Formation of Metastases in C57 BL/6 Mouse Lung (LL/2 Cells접종에 의한 C57 BL/6 Mouse 폐에서의 전이과정)

  • Comparative differences between the fine structure of cultured LL/2 cell in vitro and tumor cells in vivo which were induced in the lung by inoculation of LL/2 cells to C57 BL/6 mouse via tail vein during 21 days are not observed except for cell configuration which was changed spindle shape into oval shape. At first tumor cells appeared at lymphatic nodules and around capillary in the lung. Tumor cells divided actively by mitosis, so they became tumor nodules. The pulmonary aveoli around tumor nodules were observed somewhat flattened in shape but the cells in the aveoli appeared to be in normal condition. Furthermore the normal lung cells were observed in the tumor nodules and some apoptotic tumor cells appeared in the large tumor nodules. A lot of neutropiles were observed in the aveoli and tumor nodules of C57 BL/6 mouse lung after inoculation 22 days and 31days.

Anti-invasive Activity of Human Breast Carcinoma Cells by Genistein through Modulation of Tight Junction Function (인체유방암세포의 tight junction 기능 조절을 통한 genistein의 암세포 침윤 억제 효과)

  • Tight junctions (TJs) that act as paracellular permeability barriers play an essential role in regulating the diffusion of fluid, electrolytes and macromolecules through the paracellular pathway. In this study, we investigated the correlation between the tightening of TJs, permeability and the invasive activity of genistein - a bioactive isoflavone of soybeans - in human breast carcinoma MCF-7 and MDA-MB-231 cells. The inhibitory effects of genistein on cell proliferation, motility and invasiveness were found to be associated with the increased tightness of the TJs, which was demonstrated by an increase in transepithelial electrical resistance and a decrease in paracellular permeability. Additionally, the immunoblotting results indicated that genistein repressed the levels of the proteins that comprise the major components of TJ, claudin-3 and claudin-4, which play a key role in the control and selectivity of paracellular transport. Furthermore, genistein decreased the metastasis-related gene expressions of insulin like growth factor-1 receptor and snail, while concurrently increasing that of thrombospondin-1 and E-cadherin. In addition, we demonstrated that claudins play an important role in the anti-motility and invasiveness of genistein using claudin-3 small interfering RNA. Taken together, our results indicate a possible role for genistein as an inhibitor of cancer cell invasion through the tightening of TJs, which may counteract the up-regulation of claudins. In addition, our results indicate that this may be beneficial for the inhibition of tumor metastasis.

Antiproliferative Effect of Extracts from Corydalis heterocarpa on Human Cancer Cells (염주괴불주머니 (Corydalis heterocarpa) 추출물의 암세포 성장 억제 효과)

  • Whole plants of Corydalis heterocarpa were extracted twice with $CH_2Cl_2$ and MeOH in turn. The combined crude extracts were concentrated in vacuo and then partitioned between $CH_2Cl_2$ and $H_2O$. The organic layer was fractionated with n-hexane and 85% aq. MeOH, and the aqueous fraction was also further fractionated with n-BuOH and $H_2O$, successively. Growth inhibition effects of crude extracts and their solvent fractions were evaluated in AGS, HT1080, U-937, MCF-7 and HT-29 human cancer cells using MTT assay. The inhibitory effects of solvent fractions were increased in a dose-dependent manner. Among these tested samples, 85% aq. MeOH fraction showed the most potent inhibitory effect on the growth of human cancer cells. These results suggest that active compounds having much stronger anticancer effect can be isolated from Corydalis heterocarpa.

Inhibitory Effects on Cell Survival and Quinone Reductase Induced Activity of Aster yomena Fractions on Human Cancer Cells (쑥부쟁이 분획물의 in vitro 암세포증식 억제 및 QR 유도효과)

  • This study was performed to determine the inhibitory effects on cell survival and Quinone reductase induced activity of Aster yomena (AY) on human cancer cells which, using methanol, was extracted and fractionated into five different solvent types: hexane (AYMH), ethylether (AYMEE), ethylacetate (AYMEA), butanol (AYMB) and aqueous (AYMA) partition layers. The experiment was conducted to determine cytotoxicity of various Aster yomena partition layers on HepG2, HeLa and MCF-7 cells by MTT assay. Among various partition layers of Aster yomena, A YMEE and A YMEA showed the strong cytotoxic effects on all cancer cell lines we used. The Quinone reductase (QR) induced activity on HepG2 cells, A YMH at a does of 100 $\mu$g/mL was 2.46 times more effective compared to the control value of 1.0.

