Enhancement of Anticarcinogenic Effect by Combination of Sedum sarmentosum Bunge with Platycodon grandiflorum A. Extracts (도라지 추출물 첨가에 의한 돌나물의 항발암 상승효과)

  • Anticarcinogen is one of the major strategies for cancer control. It is well established that dietary factors play an important role in modulating the development of certain types of human cancer. We investiagted the anticarcinogenic effects of Sedum sarmentosum Bunge (SS) with Platycodon grandiflorum A. extracts on HepG2, HeLa and MCF-7 cell lines. By the MTT assay, among the five partition layers of methanol extract of SS (SSM), the ethylether partition layer of SS (SSMEE) showed the strongest cytotoxic effects on all cell lines. We also investigated the synergistic effect of the combination of SS and PG extracts on growth inhibition of the HepG2, HeLa and MCF-7 cell lines compared to the effects of five partition layers of SSM. Combination of SS and PG extracts significantly increased cytotoxic effects on all cell lines. Therefore, we were able to conclude that ethylether partition layer, SSMEE might have potentially useful cytotoxic materials on all the human cancer cells which we used. And we could suggest that the combination of SS with PG enhanced the anticarcinogenic effect on HepG2, HeLa and MCF-7 cell lines. We also determined QR activity of partition layers of SSM, among them, SSMEE on HepG2 cells showed the highest QR activity, 3.21 as control value of 1.0.

Effect of Mycelia Extracts from Lentinus edodes Mushroom-Cultured Lonicera japonica Thunberg on Anticancer and Antiallergy Activities (인동초로 배양한 표고버섯 균사체 추출물의 항암 및 알레르기 억제효과 검증)

  • This study was conducted to investigate the effect of mycelia of Lentinus edoes mushroom-cultured Lonicera japonica Thunberg (LLJ) on proliferation of the cancer cell lines (Hep3B, MCF-7 and HeLa), sarcoma 180 (S-180) and antiallergy. In an anti-cancer test using Hep3B (hepatic cancer cell), MCF-7 (breast cancer cell) and HeLa (uterine cancer cell), LLJ extract showed higher antiproliferating effect than that of LJ (Lonicera japonica Thunberg) extract. In an anti-cancer testing using Hep3B cells, LLJ extract showed growth-inhibitory effect of $85.60{\pm}4.66%$ at 3mg/rnL. In an anti-cancer testing using MCF-7 cells, LLJ and LJ extracts showed high antiproliferating effect. LLJ showed the tumor suppressive effect in mice injected with S-180 cells. The growth-inhibitory rates against tumor cells were 61% for LLJ, 37% for LJ. LLJ inhibited histamine release from rat peritoneal mast cells activated by compound 48/80. These results suggest that Lentinus edodes mushroom-cultured herb has an antiproliferating effect against cancer cell lines (Hep3B, MCF-7 and HeLa) and S-180 tumor, and will be beneficial in the treatment of allergic reaction.

Effect of Broccoli Extract on Inhibition of Cancer Cell Proliferation (브로콜리 추출물의 암세포 증식 억제에 미치는 효과)

  • This study was conducted to examine the effect of Broccoli Extract on the proliferation inhibition of human-derived cancer cells and the degree of inhibition. The three cell lines used in the experiment were respiratory system lung cancer cells A549, digestive system liver cancer cells SNU-182 and biliary tract cancer SNU-1196. All cancer cells were derived from the human body, and the CCK-8 method was used to measure the degree of inhibition of cancer cell proliferation. As a result of examining the effect on Broccoli Extract 10ug/mL, 100ug/mL, 1000ug/mL, Broccoli Extract inhibited proliferation in a concentration-dependent manner in most cancer cells, In particular, lung cancer cell A549 and liver cancer cell SNU-182 showed significant proliferation inhibition at 1000ug/mL.As a result, it can be seen that broccoli extract provides potential as a cancer preventive and therapeutic agent for tumor suppression mechanisms proven through cell experiments.

