Antioxidant and Antiinflammation Activities of Prunus persica Tree Extracts (야생 복숭아나무 추출물의 항산화 및 항염증 효과)

  • Reactive oxygen species (ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Several lines of evidence provided that ROS appears to cause to develop aging and various diseases. High level of hyaluronic acid with decreased molecular weight has been detected in patients with inflammatory diseases including rheumatoid arthritis. In this study, we have conducted to investigate the antioxidant and hyaluronidase inhibitory activities of Prunus persica Batsch var. davidiana Maximowicz in order to screen the bioactive substances which can be developed as possible anti-inflammatory agents. Among the extracts of Prunus persica Batsch var. davidiana Maximowicz, EtOAc extract exhibited the strongest effect on antioxidant experiment such as DPPH, Ferric- Thiocyanate and Rancimat. Also, EtOAc extract showed a potent hyaluronidase inhibitory activity.

Effect of Soluble EPCR on the Anti-Inflammatory Effects by Activated Protein C (수용성 EPCR에 의한 활성화된 단백질 C의 항염증 작용에 관한 연구)

  • In this study, we evaluated the effect of soluble EPCR(Soluble Endothelial Protein C Receptor, sEPCR) on the anti-inflammatory activities by activated protein C(APC) in endothelium. We demonstrated that sEPCR inhibited the barrier protective activity, the inhibition of neutrophils adhesion toward endothelial cells and the inhibition of transendothelial migration by APC in endothelial cells. Interestingly, sEPCR also blocked the mechanism by which APC inhibited the expression of cell adhesion molecules(CAM) by TNF-alpha in endothelial cells. These results suggested that the anti-inflammatory activities of APC was inhibited by sEPCR which blocked the binding motifs of Gla domain of APC to membrane bound EPCR. This finding will provide the important evidence in the development of new medicine for the treatment of severe sepsis and inflammatory diseases and good clue for understanding unknown mechanisms by which APC showed the anti-inflammatory activities in endothelium.

Anti-inflammatory effect of beluga lentil extract in RAW 264.7 macrophages (RAW 264.7 대식세포에서 벨루가 렌틸 추출물의 항염증 효과)

  • The anti-inflammatory effect of beluga lentil extract (BLE) and its underlying mechanisms were investigated in lipopolysaccharide (LPS)-treated RAW 264.7 cells. Treatment with BLE significantly decreased nitric oxide (NO) production and protein and mRNA expressions of inducible NO synthase (iNOS) in LPS-treated RAW 264.7 cells. Down-regulation of this inflammatory gene expression was not associated with NF-κB/MAPK signaling pathways, and further mechanistic studies demonstrated that BLE decreased LPS-induced iNOS expression through upregulation of the nuclear factor erythroid 2-related factor 2 (Nrf2)-mediated heme oxygenase-1 (HO-1) expression. These results suggest that beluga lentil represent a potential source of natural anti-inflammatory agents, and further studies will be necessary to determine its anti-inflammatory effects in vivo.

  • Antiinflammatory and antioxidative effects of mulberry fruit were investigated by using bioassay screening system. The extract of muberry fruits showed inhibitory effect in phospholipase $A_2$ activity and cyclooxygenase II activity. Among nine varieties of mulberry tree, the antiinflammatory activities of the Shingwangppong, Ficus, Cheongilppong and Keomseolppong were higher than that of the other varieties. Also, antioxidative activity of mulberry fruit was examined by DPPH free radical scavenging method. The radical scavenging activity of the mulberry fruit decreased as following order : Shingwangppong > Keomseolppong > tetraploid Ficus> diploid Ficus.

Effect of sword bean (Canavalia ensiformis) fermentation filtrate on the antioxidant, anti-inflammatory, and antimicrobial activities (작두콩(Canavalia ensiformis) 발효액이 항산화, 항염증 및 항균 활성에 미치는 영향)

  • In the present study, various experiments were performed to evaluate the biological activities, such as the antioxidant, anti-inflammation, and antimicrobial activities of sword bean (Canavalia ensiformis) fermentation filtrate by Lactobacillus plantarum (L. plantarum) and Lactobacillus brevis (L. brevis). Total polyphenol (TPC) and flavonoid contents (TFC) of sword bean were significantly decreased after fermentation regardless of Lactobacillus sp. (p<0.05). The DPPH radical scavenging activity of sword beans also decreased after fermentation. However, nitric oxide (NO) radical scavenging activity conspicuous increased after fermentation (p<0.001) in a treated concentration-dependent manner, and the effect for L. brevis was higher than for L. plantarum. In addition, the sword bean fermentation filtrate showed a strong inhibitory effect against Pseudomonas aeruginosa, Staphylococcus sp., and Escherichia coli. Cell cytotoxicity was not exhibited in all experimental groups (data not shown). These findings suggest that the sword bean fermentation filtrate may be used effectively in various industries due to its high anti-inflammatory and antimicrobial activities.

Anti-inflammatory Effect of Ishige foliacea in RAW 264.7 Cells (넓패추출물에 의한 RAW 264.7 세포에서의 항염효과)

  • This study was carried out to identify the anti-inflammatory effects of Ishige foliacea (I. foliacea) extract on skin using RAW 264.7 cells. The anti-inflammatory effects of I. foliacea extract on RAW 264.7 cells were assessed by cell viability assay, mRNA expressions, and nitric oxide (NO)/prostaglandin E2 (PGE2) productions. The anti-inflammatory effects of I. foliacea extract were elucidated by analysis of IL-1α/IL-1β/IL-6/TNFα gene expressions and PGE2/NO production. Quantitative real-time polymerase chain reaction showed that I. foliacea extract decreased the gene expression levels of iNOS/COX2/IL-1α/IL-1β and IL-6. Furthermore, PGE2/NO production also revealed that I. foliacea extract exhibited anti-inflammatory properties. These results suggest that I. foliacea extract is an anti-inflammatory compound. It could be a potent cosmeceutical material for anti-inflammatory effects. Further studies on the anti-inflammatory mechanisms of broadleaf extracts are expected to help identify pharmacological mechanisms related to inflammation in addition to cosmeceuticals.

