Antioxidant Capacity and Protective Effects on Neuronal PC-12 Cells of Domestic Bred Kiwifruit (국내 육성 참다래의 항산화능 및 PC-12 신경세포 보호 효과)

  • This study was conducted to comparatively evaluate antioxidant capacity (AC) of seven cultivars of kiwifruit (Actinidia spp.) and their protective effects on neuronal PC-12 cells. The contents of total phenolics (TP) and total flavonoids (TF) of kiwifruits were also examined. Five cultivars of kiwifruit, Actinidia chinensis (cv. Haehyang and cv. Haegeum), A. eriantha (cv. Bidan), A. arguta ${\times}$ A. deliciosa (cv. Mansoo), and A. arguta (cv. Chiak), were bred in Korea, while two cultivars, A. deliciosa (cv. Hayward) and A. linguiensis (accession number 041AE), originated from New Zealand and China, respectively. Skin extracts of kiwifruit showed higher TP, TF, and AC than flesh extracts. The highest levels of TP and AC were found in cv. Bidan flesh extract among cultivars studied, but the TF content of cv. Bidan flesh extract was the lowest. The kiwifruit bred in Korea had higher AC than their counterparts. AC of kiwifruit had a highly positive linear correlation with TP and TF. The flesh extracts from cv. Hayward, cv. Haehyang, and cv. Haegeum significantly (p < 0.05) prevented PC-12 cells from oxidative stress induced using $H_2O_2$ compared to a control with $H_2O_2$ only. Overall, our results suggest that kiwifruit bred in Korea may offer a good source of antioxidants and serve as functional materials.

Antioxidaive Activity of Lotus Root(Nelumbo nucifera G.) Extracts. (연근 추출물의 항산화 효과)

  • This study was investigated on the antioxidant capacity of 80% ethanol extract of lotus root in vitro. The ex-traction yields of 80 % ethanol extract was 9.14%. Lotus root ethanol extract was fractionated by the following: n-hexane, chloroform, ethylacetate and n-butanol. n-Butanol fraction showed the highest ex-traction yield of all fractions. Antioxidative activities of different fractions were examined by l.l-diphenyl-2-picrylhydrazyl(DPPH) radical generation, the Rancimat test, the nitrite scavenging ac-tivity and the thiobarbituric acid(TBA) method, and compared with the properties of the commercial antioxidant butylated hydroxytoluene(BHT). The antioxidative capacity of the ethylacetate fraction was the highest among fractions and its fraction showed higher contents of total polyphenol. Furthermore, the antioxidative capacity of the ethylacetate fraction was similar to that of BHT. In conclusion, these results suggest that lotus root may be a good candidate as a natural antioxidant source.

Comparison of the Total Phenolic and Flavonoid Contents and Antioxidant Activities of Four Kinds of Sand Dune Plants Living in Taean, Korea (태안에서 서식하는 사구식물 4종의 폴리페놀, 플라보노이드 함량과 항산화 활성 비교)

  • This study attempted to investigate the antioxidant properties of four different species of sand dune plants (Calystegia soldanella, Messerschmidia sibirica, Vitex rotundifolia and Rosa rugosa). In order to validate the antioxidant activity of these plants, we first determined the total amount of flavonoid versus phenolic contents (TFC/TPC) and extracted crude flavonoids for measuring antioxidant activities that were determined by DPPH, ABTS and FRAP assays with radical scavenging effects. We found that highest amounts of TPC were detected in R. rugose with values of 110.20 mg/g (leaves) and 65.71 mg/g (stems), while the highest amounts of TFC in V. rotundifolia with values of 38.07 mg/g (leaves) and 6.55 mg/g (stems). We further examined how closely related the amounts of TFC/TPC in antioxidant activities and found that R. rugose has the highest activities of radical scavenging with values of $63.4{\mu}g/ml$ and $51.2{\mu}g/ml$ determined by DPPH and ABTS assays compared with the value of $21.2{\mu}g/ml$ by FRAP assay. It is of note that there is a statistically significant correlation between the resulting antioxidant activities and the total ratio of TFC and TPC, suggesting that the different amounts of TFC/TPC may directly contribute to the various antioxidant activities.

