Recent research has revealed that hydrolyzed collagen peptides have beneficial effects in various diseases such as osteoarthritis and human rheumatoid arthritis and also play a protective role in skin by improving the activity of antioxidants. In this study, we investigated the effects of a novel mixture (AP-CPM01) containing collagen peptides and elastin peptides on photoaged hairless mice skin both in vivo and in vitro. To evaluate the effects of AP-CPM01 on UVBinduced skin wrinkle formation in vivo, the hairless mice were exposed to UVB irradiation and orally administered the AP-CPM01 at 333 mg/kg per day for 10 weeks. The effects on skin appearance and epidermal thickness were measured using bioengineering and histochemical methods. In addition, the influence of AP-CPM01 on collagen metabolism in human skin fibroblasts was also investigated. The skin of mice in the AP-CPM01 treated group had better appearance and less wrinkling than that of mice in the control group. In the human fibroblast cells, the amount of de novo procollagen synthesis was increased after AP-CPM01 treatment, reflecting that AP-CPM01 can induce de novo procollagen synthesis and reduce UVB-induced skin wrinkle formation. These results suggest that AP-CPM01 is a potent candidate for antiphotoaging functions.
This study is about a technique for obtaining collagen by extracting fat by treating collagen-containing liposuction effluent in the presence of supercritical fluid. Using a supercritical solvent, a collagen extract could be obtained from animal-derived fat in a short time (about 6 hours), and about 2-3% of collagen by mass compared to the raw material could be obtained. The presence of collagen in the extract obtained by supercritical extraction was confirmed by SDS-PAGE, and it was confirmed that it was type 1 collagen having a relatively large molecular weight. In addition, the growth factors of IGF-1, bFGF, VEGF and NGF were analyzed to find out which growth factors were present in the collagen obtained by supercritical extraction, and it was found that these growth factors were contained in the extract. There was no significant difference in DNA content per mg of sample before and after supercritical treatment. Further in-depth studies are likely to be needed on decellularization technology using the supercritical process. In conclusion, the extracellular matrix obtained through the solvent extraction process using a supercritical fluid contains growth factors above a certain amount even after decellularization and removal of fat, so that it was found that not only biocompatibility is greatly increased, but also tissue regeneration can be rapidly induced.

The objective of this study was to investigate the effects of ingestion of low-molecular-weight collagen peptides on lipid composition, blood glucose level, and enzyme activities in the serum of hyperlipidemic rats fed experimental diets for 5 weeks. Concentrations of total cholesterol, low-density lipoprotein (LDL)-cholesterol, free cholesterol, triglyceride (TG), phospholipid (PL), and blood glucose, the atherosclerotic index, and the cholesteryl ester ratio were higher in serum of the hyperlipidemic group (CW group), and the cholesterol-plus-collagen peptides extract group (CCP group) than in the control group (BG group basal diet plus water). However, the concentrations of total cholesterol, LDL-cholesterol, free cholesterol, TG, PL, and blood glucose, the atherosclerotic index, and the cholesteryl ester ratio in serum were lower in the CCP group than in the CW group. By contrast, the ratio of HDL-cholesterol to total cholesterol and the absolute HDL-cholesterol level in the CCP group were higher than in the CW group. The activities of alkaline phosphatase (ALP) and aminotransferases (AST, ALT) in serum were lower in the CCP group than in the hyperlipidemic CW group. The results indicate that an extract of low-molecular-weight collagen peptides effectively inhibited increases in lipid elevation, blood glucose level, and enzyme activities, in the serum of hyperlipidemic rats.
Thread lifting has become popular as a minimally invasive technique for facial rejuvenation. Commercially available threads are composed of poly-L-lactic acid (PLLA), polycaprolactone (PCL), or polydioxanone (PDO). However, the histological changes that occur in response to implanted threads are unclear. The aim of this study was to evaluate histological changes that occur in response to implantation with three types of bioresorbable threads (PLLA, PCL, PDO) in rat skin. PLLA, PCL and PDO threads were implanted in the dorsal skin of Sprague Dawley rats and tissue samples were harvested 2, 4, 8 and 12 weeks post-implantation. To evaluate histologic changes induced by bioresorbable face-lifting threads, tissue samples were stained with hematoxylin & eosin, Masson's trichrome stain and Herovici's collagen stain. All three threads induced neocollagenesis of type 3 collagen in the rat skin. The amount of collagen induced by the threads was dependent on the thread surface area. The PDO cavern-type thread was most effective in inducing neocollagenesis due to its extensive surface area. Our results suggest that type 3 collagen induced by bioresorbable threads depends on the thread surface area to uphold the dermis and contributes to facial rejuvenation.
We studied the effects of genistein obtained from glycolysis of genistin, a kind of phytoestrogen present in soybeans, on cell proliferation and type I pN collagen synthesis in normal human dermal fibroblasts(NHDF). Cell proliferation was increased significantly with genistein treatment at 54-year aged NHDF. Genistein increased cell proliferation more strongly in cells form old doner than young doner. The senescence-associated ${\beta}$-galatosidase activity was decreased in NHDF from 77-year old doner with genistein treatment. Type I pN collagen synthesis was increased with genistein treatement in UVA treated and non-treated NHDF. The increasement of collagen synthesis was more effective in aged cells than young cells. Type I pN collagen synthesis was also increased with genistein treatment in collagen matrix culture with NHDF from sun-exposed and non-exposed skin from 54-year old doner. Genistein treatment inhibited MMP-1 synthesis in old NHDF but not in young NHDF. In conclusion, genistein may be a useful agent for preventing intrinsic aging as well as photoaging.
The aim of this study was to investigate and development collagen peptide materials from skate skin. Protein and fat content of collagen peptide showed about 95% and 0.1%, respectively. Average molecular weight of collagen peptide was measured as 1,015. In the analysis of amino acid, glycine and hydroxy proline content in collagen peptide was 19.32% and 16.25%, respectively, showing a typical characteristics of the collagen peptide. In obese db/db mice ingested 500 mg/day of collagen peptide for 18 days, the amounts of food and water intake were decreased considerably, contents of triglyceride, total cholesterol were decreased significantly in white adipose tissue of db/db mice. The final yield of collagen peptide was 17.23% in the optimized process for mass production. These results indicate that collagen peptide from skate skin may serve as candidates of fat reduction in adipose tissue and could be used as functional food and cosmetic ingredients.

