Toxic Levels of Amyloid Beta Peptide Do Not Induce VEGF Synthesis

  • Alzheimer's disease is a neurodegenerative disorder associated with progressive loss of cognitive function and memory. Amyloid beta peptide ($A{\beta}$) is the major component of senile plaques and is known to exert its cytotoxic effect mainly by producing $H_2O_2$. Vascular endothelial growth factor (VEGF) is elevated in the cerebrospinal fluid (CSF) and brain of AD patients, and $H_2O_2$ is one of the factors that induce VEGF. Therefore, we tested whether $A{\beta}$ might be responsible for the increased VEGF synthesis. We found that $A{\beta}$ induced the production of $H_2O_2$ in vitro. Comparison of the amount of $H_2O_2$ required to induce VEGF synthesis in HN33 cells and the amount of $H_2O_2$ produced by $10{\mu}M\;A{\beta}_{1-42}$ in vitro suggested that a toxic concentration of $A{\beta}$ might induce VEGF synthesis in these cells. However, toxic concentrations of $A{\beta}$ failed to induce VEGF synthesis in several cell systems. They also had no effect on antioxidant enzymes such as glutathione peroxidase, catalase, and peroxiredoxin in HN33 cells. $Cu^{2+}$, $Zn^{2+}$ and $Fe^{3+}$ are known to accumulate in the brains of AD patients and promote aggregation of $A{\beta}$, and $Cu^{2+}$ by itself induces synthesis of VEGF. However, there was no synergistic effect between $Cu^{2+}$ and $A{\beta}_{1-42}$ in the induction of VEGF synthesis and $Zn^{2+}$ and $Fe^{3+}$ also had no effect on the synthesis of VEGF, alone or in combination with $A{\beta}$.

  • 본 연구는 연어 뇌 연접의 단백질구성에 대한 기초연구로서, 기억형성에 중요한 역활을 하는 NMDA 수용체의 분포와 PTK에 의한 인산화에 대하여 조사하였다. 본 실험에 사용한 연어 뇌의 PSD 분획에서는 Coomassie로 염색할 경우 20여개 분명한 단백질띠를 확인할 수 있었으며, 소량으로 존재하여 전체적으로 도말되어 보이는 펩타이드의 수는 알 수 없었다. 이들 중 180kD 크기의 단백질은 phosphotyrosine 특이성 항체에 상대적으로 잘 인식이 되었다. 또한 이 180 kDa의 단백질들은 쥐에 있어서의 NR2A 및 NR2B의 위치인 분자량 약 180kD의 위치와 동일한 것으로 보아 연어에 있어서도 후각기능에 관계하는 NR2A 및 NR2B가 존재함을 추정할 수 있다.

  • 활성있는 새로운 농약을 창출하기 위해, saccharin 모체에 imidazolidine-2,4,5-trione과 2-thio-imidazolidine-4,5-dione 기를 도입시켰다. 각 saccharin 유도체들은 1-치환된 urea (혹은 1-치환된 thiourea)와 oxalyl chloride의 반응을 시작으로 4단계를 거쳐 합성하였다. 1-치환된 urea를 사용해서 1-치환된-3-(1,1,3-trioxo-1,3-dihydro-1$^{6}$-benzo[d]isothiazol-2- ylmethyl)-imidazolidine-2,4,5-trione 5a, 5b, 5c를 합성하였고, 1-치환된 thiourea를 사용하여 1-치환된-2-thioxo-3-(1,1,3-trioxo-1,3-dihydro-1$^{6}$-benzo[d]isothiazol-2-ylmethyl)-imidazolidine-4,5-dione 12a,12b, 12c를 합성하였다.

길항세균 Bacillus amyloliquefaciens A-2를 이용한 토마토 잎곰팡이병 방제용 미생물 제제 (Formulation of Bacillus amyloliquefaciens A-2 and Its Efficacy to Control Tomato Leaf Mold Caused by Fulvia fulva)

