Anti-Cancer Activity of the Leave Extracts of Rodgersia podophylla through β-catenin Proteasomal Degradation in Human Cancer Cells (도깨비부채(Rodgersia podophylla) 잎 추출물의 인간 암세포의 β-catenin 분해 유도 활성)

  • In this study, we evaluated the effect of Rodgersia podophylla leave extracts (RPL) on ${\beta}-catenin$ level in human cancer cells. RPL dose-dependently inhibited cell proliferation in SW480, A549, MDA-MB-231, PC-3 and AsPC-1 cells. RPL dramatically decreased ${\beta}-catenin$ protein level in all cancer cells. However, decreased level of ${\beta}-catenin$ mRNA expression was observed in A549 and AsPC-1 cells. In addition, RPL dramatically attenuated cyclin D1 mRNA expression in all cancer cells. MG132 decreased the downregulation of ${\beta}-catenin$ protein level induced by RPL in all cancer cells, while RPL-induced downregulation of ${\beta}-catenin$ was inhibited by the inhibition of $GSK-3{\beta}$ by LiCl in MDA-MB-231 cells. RPL phosphorylated ${\beta}-catenin$ and $GSK-3{\beta}$. In addition, the inhibition of $GSK-3{\beta}$ by LiCl attenuated RPL-induced ${\beta}-catenin$ phosphorylation. Based on these findings, RPL may be a potential candidate for the development of chemopreventive or therapeutic agents for human cancer.

Effect of the Extracts from the Leaves and Branches of Sageretia thea on β-catenin Proteasomal Degradation in Human Colorectal and Lung Cancer Cells (상동나무(Sageretia thea) 잎과 가지추출물의 대장암과 폐암세포의 β-catenin 분해 유도 활성)

  • In this study, we evaluated the effect of branch (STB) and leave (STL) extracts from Sageretia thea on ${\beta}-catenin$ level in human colorecal cancer cells, SW480 and lung cancer cells, A549. STB and STL dose-dependently suppressed the growth of SW480 and A549 cells. STB and STL decreased ${\beta}-catenin$ level in both protein and mRNA level. MG132 decreased the downregulation of ${\beta}-catenin$ protein level induced by STB and STL. However, the inhibition of $GSK3{\beta}$ by LiCl or ROS scavenging by NAC did not block the reduction of ${\beta}-catenin$ protein by STB and STL. Our results suggested that STB and STL may downregulate ${\beta}-catenin$ protein level independent on $GSK3{\beta}$ and ROS. Based on these findings, STB and STL may be a potential candidate for the development of chemopreventive or therapeutic agents for human colorectal cancer and lung cancer.

  • This study analyzes emerging earthquake and space use technologies from the latest Korean and Japanese scientific and technological foresights in 2022 and 2019, respectively. Unlike the earthquake prediction and early warning technologies presented in the 2017 study, the emerging earthquake technologies in 2022 in Korea was described as an earthquake/complex disaster information technology and public data platform. Many detailed future technologies were presented in Japan's 2019 survey, which includes largescale earthquake prediction, induced earthquake, national liquefaction risk, wide-scale stress measurement; and monitoring by Internet of Things (IoT) or artificial intelligence (AI) observation & analysis. The latest emerging space use technology in Korea and Japan were presented in more detail as robotic mining technology for water/ice, Helium-3, and rare earth metals, and manned station technology that utilizes local resources on the moon and Mars. The technological realization year forecasting in 2019 was delayed by 4-10 years from the prediction in 2015, which could be greater due to the Corona 19 epidemic, the declaration of carbon neutrality in Korea and Japan in 2020 and the Russo-Ukrainian War in 2022. However, it is required to more active research on earthquake and space technologies linked to information technology.

  • This research aims to discuss the impact of on regional development as a place brand and glocal heritage. Based on understanding of the changes and influences of local heritages in the globalization era as a key component of place branding, this study explores how is used to develop a place branding strategy for Lijiang. The research methods are both a literature review and a field research related to Lijiang and its culture. Also, the resources from news, internet, and YouTube are used to analyze the impact of . The performance has been attracting tourists from both Chineses and foreigners and contributed to increase the economic profits of local tourism industry as one of the representative identities of Lijiang. Also, in the process of preservation and recreation of cultural heritages of Lijiang such as , the participation of local residents and on-going interactions between the residents and global tourists highly influence on a transition from place marketing to place branding. By applying local cultural heritages to place branding strategies, the regional values of Lijiang strengthen its place identity from a place of preserving a minority's heritage to that of flourishing cultural exchanges and hybridization from the world.