Eogeul-tang is a traditional Korean soup dish made from dried pollack, minced beef, and tofu. This comparative study analyzed the quality characteristics of Eogeul-tang by varying the added amounts of cow gristle. The objective of this study was to analyze Eogeul-tang to promote its functionality and increased consumer preference. Collagen contents were $18.08{\pm}0.13g$ and $22.17{\pm}0.14g$ per 100 g of dried pollack skin and knee cartilage of cow, respectively. The overall collagen content was higher in the knee cartilage of cow. Different amounts of cow gristle (25%, 50%, 75%, and 100%) were added to traditionally cooked Eogeul-tang, and the general composition of Eogeul-tang was analyzed. Significant differences were exhibited in the amounts of water content, crude protein, crude fat, and crude ash depending on the added amounts of cow gristle. Moreover, the collagen content also significantly increased as the amount of gristle content increased. In particular, significant increases in the amounts of collagen components, including proline, glycine, and alanine were observed with increasing amount of gristle. When a sensory test was conducted on Japanese, overall preference values were highest for Eogeul-tang composed of 25% beef and 75% gristle compared to traditionally cooked Eogeul-tang. In conclusion, the study results promote the functionality of collagen as well as the increased quality of Eogeul-tang added with cow gristle manufactured by traditional cooking methods.

This study was conducted to examine the effects of soluble collagen peptides extract supplementation on the improvement of blood glucose, lipid components and enzyme activities in the sera of streptozotocin (STZ, 55 mg/kg BW, IP injection)-induced diabetic rats fed on experimental diets for 5 weeks. The concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglycerides (TG) and phospholipids (PL) in serum were remarkably higher in the diabetic group (group SW) and STZ (IP)+collagen peptides extract supplementation group (group SFW) than those in the control group (group CG, basal diet + water). However the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, TG and PL in serum were lower in the SFW group than in the SW group, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the SFW group were higher than in the SW group. The activities of alkaline phosphatase (ALP) and aminotransferase (AST, ALT) in serum were lower in the SFW group than in the diabetic SW group. The results shown above suggest that soluble collagen peptides extract supplementation effectively improves blood glucose, lipid compositions and enzyme activities in the sera of STZ induced diabetic rats.

To research the effects of marigold extract, which is used mixed with calendula extract, on collagen growth and MMP-1 expression in human fibroblast, we measured cytotoxicity, collagen growth and MMP-1 expression by using HDF cells. The result of measurement showed over 80% cell survival rate in $5{\sim}100{\mu}g/mL$ concentration of marigold extract and calendula extract for HDF cells, which indicates there is no cytotoxicity. The result of measuring collagen synthetic abilities showed both types of extract had collagen synthetic ability increase dose dependently, by 25% in $100{\mu}g/mL$ concentration of marigold extract, and by 7% in $100{\mu}g/mL$ concentration of calendula extract. The result of experimenting the effect on MMP-1 expression showed that both types of extract suppress MMP-1 expression. The result of observing phosphorylation of p-JNK and p-ERK, which are known to be involved with MMP-1 expression, revealed that marigold extract effectively suppresses MMP-1 expression through signaling pathway of p-JNK and p-ERK. The above results confirm the wrinkle improvement effect of marigold extract, and furthermore, it can be used as a cosmetic ingredient for anti-aging.

Collagen hydrolysate (CH) is known to prevent skin aging by stimulating skin dermal fibroblasts to promote synthesis of extracellular matrix such as collagen and elastin. Recently, among the various factors that cause skin aging, advanced glycation end products (AGEs) have received particular attention. However, the effect of CH on AGE accumulation has not been studied. Since CH could affect AGE accumulation by promoting production of skin structural proteins, clinical trial was performed using low molecule collagen peptide (LMCP), which were CH containing 25% tripeptide and 4% Gly-Pro-Hyp. Skin autofluorescence (SAF) values were measured using an AGE reader to evaluate accumulation of AGE in skin. As a result of applying 0.5% and 1.0% LMCP solutions to the subject's forearm for 8 weeks, the SAF value at the test site significantly decreased compared to the control site. Additionally, in vitro test was performed using CCD-986sk to evaluate the promotion of collagen synthesis in skin fibroblasts by LMCP. As a result, 800 ㎍/mL of LMCP significantly increased synthesis of human pro-collagen Iα1 (COL1A1) in CCD-986sk. Through this study, we have confirmed that tropical LMCP applications can promote collagen synthesis to help anti-glycation effects, suggesting that LMCP has potential as an anti-aging cosmetic material.
