고지방식이로 유도된 비만 생쥐모델에서 열다한소탕의 항비만효과 (Anti-obesity Effect of Yeoldahanso-tang on Obesity Mice)

  • Objectives This study was conducted to evaluate the effects of Yeoldahanso-tang on obesity in rats induced by high fat diet experimentally. Methods The experiment was conducted with rats divided into 5 groups. Rats were evaluated for change of weight, hematologic and serum biochemical parameters. Results Yeoldahanso-tang group showed significant reductions in FER, body weight, adipose tissue weight and size. The level of Creatinine, glucose, ALP, T-cholesterol, Triglyceride, LDL-cholesterol, Leptin and IGF-1 of Yeoldahanso-tang group was significantly lower than those in HFD-CTL group. The level of HDL-cholesterol, free fatty acid and Adiponectin in Yeoldahanso-tang group was significantly higher than those in HFD-CTL group. As compared with HFD-CTL group, AMPK-${\alpha}1$, UCP2 and adiponectin mRNA in liver of Yeoldahanso-tang group were significantly increased and AP2/FABP4, AMPK-${\alpha}2$ and PPAR-${\gamma}$ mRNA in liver of Yeoldahanso-tang group was significantly decreased. Conclusion These results suggest that Yeoldahanso-tang has inhibitory effects on obesity in high fat diet induced obese mice.

Anti-aging Activity of Aralia Cordata Thunb. by Inhibiting Oxidized Low-dencity Lipoprotein Production in Rats

  • Aralia cordata Thunb. (Araliaceae, ACT) is an remarkable herbal plant that has been widely used in traditional oriental medicine for the treatment of inflammatory diseases and cardiovascular disorders. In this study, we have established a vascular aging model in rats by orally administrating excessive vitamin $D_2$ (500,000 IU/kg/day) for 4 days followed by feeding high cholesterol diet for 16 weeks and then rats were randomly divided into control group, high cholesterol diet (HCD) group, HCD+ACT (30 mg/kg) and HCD+ACT (60 mg/kg) group. ACT (30, 60) significantly reduced total cholesterol (TC) content compared with HCD, but no significant differences in the serum lipids. Secondly, we measured the serum levels of Oxidized Low-dencity Lipoprotein (OxLDL) and malondialdehyde (MDA) in order to further investigate the anti-vascular aging mechanism of ACT. The results, ACT (30, 60) treatments decreased OxLDL, MDA content and increased Cu/Zn superoxide dismutase activity compared with HCD treatments. The results suggested that ACT inhibited OxLDL production rather than serum lipids lowering and that ACT could be used as potential anti-atherosclerotic agent in aged cells.

흰쥐 건성피부에 대한 목이버섯의 물추출물과 에탄올추출물의 생리적 효능 (Physiological Effects on Dried Skin in Rat by Water & Ethanol Extracts of Auricularia Auricula U.)

  • It has been well known that mushroom polysaccharide extracts have superoxide and hydroxyl radical scavenging activities. So we studied the efficacy and anti-oxidative effect of mushroom(Auricularia auricula U.) in rats treated with water and ethanol extracts of Auricularia auricula U after inducing dryness of skin by the application of detergent. The results of the research for of the efficacy and anti-oxidative effect Auricularia auricula U. are as follows thats. Weight gain and feed intake in animals treated with water extracts and ethanol extracts are significantly higher(p<0.05) than animal treated with detergent. Animals treated with water extracts of Auricularia auricula U. compared to animals treated with ethanol extracts have the higher HDL-cholesterol levels in the blood and the lower LDL-cholesterol levels in the blood. SOD-like activity of the ethanol extracts of Auricularia auricula U. was $27.39{\pm}1.61%$ at 2000ppm.

  • The anti-hyperlipidemic effect of Houttuynia cordata was assessed in poloxamer-407 induced hyperlipidemic mice model. The butanol fraction and its isolated compound, quercetin-3-O-rhamnoside, significantly reduced the blood triglyceride and total-cholesterol level and increased the blood HDL-cholesterol level. They also showed the significant reductive effect on the blood AST and ALT level, rising in proportion to the liver damage, in hyperlipidemic mice.