Empirical Assessment of International Entry Strategy for Large Construction Companies (주요 전략지수별로 살펴 본 국내 대형건설업체의 해외건설 진출전략 효과에 대한 실증적 연구)

  • Although international construction market gradually takes a important position among the export industries, the previous studies about international construction entry strategy have just focussed on trend investigation or suggestion for revitalization. Moreover, in order to prepare the market uncertainty such as world financial crisis and to plan the long term strategy, specific strategy studies based on corporate level are required. Therefore, this study estimates the nine strategic index and four financial index of 31 companies that performed 1920 international projects from 1993 to 2007 and evaluate the performance as three periods by multi-regression analysis. Also, this study analyze dynamic correlation between these index and the performance considering times. this study verifies that market diversification, product diversification, localization and decrease of debt to asset ratio make a good effect on the international order as long term strategy and shows that collaborated entry with domestic corporations, alliance entry with host country's company, alliance entry with third country's company, portion of labor cost and portion of management expense differently make a influence on the performance as times. these results will be helpful for the construction companies to plan the international entry strategy reasonably and specifically.

Anti-inflammatory Effect of Berchemia berchemiaefolia Leaves through Inhibition of NF-κB and MAPK Signaling Activation in LPS-stimulated RAW264.7 Cells (망개나무 추출물의 NF-κB 및 MAPK 신호전달 억제를 통한 항염증 효과)

  • Berchemia berchemiaefolia (Makino) Koidz. is found not only in korea, but also in japan and is known as a rare plant all over the world. In this study, we evaluated anti-inflammatory effect of B. berchemiaefolia (Makino) Koidz. leaves (BBK-L) in LPS-stimulated RAW264.7 cells. BBK-L inhibited the generation of NO through the suppression of iNOS experession. BBK-L attenuated the expression of iNOS, COX-2, TNF-α, IL-1β, IL-6 induced by LPS. BBK-L decreased LPS-mediated IκB-α degradation inhibite which resulted in the inhibition of NF-κB activation in RAW264.7 cells. In addition, BBK-L suppressed ERK1/2, JNK phosphorylation induced by LPS. BBK-L showed anti-inflammatory effect through blocking the generation of the inflammatory mediators such as NO, iNOS, COX-2, TNF-α, IL-1β, IL-6 via the inhibiting of NF-κB and MAPK signaling activation. These results suggests that BBK-L may have great potential for the development of anti-inflammatory drug to treat acute and chronic inflammatory disorders.

Inhibitory Activity of Lonicera caerulea Against Cell Proliferation in Human Colorectal Cancer Cells (댕댕이나무(Lonicera caerulea)의 대장암세포 생육억제 활성)

  • In this study, we evaluated the effect of the extracts from Lonicera caerulea leaves (LCLE), branches (LCBE) and fruits (LCFE) on the cell growth and migration in human colorectal cancer cells, HCT116 and SW480 cells. LCLE and LCBE dose- and time-dependently inhibited the proliferation of HCT116 and SW480 cells. However, LCFE did not affect the proliferation of HCT116 and SW480 cells. In addition, LCLE and LCBE dramatically cell migration and wound healing in HCT116 cells. LCLE and LCBE decreased β-catenin protein level but not mRNA level in HCT116 and SW480 cells. Furthermore, LCLE decreased TCF4 level in both protein and mRNA level in HCT116 and SW480 cells. However, LCBE decreased TCF4 protein level but not mRNA level in HCT116 and SW480 cells. Based on these findings, LCLE and LCBE may inhibit the cell proliferation and migration through blocking Wnt signaling activation in human colorectal cancer cells. Therefore, LCLE and LCBE may be a potential candidate for the development of chemopreventive or therapeutic agents for human colorectal cancer.

Land Use Change Detection at Kyeryongsan National Park by Using Remote Sensing and Geographical Information Systems (원격탐사와 GIS를 이용한 계룡산국립공원의 토지이용변화)

