were diverse as those useful parts. Leaves, fruits and seeds were used as edible and medicinal; sprouts (young shoots) were mainly used as edible; but roots and flowers were used as medicinal. The study does not only provides examples of traditional uses of native plants, but also facilitates sustainable managements and developments of ethnobotanical knowledge for the contemporary society.

Occurrence and antibiotic resistance of Enterococcus spp. from retail fresh-cut products in Korea (국내 신선 편이식품으로부터 분리한 Enterococcus의 항생제 저항성)

  • Enterococcus spp. have been considered major indicator organisms for antibiotic resistance due to their ability to easily acquire and to harbor antibiotic resistance. In this study, Enterococcus spp. were isolated from 174 retail fresh-cut products (fresh vegetable salads, microgreens, and sprouts) in Korea. Among the 20 Enterococcus isolates obtained, 18 (90.0%) were Enterococcus faecalis and 2 (10.0%) were Enterococcus faecium. The patterns of antibiotic resistance against nine antimicrobials were analyzed. Most of the isolates (85.0%) were resistant to quinupristin/dalfopristin, 40.0% and 50.0% of the isolates showed intermediate resistance to two critically important antimicrobials for human medicine, ciprofloxacin and erythromycin, respectively. Vancomycin-resistant enterococci were not detected in this study. Given the importance of antimicrobial resistance of enterococci in food safety as well as in public health, our results regarding the occurrence (level of contamination) and antimicrobial resistance of Enterococcus spp. could provide useful information that aids the risk analysis of antibiotic resistance.

High School Student's Attitude about Kimchi and Development of Kimchi as a Menu Item for Meal Service (고등학생의 김치이용 태도 및 급식 메뉴 개발 연구)

  • Kimchi is the most well-known Korean traditional food, but it is also the main leftover of school lunch and dinner menus. This study aimed to familiarize teenagers with kimchi through school meals and to increase their daily kimchi intake, ultimately by appealing to the young generation's taste. A questionnaire survey was conducted in the Ansan area to examine student's acceptability of kimchi and their attitudes toward kimchi. Approximately 65% of males and 67% of female students liked the moderately fermented and pungent taste of kimchi. Kimchi served in school meals was regarded as nutritional but cheap. Approximately 72% of male and 82% of female students responded that they liked menu items using kimchi. Approximately 48% of students responded that menu items using kimchi in schools are not diverse. Students preferred meat as an ingredient in kimchi. The preferred cooking methods were stir-frying and frying, whereas boiling was the least favorite. Based on the survey results, ten kimchi menu items had been developed. The suitability of the menu was evaluated by students and cooks. Six kimchi items, including Kimchi mixed with rice, chicken, soybean sprouts, Kimchi cheese rice, stewed beef ribs with kimchi, rice topped with kimchi curry, kimchi cheese meat roast, and kimchi udong were considered appropriate for school meals, whereas kimchi kangchong, kimchi topokki, kimchi stew with surimi, and frozen Pollack kimchi soup were not suitable as menu items. Kimchi topokki was not accepted by students, while kimchi kangchong was not accepted by cooks. Cooks judged the suitability of a menu item by the cooking process and cooking times, whereas students judged an item by its sensory preference. Approximately 63% of students responded that kimchi intake has increased by participating in the development of kimchi dishes.

  • Pharmacological studies and clinical practices have indicated that Radix Astragali, a dried root of Astragalus membranaceus possesses a lot of biological activities, including antioxidant, hepatoprotective, anti-diabetic, tonic, diuretic, antimicrobial, antiviral, and immunological activities. These biological activities approved by the modern pharmacological studies are mainly due to the constituents of Astragalus membranaceus including polysaccharides, saponins, flavonoids, amino acids, and trace elements. In resent, the main constituents in the root part showing a lot of biological activities has been isolated also from the aboveground parts such as leaves and sprouts in our laboratory. However, the safety evaluation for the aboveground parts of Astragalus membranaceus should be checked before expanding their application as one of food. In the study, a 90-day rat oral gavage study has been conducted with the extracts from Astragalus membranaceus-above-ground parts at doses of 1000, 3000, and 5000mg/kg/day. The following endpoints were evaluated: clinical observations, body weight, gross and microscopic pathology, clinical chemistry, and hematology. Based on the analysis of these endpoints, it was estimated that NOEL (no observed effect level) for male rats and NOAEL (no observed adverse effect level) for female rats are 5000mg/kg/day of the water-extracts from Astragalus membranaceus-aboveground parts.