Total Phenolics, Total Flavonoids, and Antioxidant Capacity in the Leaves, Bulbs, and Roots of Allium hookeri (삼채의 잎, 인경, 뿌리의 총페놀 함량, 총플라보노이드 함량 및 항산화능)

  • To quantitatively evaluate the total phenolics, total flavonoids, and antioxidant capacity in the leaves, bulbs, and roots of fresh Allium hookeri, they were extracted using various solvents including water, aqueous methanol (20, 40, 60, and 80%; v/v), and absolute methanol. The leaves had the highest levels of total phenolics (240.4-276.6 mg gallic acid equivalents/100 g) and total flavonoids (9.7-34.1 mg catechin equivalents/100 g). The highest antioxidant capacities of 78.7- 103.4 mg vitamin C equivalents (VCE)/100 g, 24.4-59.0 mg VCE/100 g, and 1,798.8-2,169.7 mg VCE/100 g in the leaves were also observed using 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), and oxygen radical absorbance capacity (ORAC) assays, respectively. The total phenolics had a higher linear correlation with antioxidant capacity than the total flavonoids. In general, 60% (v/v) aqueous methanol extract had higher levels of total phenolics and flavonoids, and higher antioxidant capacity than any other solvents used. This study suggested that A. hookeri might be a good source of phenolics and antioxidants.

Effects of Fermentation on the Metabolic Activities of Pine Needle Juice (발효과정이 솔잎 착즙액의 항산화, alpha-Glucosidase 및 Angiotensin Converting Enzyme 저해 활성에 미치는 영향)

  • The objective of this study was to compare the content and metabolic activities between fresh pine needle juice (PNJ) and fermented pine needle juice (FPNJ). A variety of factors were measured, including total phenolic content (TPC), antioxidant activity [DPPH radical scavenging activity (RSA), total radical-trapping antioxidant potential (TRAP), oxygen radical absorbance capacity (ORAC), cellular antioxidant capacity (CAC)], anti-genotoxic activity, ${\alpha}$-glucosidase inhibitory activity, and angiotensin converting enzyme (ACE) inhibitory activity. The TPC was $17.3{\pm}0.2$ and $4.6{\pm}0.0$ mg GAE/g in PNJ and FPNJ, respectively. The DPPH RSA, TRAP, and ORAC values increased in a dose-dependent manner for both PNJ and FPNJ, with significantly higher activities in PNJ than FPNJ. The CAC against AAPH-induced oxidative stress in HepG2 cells was protected by both PNJ and FPNJ. Pretreatment with PNJ and FPNJ in human leukocytes produced significant reductions in $H_2O_2$-induced DNA damage at a concentration of $50{\mu}g/mL$. ${\alpha}$-Glucosidase inhibitory activity was significantly higher in FPNJ than PNJ. The ACE inhibitory activity was about 87.1% and 60.0% in 1:1 diluted PNJ and FPNJ, respectively. This study suggests that the fermentation of PNJ could enhance the regulation of blood glucose metabolism and both PNJ and FPNJ might be a new potential source of natural antioxidant, anti-diabetic, and anti-hypertensive agents applicable to food.

Studies on the Antioxidative and Antimicrobial Effects of Chondria crassicaulis (서실의 항산화, 항균효과 연구)

  • In this study, we investigated the biological activity of antioxidant and antimicrobiological effect of Chondria crassicaulis (CC), which, using methanol, dichlolometane and ethanol, were extracted and fractionated into four different types: hexane(CCMH), methanol (CCMM), butanol (CCMB), and aqueous (CCMA) partition layers. The reducing activity on the 1,I-diphenyl-2-picrylhydrazyl (DPPH) radical and $O^{2-}$ and $O_{2}$$O_{2}$radical scavenging potential, in search for antioxidation effects of CC partition layer, were sequentially screened. Among the four fractions, CCMM had the highest antioxidative activity. The antimicrobial activity was increased in proportion to its concentration by the paper disc method. Among the various solvent layers, the CCMB, CCMH and CCMM showed relatively strong antimicrobial activities in the order.

