Quinic Acid Alleviates Behavior Impairment by Reducing Neuroinflammation and MAPK Activation in LPS-Treated Mice

  • Compared to other organs, the brain has limited antioxidant defenses. In particular, the hippocampus is the central region for learning and memory and is highly susceptible to oxidative stress. Glial cells are the most abundant cells in the brain, and sustained glial cell activation is critical to the neuroinflammation that aggravates neuropathology and neurotoxicity. Therefore, regulating glial cell activation is a promising neurotherapeutic treatment. Quinic acid (QA) and its derivatives possess anti-oxidant and anti-inflammatory properties. Although previous studies have evidenced QA's benefit on the brain, in vivo and in vitro analyses of its anti-oxidant and anti-inflammatory properties in glial cells have yet to be established. This study investigated QA's rescue effect in lipopolysaccharide (LPS)-induced behavior impairment. Orally administering QA restored social impairment and LPS-induced spatial and fear memory. In addition, QA inhibited proinflammatory mediator, oxidative stress marker, and mitogen-activated protein kinase (MAPK) activation in the LPS-injected hippocampus. QA inhibited nitrite release and extracellular signal-regulated kinase (ERK) phosphorylation in LPS-stimulated astrocytes. Collectively, QA restored impaired neuroinflammation-induced behavior by regulating proinflammatory mediator and ERK activation in astrocytes, demonstrating its potential as a therapeutic agent for neuroinflammation-induced brain disease treatments.

The Effects of Red Ginseng and Fermented Red Ginseng on Neurotoxicity in Mice Induced by MPTP (홍삼과 발효홍삼이 MPTP에 의해 유도된 생쥐의 신경독성에 미치는 영향)

  • This research observed the interrelations between the active components found specifically in red ginseng and fermented red ginseng from among the variety of ginseng variations and the protective effect and anti-oxidant effect exercised on brain cells in the animal model for MPTP-induced neurotoxic Parkinson's Disease and obtained the following conclusions. The results above comprehensively demonstrated that the fermented red ginseng extract exercised greater protective effects against oxidant brain damage by MPTP when compared to the group administered with the red ginseng extract. This was induced an increase in TH protein expression, and further raised the efficiency of the anti-oxidant enzyme defensive system against neurotoxicity, thereby restraining the lipid peroxidation caused by the active oxygen generated during the course of MPTP metabolism and enhancing the body's defensive capacities in response to tissue damage, thereby demonstrating a protective effect against MPTP induced neurotoxicity.

The Literature Study of Research Trend of Alismatis Rhizoma and Relationship Between the Herbology and KCD (택사(澤瀉)의 국내·외 연구동향과 『본초학』, 한국표준질병사인분류의 상관관계에 대한 연구)

  • Objectives : This study was aimed to analyze the correlation between the Herbology and contemporary research results, KCD and terms of Alismatis Rhizoma. The study will present information that can be used to find the direction of further researches and be applied to the education of Herbology.Methods : Papers were searched in Riss4U and PubMed, and categorized. Medicine or pharmacy articles about Alismatis Rhizoma were matched with the Herbology treatment and KCD. KCD-codes and terms were arranged by the Herbology treatment. The Degree of Herbology research (HDR) was calculated by numbers of papers, study method, citation rates.Results : There were 392 articles about Alismatis Rhizoma. Among these there were 141 medicine and pharmacy articles about Alismatis Rhizoma, and 51 articles were matched to the Herbology treatment. Studies about Phlegm-Fluid Retention and Dizziness, Edema and Distention and Fullness was more active than others. Analysis of other articles showed that studies about anti-cancer, anti-biotic effect, anti-diabetes mellitus, anti-oxidant effect, anti-inflammation, hepatoprotective effect were active, too. In HDR, Phlegm-Fluid Retention and Dizziness was scored by 181, Edema- Distention and Fullness by 173, Hyperlipidemia by 83, Dysuria 64, Heat strangury pain by 18, Diarrhea and decreased urine volum by 0.Conclusions : 141 articles about Alismatis Rhizoma were analyzed and 51 articles were matched to the Herbology treatment. Studies about Phlegm-Fluid Retention and Dizziness, Edema and Distention and Fullness were more active than others. And studies about anti-cancer, anti-biotic effect, anti-oxidant effect, anti-inflammation, Osteoporosis should be more studied and be more discussed among Herbology researchers.