Using Air Temperature and Sunshine Duration Data to Select Seed Production Site for Eleutherococcus senticosus Max (기온과 일도시간 분석에 의한 가시오가피의 파종적지 선정)

  • It was very hard to gather the seeds of Eleutherococcus senticosus Max. known as a medicinal plant for they tend to drop under the high temperature condition during the summer period in Korea. Therefore, this study was conducted to select seed production site for Eleutherococcus senticosus in Korea, comparing the climate of Hokkaido of Japan, in which the seeds have been produced, with that of various place in this country. It was low that the average maximum temperature during the hottest summer two months (July and August) as a 24.4$^{\circ}C$ in Hokkaido and 21.2$^{\circ}C$ in Daegwanryeong compared with 27.4$^{\circ}C$ in Changsu. Especially in Daegwanryeong, average maximum temperature from June to September remained as low as 21$^{\circ}C$. Effective accumulated temperature(>5$^{\circ}C$) was 807$^{\circ}C$ in Hokkaido and 964$^{\circ}C$ in Daegwanryeong during the ripening period. Monthly sunshined hours from July to August were 121.7~128 hours in Daegwanryeong and 83.5~85.4 hours in Hokkaido. The Eleutherococcus senticosus sprouts at 8.5$^{\circ}C$, comes to flowering season in mid-August, and ripens during late-August and October in Hokkaido, the climate of which is similar to that of Daegwanryeong.

An Investigation of Side-dishes found in Korean Literatures before the 17th Century (17세기 이전 조선시대 찬물류(饌物類)의 문헌적 고찰)

  • In this study, we investigated e kinds and names of side dishes along with their recipes and ingredients occuring in Korean cookbooks published before the 17th century. The side dishes were classified 79 kinds of Guk, 23 kinds of Jjim and Seon, 15 kinds of Gui, 3 kinds of Jeon, 7 kinds of Nureumi, 3 kinds of Bokkeum, 30 kinds of Chae, 11 kinds of Hoe, 7 kinds of Jwaban, 6 kinds of Mareunchan, 12 kinds of Pyeonyuk and 5 kinds of Jeonyak, Jokpyeon and Sundae. The earliest records were found on Guk, Jjim, Jwaban, Po and Pyeonyuk Gui, Namul and Hoe were recorded after the 1500's and Nureumi, Jeon, Jeonyak, Jokpyeon and Sundae were developed relatively late in the late 17th century. As to the kinds of side dishes, Guk was the most common. Guks cooked before the 17th century used different recipes and more types of ingredients than today, including some that are not used today. For Jjim, various seasonings were added to main ingredients such as poultry, meat, seafood and vegetable. Most of the records found for Jjim used chicken as the main ingredient. Gui was recorded as Jeok or Gui and there weren't many ingredients for Gui before the 17th century. Gui was usually seasoned with salt or soy bean sauce and broiled after applying oil. Vegetables were broiled after a applying flour-based sauce. The Jeon cooked at that time was different from the one that is cooked today in that cow organs or sparrows were soaked in oily soy bean sauce before being stewed. Nureumi, which was popular in the 17th century, but rarely made today, was a recipe consisting of adding a flour or starch-based sauce to stewed or broiled main ingredients. Chae was a side dish prepared with edible plants, tree sprouts or leaves. Chaes like Donga and Doraji were colored with Mandrami or Muroo. Hoe was a boiled Hoe and served after boiling seafood. Jwaban was cooked by applying oil to and then broiling sparrows, dudeok, and mushrooms that had been seasoned and dried. For dried Chans, beef or fish was thin-sliced, seasoned and dried or sea tangle was broiled with pine nuts juice. There are some recipes from the 17th century whose names are gone or the recipes or ingredients have changed. Thus we must to try to rebuild three recipes and develop recipes using our own foods of today.

Study of germination for none seasonal pruning mulberry farm and their insective diseases (순상전 춘기말벌채고조의 발아상태와 충해 발생 관계)

  • 1. In the mulberry groves for both rearing of spring and fall, the rate of germination was 74.3 percent and the rate of the unfinished bud was 25.7 percent. 2. When the summer pruning after spring rearing was not worked, new sprouted leaves from the axillary buds of an age old branches was 74.88 percent of germination and the rest from unfinished bud during the spring showed 28.12 percent. 3. In the furrows running from north to south, new buds facing the due north were the worst but towards south-east and north-east were the best in their sprouts. 4. One year-old branches which had been raised without the summer pruning in the bush types of Morus Lhou (ser) Koidz offered a habitat where Apriona Rugico1lis Cherr could live. 5. On the grove of Morus Lhou Koidz cultivated at lower zone under a higher humidity and the shortage of sun light the appearance of Anomoneura Mori Schw was likely to happen very much. 6. One year old branches without the summer pruning produced more leaves owing to a long period of growth than branches with the summer pruning in the fall crops. 7. It is necessary to reconsider a certain distance of planting because of causing, when the summer pruning was not made in the bush plantation, a obstacle of fertilizer operation or the forth coming year's management. 8. It can not be thought the ideal method for us to raise mulberry tree such as Morus Lhou Koidz under natural form and at a short distance of planting (one another). 9. Among the species, Morus alba L. had a great deal of leaves and of the rate of germination per unit length of branch.

