생약재 및 향신료의 Streptococcus mutans 증식 억제 효과 (Antimicrobial Activity of Some Medical Herbs and Spices against Streptococcus mutans)

  • In order to find out an anticariogenic substance, antibacterial activities of some medical herbs and spices against Streptococcus mutans were investigated. The essential oils of oregano, thyme, sage, fennel, nutmeg, rosemary, calamus and cassia cortex were shown antibacterial effect against S. mutans. The essential oil of oregano among other tested medical herbs and spices showed the most active antimicrobial activity. The essential oil of oregano inhibited the activity of glucosyltransferase from S. mutans. The minimal inhibition concentration of oregano essential oil was determined as 0.05nl/ml. The activity of glucosyltransferase from S. mutans was inhibited in the presence of oregano essential oil.

Preparation of Shark Byproduct Extract and Gellan Gum based Antibacterial Film Containing Green Tea Extract

  • In this study, we tried to examine the possibility of developing a dental product such as tooth decay prevention and oral hygiene by manufacturing a natural polymer film for oral use. Natural polymer films were prepared from shark byproduct extract (SBE) and gellan gum (GG). As an antibacterial substance, the antibacterial activity of green tea extract against tooth decay-causing bacteria was measured. An film was prepared by adding green tea extract to the composition of SBE and GG. The mechanical, solubility, moisture content and antibacterial function of the prepared film were investigated in detail. Also, the incorporation of GTE into the SBE/GG film improved the physical performance of the film. Increasing the content of GTE improved the antioxidant and antibacterial properties of the film. Formulation of antimicrobial SBE/GG film containing green tea extract was established and these results evidently showed potential for cavity prevention products application.

갓(Brassica juncea)의 주 항균물질의 구조 분석 (Structural Analysis of Major Antimicrobial Substance Obtained from Leaf Mustard(Brassica juncea))

  • A major component(compound A) in the ethylacetate fraction exhibited a strong antimicrobial activity was identified by UV, IR, FABMS and NMR. The compound A showed strong absorbance at 209, 259 and 359nm, indicating a flavonoid ring structure. IR spectrum possessed absorbance of OH at 3400∼3300cm-1, ketone at around 1650cm-1, and aromatic C=C at around 1660cm-1. Molecular weight of the compound A calculated as 478 from the information of m/z 479(M+H)+ and m/z 477(M-H)+ in the FABMS spectrum. Molecular formula of this compound was found to be C22H22O12 from m/z 479.1220(+3.1mmu for C22H23O12) of HRFABMS spectrum and from 13C-NMR spectrum. 1H-NMR and 13C-NMR spectra of the compound A revealed aromatic proton and benzene rings. Distortionless enhancement by polarization transfer(DEPT) exhibited that the compound A possessed 10 quaternary carbons and 3 substituted benzene rings including a methoxy group substitution. The compound A was identified as isorhamnetin 3-O-β-glucopyranoside by spectrophotometric methods in conjunction with 1H-1H COSY, 1H-13C COSY and HMBC, which revealed a flavone with OH group at 3, 5, 7, and 4' and methoxy group at 3' positions esterified to glucose.

  • Anaerobic bacteria are suggested to be potential source for new antibiotics. In order to search for antibiotics from domestic origin, we collected 800 soil samples across Korean locations and could isolate as many as 989 anaerobic strains. Among them 10, strains were found to have good producing capacity of antibiotics. An anaerobe was finally selected due to secreting antibiotics having high antimicrobial activity towards multiple resistant microorganism(E coli JM 83) transformed by genetic engineering technique. Its morphological, physiological and biochemical charateristics were investigated, together with antimicrobial spectrum therefrom. On antimicrobial spectrum study, substance secreted from this strain, had no activities to fungus and yeast. The selected strain showed G(+) and coccal shape, on Gram, staining and electron scanning microscopy, respectively. Biochemically this strain utilized glucose, fructose lactose, sucrose, but did not arabinose, cellulose, rhamnose, sorbitol, trehalose, mannitol. Catalase test showed negative property. Optimal growth temperature was $37^{\circ}C$. The results obtained above suggest this strain Streptococcus faecium subspp. and we named it Streptococcus sp. An-21-1.

Characterization of Bacteriocin Produced by Lactobacillus bulgaricus

  • The antimicrobial substance produced by Lactobacillus bulgaricus was inactivated by protease. It showed inhibitory activity against Staphylococcus aureus ATCC6538, Streptococcus agalactiae ATCC14364, some Gram-positive and Gram-negative bacteria, and characteristics of a bacteriocin. The optimal temperature and culture time for the production of bacteriocin were $30^{\circ}C$ and 10 h, respectively, in the culture of L. bulgaricus. The bacteriocin production started in the exponential phase and reached a maximum at the early stationary phase. Using Staph. aureus ATCC6538 and Strep. agalactiae ATCC14364, known as common bovine mastitis pathogens, as indicator strains for determination of the bacteriocin activity, the antimicrobial activity of the bacteriocin was found to be stable in acidic and neutral pH's (2- 7) even at lOOT, whereas it was lost at high pH (10- 11) and $100^{\circ}C$. The mode of action for the antimicrobial activity was bacteriocidal, and the molecular weight determined by SDS-PAGE and overlay method was 14 kDa.

