The aim of this study was to determine the optimal cooking conditions for astringent persimmon concentrates. With increasing time of concentration, the moisture contents and L-, a- and b-values all decreased, whereas the brix level and viscosity increased. The crude protein, fiber and ash increased and the vitamin C decreased by concentrating. The pH was the lowest at 23 hrs of boiling concentration. With longer boiling concentration time, the fructose, glucose, and sucrose levels increased but the maltose level decreased. The DPPH radical scavenging effects of astringent persimmon concentrates were more than 92%. The total phenolics, flavanol tannin, leucoanthocyanin, and chlorogenic acid levels increased with longer boiling concentration time. In the sensory evaluation of the astringent persimmon concentrates, overall acceptability was the best at the 23-hr boiling concentration. From the above results, the 23-hr boiling, astringent persimmon concentrates could be useful for cooking in terms of obtaining the high amounts of phenolic compounds as functional compounds and overall acceptability.
In postmenopausal women, estrogen deficiency can be associated with metabolic diseases, such as obesity, diabetes, and cardiovascular diseases. The purpose of this study was to investigate factors related to lipid, glucose and antioxidnat metabolism. Sprague-Dawley female rats were subjected to either bilateral ovariectomy or sham operation and randomly divided into 3 dietary groups, (n=8) sham-operated fed with normal diet (SHAM), ovariectomized fed with normal diet (OVX), and ovariectomized fed with normal diet supplemented with Eucommia ulmoides extract (OVX-EU). The OVX-EU group showed significantly lower body weight, triacylglycerol, and total cholesterol than that showed by the OVX group. In addition, the OVX-EU group showed improved lipogenesis, glucose-regulating enzyme activities, adipokine and antioxidant enzyme activities. These findings demonstrate that extracts from E. ulmoides extract can be used as a functional food.
Products of ginseng browning reaction were investigated to determine the nature of their antioxidant activity using a model system, White Ginseng and Red Ginseng extracts. In the simulated ginseng model system, the brown color was intensified with an increase in the length of reaction time and the antioxidant activity initially increased in proportion the length of reaction time for up to 17 hrs and then leveled off thereafter. Pararell results were obtained manufacturing of Red Ginseng, Comparison of the antioxidant activity of the inner and outer solutions after dialysis of browning solution showed that the outer solution had a stronger antioxidant activity than the inner one. For further analyses, browning reaction products were fractionated into three peaks on Amberlite CG-120 type I ionexchange resin and designated as fractions I, II and III in order of elution. Partial characterization of the fractions revealed that floe most intense brown fraction (Fraction II) had the strongest antioxidant activity and also exhibited reducing power for Somogyi-Nelson reagents and ninhydrin positive reaction. Both the browning reaction products and Red Ginseng extracts were found to possess potent reactivity with ${\alpha}$,${\alpha}'$-diphenyl-${\beta}$-picrylhydrazyl, whereas White Ginseng extracts showed negligible reactivity.
The demand for fuel and energy resources continues to grow due to increased consumption and emerging economies in all parts of the world. With this increase in demand, crude oil prices in the international market has jumped dramatically. Global warming, which is a consequence of increasing greenhouse gas (GHG) emissions, has become scientific, social, and political concerns. To cope with global warming and energy crisis, cost-competitive biofuels are urgently needed. In addition, development of an infrastructure, which supplies energy stably and diversifies energy resources, as well as new cost-saving technologies should be developed to reduce the costs of producing biofuels. Due to high oleic acid content, rapeseed (Brassica napus L.) is currently the potential feedstock for biodiesel production in temperate zone region and the production and use of rapeseed oil is already commercialized in Europe. In Korea double-cropping (rice and rapeseed) became more prevalent because it reduces competitions from land constraints. Production of rapeseed as a biodiesel feedstock may reduce the influence of rising oil prices and nation's dependence on imported petroleum and increase job opportunities and farm incomes.
This study investigated the properties and antioxidative activities of phenolic acid concentrates of rice bran. Rice bran contains bioactive substances such as phenolic compounds, which can provide health benefits as natural antioxidants. This study examined how levels of phenolic acids can be obtained efficiently through various extraction methods. The extractions of defatted rice bran were followed by using ethylacetate (RBE-I), ethylacetate after alkaline hydrolysis (RBE-II), and 80% methanol (RBE-III). For all extracts, yields (%), total polyphenol contents (TPC), various phenolic acids and antioxidative activities were estimated. RBE-II had the highest total polyphenol contents (526.72 mg/100 g rice bran) and showed high antioxidative activity (74.7%). To concentrate the phenolic acids, RBE-II was passed through Sep-pak $C_{18}$ Vac cartridge and F1-RBE-II was collected by the elution of 50% methanol. The total phenolic content of F1-RBE-II (736.8 mg/100 g rice bran) was higher than that of RBE-II (367.1 mg/100 g rice bran), and the ratios of ferulic acid (73%) and sinapic acid (14%) increased. As RBE-II was analysed by HPLC, 6 different phenolic acids were found via chromatography, whereas F1-RBE-II showed 5 different peaks and the major phenolic acid was identified as ferulic acid. The ABTS radical scavenging activity of F1-RBE-II was the highest among the rice bran extracts. In a ${\beta}$-carotene-linoleic acid model system, linoleic acid oxidation was reduced by F1-RBE-II (73%) and RBE-II (35%).