Anti-inflammatory effect of CGT in atopic dermatitis model mice (아토피피부염을 유발한 마우스에서 청기해독탕의 항염증 효과)

  • In order to investigate the effect of CGT on atopic dermatitis, various anti-inflammatory factors were studied. In-vitro, inflammatory mediators, such as MTT and nitric oxide and ROS were detected after the addition of LPS with or without CGT in RAW 264.7 cells. In-vivo, in order to verify the effectiveness of CGT in atopic dermatitis animal model, its role in inflammation factors and histological changes were observed in NC/Nga mice. CGT showed cell viability of 100% or higher in all concentration in RAW 264.7 cells. CGT inhibited LPS-induced productions of inflammatory mediators nitric oxide and antioxidant activity reactive oxygen species production in RAW 264.7 cells. CGT treated group showed significant decrease in serum of the expression of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ by 53%, 43% and 57% respectively. And CGT treated group showed decrease in serum of the expression of IgE by 56% respectively. Also, infiltration of adipocytes into skin was suppressed and the thickness of epidermis and dermis were relatively decreased in the CGT treated group. As a result, CGT has an anti-inflammatory effects in NC/Nga mouse. Thus, these results suggested a beneficial effect of CGT in treatment with Atopic dermatitis and inflammatory.

The chemical constituents, antioxidant activity and anti-inflammatory activities of Solanum nigrum Linne by different harvest time (수확시기별 까마중 전초의 성분, 항산화활성 및 항염증 활성)

  • In this study, we investigated the variation in free sugars, amino acids, antioxidant activity, and anti-inflammatory activity of Solanum nigrum Linne based on harvest time. Major amino acids identified by HPLC analysis were proline, histidine, and serine. The highest content of total amino acids were found in S. nigrum aerial parts and roots harvested on July 10th and August 10th. Four kinds of free sugars (fructose, glucose, sucrose, maltose) were detected in S. nigrum, and the free sugar content varied significantly with harvest time. The fructose content of S. nigrum decreased with as harvest time increased. The total polyphenol content of S. nigrum was highest in those harvested on August 30th. The antioxidant activity of ethanol extract from S. nigrum collected at different harvest times were measured by DPPH and ABTS radical scavenging assays. The anti-inflammatory activity of these extracts were assayed via nitric oxide suppression in C6 glioma cells with a lipopolysaccharide (LPS)-induced inflammatory response. The anti-inflammatory activity and antioxidant effects were the highest in the extract from S. nigrum collected on August 30th. Good correlations were observed between antioxidant and anti-inflammatory activities in ethanol extract of S. nigrum roots harvested on August 30th.

A Study on Anti-oxidative, Anti-inflammatory, and Melanin Inhibitory Effects of Chrysanthemum Sibiricum Extract (구절초 꽃 추출물의 항산화, 항염증 및 멜라닌 생성 억제 효과에 관한 연구)

  • The purpose of this study is to observe how Chrysanthemum Sibiricum Extract has anti-oxidant activity, cytotoxicity for skin cells, and anti-inflammatory and melanin inhibitory effects in order to find the development possibility of Chrysanthemum Sibiricum Extract as the ingredient of a functional cosmetic product. According to the analysis, Chrysanthemum Sibiricum Extract was found to have high contents of polyphenols and flavonoids and excellent DPPH radical scavenging activity. The extract had no significant cytotoxicity for Raw 264.7 cell and B16F10 cell and significantly inhibited the creation of NO induced LPS in Raw 264.7 cell so as to present anti-inflammatory effect. In B16F10 cell, melanin creation was induced with ${\alpha}$-MSH, and then melanin biosynthesis inhibition was measured. As a result, melanin creation was inhibited in the concentration dependence way. Given the results, it is considered that Chrysanthemum Sibiricum Extract is applicable as the ingredient of a functional cosmetic product that has low cytotoxicity for skin cells, high anti-oxidant activity, anti-inflammatory effect, and whitening effect.

Effect of Sorghum nervosum extract on an anti-inflammatory and cytoprotective (고량(Sorghum nervosum)추출물의 항염증 및 세포 보호 효과)

  • This study was intended to test anti-inflammation and cytoprotective effect against UVB after Sorghum nervosum was extracted with 70% ethanol. The efficacy of Sorghum nervosum was assessed regarding cell viability analysis, reactive oxygen species measurement, anti-inflammation, a change in COX-2 protein, and cytoprotective effect against UVB. According to the results of experiment, the cell viability of 97% or higher was shown at all concentrations of Sorghum nervosum in RAW264.7 macrophage, HaCaT cell. And in anti-inflammatory NO inhibitory activity, a concentration-dependent inhibitory effect was shown. And COX-2 protein expression was also significantly (p<.001) inhibited at 25, $50{\mu}g/mL$. With regard to cytoprotective effect against UVB, in the quantitative analysis results of reactive oxygen species within the cell, it was verified that Sorghum nervosum extract had an effect on an decrease in the total amount of ROS. When the results of study are considered comprehensively, it is thought that there is possibility of Sorghum nervosum development as raw materials for cosmetics showing an anti-inflammation and cytoprotective function against UVB.
