Mutation of Canine Tumor Suppressor Gene p53 in a Mammary Gland Adenocarcinoma and a Malignant Mast Cell Tumor (개의 유선암종과 악성 비만세포 종양에서 발생한 종양억제 유전자 p53의 변이)

  • To identify mutations in exons 5 to 8 of the p53 tumor suppressor gene, we have analysed in 12 spontaneous canine tumors. In a malignant mast cell tumor, a 1 base pair alteration AGT $\longrightarrow$AGC (silent point mutation, serine) in codon 249 in exon 8 was detected. And the mammary gland adenocarcinoma was found to have a mis-sense point mutation (CCT $\longrightarrow$ TCT) in codon 285 in exon 8.

Isolation and Genomic Characterization of the T4-Like Bacteriophage PM2 Infecting Pectobacterium carotovorum subsp. carotovorum

  • In order to control Pectobacterium carotovorum subsp. carotovorum, a novel virulent bacteriophage PM2 was isolated. Bacteriophage PM2 can infect 48% of P. carotovorum subsp. carotovorum and 78% of P. carotovorum subsp. brasilliensis but none of atrosepticum, betavasculorum, odoriferum and wasabiae isolates had been infected with PM2. PM2 phage belongs to the family Myoviridae, and contains a large head and contractile tail. It has a 170,286 base pair genome that encodes 291 open reading frames (ORFs) and 12 tRNAs. Most ORFs in bacteriophage PM2 share a high level of homology with T4-like phages including IME08, RB69, and JS98. Phylogenetic analysis based on the amino acid sequence of terminase large subunits confirmed that PM2 is classified as a T4-like phage. It contains no integrase- or no repressor-coding genes related to the lysogenic cycle, and lifestyle prediction using PHACT software suggested that PM2 is a virulent bacteriophage.

  • As one of methodological approaches for objective and systematic evaluation for rural resources, a sequential system for significance determination of resources items was formulated using AHP (Analytic Hierarchy Process) as the base technique. By the system constructed in this study, the significance values were calculated for all items of standard goal system which had been presented in the first phase- paper of research works for rural resources evaluation. The results analysed and obtained in the study are summarized as follows; 1. In case that the number of resources items for evaluation are relatively large, stepwise matrix method is considered as the better one for pair-comparison works in view of evaluator's convenience and consistency. 2. The significance valuing results for most resources items showed a considerable difference between characteristic areas, especially in aspects of land and human resources, so being considered as wet reflecting the specific areal varieties, 3. On land resources, the items for industrial development and land use showed higher significance value, while natural environment resources the items for ecological varieties and on human resources the items for artificial facilities. However, lower-valued ones were the items for mineral stocks in land resources, for control and protection in natural environment resources and for lower-level culture heritage respectively.

  • Simmondsia chinensis (Link) Schneider, a multipurpose and monogeneric dioecious shrub from arid zones, has emerged as a cash crop all over the globe. Its seed propagation poses severe problems due to its male-biased population: the male:female ratio is 5:1. Investigations have been carried out to generate a sex-specific Inter-simple sequence repeat (ISSR) marker for the early detection of male and female plants. Of the 42 primers analysed with a bulk sample of pooled male DNA and a bulk sample of pooled female DNA, only one primer, UBC-807, produced a unique ~1,200 base-pair fragment in the male DNA. To validate this observation, this primer was re-tested with individual male and female samples from eight cultivars. A similar unique ~1,200 bp fragment was present in the male individuals of all eight cultivars and completely absent in the female individuals tested. This is the first report of the use of ISSR markers to ascertain sex in physiologically mature S. chinensis plants.

  • The purpose of this study was to establish early diagnosis for bovine virus diarrhea-mucosal disease(BVD-MD) Two Holstein among 22 breeding cows were shown ulcer in the mouth and watery diarrhea. Diarrheal feces and ulcerous lesion of the mouth from 2 cows were sampled for detection of viral antigen. BVD virus was isolated by inoculation of the samples to MDBK cells, and the cytopathic effects were observed in cultured MDBK cells which inoculated with virus isolates from the feces. Viral antigens were detected in the feces and ulceruous lesion by immunogold staining. The serum neutralization titers were shown 1 : 64 or greater in 8 blood samples by using BVD virus (NADL strain). By the RT-PCR, using reverse primer 5'-ACTCCATGTGCCATGTACAG-3', forward primer 5'-ACTCCATGTGCCATGTACAG-3', 285 base pair band specific to BVD virus was detected. In conclusions, the results of above tests which executed using the diarrheal feces and ulcerous lesion of the mouth and the isolates were conformed as BVD virus.

