Objectives This study aims to evaluate the wound healing effects of oral administered hot water extracts of Korean black ginseng (KBG). Methods 40 C57BL/6 mice were divided into five groups; normal, control, vitamin E 200 mg/kg, KBG 100 mg/kg, KBG 200 mg/kg, each n=8. Skin wounds were made in the back of all mice except normal group using biopsy punches. Wounds were observed on days 7 and 14 after injury. The anti-oxidant and inflammatory protein levels were evaluated using western blotting. Skin tissue was analyzed by hematoxylin & eosin and Masson's trichrome staining method. Results KBG significantly accelerated reducing wound area. KBG significantly decreased myeloperoxidase activity. KBG significantly decreased oxidative stress factors such as NADPH oxidase-4 and p22phox and increased antioxidant enzymes including nuclear factor erythroid 2-related factor2, kelch-like ECH-associated protein-1, heme oxygenase-1, superoxide dismutase, catalase and glutathione peroxidase-1/2. Moreover, KBG significantly decreased inflammation factors including nuclear factor-κB, phosphorylated inhibitor of κBα, cyclooxygenase-2, inducible nitric oxide synthase, tumor necrosis factor-α and interleukin (IL)-6 and increased anti-inflammation cytokine such as IL-4 and IL-10. In addition, KBG significantly increased tight junction proteins including claudin-1, claudin-3, claudin-4. In histopathologic, KBG made the epithelium thin and uniform, and accelerated the remodeling of collagen. Conclusions The results suggest that KBG has healing effects on skin wound in mice by anti-inflammatory and antioxidant activity.
This study was designed to analyze the election donating ability(EDA), superoxide dismutase(SOD)-like activity, nitrite scavenging ability and xanthine oxidase for food nutritional evaluation and excavation of functional materials in fruits of Elaeagnus multiflora THUNB. The electron donating ability of ethanol extracts in the 1.0 mg/ml concentration of extract was higher in extract of unripe fruits than others. The superoxide dismutase (SOD)-like activity was increased according to increasing of the extract concentration. The SOD-like activity was highest in the extract of unripe fruits among the extracts. The nitrite scavenging ability of unripe fruits ethanol extracts in 1.0 mg/ml of extraction solution in pH 1.2 was higher than extract of ripe and Ovenipe fruits. The nitrite scavenging ability was decreased according to increasing of pH. Also, inhibition of xanthine oxidase activity were higher in extracts of unripe fruits than those in extract of ripe and over-ripe fruits.
In this study, Coffea arabica ethanol extract (CAE) was tested for possible functional cosmetic agent. Whitening effect was measured by tyrosinase inhibition assay, and anti-oxidant activity was checked by SOD-like activity. SOD-like activity of CAE showed $94.8{\pm}6.2%$ at $500{\mu}g/mL$. The anti-bacterial activities CAE was evaluated against three different gram-positive bacteria and six gram-negative bacteria including MRSA strains. CAE exhibited in vitro broad spectrum antimicrobial activities of gram-negative bacteria without antifungal activity. CAE was strong exhibited against MRSA CCARM3561. The tyrosinase and L-DOPA inhibitory activities of the CAE lower than those positive control arbutin. CAE reduced melanin contents of B16-F10 melanoma cell in a dose dependent manner and decrease about 89.2% at a concentration $100{\mu}g/mL$. These result highlight the potential of coffee extract as a naturally active and non-toxic antibacterial suitable for cosmetic applications.

We evaluated the effects of prebiotic (mannan oligosaccharides, Mos) and probiotic diet supplements on growth performance, innate immunity, antioxidant activity, and intestinal changes in the microbial flora of red seabream Pagrus major. A basal diet (Con) was formulated to meet the nutrient requirement of red seabream. The dietary starch in Con was replaced with 0.6% Mos, Lactobacillus plantarum, Bacillus subtilis, B. licheniformis and probiotic mixture (labeled as Mos, Pro-LP, Pro-BS, Pro-BL and Pro-Mix, respectively). We stocked 450 fish in 18 polypropylene tanks (400 L) in triplicate groups per dietary treatment. The fish were fed one of the diets twice (08:30, 18:30 h) a day for 63 days. Lysozyme activity was significantly higher in all the supplemented groups than that of the Con group. The immunoglobulin level of Pro-Mix, anti-protease activity of Pro-BL, and glutathione peroxidase and superoxide dismutase activity of Pro-BS, Pro-BL and Pro-Mix groups were significantly higher than those of the Con group. The ratio of total Vibrio/heterotrophic marine bacteria counts was significantly lower in Pro-LP, Pro-BL and Pro-Mix groups than that of the Con group. Therefore, dietary supplementation of Mos and probiotics to improves immune response and antioxidant enzyme activity and inhibits Vibrio bacteria in the intestine.