Effects and Molecular Mechanisms of Eupatorium chinensis var. simplicifolium Extract on Abnormal Proliferation of Vascular Smooth Muscle Cells (등골나물추출물의 혈관 평활근 세포의 비정상 증식에 대한 억제 효과 및 분자기작)

  • Eupatorium chinensis var. simplicifolium (EUC) has anti-inflammatory and antioxidant effects. Young sprouts of EUC have been used as food for a long time, and the whole EUC plant has been used as an herbal remedy in oriental medicine. Arteriosclerosis, or chronic inflammation in arterial vessels, is a cardiovascular disease and is involved in various disorders. Cardiovascular diseases such as restenosis and neuropathic hyperplasia are mainly caused by abnormal growth and movement due to multiple growth factors in vascular smooth muscle cells (VSMCs). Platelet-derived growth factor (PDGF) is a mitogen released from damaged vessel walls and is involved in the proliferation and migration of VSMCs. To determine the effects of EUC on the abnormal proliferation and migration of VSMCs, the present study investigated intracellular signaling pathways in PDGF-BB-induced VSMCs treated with and without EUC. Pretreating PDGF-BB-induced VSMCs with EUC tended to effectively decrease cell proliferation and migration. Subsequently, the intracellular growth-related signaling pathways of AKT, phospholipase C gamma (PLC-γ), and mitogen-activated protein kinase (MAPK) were investigated using western blotting to confirm inhibited phosphorylation. Furthermore, flow cytometry data showed that EUC blocked the cell cycle of VSMCs. These results suggest that EUC can inhibit the proliferation and migration of VSMCs by controlling the cell cycle and growth factor receptors. Furthermore, this indicates that EUC can be used as a preventative against cardiovascular disease resulting from abnormal proliferation and migration of VSMCs.

Anti-Diabetic Effects of Sprouts in High-Fat Diet and Streptozotocin-Induced Type II Diabetes Mellitus Mice (고지방식이와 STZ 유도 제2형 당뇨 마우스에서 새싹의 항당뇨 효과)

  • Sprout vegetables containing various types of polyphenols and flavonoids, are known to have anti-inflammatory, antioxidant, cholesterol-lowering, and anti-obesity activities. However, there have been few reports on the anti-diabetic efficacy of sprout vegetables. Here, we investigated the anti-diabetic effects of sprout extract obtained from buckwheat, beet, rape, broccoli, kohlrabi, red young radish, and dachai, in high fat diet (HFD) and streptozotocin (STZ)-induced type II diabetes mellitus mice. The mice were fed a HFD (60% calories as fat) for 8 weeks prior to intraperitoneal injection with STZ (75 mg/kg). The diabetic mice were divided into four groups: standard diet (STD, 10% calories fat), HFD, HFD with sprout extract (SPE) and HFD with metformin (MET). After 4 weeks, body weight gain was much lower in both SPE and MET groups than in HFD group. In contrast, there was no difference experiment groups regarding food intake ratio. The level of fasting blood glucose was significantly lower in the SPE and MET groups compared to the HFD group. Oral glucose tolerance and insulin tolerance in the SPE and MET groups were significantly ameliorated in comparison to the HFD group. The concentrations of serum total cholesterol, triglycerides, and LDL cholesterol in the SPE and MET groups were remarkably reduced in comparison to the HFD group, and HDL cholesterol concentration was higher in the SPE and MET groups than in the HFD group. Glutamate oxaloacetate transaminase and glutamate pyruvate transaminase levels were between SPE and HFD groups. The serum insulin and leptin concentrations were significantly reduced in both the SPE and MET groups compared to the HFD group. Therefore, these results indicate that sprout extract could improve insulin resistance and attenuate blood glucose level in HFD/STZ-induced type II diabetes mellitus mice. We conclude that this study may provide positive insights into sprout extract as a functional food ingredient for treatment of type II diabetes mellitus.

Comparison of the Standard Culture Method and Real-time PCR for the Detection of Vibrio parahaemolyticus in Seafoods and Vegetables (해산식품과 채소에서 Vibrio parahaemolyticus 검출을 위한 배지배양법과 real-time PCR의 비교검증)

  • Vibrio parahaemolyticus (V. parahaemolyticus), which is commonly found in raw seafood, causes gastroenteritis in humans. Rapid and effective methods have been developed as culture methods require up to 5-7 days. In this study, real-time PCR was compared with the standard culture method for detecting V. parahaemolyticus in seafood and radish sprout samples. Five hundred grams of the samples were artificially contaminated with V. parahaemolyticus then divided into 20 samples. The samples were incubated in alkaline peptone water and then streaked onto thiosulfate-citrate-bile saltssucrose agar. Biochemical tests for suspicious colonies were performed using the API 20NE strip. In parallel, real-time PCR was performed targeting the toxR gene using the enrichment broth. The real-time PCR was sensitive in discriminating V. parahaemolyticus from other foodborne pathogens. The detection limit of the real-time PCR was $10^3\;CFU/mL$ in phosphate-buffered saline. Although the real-time PCR detected more positive samples (76 out of 180, 42%) than the culture method (66 out of 180, 37%), there was no significant statistical difference (p>0.05) between the two methods. In conclusion, real-time PCR assays could be an alternative to the standard culture method for detecting V. parahaemolyticus in seafood and radish sprouts, which has many advantages in terms of detection time, labor, and sensitivity.