  • 치자(Gardenia jasminoides Ellis) 열매의 methanol추출물은 Gram양성 세균, Gram음성 세균, 효모 등의 미생물에 대하여 항미생물 활성을 보여, solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, ODS column chromatography, HPLC등에 의해 얻어진 활성 물질을 MS, GC-MS, $^{1}H-NMR,\;^{13}C-NMR$ 그리고 2D-NMR 등의 기기 분석에 의해 3,4-dihydroxybenzoic acid는 동정하였다. 치자 열매에서 3,4-dihydroxybenzoic acid가 항미생물 활성물질로 분리 동정된 것은 처음으로 생각된다. 3,4-dihydroxybenzoic acid는 건조 치자열매에 g당 $32.7\;{\mu}g$ 정도 함유되어 있었다.

  • 두릅나무(Aralia elata $S_{EEMANN}$) 껍질의 MeOH 추출물이 세균과 효모, 곰팡이에 대해 항미생물 활성을 보여 solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 column chromatography, silica gel partition chromatography, HPLC, TLC 등의 기법으로 주활성본체를 분리하였다. MS, GC-MS, IR, 1^H-NMR, 13^C-NMR 등의 기기분석을 통해 항미생물 활성의 본체구명을 시도한 결과 3,4-dihydroxybenzoic acid로 동정되었다. 3,4-dihydroxybenzoic acid가 항미생물 활성본체로서 분리된 것도 처음으로 생각되며 두릅에서 이 물질이 분리, 동정된 것도 처음으로 생각된다. 건조 두릅껍질 g당 3,4-dihydroxybenzoic acid의 함량은 0.869mg 정도이었다.

소 장관 유래 Enterococcus faecium의 Enterobacter sakazakii에 대한 생육저해활성 (Growth Inhibitory Activity of Enterococcus faecium Isolated from Bovine Intestinal Tract against Enterobacter sakazakii)

  • 소 장관으로부터 분리한 유산균 중에서 Enterobacter sakazakii에 생육저해활성을 나타내는 균주를 분리한 후 동정한 결과 Enterococcus faecium으로 동정되었고 E. faecium JH95로 명명하였다. 본 균주는 kanamycin과 streptomycin에 대해 $100{\mu}g/mL$가지 매우 높은 내성을 나타내었다. 본 균주의 배양액은 L. monocytogenes, C. perfringens와 E. sakazakii에 대하여 높은 항균 활성을 나타내었으며, S. typhimurium, S. aureus와 E. coli O157:H7에 대해서도 항균활성을 나타내었다. 본 균주의 배양 상등액은 항균활성을 보이지 않았으며, 배양액이 지니고 있는 항균활성은 $100^{\circ}C$에서 5분간 가열하거나 단백질 가수분해효소 처리에 의해 소실되어 식중독 미생물에 대한 생육저해물질이 단백질인 것으로 추정되었다.

백작약 추출물이 식중독 유발 세균의 증식에 미치는 영향 (Effect of Extracts from Paeonia japonica on the Growth of Food-borne Pathogens)

  • 본 연구에서는 과거부터 민간이나 한방에서 널리 사용되어져 온 백작약의 식중독 유발 세균에 대한 항균 활성을 조사해 보고자하였다. 백작약을petroleum ether. chloroform, ethylacetate 및 methanol의 4가지 유기용매 및 열수에 추출하고, Gram(+)세균인 Staphylococcus. aureus 및 Staphylococcus. epidermidis와Gram(-)세균인 Salmonella typhimirium, Salmonella paratyphimirium, Shigella dysenteriae, Shigella flexneri, Shigella sonnei 및 Pseudomonas aeruginosa에 대한 성장 억제 실험을 해 본 바, 백작약의 methanol 추출물이 10 mg/mL 농도에서 Salmonella typhimurium에 대해 가장 높은 항균력을 나타내었다. Staphylococcus aureus에 대한 항균력은 백작약의 ethylacetate 추출물이 5 mg/mL에서 가장 큰 항균 효과를 보였으며, 액체 배지에 배양한 경우 5~10 mg/mL 농도에서 18시간까지 성장 억제 효과가 나타났다.

밤나무 잎의 항미생물 효과 및 활성물질 (Active Compounds and Antimicrobial Effects from Castanea crenata Leaf)

  • 밤나무 잎에 함유된 기능성 물질 검색의 일환으로 항미생물 효과를 검정하고 활성물질의 분석을 시도하였다. 예비실험을 통해 추출물의 활성을 확인한 후 시료를 n-hexane, EtOAc, MeOH로 순차 추출하여 정제한 다음 활성이 강한 MeOH 분획의 0.2 g, 0.5 g에 해당하는 추출물로 13종의 미생물에 대해 활성을 측정한 결과, Cram 양성 세균 중에서 S. aureus와 S. epidermidis는 대조구로 사용한 benzoic acid보다 강한 활성이 나타났고, M. luteus와 L. mesenteroides, B. subtilis에서도 활성이 인정되었으며, Gram 음성 세균인 E. coli, S. typhimurium, P. aeruginosa에서도 강한 활성이 나타나 밤나무 잎 추출물은 비교적 넓은 항균 spectra와 활성이 있음이 확인되었다. 이에 활성물질을 규명하고자 시료를 solvent fractionation, silica gel adsorption column chromatography, Sephadex LH-20 column chromatography, HPLC 등으로 정제하고 활성이 나타난 산성 획분과 페놀성 획분의 물질을 분리한 후 MS, $^1H-NMR,\;^{13}C-NMR$ 등으로 분석한 결과, 산성 획분에서는 stigmast- 5-en-3-ol($\beta$-sitosterol)이 활성을 갖는 물질로 나타났다.