  • 국립공원의 뛰어난 경승지와 생태적으로 보존가치가 높은 지역은 다음 세대에게 물려줄 국가의 귀중한 자원으로서 잘 보존되고 관리되어야 하나 한국의 국립공원에서는 공원의 자연환경을 해치는 개발행위가 이뤄졌거나 제안되고 있다. 이러한 개발행위를 효과적으로 평가하기 위해서는 관련 환경자원의 활용이 필요하고 특히 토지이용변화의 파악이 요구되나 한국의 국립공원은 적절한 토지이용도가 결여되어 있다. 그러므로, 본 연구의 목표는 토지이용변화도를 제작하여 향후 국립공원에서 제안되는 개발행위가 해당국립공원의 토지이용변화에 미친 영향을 파악함으로써 국립공원의 환경 및 경관 관리에 기여하는 데에 있다. 본 연구는 계룡산국립공원을 대상으로 1988년도부터 1998년까지의 토지이용변화를 파악하기 위해 원격탐사 자료를 이용하여 2개년도의 Landsat TM 영상을 기하보정하여 토지피복분류를 추출하였다. 이를 바탕으로 항공사진과 현지조사를 통해 확인하여 작성한 토지이용현황도와 토지이용변화도를 GIS에 입력하였으며 GIS의 분석기능을 이용하여 10년간의 토지이용변화에 대해 파악하였다. 토지이용변화 파악 결과, 농지와 나대지가 계룡산국림공원 전체에서 43.7ha, 102.2ha 각각 감소하였고 산림과 개발지는 121.Oha, 24.8ha. 각각 증가하였다. 산림의 증가는 1988년 영상분류당시 산림의 가장자리 유령림이 농경지로 분류되었던 것이 10년 뒤 영상에서는 산림으로 분류된 결과로 파악되며 개발지의 증가는 계룡산국립공원 동학사 제2집단시설지구의 개발로 기존 농경지가 감소, 전용되었고 나대지는 개발로 인해 감소되었다. 개발지의 증가는 취락지구와 집단시설지구에서 두드러졌으며 이들 두 지구에서 산림, 농경지, 나대지 모두 감소하고 대신 개발지가 증가하였다. 본 연구를 수행한 결과 계룡산 국립공원의 가장 큰 토지이용변화는 건물 신축이었으며 이는 집단시설지구의 신설에 기인하고 있다. 취락지구도 개발 행위가 증가하고 있어 집단시설지구와 취락지구의 이용후 평가가 이뤄져야 한다고 생각한다. 본 연구에서 작성한 토지이용변화도는 향후 국립공원의 경관 변화를 파악하는 데에 효과적으로 이용될 수 있다고 판단된다.

A Study on the Value of Island Landscape as Scenic Site Resource - Focus on the Raising Fine Village(Gwanmaedo, Youngsando) - (도서 경관의 명승자원으로서의 가치연구 - 명품마을(관매도, 영산도)을 중심으로 -)

  • Despite the fact that the importance of islands and oceans is increasingly being emphasized as they are recognized as alternative spaces for the future, some islands that have not been designated as cultural assets have lost their natural and cultural landscapes to development projects and other plans for turning islands into tourist resources, and are still in the process of being destroyed. Unlike old perceptions of islands, islands, in the minds of people living in the modern age, have become places for taking a quiet rest or enjoying and appreciating the undamaged beauty of nature itself. Keeping up with the trend of people increasingly visiting the islands these days, it is high time to prepare plans for the promotion, preservation management, and usages of islands based on researches of islands with excellent natural landscapes and through the designation of these islands as cultural properties. As the first step of studying island landscape resources as resources of cultural assets, the current study includes literature reviews and field investigations of Gwanmaedo and Yeongsando, two islands that have been selected as prestigious villages and are part of the Dadohaehaesang National Park. Based on these preliminary investigations, landscape resources showing distinctive natural landscapes and cultural sceneries were selected and analyzed in detail, thus presented for their value as resources of Scenic site and future research.

Extract from the branches of Rhamnus yoshinoi exerts anti-cancer effects on human prostate cancer cells through Wnt/β-catenin proteasomal degradation and identification of compounds by GC/MS (짝자래나무[Rhamnus yoshinoi] 가지 추출물에 의한 전립선암세포의 Wnt/β-catenin 분해 유도 활성 및 GC/MS 분석)

  • We evaluated the anti-cancer activity against human prostate cancer cells and the associated molecular mechanism of extracts from the branches of Rhamnus yoshinoi (RYB). Treatment with RYB suppressed viability of human prostate cancer cells (PC-3) and decreased protein levels of both β-catenin and T-cell factor 4 (TCF4). This was reflected in reduced TCF4 mRNA, but not decreased β-catenin mRNA. PC-3 cells were pretreated with the proteosome inhibitor MG132 before treatment with RYB, which blocked RYB-mediated down regulation of β-catenin in PC-3 cells, thus confirming that RYB promotes the proteasomal degradation of β-catenin. RYB induced β-catenin phosphorylation, and GSK-3β inhibition by LiCl blocked the phosphorylation and proteasomal degradation of β-catenin by RYB. These results suggest that GSK-3β may be an important upstream kinase for RYB-mediated regulation of β-catenin. Finally, GC/MS analysis of RYB identified 18 compounds. Based on these findings, RYB shows potential for development as a therapeutic agent for prostate cancer.