Effects of Chitosan on Anti-tumor Activity in Mice (키토산이 암세포성장에 미치는 효과)

  • Cytotoxic anticancer chemotherapeutic agents generally produce severe side effects, while reducing host resistance to cancer and infections, especially through the destruction of lymphoid and bone marrow cells. In this study, we have investigated the effect of chitosan on cytotoxic activity against cancer cells and life span in mice. The direct cytotoxicity of chitosan or chitosan-combinated chemotherapeutic agents for tumor cells was observed. In addition, the effect of life span extention was counted on sarcoma 180 mice injected with chitosan-combinated mitomycin C. The effect of growth inhibion for cancer cells, K562 and Yac-1 was shown in the cytotoxicity test of chitosan or chitosan-combinated chemotherapeutic agents. Also, the effect of life span extension was observed on sarcoma 180 mice injected with chitosan-combinated mitomycin C. Our results suggest that life span extension in sarcom 180 mice exposed with chitosan-combinated chemotherapeutic agents showed the probability of its usefulness for cancer therapy if more research results were accumulated.

Chivosazole F, An Efficient Inhibitor of Multidrug-Resistant Cancer Cells Isolated from Sorangium Cellulosum [Myxobacteria] (Chivosazole F: 점액세균 Sorangium Cellulosum이 생산하는 다제내성 암세포의 생장억제물질)

  • In the course of our search for compounds effective to multidrug-resistant cancer cells from myxobacteria with the adriamycin-resistant cancer cell line CL02, we found cytotoxic activity against the CL02 cells in culture extract of Sorangium cellulosum JW1045. Activity-guided fractionation of the culture extract led to the isolation of an active principle, chivosazole F, This compound showed high cytotoxic activity against cultured human cancer cells. The $IC_{50}$ values, measured by a SRB assay with different cell lines, ranged from 0.1 to 10 ng/ml. Furthermore chivosazole F was as active against drug-resistant cancer cells CL02 and CP70 as against the corresponding sensitive cells.

The Inhibitory Effects of Melittin on Human Prostate Cancer Cell PC-3 in vivo and in vitro (Melittin의 전립선암세포 증식에 대한 억제 효과)

  • 목적 : 이 연구는 봉독의 주요 성분인 낮은 농도의 melittin이 in vitro에서 세포자멸사 관련 단백질과 전립 선암세포 PC-3 증식 관련 수용체의 발현 조절을 통하여 세포자멸사(Apoptosis)를 유도하는지 in vivo에서 또한 전립선 암세포주인 PC-3 세포의 성장을 억제하는지 살펴보고자 하였다. 방법 : Melittin을 처리한 후 전립선암세포 PC-3의 성장억제를 관찰하기 위해 WST-l assay와 morphology analysis를 시행하였고, 세포자멸사 관련 MAP kinase 계열의 대표인 ERK1/2과 전립선암세포 증식관련 수용체인 PDGF-BB receptor ${\beta}$의 활성 변화 관찰에는 western blot analysis 및 Immunofluorescence Staining , Confocal immunocytochemistry를 시행하였으며, 전립암세포의 종양형성에는 흉선을 제거한 쥐에 Tumorigenecity study를 시행하였다. 결과 : 1. PC-3 세포에서 Melittin 처리 후 세포증식이 억제되었고 세포의 형태는 세포자멸사의 특징을 나타내었다. 2. PC-3 세포에서 Melittin 처리 후 ERKl/2과 PDGF-BB receptor ${\beta}$의 활성이 억제되었다. 3. PC-3 세포에서 Melittin과 AG1296을 함께 투여시 PDGF-BB receptor ${\beta}$ 활성억제의 상승효과가 나타났다. 4. 흉선 제거 후 전립선암세포주를 이식한 쥐에서 Melittin을 피내로 주입한 결과 전립선암의 크기와 무게가 유의하게 감소하였다. 결론 : 이상의 결과는 Melittin이 ERKl/2과 PDGF BB receptor ${\beta}$의 활성 억제를 통하여 인간 전립선암세포주인 PC-3의 세포자멸사를 유발함으로써 증식억제 효과가 있음을 입증한 것이며, 이를 재확인한 생처 연구에서의 긍정적인 결과는 향후 Melittin의 전립선암 예방과 치료에 대한 효과적인 치료제 개발에 초석이 될 것으로 기대된다.