This study was investigated to analyze the contents of flavonoid compounds and the effects of fermentation on the physiological activities of medical plants, also known as SanYa (SY). Antioxidative activity of the fermented SanYa (FSY) was measured by using DPPH radical scavenging and SOD-like activity. DPPH radical scavenging and SOD-like activity of FSY were 94.3% and 45.0%, respectively. Nitric oxide (NO) synthesis was increased 11 times through the addition of FSY. However, NO production of the macrophages RAW264.7 cells stimulated with lipopolysaccharide (LPS) was reduced to 56% through the addition of FSY. FSY showed fibrinolytic activity and indicated about 69.8% and 73.7% of xanthine oxidase and angiotensin converting enzyme inhibitory activities, respectively. These results suggested that FSY plays a significant role in fibrinolytic activity and have strong xanthine oxidase and angiotensin converting enzyme inhibitory activities.
The biological activity of hot water extract from Ecklonia cava Kjellman (ECE) was investigated to assess antioxidative, anti-skin aging, and nitrite scavenging abilities, as well as alcohol metabolizing activities. Antioxidant activity of ECE was measured by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity. DPPH radical scavenging activity and SOD-like activity of ECE increased in a remarkably dose-dependent manner, and were about 91.4% and 75% at 1 mg/ml, respectively. The xanthine oxidase inhibitory activity was indicated to be about 70% at 1 mg/ml of ECE. Nitrite scavenging ability of ECE showed to be 93.6% at 1 mg/ml and pH 1.2. The influence of ECE on alcohol metabolism was demonstrated through the generating activity of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). The facilitating rate of ADH and ALDH activity by ECE was 167.2% and 334% at 10 mg/ml, respectively. In addition, tyrosinase and elastase inhibitory activities of ECE were 58% and 72% at 10 mg/ml, respectively. These results indicated that ECE has valuable biological attributes owing to its antioxidant, nitrite scavenging, alcohol metabolizing, and elastase and tyrosinase inhibitory activities.
The biological activities of Smilax china L. rhizome (SCR), hot water (SCRW) and 70% ethanol extract (SCRE) were analyzed. The total phenolic contents of SCRW and SCRE were 51.7 and 100.5 mg/g, respectively. The measured flavonoid content of SCRW ($67.7{\mu}g/g$) was almost double that of SCRE ($31.7{\mu}g/g$). SCRE ($IC_{50}=42.4{\mu}g/mL$) exhibited stronger antioxidant activity in the DPPH system than the positive control ${\alpha}$-tocopherol ($71.3{\mu}g/mL$) or butylated hydroxy anisole ($53.8{\mu}g/mL$) did. SCRE ($IC_{50}=50.3{\mu}g/mL$) also showed stronger ABTS radical scavenging activity, as did ${\alpha}$-tocopherol ($67.1{\mu}g/mL$). The SOD-like activity and Tyrosinase inhibition activity of SCRW and SCRE showed almost the same pattern. The best SOD-like activity and tyrosinase inhibition activity were measured as 24.9% and 20.3% in SCRW at $1,000{\mu}g/mL$, respectively. The cytotoxic effects of the SCR extracts were analyzed via MTT assay on human cancer and normal cells. SCRW and SCRE did not show cytotoxicity up to the concentration of $1,000{\mu}g/mL$ against the normal human cell line HEK293. Against human breast cancer cells (MCF-7), SCRW inhibited MCF-7 growth (by 27.6%) better than the anticancer drug cyclophosphamide (15.5%) at $1,000{\mu}g/mL$. SCRE ($1,000{\mu}g/mL$) inhibited the growth of human lung cancer cells A549 (37.6%) and human stomach cancer cells AGS (53.6%) more effective than did SCRW (21.0% and 35.4%) or CPA (22.2% and 31.7%). These results suggest the potential use of SCRE and SCRW as an excellent antioxidant and antiproliferative substance, respectively.
The purpose of this study was to investigate the chemical composition of the freeze-dried stems and fruit of the cactus Cheonnyuncho. The analysis showed that powdered stems have the highest fat content (1.91%) and the powdered fruits have he highest protein content (2.62%). The K content of the fruits higher than that of the stems, while the Ca, Mg, Na and P contents of the stems were higher than those of the fruits. Both the stems and fruits powders contained high levels of the amino acids glutamic acid and aspartic acid. The free sugars such as sucrose, fructose, and glucose were detected in both the stems and fruits. The 75% ethanol (EtOH) extract showed a relatively high antioxidative activity compared to those of the water and 75% methanol (MeOH) extracts. Furthermore, the 75% EtOH extract of the stem powder exhibited a total polyphenol content of 3.60 g/100 g, and a total flavonoid content of 2.00 g/100 g. The antioxidant activities of the stem and fruit powder extracts, measured in DPPH radical scavenging experiments, were higher than that of the control group.
This study was carried out to evaluate the agronomic characteristics and biological activities of $Sorghum$$bicolor$ germplasms introduced from USA. Plant height, stem diameter, tiller number, leaf length, leaf width, leaf vein color, ear type, ear length and ear width have different from the cultivated accessions. Sweet-N-Sterile (#4) showed the tallest height, widest ear-width and stem diameter. Most of $Sorghum$$bicolor$ plants may be available to use for bio-energy from these results showing big biomass. Antioxidant activities of 11 cultivars collected from USA were examined by DPPH assay and reducing power. Among the cultivars, Premium stock (#1), Early Sumac (#7), SS Silage (#9) and WGF Grain Sorghum (#11) showed a significantly higher antioxidant activity in comparison to others. Early Sumac (#7) and SS silage (#9) showed more strong reducing power activities than ${\alpha}$-tocopherol, a positive control. Premium stock (#1), Sweet-N-Sterile (#4), Early Sumac (#7) and SS Silage (#9) were also showed high antioxidant activities by DPPH assay and reducing power experiment. BMR Gold I (#3) displayed strong antimicrobial activity against $Escherichia$$coli$ at minimum inhibitory concentrations (125 ${\mu}g$/ml).
