Suppressive Effect of Water Extract from Leaf of Cirsium japonicum var. ussuriense on Collagen-induced Arthritis (엉겅퀴 잎 수용성 추출물의 콜라겐 유도 관절염 억제효과)

  • Cirsium japonicum var. ussuriense is often used in treatment of human disease such as hemorrhage, blood congestion and inflammation. The present study has been undertaken to investigate the effect of the leaf extract from C. japonicum var. ussuriense (CLE) on the development of collagen-induced arthritis (CIA) in DBA1/J mice. CLE administration suppressed markedly the arthritis incidence and arthritis score in CIA mice. Also, CLE significantly suppressed the release of $PGE_2$, TNF-${\alpha}$, IL-4 and IL-6 in CIA mice. However, CLE significantly increased the production of IL-10, but not IL-4. These results suggest that CLE suppress inflammatory mediators and regulates Th1 and Th2 cytokines. These properties may contribute to the anti-arthritis action of CLE.

  • Inflammatory reaction is characterized by over-production of inflammatory mediators due to an up-regulation of inflammatory pathways, which produce pro-inflammatory mediators, such as interleukin-1beta (IL-$1{\beta}$), IL-6, tumour necrosis factor alpha (TNF-$\alpha$), prostaglantin $E_2$ ($PGE_2$), and nitric oxide (NO) in U937 cells. We investigate the anti-inflammatory effects of water extracts from Lonicerae Flos and Scutellariae Radix in lipopolysaccharide (LPS)-stimulated U937 cells. Each extract suppressed the production of inflammatory mediators (NO, IL-$1{\beta}$, TNF-$\alpha$, and $PGE_2$) and the expression of inducible NO synthase and cyclooxygenase-2 in LPS- stimulated U937 cells in a dose-dependent manner. These suppressive effects were synergistically increased by their combination. Their combination extract also inhibited NF-${\kappa}B$-DNA complex of NF-${\kappa}B$ binding activity and translocation of NF-${\kappa}B$ from cytosol to nucleus. These results suggest that the combination of water-extractable components of Lonicerae Flos and Scutellariae Radix may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

  • Purpose: This study was performed to detect various chemicals such as metal elements and volatile organic compounds which contained in mascara and we analyzed 5 sorts of mascaras frequently used by people. Methods: We purchased five sorts of mascaras commercially available and analyzed for detecting metal elements and volatile organic ingredients. Results: We found somewhat high concentration of aluminum and manganese and several organic compounds in some samples. Conclusions: The results suggest that mascara has metal elements and organic compounds which can influence to eye.

  • Objectives : The water extract of Hwangnyeonhaedok-tang (HHT), composed of the Scutellariae Radix, Coptidis Rhizoma, Phellodendri Cortex and Gardeniae Fructus has been traditionally used to treat fever, inflammation, gastritis and hypertension in east asia. However, little is known about the ameliorative effects of HHT on atopic dermatitis. Therefore, the purpose of this study was to investigate the therapeutic effect of HHT on atopic dermatitis Methods : We investigated the inhibitory effects of HHT on the production of proinflammatory cytokines in rat peritoneal mast cells (RPMCs), on the scratching behavior in ICR mice, and on atopic dermatitis symptoms in 2,4-dinitrofluorobenzene (DNFB)-induced atopic dermatitis-like model hairless mice. Results : Levels of TNF-${\alpha}$ and IL-$1{\beta}$ increased by PMA plus A23187 co-treatment were significantly inhibited by HHT in a dose-dependent manner. HHT also inhibited the histamine release from RPMCs stimulated by compound 48/80, which promotes histamine release. The oral administration of HHT reduced the scratching behavior induced by compound 48/80 and histamine in ICR mice. Furthermore, the intradermal treatment of HHT reduced the ear edema, skin lesions, and atopic molecular marker (IgE and IL-4) in DNFB-induced atopic dermatitis model mice. Conclusions : These results suggest that HHT may be used as a potential treatment for AD as a prescription for treatment of atopic dermatitis.

Ginsenosides Prevent High Glucose-induced Apoptosis in HT22 Cells (해마 세포주에서 인삼의 고포도당에 의한 세포사멸 차단효과)

  • Diabetic neuropathy is characterized by the decrease of cell viability in neuron, which is induced by the hyperglycemia. HT22 cell is the neuron cell line originated from hippocampus. Ginsenosides have been reported to retain anti-diabetic effect. However, the preventive effect of ginsenosides in the condition of diabetic neuropathy was not elucidated. Thus, this study was conducted to examine the protective effect of ginsenoside total saponin (GTS), panoxadiol (PD), and panoxatriol (PT) in the high glucose-induced cell death of HT22 cells, an in vitro cellular model for diabetic neuropathy. In present study, high glucose increased lactate dehydrogenase(LDH) activity, the lipid peroxide(LPO) formation and induced the decrease of cell viability. These effects were completely prevented by the treatment of GTS, but partially prevented by the treatment of PD and PT. High glucose also increased the expression of Bax and cleaved form of caspase-3 but decreased that of Bcl-2. These effects of high glucose on Bax, Bcl-2 and cleaved form of caspase-3 were completely prevented by the treatment of GTS, but partially prevented by the treatment of PD and PT in HT22 cells. In conclusion, ginsenosides prevented high glucose-induced cell death of hippocampal neuron through the inhibition of oxidative stress and apoptosis in HT 22 cells.