Effects on Hot Water Extract of Schizandra chinensis on Colon Cancer (오미자 열수추출물의 대장암세포 증식억제 효과)

  • The anticancer activity of Schizandra chinensis Baillon was investigated for the development of functional food resources. The antiproliferative activity of hot water extracts of Schizandra chinensis Baillon in human colon cancer cell line (HT-29) were identified using cell viability, morphology study, cell cycle and RT-PCR analyses. HT-29 cells were cultured in several concentrations (0, 1.0, 2.0, 4.0 mg/mL) of water extracts of Schizandra chinensis Baillon. In our study, colon cancer cell growth could be inhibited by hot water extracts of Schizandra chinensis Baillon in a dose-dependent manners. It was associated with morphological changes and apoptotic cell death with cell shrinking, chromatin condensation, apoptotic bodies and cell cycle analysis. These results suggest that Schizandra chinensis Baillon may inhibit the growth of human colon cancer cells by various apoptosis-aiding activities as well as apoptosis itself.

Effect of Methanol Extracts of Red Colored Rices on Antioxidant Activity and Growth Inhibitory Activities of Cancer Cells (적미 추출물과 분획물의 항산화 활성 및 암세포 성장억제효과)

  • The inhibitory effects of the water and methanol extracts of Jakwangdo and Honghyangmi on the rat microsome lipid peroxidation and growth of four human cancer cells such as HepG2 (liver cancer), SNU-1 (stomach cancer) MCF-7 (breast cancer) and SNU-C4 (colon cancer) were examined. The methanol extracts of red colored rices showed the antioxidant activity and growth inhibitory effects of cancer cells. However, water extracts did not show the activities. Inhibitory activities of methanol extracts of Jakwangdo and Honghyangmi against lipid peroxidation of rat microsome was 80% and 68%, respectively, at the concentration of 1 mg/assay. Jakwangdo methanol extracts showed the highest growth inhibitory activity in MCF-7 cells among the cancer cells tested. The methanol extracts of red colored rices were further fractionated with hexane, chloroform, ethyl acetate and butanol. Both chloroform and hexane fractions showed strong growth inhibitory activity in HepG2 and MCF-7 cells.

Comparison of Anticacer Activities from the Culture and Extraction Conditions of the Spirulina platensis (Spirulina platensis의 배양 및 추출조건에 따른 항암활성 비교)

  • A extract from Spirulina platensis of seawater and freshwater was obtained by using the water and ethanol. Extraction yields of seawater S. platensis were observed about 3% higher than freshwater S. platensis. Cytotoxicity (HEK293) and inhibition ratio of cancer cell line (A549, AGS, MCF7, Hep3B) in adding of the extracts from the S. platensis of seawater and freshwater were measured by SRB assay. Cytotoxicity of all of the extracts in adding 1.0 mg/ml was below 26%. Ctotoxicity of the extracts from the seawater S. platensis were about 6% less than freshwater S. platensis. Inhibition ratio of cancer cell growth was inhibited by adding 1.0 mg/ml of the extracts that was obtained about 80%. Inhibition effect of cancer cell growth in adding seawater S. platensis was observed higher than freshwater S. platensis. Differentiation ratio of HL-60 cells in adding the extracts of seawater S. platensis was observed highly that was 160.9%.