  • 토마토 시설재배에서 증가하는 토마토 잎곰팡이병을 방제할 미생물 제제를 병원균 Fulvia fulva에 길항력이 강한 미생물 Bacillus amyloliquefaciens A-2 균주를 이용하여 개발하기 위해 본 연구를 수행하였다. 미생물 A-2 균주를 이용하여 개발하기 위해 본 연구를 수행하였다. 미생물 A-2 균주를 현미유가 첨가된 배지에서 대량 발효배양하고 각종 전달매체와 첨가제를 혼합하고 건조하여 제조흰 미생물 제제들의 효과를 검정하였다. 제조된 제제 A-2H가 방제 효과가 가장 우수하였으나 처리에서 단점으로 인해 이를 보완하며 효과가 통일한 제형인 A-2MP를 선발하였다. 선발된 A-2MP는 100배, 500배 희석처리 후 생육상에서 토마토에 처리한 경우 화학농약과 대등한 방제효과를 나타 내었다. 더욱이 1,000배 희석처리에서도 화학농약보다 효과가 감소하였으나 72% 정도의 방제효과를 보였다. 토마토를 토경재배한 용실에서 A-2MP의 방제효과를 검정한 결과 100배 희석 처리구의 경우 79.4%의 방제효과를 나타내 화학농약 트리후미졸 처리구의 79.6%의 방제효과와 차이가 없었다. A-2MP 제제를 62.2%의 자연발병한 농가의 온실에서 방제효과 검정한 결과 100배 희석액을 일주일 간격으로 3회 처리한 경우 60%의 방제가로 화학농약 방제가 81.6%보다 낮았으나 화학농약 효과의 73% 수준으로 나타났다. 한편, A-2MP제제를 $4^{\circ}C$$25^{\circ}C$ 보관한 경우 모두 10개월동안 생균수 및 방체효과과 안정적으로 유지되었다. 본 연구결과는 저렴한 원료와 길항균 B.amyloliquefaciens A-2 균주를 활용한 A2-MP제제가 토마토 잎공팡이병 방제용으로 쓰일 수 있음을 보여준다.

DISJOINT PAIRS OF ANNULI AND DISKS FOR HEEGAARD SPLITTINGS

  • We consider interesting conditions, one of which will be called the disjoint $(A^2,\;D^2)-pair$ property, on genus $g{\geq}2$ Heegaard splittings of compact orient able 3-manifolds. Here a Heegaard splitting $(C_1,\;C_2;\;F)$ admits the disjoint $(A^2,\;D^2)-pair$ property if there are an essential annulus Ai normally embedded in $C_i$ and an essential disk $D_j\;in\;C_j((i,\;j)=(1,\;2)\;or\;(2,\;1))$ such that ${\partial}A_i$ is disjoint from ${\partial}D_j$. It is proved that all genus $g{\geq}2$ Heegaard splittings of toroidal manifolds and Seifert fibered spaces admit the disjoint $(A^2,\;D^2)-pair$ property.

침전법으로 제조한 $Al_2O_3$-$ZrO_2$계 세라믹스의 미세구조 및 기계적 특성 (Microstructures and Mechanical Properties of $Al_2O_3$-$ZrO_2$ Ceramics Prepared by a Precipitation Method)

  • A precipitation method, one of the most effective liquid phase reaction methods, was adopted in order to prepare high-tech Al2O3/ZrO2 composite ceramics. Al2(SO4)3.18H2O, ZrOCl2.8H2O and YCl3.6H2O were used as starting materials and NH4OH as a precipitation agent, various types of metal hydroxides were obtained by single precipitation(series A) and co-precipitation(series B) method at the pH condition between 7 and 11. Fine Al2O3-ZrO2 powders were prepared at optimum calcination condition and the effects of ZrO2 on microstructures and mechanical properties of Al2O3 were investigated. The composition of Al2O3/ZrO2 composites wax fixed as Al2O3-15 v/o ZrO2(+3m/o Y2O3). ZrO2 limited the grain growth of Al2O3 and increased grain size homogeneity of Al2O3 more effectively than MgO.Flexural strength values in Al2O3 and Al2O3/ZrO2 composites were 340-430 MPa and 540-820 MPa, respectively, and the effect of strength improvement showed 20-50% by adding ZrO2 to Al2O3. Fracture toughness of Al2O3/ZrO2 composites was improved by stress-induced phase transformation of tetragonal ZrO2 and toughening effect by microcrack was not observed. Also, ZrO2 particles located at Al2O3 grain junction contributed to toughening, while spherical ZrO2 particles located within Al2O3 grain did not contribute to toughening. Weibull moduli of Al2O3 ceramics and Al2O3/ZrO2 composites of series A and series B were 4.34, 5.17 and 9.06, respectively. Above 0.5 of failure probability, strength values in Al2O3 ceramics and Al2O3/ZrO3 composites of series A and series B were above 400 MPa, 700 MPa and 650 MPa, respectively.

