Screening of Cytotoxicity and Antimicrobial Effects of Extracts from Atractylodes macrocephala Koidz (백출 추출물의 세포독성과 항균효과검색)

  • This study was carried out to evaluate cytotoxic effects of Atractylodes macrocephala Koidz. (A. macrocephala Koidz.) extract on NIH 3T3 fibloblast. SK-MEL-3 (HBT 69) and KB (ATCC No, OCL 17) cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-y1)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. 10.2 mg/ml Concentration of A. macrocephala Koidz. extracts in SK-MEL-3 showed that their susceptibility (sensitivity) to these compounds decreased in the following order ; adriamycin > H₂O > ethyl acetate > ethyl alcohol > chloroform > n-hexane in SK-MEL-3 cell lines ; 5-FU > H₂O > n-hexane > ethyl acetate > ethyl alcohol > chloroform in KB cell lines. In order to develop an antimicrobial agent, A. macrocephala Koidz. was extracted with solvents. The minimal inhibitory concentrations (MICs) of each solvent extract of A. macrocephala Koidz. against microogranisms were also examined. Antimicrobial activities of ampicillin and ketoconazole as references were compared to those of each solvent extract of A. macrocephala Koidz. The antimicrobial activity of the ethyl acetate soluble extract of A. macrocephala Koidz. had growth inhibition activity against S. mutans and P. putida (MICs. 500 ㎍/ml). These results suggest that the ethyl acetate soluble extract of A. macrocephala Koidz. possessed antitumorous and antimicrobial agents

Inhibitiory Effects of Mixture of Atractylodes Macrocephala and Amomum Villosum Extracts on Adipocyte Differentiation in OP9 Cells (백출과 사인 추출 혼합물의 지방세포분화 억제 효과)

  • In this study, we investigated the inhibition effects of mixtures of Atractylodes macrocephala (AM) and Amomum villosum (AV) water extracts on adipocyte differentiation. Treatment with mixtures of AM and AV extracts in a ratio of 3:1 for 24 and 48 hours did not show any cytotoxicity in OP9 cells. Mixtures of AM(3) and AV(1) extracts inhibited adipocyte differentiation, expression of peroxisome proliferator-activated receptor gamma (PPARγ) and CCAT/enhancer-binding protein alpha (C/EBPα), the major transcription factors of differentiation. It also inhibited the expression of lipoprotein lipase (LPL), adipocyte protein 2 (aP2), which are PPARγ-target genes in adipocyte. We also checked the inhibition effects on cell proliferation during the early stage of differentiation by treatment with mixtures of AM(3) and AV(1) extracts. It markedly inhibited adipocyte proliferation after 48 hours, and also the phosphorylation of ERK1/2 and Akt after 10 min or 3 hour. These results identify a possible mechanism of action of mixtures of AM(3) and AV(1) extracts, suggesting that the mixtures of AM(3) and AV(1) extracts-induced inhibition of ERK and Akt phosphorylation suppresses adipogenesis by inhibiting other signaling cascades that include PPARγ and C/EBPα during the process of OP9 adipocyte differentiation.

Effect of Mixture of Atractylodes macrocephala and Amomum villosum Extracts on Body Weight and Lipid Metabolism in High Fat Diet-Induced Obesity Model (고지방식이 유도 비만 모델에서 백출과 사인 추출 혼합물이 체중 및 지질대사에 미치는 영향)

  • In this study, we investigated the dose-dependent effects of mixtures of Atractylodes macrocephala (AM) and Amomum villosum (AV) water extracts in a ratio of 3:1 on high fat diet (HFD)-induced obesity model. Oral administration of various concentrations with mixtures of AM and AV extracts in a ratio of 3:1 for 6 weeks inhibited HFD-induced increases of body, liver and epididymal fat weights in a dose-dependent fashions. Those effects may be mediated by decreased expressions of lipogenesis-related genes such as acetyl coA carboxylase (ACC) and fatty acid synthase (FAS) in liver. Also, increase of insulin and decrease of adiponectin in serum by HFD supply were inhibited by three different dosages of mixtures of AM and AV extracts in a ratio of 3:1. HFD supply induced increases of serum total cholesterol, triglyceride and LDL cholesterol. However, hyperlipidemia was significantly decreased in dose-dependent manners by treatment with mixtures of AM and AV extracts. Based on the results of the present study, hypolipidemic and anti-obesity effects by mixtures of AM and AV extracts were found in HFD-induced obesity model. Further clinical investigation is needed to develop anti-obesity therapeutic or preventive agents by using mixtures of AM and AV extracts.

Ameliorating Effects of Atractylodes macrocephala Koidzumi on TNF-α-induced 3T3-L1 Adipocyte Dysfunction (백출추출물이 TNF-α 유도 지방세포염증과 인슐린저항성 회복에 미치는 영향)

  • Objectives : The present study was designed to investigate effects and molecular mechanisms of Atractylodes macrocephala Koidzumi extracts(AMK) on the improvement of adipocyte dysfunction induced by TNF-${\alpha}$ in 3T3-L1 adipocytes. We examined whether AMK could directly influence the inflammation and insulin resistance in 3T3-L1 adipocytes. Methods : Potential roles of AMK in the lipolysis, production of inflammatory adipokines and ROS, expression and phosphorylation of ERK, JNK, and $I{\kappa}B{\alpha}$ protein, and expression of $PPAR{\gamma}$ and C/EBP${\alpha}$ were investigated in this study. Results : Our data demonstrated that TNF-${\alpha}$ significantly increased lipolysis, levels of MCP-1, IL-6, and ROS and phosphorylation of ERK, JNK, and $I{\kappa}B{\alpha}$ protein, while TNF-${\alpha}$ reduced the expression of $PPAR{\gamma}$ and C/EBP${\alpha}$ in adipocytes, suggesting that TNF-${\alpha}$ induced a condition with the occurrence of inflammation and insulin resistance. Those alterations induced by TNF-${\alpha}$ were prevented by the treatment of AMK. AMK down-regulated the phosphorylation of ERK, JNK, and $I{\kappa}B{\alpha}$ protein and up-regulated the expression of $PPAR{\gamma}$ and C/EBP${\alpha}$ on TNF-${\alpha}$-induced inflammation and insulin resistance. Conclusions : Thus, our results indicate that AMK can be used to prevent from the TNF-${\alpha}$-induced adipocyte dysfunction through MAPK, $NF{\kappa}B$ and $PPAR{\gamma}$ pathways.