The water fern Azolla belongs to the Azollaceae and forms a symbiotic association with a $N_2$-fixing cyanobacterium, referred to as Anabaena azollae, and this association has currently been demonstrated to have potential as a nitrogen source for rice production. Because of that, Azolla fern has been used not only as organic manure in southern China and northern Vietnam for a long time but also as food for animate creatures in the underwater and decontaminant in the water. However, the phenolic compounds and active materials of Azolla have not been examined in detail in the past studies. In the present study, anti-oxidant ability test and experiment to find a particular active material of Azolla imbricata and Azolla imbricata fraction (AIF) were performed. In anti-oxidant test such as DPPH test and lipoxygenase inhibition test, the value of test represented high activities compared with authentic sample - green tea and NDGA (nordihydroguaiaretic acid). In MMP-1 test, related to collagen protection and elasticity of skin, its inhibitory effect was measured over 75 %, and the phenolic compounds of AIF related with this activity were confirmed luteolin derivatives by using FT-IR spectroscopy, element analyzer (EA) and Liquid chromatography-MASS spectroscopy.
Some flowers have high sensual appealability due to their unique shapes, colors, smells, and tastes. Such edible flowers receive social attention as a noble ingredient of functional teas. In this study, methanol extracts of 23 commercial flower teas (CFTs) were prepared, and their color differences were compared. No tar color pigments were detected in the 23 CFT. The average content of total polyphenol of the 23 CFTs was 80.2±50.92 mg/g and the extracts of jin-dal-rae (Rhododendron mucronulatum Turcz), mae-hwa (Prunus mume), mae-mil (Fagopyrum esculentum), mok-ryun (Magnolia kobus), and sal-gu (Prunus armeniaca var. ansu Maxim) flowers showed total polyphenol contents greater than 150 mg/g. The average content of total sugar of the 23 CFTs was 187.4±166.5 mg/g and the extracts of chamomile (Chamaemelum nobile), kuk-hwa (Chrysanthemum morifolium), dong-baek (Camellia japonica L.), and won-chu-ri (Hemerocallis fulva) flowers showed total sugar contents greater than 400 mg/g. Among the 23 CFTs, the extract of jang-mi (Rosa hybrida hortorum) flower has prominent anti-thrombosis activity, and the extracts of dal-ma-ji (Oenothera lamarckiana), dong-baek, hibiscus (Hibiscus syriacus), and mae-mil flowers showed strong inhibitions against thrombin and blood coagulation factors. Also, the extracts of jang-mi, kum-jan-hwa (Tagetes erecta L.), mae-mil, mok-ryun dong-baek, and jin-dal-rae flower showed strong radical scavenging activities against DPPH, ABTS, and nitrite and reducing power. Our results suggest that the flowers of jang-mi, mae-mil, and dong-baek can be developed as promising anti-thrombosis treatments.
BACKGROUND/OBJECTIVES: Citrus flavonoids have a variety of physiological properties such as anti-oxidant, anti-inflammation, anti-cancer, and anti-obesity. We investigated whether bioconversion of Citrus unshiu with cytolase (CU-C) ameliorates the anti-adipogenic effects by modulation of adipocyte differentiation and lipid metabolism in 3T3-L1 cells. MATERIALS/METHODS: Glycoside forms of Citrus unshiu (CU) were converted into aglycoside forms with cytolase treatment. Cell viability of CU and CU-C was measured at various concentrations in 3T3L-1 cells. The anti-adipogenic and lipolytic effects were examined using Oil red O staining and free glycerol assay, respectively. We performed real time-polymerase chain reaction and western immunoblotting assay to detect mRNA and protein expression of adipogenic transcription factors, respectively. RESULTS: Treatment with cytolase decreased flavanone rutinoside forms (narirutin and hesperidin) and instead, increased flavanone aglycoside forms (naringenin and hesperetin). During adipocyte differentiation, 3T3-L1 cells were treated with CU or CU-C at a dose of 0.5 mg/ml. Adipocyte differentiation was inhibited in CU-C group, but not in CU group. CU-C markedly suppressed the insulin-induced protein expression of CCAAT/enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) as well as the mRNA levels of $CEBP{\alpha}$, $PPAR{\gamma}$, and sterol regulatory element binding protein 1c (SREBP1c). Both CU and CU-C groups significantly increased the adipolytic activity with the higher release of free glycerol than those of control group in differentiated 3T3-L1 adipocytes. CU-C is particularly superior in suppression of adipogenesis, whereas CU-C has similar effect to CU on stimulation of lipolysis. CONCLUSIONS: These results suggest that bioconversion of Citrus unshiu peel extracts with cytolase enhances aglycoside flavonoids and improves the anti-adipogenic metabolism via both inhibition of key adipogenic transcription factors and induction of adipolytic activity.