Inhibitory effects of the Juglans sinensis Walnut Ethanol Extract on Carbon Tetrachloride-induced hepatic fibrosis in Rats (호두인 에탄올 추출물의 사고화탄소 유도 간섬유화 억제 효과)

  • The inhibitory effects of the Juglans sinensis walnut ethanol extract (JSWE) on carbon tetrachloide (CCl₄)-induced fibrosis, serum transaminases (GOT and GTP), hepatic glutathione (GSH), serum pro-inflammatory cytokines (IL-1β and IL-6) were investigated in rats. JSWE significantly inhibited on CCl₄-induced fibrosis in dose-dependent manner. Moreover, JSWE significantly inhibited on the serum levels of GOT, GTP, IL-1β, and IL-6 in dose-dependent manner in CCl₄-induced fibrosis rat. However, JSWE markedly increased the production of hepatic GSH in a dose-dependent manner. These results show that JSWE may explain some known biological activities of Juglans sinensis walnut including their anti-fibrotic and anti-inflammatory effects, and is of considerable benefit in the treatment for live diseases.

  • The study performed a series of flexural tests on Closed Cold-Formed Steel Sections for stud, joist, and roof truss. Results were compared with analytical values. Each 2.4-m long and 0.9-m wide specimen consisted of two steel beams set at 0.46 m interval. The steel beams were attached to the specimens using either plaster board or ply wood. Another specimens did not use any attachment material. Positive and negative bending tests were conducted to investigate the composite behavior, including the effects of plaster board or ply wood on the buckling behavior of steel beam. Full-scale roof truss tests were also performed to study the buckling behavior and failure mode of the truss members.

Immunosuppressive Activity of Wogonin from Scutellaria Radix on DNFB-induced Delayed Type Hypersensitivity in Mice (마우스에서 황금 유래 Wogonin의 Th1과 Th2 사이토가인 조절 효과)

  • In the present study, wogonin, a major flavone isolated from Scutellaria Radix were examined for its imunosuppressive activity on the 2,4-dinitro-fluorobenzene (DNFB)-induced delayed type hypersensitivity (DTH) in C3H/HeN mice. This compound inhibited selectively $TNF-\alpha$ and IL-4, but not IL-6, IL-10 and $IFN-\gamma$ on DNFB-induced Th1 and Th2 cytokines in a concentration-dependent manner. Interestingly, this compound increased the expression of heme oxygenase-1 (HO-1) in a concentration-dependent manner The above results reveal that wogonin possesses anti-inflammatory, humoral and cellular immunomodulatory, and stress reducing activities on the DNFB-induced DTH in mice and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

Anti-Inflammatory Effect of Ethanol Extract of Angelica uchiyamana in Activated Murine RAW 264.7 macrophages (활성화된 설치류 RAW 264.7 대식세포에서 당귀 에탄올 추출물의 항염증 효과)

  • The inhibitory effects of ethanol extract of Radex Angelica uchiyamana, on LPS- or IFN- plus LPS-induced production of NO, and $TNF-{\alpha}$, and expression of iNOS and COX-2 were investigated in the activated RAW 264.7 cells. This extract significantly inhibited the production of NO and $TNF-{\alpha}$, and suppressed the expression of iNOS and COX-2 in a dose-dependent manner. These results show that ethanol extract of Radex Angelica uchiyamana may explain some known biological activities including anti-inflammatory effect, and is of considerable benefit in the treatment for NO and pro-inflammatory cytokine overproduction related immunological diseases.

Development of a Scale to Measure Self-Care for Korean Patients with Chronic Obstructive Pulmonary Disease (만성 폐쇄성 폐질환자의 자가관리측정도구 개발)

  • Purpose : The objective of this study was the development and validation of a scale to measure the self-care of patients with chronic obstructive pulmonary disease(COPD) in Korea. Method: Self-care scale was developed based on the self-care activities patients had to carry out in order to manage their COPD. The original scale contained 34 items rated along a five-point Likert scale and was reviewed by 18 professional nurses and 10 Korean patients with COPD for content validity. Subsequently, patients with COPD were asked to complete this 23-item scale and further tests were done with the 125 useable responses. Result: Factor analysis identified eight factors-'maintaining a clean air way', 'taking medication', 'support from family', 'preventing infection', 'managing symptoms', 'breathing exercising', and 'taking in nutrition'. The internal consistency of the total scale was Cronbach's α=0.7226. These eight factors explained 60.8% of total variance. There was correlation among Korean Self-Care Scale score, administration level, and knowledge level but there was no correlation to patients' satisfaction with medical services. Conclusion: The 23 item questionnaire positively identified 8 areas defined important for COPD patients. Further studies are required to see how these can be integrated into patient education.