  • From February 2000 to December 2001, A total of 1,785 samples was taken from beef and pork carcasses in Seoul. Seven(0.69%) Listeria spp. were isolated from the 1,014 of beef carcasses, and five(0.65%) were isolated from the 771 of pork carcasses. The isolates were identified L monocytogenes by API listeria, and VIDAS LMO kit, serological test and PCR assay were preformed. A total 12 strains of L monocytogenes were isolated form samples tested and all of the strains were classified into serotype 1. PCR primers are selected to amplify a 520-base pair(bp) DNA fragment from the listeolysin O gene(hlyA) of Listeria monocytogenes. A 520-bp product was detected in PCR with DNA from L monocytogenes, but not from the other Listeria species tested.

Understanding Rifampicin Resistance in Tuberculosis through a Computational Approach

  • The disease tuberculosis, caused by Mycobacterium tuberculosis (MTB), remains a major cause of morbidity and mortality in developing countries. The evolution of drug-resistant tuberculosis causes a foremost threat to global health. Most drug-resistant MTB clinical strains are showing resistance to isoniazid and rifampicin (RIF), the frontline anti-tuberculosis drugs. Mutation in rpoB, the beta subunit of DNA-directed RNA polymerase of MTB, is reported to be a major cause of RIF resistance. Amongst mutations in the well-defined 81-base-pair central region of the rpoB gene, mutation at codon 450 (S450L) and 445 (H445Y) is mainly associated with RIF resistance. In this study, we modeled two resistant mutants of rpoB (S450L and H445Y) using Modeller9v10 and performed a docking analysis with RIF using AutoDock4.2 and compared the docking results of these mutants with the wild-type rpoB. The docking results revealed that RIF more effectively inhibited the wild-type rpoB with low binding energy than rpoB mutants. The rpoB mutants interacted with RIF with positive binding energy, revealing the incapableness of RIF inhibition and thus showing resistance. Subsequently, this was verified by molecular dynamics simulations. This in silico evidence may help us understand RIF resistance in rpoB mutant strains.

Ab Initio Studies on Proton Affinities of Substituted Furans (치환 퓨란의 양성자 친화도에 대한 Ab Initio 연구)

  • The geometry of furan, relevant to the binding of bis-furan lexitropsin that contains this ring to the base pair of minor groove of DNA, is optimized by semiempirical (MNDO) and ab initio (Hartree-Fock) methods. The proton affinity and electronic structure are evaluated at the 6-31G and $6-31G^{\ast}$ level of theory for the optimized geometry. The proton affinities are also studied for various substituted furans with the electrondonating and -withdrawing groups to estimate the substituent effect on the proton affinity of furans. It has been found that the electron-donating substituents increase the proton affinity of furan, whereas the electron-withdrawing substituents decrease it. This result can be explained with atomic charge and electron density at oxygen of substituted furans.

  • Due to the building energy consumption of total energy consumption of Korea takes over 24%, economizing building energy and using renewable energy resources is being required. To suggest the prototype of zero energy house of country house, the passive systems and active systems are applicated and simulated. In case of wall insulation system is applicated, the heating load of building is reduced. Also, clear triple pair glazing system reduced 2.1% of heating load of building. The amount of reducing heating load by infiltration is depending on the Heating system. In this model, the 0.3ACH made 14.6% saving on heating load from base infiltration 0.82ACH. The solar thermal system of active system could save 80% of DHW and PV system supplies electric power more than average consumption of year. Through the optimum process, the end use of zero energy house of country house is 36kWh/m2.yr and total energy consumption is reduced about 74.2%.

Effects of Lonicerae Flos' Ethyl Acetate Fraction on Mutagenicity (금은화(Lonicerae Flos)의 Ethyl Acetate 분획이 돌연변이원성에 미치는 영향)

  • Based on the following tests with fractions extracted from organic solvents such as benzene, chloroform, ethyl acetate and methanol, this study is to analyse the antimutagenicity of Lonicerae Flos. When carrying out Ames test with Salmonella typhimurium strain, it seemed that there was stronger antimutagenicity in TA 100 treated by MNNG than did one in TA 98 by NPD, and that there was stronger antimutagenicity through base pair exchange than one through frame shift. In the umu test, each fraction tended to inhibit the activity of ${\beta}-galactosidase$ induced by AF-2. As shown in these above tests, the ethyl acetate fraction was the strongest among four fractions. On the other hand, its component consisted of luteolin, apigenin, chlorogenic acid and five unknown ingredients. Of these unknown ingredients, E, F strongly tended to inhibit the activity of ${\beta}-galacosidase$. In addition,there was also the decrease in its activity of apigenin, luteolin and chlorogenic acid.