Antioxidant Activity of Korea Traditional Fermented Sauces Made with Asparagus (아스파라거스를 이용한 전통장류의 항산화 효과)

  • Asparagus have anti-cancer, antioxidant, and anti-inflammatory properties. Since the high functionality of Korean traditional sauces was discovered, various studies have been conducted using asparagus. In this study, the antioxidant activity of traditional sauces containing asparagus was confirmed. The antioxidant capacity of asparagus was independent of the concentration and showed the highest activity at 70 mg/ml. For instance, concerning Meju containing asparagus, high antioxidant activity was observed in water and ethyl acetate extracts; the total polyphenol content was equivalent to 1.4 mg/g and 0.7 mg/g of tannic acid in the water and ethyl acetate fractions respectively. The DPPH and ABTS radical scavenging activities were 27.6% and 9.04%, respectively, in the water layer, and 10.7% and 52.4%, respectively, in the ethyl acetate layer. Additionally, the antioxidant activity of asparagus-containing traditional sauces was measured using 95% ethanol extracts; soy sauce showed the highest activity, retained in a wide range of concentrations.

Effects of Heat and pH Treatments on Antioxidant Properties of Ishige okamurai Extract (패 추출물의 항산화능에 미치는 열과 pH의 영향)

  • This study was carried out to determine the optimum extraction conditions for Ishige okamurai by comparing the yields, total phenolic compound content (TPC), and antioxidant properties of its 95%, 70%, 50% fermented ethyl alcohol and water extracts. Additionally, the effects of heat and pH treatments on the antioxidant properties of the extracts were evaluated by their TPC and 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging capabilities. The yields of the extracts were greatest in the order of water > 50% > 70% > 95% fermented ethyl alcohol, and the TPC of the 70% (26.18%) and 50% fermented ethyl alcohol (27.56%) extracts were higher than those of the others. However, in terms of DPPH radical scavenging and ferrous-reducing power, the 70% fermented ethyl alcohol extract of Ishige okamurai showed the highest antioxidant effects. Additionally, in the results for the heat and pH treatments, the antioxidant properties of the 70% fermented ethyl alcohol extract were not influenced by the treatment conditions except at pH 10.

The Antioxidation Effect of Salsola komarovii Extract and Its Influence on Cell Bio activity (수송나물(Salsola komarovii)의 항산화, 항염 및 미백 활성)

  • S. komarovii is halophyte that grows in soil or waters of high salinity, such as in saline semi-deserts, sloughs and seashores. Traditionally, S. komarovii has been used for food and medicinal purposes in Korea. S. komarovii was extracted in 70% ethanol to measure anti-oxidative activity using DPPH and ABTS assay. The IC50 values of the S. komarovii extract against DPPH radicals and ABTS radicals were 186.10 mg/mL and 121.89 mg/mL. In addition, total polyphenol and reducing power were measured. The S. komarovii extract exhibited superior polyphenolic (22.5%) and antioxidant (28.4%) contents. Regarding cell bioactivity, MTT assay was conducted to reveal cytotoxicity of S. komarovii extract and showed the non-cytotoxicity of S. komarovii extract. Anti-inflammatory and skin whitening effects were measured at 100 ㎍/mL. Therefore, this study suggests that the S. komarovii extract can be used as a functional cosmetic product material.

The Antioxidation Effect of Mutimo cylindricus Extract and Its Influence on Cell Bioactivity (회초리말(Mutimo cylindricus)의 항산화, 항염 및 미백 활성)

  • The objective of this study was to evaluate functionality of Mutimo cylindricus as a cosmetic ingredient, one of the brown algae. The M. cylindricus 70% ethanol extract was manufactured for antioxidant measurement. DPPH and ABTS methods were used to measure antioxidants, and its EC50 values were 2.040 and 2.182 mg/mL in each experiment. The measurement of total polyphenol contents and reducing power showed total polyphenol content of 103 mg gallic acid/g extract and reducing power of 134 mg ascorbic acid/g extract. To measure cell toxicity, MTT method was used, and its result showed that the extract was not cytotoxic. And it has anti-inflammatory and whitening activity at concentrations of 100 ㎍/mL. The result confirmed that M. cylindricus extract is available as a cosmetic material with whitening and anti-inflammatory properties.