The current study was performed to investigate the changes of bio-activities of Salicornia europaea (SE) depending on the different dry methods. The ethanol extracts were prepared from the freeze-drying SE (FD), hot-air drying SE (HD), and shade drying SE (SD). Their anti-oxidant and anti-thrombosis activities were compared. The yields of ethanol extraction in FD, HD, and SD were 14.4, 13.2 and 11.9%, respectively. The highest contents of total polyphenol (4.6 mg/g) and total sugar (23.4 mg/g) were shown in FD, whereas, the highest content of reducing sugar (14.6 mg/g) was observed in HD. In anti-oxidation activity assay, the FD and HD showed similar radical scavenging activities and reducing power. However, in SD, nitrite scavenging activity and reducing power were severely decreased. In anti-coagulation activity assay, the thrombin time (TT), prothrombin time (PT) and activated partial thromboplastin time (aPTT) of FD, HD and SD did not show significant changes at 5 mg/mL concentration. But the HD at 7 mg/mL had strong inhibitions against thrombin and blood coagulation factors. The platelet aggregation and hemolysis activities were not affected by dry methods. Our results suggest that both FD and HD are effective to maintain the functional ingredient of SE, and HD is economic and efficient dry process for production of functional food.

In this study, the anti-inflammatory and anti-oxidant efficacy of camellia subcritical water extracts (SWE, 135 ~ 180 ℃, 70 bar) was compared with 70% ethanol and hot water extracts. Among these extracts, the yield (57.9%) of the subcritical water extract, which was extracted under the condition of 180 ℃ and 70 bar was the highest, which increased the extraction yield by more than two times compared to the hot water extract (28.1%). The results of the nitric oxide (NO) production inhibition activity experiment using RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS) showed that subcritical water extracts had superior effects in inhibiting the production of NO without cytotoxicity than 70% ethanol and hot water extracts. In addition, DPPH and ABTS+ radical scavenging activity experiments showed that the radical scavenging activity of subcritical water extract was similar to that of 70% ethanol and hot water extract. Moreover, the content of gallic acid was determined by HPLC and the quantity was about 1.62 mg/g for the SWE (165 ℃, 70 bar), which was the highest among all of the extracts. Based on these results, it is concluded the SWE of C. japonica flowers could be potentially applicable as anti-inflammatory and anti-oxidative ingredients in cosmetic formulations.
Akebiae caulis have been used in folk medicines for diuretic, menstrual pain, and diuretic pain. It has been also resolved nephritis and cystitis. In this study, we isolated three phenolic compounds from 70% methanol extract using the open column chromatography. These isolated compounds which were 2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside, 5-O-caffeoylquinic acid, and syringoylglycerol 2-O-${\beta}$-D-glucoside were determined by physico-chemical apparatus. Furthermore, we conducted DPPH and ABTS assay in order to screen the antioxidant activity of isolated three compounds. Also, we developed a rapidly HPLC-UV analysis method of two phenolic compounds (2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside, 5-O-caffeoylquinic acid) for evaluating the Akebiae Caulis collected 30 samples from different regions. From the experiments, all three isolated compounds showed the significant antioxidant activity. We suggested that the content criteria of marker compounds were shown by a simple and rapid HPLC-UV method. The contents respectively were 0.009% (2-(3,4-dihydroxyphenyl)-ethyl-${\beta}$-D-glucopyranoside) and 0.036% (5-O-caffeoylquinic acid).