Anti-Cancer Effects of Green Tea by Either Anti- or Pro-Oxidative Mechanisms

  • Tea derived from the leaves and buds of Camellia sinensis (Theaceae) is consumed worldwide. Green tea contains various components with specific health-promoting effects, and is believed to exert protective effects against diseases including cancer, diabetes and hepatitis, as well as obesity. Of the various tea components, the polyphenol catechins have been the subject of extensive investigation and among the catechins, (-)-epigallocatechin gallate has the strongest bioactivity in most cases. Our research group has postulated that hepatocyte nuclear factor-$4{\alpha}$, sterol regulatory element-binding proteins, and tumor necrosis factor-${\alpha}$ are targets of green tea constituents including (-)-epigallocatechin gallate for their anti-diabetes, anti-obesity, and anti-hepatitis effects, respectively. Published papers were reviewed to determine whether the observed changes in these factors can be correlated with anti-cancer effects of green tea. Two major action mechanisms of (-)-epigallocatechin gallate have been proposed; one associated with its anti-oxidative properties and the other with its pro-oxidative activity. When reactive oxygen species are assumed to be involved, our findings that (-)-epigallocatechin gallate downregulated hepatocyte nuclear factor-$4{\alpha}$, sterol regulatory element-binding proteins, and tumor necrosis factor-${\alpha}$ may explain the anti-cancer effect of green tea as well. However, further studies are required to elucidate which determinant directs (-)-epigallocatechin gallate action as an anti-oxidant or a pro-oxidant for favorable activity.

Anti-oxidative and Anti-inflammatory Activities of Polysaccharide isolated from Korean-Style Soy Sauce

  • Soy sauce is one of the representatives of traditional fermented foods in Korea. However, studies on soy sauce are relatively insufficient in Korea compared to Japan. In this study, antioxidant and anti-inflammatory activities of polysaccharides were measured by polysaccharides isolated from two different soy sauces, Korean and Japanese (KSS-0 and JSS-0). KSS-0 was purified into two fractions using gel chromatography and named them as KSS-I and KSS-II. To investigate the antioxidant activity of the polysaccharides, we measured the polyphenol content and radical scavenging activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). To investigate the anti-inflammatory activity of polysaccharides, we used RAW 264.7 macrophage cells and induced inflammation using lipopolysaccharide (LPS). Then, we measured levels of inflammatory mediators such as nitric oxide (NO) and tumor necrosis factor (TNF)-α. Among the four polysaccharides, KSS-II showed the highest antioxidant activity and had good anti-inflammatory activity; KSS-II decreased inflammatory mediators in a dose-dependent manner. In conclusion, the polysaccharide isolated from Korean soy sauce (KSS-II) showed better anti-oxidant and anti-inflammatory activities than polysaccharides isolated from Japanese soy sauce, and may be useful as substances for functional foods.

Effects of Panax Notoginseng Flos extract on Anti-oxidative, Anti-inflammatory and Acne skin (삼칠화 에탄올 추출물의 항산화, 항염증 활성 및 여드름 피부에 미치는 영향)

  • The purpose of this study is to observe how Panax Notoginseng Flos Ethanol Extract has anti-oxidant activity, cytotoxicity for skin cells, and anti-inflammatory effects and Cosmetics Panax Notoginseng Flos Ethanol Extract was applied to acne skin to analyze moisture, erythema, pores, and blackberries to identify potential as cosmetics material .According to the analysis, Panax Notoginseng Flos Ethanol Extract was found to have high contents of polyphenols and flavonoids and excellent DPPH radical scavenging activity. The extract had no significant cytotoxicity for Raw 264.7 cell and significantly inhibited the creation of NO induced LPS in Raw 264.7 cell so as to present anti-inflammatory effect. After applying an Panax Notoginseng Flos Ethanol Extract of 3% skin toner and spot Solutions to the skin of the skin of the acne for 3 weeks, Increased skin moisture, erythema, pores, black head count were found to be statistically signigicant, and reduced red spots were found in skin condition questions following management. Given the results, it is considered that Panax Notoginseng Flos Ethanol Extract is applicable as the ingredient of a functional cosmetic product that has low cytotoxicity for skin cells, high anti-oxidant activity, anti-inflammatory effect, and acne improvement effect.

Study on Anti-oxidant and Anti-inflammatory Activity of the Tissue-cultured Shoot Clumps from Raoulia australis (라울리아 신초 추출물의 항산화 및 항염 효능에 관한 연구)

  • In this study, the shoot clumps extract of tissue-cultured Raoulia australis using the bioreactor culture system was tested for use a natural cosmetic ingredient. Tissue-cultured R. australis shoot clumps extract was tested anti-oxidant and anti-inflammatory activity for a cosmetic application. R. australis is a wild herbaceous plant of the asteraceae growing in New Zealand and Australia. Previous studies have reported anti-viral activity of the inhibitory effects for the growth of viruses induced meningitis, bronchitis and respiratory diseases but other biological effects are unknown. The shoot clumps extract of tissue-cultured R. australis showed higher anti-oxidant effect and anti-inflammatory effect than the natural R. australis extract. In DPPH, NBT and ABTS assay, the shoot clumps extract of tissue-cultured R. australis enhanced radical scavenging activity (up to 10~25% at $50{\mu}L/mL$) more than the natural R. australis extract. Also, the shoot clumps extract of tissue-cultured R. australis inhibited expression of iNOS and COX-2 protein in LPS-stimulated Raw 264.7 macrophages more than the natural R. australis extract. From this study, the shoot clumps extract of tissue-cultured R. australis displayed strong possibility as a new natural cosmetic ingredient for skin-care products.