The Effects on Antimicrobial and Cytotoxicity of Brassica oleracea L. Fractions. (적채 분획물의 항균, 암세포 증식 억제효과)

  • This study was carried out to investigate the affects on antimicrobial and cytotoxicity of red cabbage (Brassica oleracea L.,BO). In the paper disc test, antimicrobial activity of BO fractions was increased in proportion to its concentration. Among the various solvent fractions of methanol extract (BOM) of BO, the ethylacetate partition layer (BOMEA) showed the strongest antimicrobial activity We also determined the cytotoxicity and Quinone Reductase induced effect of BO extract and fractions on human cancer cells. The cytotoxicity of BO fractions on HepG2, HeLa and MCF-7 cells was evaluated by MTT assay. The BOMEE and BOMEA showed strong cytotoxic effects on all cancer cell lines we used. The quinone reductase induced effect of BO fractions on HepG2 cells, the hexane partition layer (BOMH) at a dose of 200 $\mu{g}$/ml was 2.88 times more effective compared to the control values of 1.0.

Inhibitory Effect of Celeriac Extract on Cancer Cell Proliferation (셀러리악 추출물의 암세포 증식 억제 효과)

  • This study was carried out examine the effect of Celeriac Extract, which contains various anticancer ingredients, on the proliferation inhibition of human-derived cancer cells and the degree of inhibition. The five cell lines used in the experiment were lung cancer cells A549, prostate cancer cells DU-145, uterine cancer cells HeLa, breast cancer cells MCF-7, and liver cancer cells SNU-182. All cancer cells derived from the human body were used, and the inhibition of cancer cell proliferation with Celeriac Extract 10ug/mL, 100ug/mL, and 1000ug/mL was measured using the CCK-8 method. As a result of examining the inhibition of cancer cell proliferation, Celeriac Extract 1000ug/mL showed significant proliferation inhibition in lung cancer cells A549, prostate cancer cells DU-145, uterine cancer cells HeLa, and liver cancer cells SNU-182, and showed a concentration dependence. However, only a concentration-dependent decrease was observed in breast cancer cells MCF-7.In conclusion, it can be seen that the cell proliferation inhibition mechanisms of Celeriac Extract using various human-derived cancer cell lines provide the potential for cancer prevention and therapeutic development.

Inhibitory Effects on Cell Survival and Quinone Reductase Induced Activity of Aster yomena Fractions on Human Cancer Cells (쑥부쟁이 분획물의 in vitro 암세포증식 억제 및 QR 유도효과)

  • This study was performed to determine the inhibitory effects on cell survival and Quinone reductase induced activity of Aster yomena (AY) on human cancer cells which, using methanol, was extracted and fractionated into five different solvent types: hexane (AYMH), ethylether (AYMEE), ethylacetate (AYMEA), butanol (AYMB) and aqueous (AYMA) partition layers. The experiment was conducted to determine cytotoxicity of various Aster yomena partition layers on HepG2, HeLa and MCF-7 cells by MTT assay. Among various partition layers of Aster yomena, A YMEE and A YMEA showed the strong cytotoxic effects on all cancer cell lines we used. The Quinone reductase (QR) induced activity on HepG2 cells, A YMH at a does of 100 $\mu$g/mL was 2.46 times more effective compared to the control value of 1.0.

  • The present sturdy was designed to determine the effect of the antiproliferation in human cancer cells using the ordinary vegetable soup (VS), the soup with broccoli (VSB) and the soup with tomatoes(VST). Human cancer cells identify the cancer cell growth with MTS, using stomach cancer cell line(AGS), human promyelocytic leukemia (HL-60) and lung cancer cell line (A549). VSB and VST are effective on the cancer cell growth inhibition activities of AGS and have a significance compared with VS. VST has a significance with HL-60 and VSB works well in A549 more than VS. Mixed vegetable soup is considered to applicate with functional materials and using for wellness life.