Herbal Black Vinegar and the Anti-obesity Complications in vivo

  • Black vinegar has been traditionally used for supplemental flavoring on food, and commercialized beverages. Here, to investigate the effects on in vivo anti-obesity complications of black vinegar produced with herbal extracts, we evaluated on the biochemical effects of high-fat diet (HFD) induced mice compared to control fed ones. After a 84-day experiment HFD mice had higher (P < 0.05) weight gains, relative abdominal-fat pads, blood glucose level, serum/liver lipids, and serum nephron indices. Continuous oral treatment of three different concentration of herbal black vinegar (HBV; stock, 2-fold, and 4-fold diluted solution) to HFD mice showed that HBV reduced marked obesity (fat depositions, adipocyte hypertrophy), hyperglycemia, hyperlipidemia (serum total cholesterol, triglyceride, LDL-cholesterol levels), enhanced liver function (AST/ALT), and kidney function (BUN, creatine levels), respectively. Thus, HBV is expected to serve as an efficient and functional supplemental ingredients or food for the alleviation of obesity syndrome.

The Functional Properties of Preserved Eggs: From Anti-cancer and Anti-inflammatory Aspects

  • Preserved egg, a kind of alkaline-fermented food, is a traditional egg product in China. Here, we investigated the nutritional functions of preserved eggs by in vivo and in vitro experiments. The results of in vivo studies showed that the levels of triglycerides (TG), total cholesterol (TCHO) and low-density lipoprotein cholesterol/high density lipoprotein cholesterol (LDL-C/HDL-C) were significantly decreased (p<0.05) in the liver of rats treated with preserved eggs. Meanwhile, the levels of two important cancer markers, interleukin-6 (IL-6) and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), were also significantly decreased (p<0.05) in treated rats. In vitro studies were performed on Caco-2 cells, a human epithelial colorectal adenocarcinoma cell line. It demonstrated that the gastrointestinal (GI) digests of preserved eggs significantly accelerated (p<0.05) the apoptosis by upregulating caspase-3 in the Caco-2 cells. Besides, after treated with preserved eggs, the half maximal inhibitory concentration (IC50) of preserved eggs digests to Caco-2 cells was 5.75 mg/mL, indicating the significant inhibition of cell proliferation provided by preserved eggs (p<0.05). The results shown in this study demonstrated that preserved eggs may be a novel functional food involved with antilipemic, anti-inflammatory activity as well as the effect on accelarating the apoptosis of Caco-2 cells.

흰쥐의 혈중 콜레스테롤 및 HDL-콜레스테롤 함량에 미치는 Clotrimazole의 영향에 관한 연구 (Influences of Clotrimazole on the Blood Cholesterol and HDL-Cholesterol level in Rats)

  • Influences of clotrimazole on the blood cholesterol and HDL-cholesterol level were studied in rats. Rats were provided food and water ad libitum and clotrimazole and methylcellulose were gavaged for 6 days. Clotrimazole was suspended in 1% methylcellulose solution as and administered at concentration 20mg/Kg, 40mg/Kg, 60mg/Kg. Body weight gain and liver weight/body weight ratio, serum cholesterol level, serum HDL-cholesterol level, serum triglyceride level, the activity of cytochrome p450 and erythromycin demethylase were determined at 6th day. Clotrimazole decreased the body weight gain a little as compared with control group and did not show any influence on liver weight/body weight ratio. Clotrimazole increased the serum HDL-cholesterol and serum triglyceride level significantly. Clotrimazole increased the microsomal cytochrome P450 significantly and increased the erythromycin demethylase (cytochrome P450 IIIA) significantly too. It might be conclued that clotrimazole showed a little influence on body weight and increased the serum lipid, especially HDL-cholesterol level. It also increased microsomal cytochrome P450 IIIA significantly. It might be concluded that clotrimazole showed a corelative influence between HDL-cholesterol and cytochrome P450 IIIA. In these results clotrimazole can be used as an anti-atherosclerotic agent by increasing the HDL-cholesterol but it is necessary that cloreimazole will show any adverse or side action on body or not.