Highly Selective Transport of Ag+Ion through a Liquid Membrane Containing 2-Mercaptobenzothiazole as a Carrier

  • 2-Mercaptobenzothiazole was used as a highly selective and efficient carrier for the uphill transport of silver ion through a chloroform bulk liquid membrane. In the presence of thiosulfate ion as a suitable metal ion acceptor in the receiving phase, the amount of silver transported across the liquid membrane after 180 min was 90 ± 3.0%. The selectivity and efficiency of silver ion transported from aqueous solutions containing equimolar mixtures of $Zn^{2+}, Cu^{2+}, Co^{2+}, Ni^{2+}, Cd^{2+}, Pb^{2+}, Bi^{3+}, Fe^{2+}, Fe^{3+}, Pd^{2+}, Mn^{2+}, Hg^{2+}, Sn^{2+}, Ca^{2+}, Mg^{2+}, K^+, Na^+ and Li^+$ were investigated.

  • In this article, we show a generalization of Clement's theorem on the pair of primes. For any integers n and k, integers n and n + 2k are a pair of primes if and only if 2k(2k)![(n - 1)! + 1] + ((2k)! - 1)n ${\equiv}$ 0 (mod n(n + 2k)) whenever (n, (2k)!) = (n + 2k, (2k)!) = 1. Especially, n or n + 2k is a composite number, a pair (n, n + 2k), for which 2k(2k)![(n - 1)! + 1] + ((2k)! - 1)n ${\equiv}$ 0 (mod n(n + 2k)) is called a pair of pseudoprimes for any positive integer k. We have pairs of pseudorimes (n, n + 2k) with $n{\leq}5{\times}10^4$ for each positive integer $k(4{\leq}k{\leq}10)$.

pT7MT, a Metallothionein 2A-Tagged Novel Prokaryotic Fusion Expression Vector

  • In the present article, a novel fusion expression vector for Escherichia coli was developed based on the pTORG plasmid, a derivative of pET32a. This vector, named pT7MT(GenBank Accession No DQ504436), carries a T7 promoter and it drives the downstream gene encoding Metallothionein 2A(MT2A). There are in-framed multiple cloning sites(MCS) downstream of the MT2A gene. A target gene can be cloned into the MCS and fused to the C-terminal of the MT2A gene in a compatible open reading frame(ORF) to achieve fusion expression. The metal-binding capability of MT2A allows the purification of fusion proteins by metal chelating affinity chromatography, known as $Ni^{2+}$-affinity chromatography. Using this expression vector, we successfully got the stable and high-yield expression of MT2A-GST and MT2A-Troponin I fusion proteins. These two proteins were easily purified from the supernatant of cell lysates by one-step $Ni^{2+}$-affinity chromatography. The final yields of MT2A-GST and MT2A-Troponin I were 30mg/l and 28mg/l in LB culture, respectively. Taken together, our data suggest that pT7MT can be applied as a useful expression vector for stable and high-yield production of fusion proteins.

Nonstoichimetry (x) of Nuclear Materials UO2+x and $(Ce_yU_{1-y})O_{2+x}$ at High Temperatures as a Function of Oxygen Activity$(a_o_2)$

  • The oxygen nonstoichiometry (x) of UO$_{2+x}$ and $(Ce_yU_{1-y})O_{2+x}$ (y=0.05, 0.25) has been measured as a function of oxygen activity (a02) at 100$0^{\circ}C$ by a solid state coulometri titration technique. The results for UO$_{2+x}$ are in a good agreement with literature data and the ao2-dependence of the nonstoichimetry has been well explained with (2:2:2) cluster model. The equilibrium oxygen activity of $(Ce_yU_{1-y})O_{2+x}$ increases with Ce-content (y) for given nonstoichiometry (x), which is ascribed to the fact that Ce is present as Ce$^{3+}\; and\; Ce^{4+}$, and, therefore, the addition of Ce reduces the oxidation capacity of UO$_{2+x}$. From the oxygen activity dependence of x in $(Ce_yU_{1-y})O_{2+x}$ the defect structure of $(Ce_yU_{1-y})O_{2+x}$ is discussed.