Inhibitory Effect of Linum usitatissimum and Perilla frutescens as Sources of Omega-3 Fatty Acids on Mutagenicity and Growth of Human Cancer Cell Lines (식물성 오메가-3계 지방산 급원인 아마씨 및 들깨의 항돌연변이 및 암세포 증식 억제 효과)

  • It has been known that Linum usitatissimum and Perilla frutescens are dietary sources of possible chemopreventive compounds such as lignans and $\alpha$-linolenic acid. Here, we investigated and compared the inhibitory effects of methanol extracts from Linum usitatissimum and Perilla frutescens on mutagenicity using the Ames test, and growth of human cancer cells (AGS human gastric adenocarcinoma, HT-29 human colon cancer, Hep 3B hepatocellular carcinoma cells). In the Ames test system using Salmonella typhimurium TA100, aflatoxin $B_1$ ($AFB_1$)-induced mutagenicity was significantly inhibited by treatment with the methanol extract from either Linum usitatissimum or Perilla frutescens (p<0.05) in a dose dependent manner. As for N-methyl-N'-nitro-N-nitrosoguamidine (MNNG)-induced mutagenicity, the methanol extracts (5 mg/assay) from Linum usitatissimum and Perilla frutescens showed 63% and 78% inhibitory rates, respectively, indicating that Perilla frutescens possessed stronger antimutagenic activity than did Linum usitatissimum. Inhibitory effects of methanol extracts from Linum usitatissimum and Perilla frutescens on the growth of human cancer cells (AGS, HT-29 and Hep 3B) appeared to increase dose dependently, and the inhibition was more effective against AGS and HT-29 compared to Hep 3B cells. Our results suggested that the methanol extract from Perilla frutescens showed stronger antimutagenic activity than that from Linum usitatissimumas assayed by the Ames mutagenic test, whereas the methanol extract from Linum usitatissimum was more effective than its counterpart for growth inhibition of human cancer cells. It is concluded that intake of Linum usitatissimum and Perilla frutescens as sources of omega-3 fatty acids will be beneficial for preventing cancer.

Inhibitory Effects of Methanol Extracts from Korean Orysa sartiva and Coix lachryma-jobi var. ma-yuen on Mutagenicity and Growth of Human Cancer Cells (한국산 현미 및 율무 추출물에 의한 돌연변이 및 인체 암세포주 증식 억제 효과)

  • This study was carried out to determine the inhibitory effects of methanol extracts from Korean Orysa sartiva and Coix lachryma-jobi var. ma-yuen on mutagenicity using Ames test and growth of AGS human gastric adenocarcinoma and HT-29 human colon cancer cells. Both treatments of methanol extracts (5 mg/assay) from Orysa sartiva and Coix lachryma-jobi var. ma-yuen to Ames test system inhibited aflatoxin $B_1$ ($AFB_1$) induced mutagenicity by 76%. In case of N-methyl-N'-nitro-N-nitrosoguamidine (MNNG) induced mutagenicity, the methanol extracts (5 mg/assay) from Orysa sartiva and Coix lachryma-jobi var. ma-yuen showed 79% and 69% inhibitory rate, respectively and the inhibitory effect was a little stronger in Orysa sartiva Inhibitory effects of methanol extracts from Orysa sartiva. and Coix lachryma-jobi var. ma-yuen on the growth of AGS and HT-29 human cancer cells were increased as dose dependent patterns and the inhibitory effects on AGS and HT-29 cells were similar. The above results indicate that the consumption of these cereals, which contain many nutrients with good quality, may be recommended as potent functional foods for improving health.

Synergistic Effect of Panax ginseng and Cinnamoum Blume Mixture on the Inhibition of Cancer Cell Growth in vitro (인삼과 계피 혼합물에 의한 in vitro에서 암세포 증식억제의 상승 효과)

  • The effects of ginseng and cinnamon extract alone or mixture on the various cancer cell lines in vitro have been examined. Human colon cancer cell line (HT-29), human rectal cancer cell line (HRT-18) and human hepatoma cell line (HepG2) were used for the experiment. When given separately, the proliferation of all cancer cell lines was inhibited in proportion to the concentration of ginseng or cinnamon extract, respectively. Based on the cytotoxic activity, mixture of ginseng and cinnamon extract demonstrated a synergistic inhibition of cancer cell growth. The progression of cell cycle from G1 to S phase was significantly inhibited by ginseng and cinnamon mixture in the HT-29 and HRT-18 cell lines.