Anti-hyperlipidemia and Anti-arteriosclerosis Effects of Laminaria japonica in Sprague-Dawley Rats

  • The anti-hyperlipidemic effects of dietary supplementation with sea tangle Laminaria japonica were investigated using an animal model in which normal rats were fed either sea tangle, sea tangle ethanol extract (EE-ST) and sea tangle extracted residue (ER-ST). Total lipid and triglyceride levels in the serum were significantly (P < 0.05) reduced in rats fed ER-ST at a dose of 200 mg/kg body weight when compared to hyperlipidemic control rats. Significant decreases in serum total cholesterol and low density lipoprotein-cholesterol levels also occurred in rats fed ER-ST at 200 mg/kg body weight. In addition, the atherosclerosis index and superoxide dismutase in blood lipids were significantly (P < 0.05) lowered in rats fed ER-ST at 200 mg/kg body weight as compared to control rats. In conclusion, sea tangle and ER-ST exhibited beneficial anti-hyperlipidemic and anti-arteriosclerosis effects.

갈근이 비만 랫드 간조직의 비만관련 유전자 발현에 미치는 영향 (The effects of Pueraria lobata extract on gene expression in liver tissue of rat with estrogen-deficient obesity)

  • Objective : It is known that Pueriaria lobata has an anti-osteoporetic effect, anti-cancer effect, anti-pyretic effect, and anti-diabetic effect. The aim of this study was to evaluate anti-obesity effect of Pueriaria lobata extract (PLE), and elucidate the effect of it on gene expression related to lipid metabolism. Method : The experiments were performed with the use of ovariectomized rats as estrogen-deficient obesity model. They were grouped NC (normal control), OC (estrogen-deficient control), PLH (100mg/kg of PLE), PLL (20mg/kg). PLE was orally administered for 6 weeks. Body weights and serum lipid level were estimated, and real-time PCR was performed to investigate the effect of PLE on gene expression in liver. Results : PLE decreased the body weight and serum cholesterol and triglyceride, but increased HDL-cholesterol. And PLE increased leptin, CYP27, CPT1, CYP8B1, ACAT2, LDLR, and SCD1, but reduced $PPAR{\gamma}$, PGC1A, HMG-CoA-R, ACAT1, SCD1, and APoB gene expression in liver tissue of rat with estrogen-deficient obesity. Conclusion : It is concluded that Pueriaria lobata reduced body weight, and its effect was expressed by regulation of gene expression related to lipid metabolism in rats with estrogen-deficient obesity.

고지방식이로 유도한 비만마우스에서 berberine과 silibinin 복합투여를 통한 지질대사 개선과 항비만 효능 증진 (Combination of berberine and silibinin improves lipid metabolism and anti-obesity efficacy in high-fat diet-fed obese mice)

  • 본 연구에서는 고지방식이(HFD)를 급식하여 제조한 비만마우스 모델을 사용하여 berberine (BBR)과 silibinin (SBN) 복합투여가 혈중 지질대사 및 항비만 개선 효능에 유의적인 시너지 효과가 있는지 조사하였다. HFD로 유도된 비만마우스를 8 주 동안 HFD의 지속적인 제공와 함께 BBR 및 SBN (BBR-SBN) 조합을 투여하였다. 실험이 진행되는 동안 체중과 식이량을 측정하였고 혈중 총 콜레스테롤, 중성지방 및 HDL 콜레스테롤 수준을 분석하였다. HFD를 제공한 마우스는 정상 대조군(NC) 그룹에 비해 체중과 총 콜레스테롤 및 중성지방 수치가 급격히 증가했다. 그러나 이러한 비만마우스에 BBR-SBN조합을 투여하였을 때 체중 증가가 현저하게 감소하였고 HDL 콜레스테롤 수치가 증가하였으며 총 콜레스테롤 및 중성지방 수치는 유의하게 억제되었다. HFD그룹의 복부지방 무게는 유의하게 증가했으며 부고환 지방조직 내의 지방세포의 크기가 NC 그룹에 비해 크게 확장된 것으로 나타났다. 그러나 BBR-SBN 그룹에서는 지방세포의 크기가 NC 그룹의 크기와 비슷했으며 복부지방 무게가 현저하게 감소하였다. 더불어, HFD 그룹에서 보이는 간 조직의 거대 소포성 지방구의 축적은 BBR-SBN 그룹에서 크게 감소되었다. 이러한 결과는 BBR-SBN 조합이 HFD 유발 비만마우스에서 체중 및 복부 지방 증가를 현저하게 감소시키는 경향이 있으며 혈청 내의 총 콜레스테롤 및 중성지방 수준을 낮추어 항비만 효능을 개선시킬 수 있는 가능성을 보여주는 것으로 앞으로 항비만 치료 및 개선제제로서의 잠재력을 가지고 있음을 시사한다.