The Anti-oxidant and Anti-microbial Activities of Purified Syringin from Cortex Fraxini (진피로부터 정제한 Syringin의 항산화 및 항균 활성)

  • The purpose of this study is to evaluate the anti-oxidant and anti-microbial activity of syringin isolated from Cortex Fraxini to investigate their potential for use as safe natural compounds. Purified syringin was dissolved in distilled water for each concentration and used in each experiment. Syringin showed higher 2,2-Diphenyl-1-picrylhydrazyl radical scavenging than butylated hydroxytoluene (BHT) at a concentration of 50 ㎍/ml. In 2,2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging, activity was similar to that of BHT at all concentrations. In antioxidant protection factor measurement, activity of syringin slightly increased as the concentration increased, as did the inhibitory effect of thiobarbutric acid reactive substances. In evaluating anti-microbial activity, the clear zones of Listeria monocytogenes KCTC 13064, Staphylococcus aureus KCTC 1916, Escherichia coli KCTC 2571, and Helicobacter pylori HPKCTC B0150 at a concentration of 200 ㎍/ml were found to be 17.8 mm, 20.45 mm, 17.05 mm, and 16.8 mm, respectively, but no clear zone was observed in the case of Candida albicans ATCC 10231. The activity against water-soluble antioxidants was therefore superior to that against lipid-soluble antioxidants. Anti-microbial activity was examined by inhibiting growth against gram-positive and -negative strains, and anti-fungal activity was not observed. Based on the results of this study, syringin has possible applications as a natural anti-oxidant and anti-microbial material.

Availability of Passiflora Caerulea Extract as Inner Beauty Material (이너뷰티 소재로서의 시계꽃 추출물의 활용 가능성)

  • This study attempted to investigate the physiological activities of Passiflora caerulea extract and provide basic data needed to verify its availability as an inner beauty material. To examine its anti-oxidant, anti-inflammatory and anti-aging effects, the followings were measured: total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity, cytotoxicity and NO inhibition effects in RAW 264.7 macrophage cells and HDF cells and MMP-1 inhibitory effects in HDF cells. The results found the followings: First, polyphenol and flavonoid contents were great with 157 mg/g and 173.5 mg/g respectively at 10mg/mL, and DPPH radical scavenging activity was confirmed. In RAW 264.7 macrophage cells and HDF cells, no cytotoxicity was observed for Passiflora caerulea extract. In addition, 69.3% anti-inflammatory effects were found at 100 ㎍/mL through NO inhibition in RAW 264.7 macrophage cells. In addition, a significant inhibition of MMP-1 expression in HDF cell was confirmed. The above results confirmed the anti-oxidant and anti-inflammatory effects of Passiflora caerulea extract, low toxicity on skin cells and its anti-aging effects by inhibiting MMP-1 expression. Therefore, it appears that Passiflora caerulea extract would be available as diverse inner beauty materials.

Anti-oxidant and Anti-inflammatory Effects of Ethanol Extracts from Aerial Part of Coriandrum sativum L. (고수(Coriandrum sativum L.) 지상부 추출물의 항산화 및 항염증 활성 효과)

  • Coriandrum sativum L., an annual herbaceous plant of Apiaceae family. The present study evaluated the anti-oxidant activities and anti-inflammatory effects of ethanol extracts of C. sativum. The anti-oxidant activities of C. sativum were measured by total contents of polyphenol, flavonoid, DPPH and ABTS radical scavenging and reducing power activity. And anti-inflammatory effects of C. sativum were measured by LPS-induced RAW 264.7 cells. The results showed that the contents of total polyphenol and flavonoid were 76.03 ± 1.36 mg of gallic acid equivalents/g and 182.23 ± 4.32 mg of rutin equivalents/g at concentration 1 mg/mL of C. sativum. The DPPH radical scavenging activity was found to be 52.8% at 500 ㎍/mL. The ABTS radical scavenging activity was shown in 58.3% after exposure to 1,000 ㎍/mL. Reducing power activity was found to be 66.8% at 2,000 ㎍/mL. The inhibitory effect of NO production was found to be 65% concentration 500 ㎍/mL. In the generation quantity of inflammatory cytokines such as TNF-α and IL-1β in cell culture medium, the expression levels of inflammatory proteins in cells were showed decrease with the increase of concentration. Therefore, we suggest that the C. sativum should be a potential source of alternative anti-inflammatory drug with good anti-inflammatory effects.