The Effect of Transplanting Time on Growth and Yield of Atractylodes macrocephala Koidz. (정식시기가 큰꽃 삽주의 생육 및 수량에 미치는 영향)

  • This study was conducted to determine the appropriate transplanting time for growing one year old rhizome of Atractylodes macrocephala Koidz. Transplanting on March 20 and April 5 showed the highest in plant height and shoot fresh weight but dry rhizome weight was the highest in the transplanting time of April 5. As the growth time of 209 days showed was the highest yield of dry rhizome. As a result, it seemed that the appropriate planting time was April 5 to 10 in the north area of the Kyongsangbuk-do. The shortest emergence time was that the accumulated temperature of early growth period was 267$^{\circ}C$, and that of highest dry rhizome yield was 3$600^{\circ}C$ for whole growth period.

The Mixed Herbal Extract, CAPA, Prevents Obesity and Glucose Intolerance in Obese Mice (비만형 동물모델에서 복합 한약 추출물 CAPA의 비만 및 당대사 개선 효능)

  • Objectives: This study investigated the effects of the mixed herbal extract from Cinnamomum cassia Blume, Atractylodes macrocephala Koidzumi, Pueraria lobata Benth, and Aconitum carmichaeli Debx (CAPA) on obesity and glucose tolerance in obese mice. Methods: Animals were divided in 6 groups, normal diet, high fat diet (HFD), HFD with CAPA 100 mg/kg (CAPA 100), CAPA 300 mg/kg (CAPA 300), and metformin 200 mg/kg or lorcaserin 10 mg/kg as positive controls, and treated for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after treatment. The oral glucose tolerance test was also carried out after treatment. Results: Compared to HFD, CAPA extract treated mice showed significant decreases in body weight, adipose tissue weight, lipid accumulations in liver and serum lipid levels without a reduction of food intake. And fasting glucose and glucose tolerance were all improved in the CAPA treated mice. Conclusions: Our results suggest that CAPA extract can prevent diet induced obesity and glucose intolerance without a reduction of energy intake in obese mice.

Atractylenoide II Isolated from Atractylodes macrocephala Inhibited Inflammatory Responses in Lipopolysaccharide-induced RAW264.7 Macrophages and BV2 Microglial Cells (백출에서 분리된 Atractylenolide II의 RAW264.7 대식세포와 BV2 미세아교세포에서의 항염증 효과)

  • Atractylodes macrocephala is a perennial herb and is a member of the Compositae family. This plant is known to contain various bioactive constituents indicating anti-inflammatory, neuroprotective, anti-oxidant, immunological enhancement, and gastroprotective effects. In this investigation, we isolated four compounds with similar chemical structures from A. macrocephala, and evaluated their anti-inflammatory effects. Among the four compounds, compound 2(atractylenolide II) showed the second-best inhibitory effect on the lipopolysaccharide(LPS)-induced production of nitric oxide in RAW264.7 macrophages and BV2 microglial cells. Compound 2 also inhibited the LPS-induced the production of prostaglandin E2(PGE2), and the expression of inducible nitric oxide synthase(iNOS) and cyclooxygenase(COX)-2 proteins in both cells. In addition, compound 2 suppressed the production of pro-inflammatory cytokines including interleukin(IL)-1β, IL-6, and tumor necrosis factor(TNF)-α. These inhibitory effects were contributed by inactivation of nuclear factor kappa B(NF-κB) and mitogen-activated protein kinases(MAPKs) pathways by treatment with compound 2. This compound did not induce the expression of heme oxygenase(HO)-1 protein indicating that the anti-inflammatory effect of compound 2 was independent with HO-1 protein. Taken together, these results suggested that atractylenolide II can be a candidate material to treat inflammatory diseases.

Growth and Yield by the Different Seeding Methods and Cultivating Root Weight in Atractylodes macrocephala Koidz (백출(Atractylodes macrocephala Koidz)의 파종방법과 종근중에 따른 생육과 수량)

  • This study was conducted to investigate the effects on the growth and yield by the different seeding methods and cultivating root weight in Atractylodes macrocephata Koidz. Seeding distances were different such as 5${\times}$5 cm, 5${\times}$10 cm, 10${\times}$10 cm, 10${\times}$15 cm by the hand seeder and 15 cm seeding in drill. Emergence date, plant height were not significantly changed with seeding space, but the number of leaf, fresh weight of above-ground part and fresh weight of root were increased in the sparse seeding compared with the dense seeding. The highest fresh root yield was 1,012 kg/10 a at the 5${\times}$5 cm. Roots yield was increased in the sparse seeding compared with the dense seeding, but the highest number of roots above 16 g yield was observed at the 10${\times}$10 cm seeding distance. Emergence date was faster 1∼3 day root weight above 6 g than that root weight 5 g. The plant height, number of stem and fresh weight of above-ground part were more increased as the root weight was heavier. The growth of underground part were more increased as the root weight was heavier, yield was increased about 27% to 112% compared with root weight above 6 g than that 134.6 kg/10 a with root 5 g.
