Antimicrobial Effect of Extract of Glycyrrhiza uralensis on Methicillin-Resistant Staphylococcus aureus (감초 추출물이 항생제 내성균주의 항균활성에 미치는 영향)

  • Antimicrobial drug-resistance is natural response to antimicrobial stress based on selection, which weakens chemotherapy effect. Introduction of large numbers of chemotherapeutic agents to clinical practice has generated strains of microorganisms that survive and multiply in vivo with high-drug concentrations. Methicillin-resistant Staphylococcus aureus (MRSA), bacteria found in normal daily life, can be easily ingested through milk vegetables, and meats, etc. MRSA emerged in many port of the world, increasing complex clinical problems. Therefore, new agents are needed to treat MRSA. Glycyrrhiza uralensis was extracted using 80% MeOH to investigate its antimicrobial activity against MRSA stains KCCM 11812, 40510, and 40512 through bacterial measurement, disc diffusion, and O.D. methods, MIC values, MRSA gene expression investigation, and scanning electron microscope observation. Results revealed MecA, Mecl, MecRI, and FemA were the most highly manifested MRSA genes. Methanolic extract of G. uralensis significantly inhibited MRSA and thus could be used in development of antibacteria.

Anti-inflammatory Effect of Castanopsis cuspidata Extracts in Murine Macrophage RAW 264.7 Cells (Murine Macrophage RAW 264.7 세포에서 구실잣밤나무 추출물의 항염증 효과)

  • This study describes a preliminary evaluation of the anti-inflammatory activity of Castanopsis cuspidata extracts. C. cuspidata was extracted using 80% ethanol and then fractionated sequentially with n-hexane, dichloromethane, ethylacetate, and butanol. To screen for anti-inflammatory agents effectively, we first examined the inhibitory effect of the C. cuspidata extracts on the production of pro-inflammatory factors and cytokines stimulated with lipopolysaccharide. In addition, we examined the inhibitory effect of C. cuspidata extracts on pro-inflammatory mediators (NO, iNOS, COX-2) in murine macrophage RAW 264.7 cells. The amounts of protein levels were determined by immunoblotting. Of the sequential solvent fractions of C. cuspidata, the n-hexane, dichloromethane and ethylacetate fractions inhibited the mRNA expression of pro-inflammatory cytokines (IL-$1{\beta}$ and IL-6), production of NO, and the protein level of iNOS and COX-2. These results suggest that C. cuspidata may have significant effects on inflammatory factors and may be provided as a possible anti-inflammatory therapeutic plant.

A Tablet PC-Based Music-Making Program for Improving Executive Function of Adolescents With Intellectual Disabilities (지적장애 청소년의 집행기능 향상을 위한 태블릿 PC 기반 음악 만들기 활동)

  • This study examined the effects of a tablet PC-based music-making program on the executive function of adolescents with intellectual disabilities. Four adolescents with intellectual disabilities participated in this program. Each participant received 45-minute individual sessions twice a week for a total of 16 sessions. The music-making program was designed in the sequence of planning; learning table PC operations; exploring musical elements; making rhythm, melody, and lyrics; composing loop sections; and presentation of the completed music. The Stroop test, Children's Color Trails Test, and Digit Span and Letter-Number sequencing tests were measured at pretest, midtest, and posttest in order to examine changes in executive function. The participants showed increased scores on all three tests. The participants' attention span also increased and their attempts to correct errors during tasks occurred more frequently at posttest. This study supports the effects of the technology-based program on the executive function of adolescents with intellectual disabilities and presents its expanded applicability for adolescents who show low cognitive function and limited motivation for cognitive engagement.

Antithrombin and Thrombosis Prevention Activity of Buckwheat Seed, Fagopyrum esculentum Moench (메밀 종자의 항트롬빈 활성과 혈전증 예방효과)

  • Direct thrombin inhibitor, which is effective to prevent or cure the thrombosis, has been investigated in worldwide. In this study, we tried to screen antithrombosis agent from edible or medicinal plant. A strong antithrombin activity was identified from methanol or $95\%$ ethanol extract of buckwheat seeds. The solvent fractionation of buckwheat extracts using hexane, ethylacetate, butanol revealed that the butanol fraction has a prominent antithrombin activity. Thrombin time (blood-clot formation time) exceeded to over $2,000\%$ by addition of the butanol fraction at concentration of $312.5{\mu}g/mL$, whereas thrombin time extended to $336\%$ by addition of aspirin at concentration of $1,500{\mu}g/mL$. The butanol fraction showed anthrone-positive and ninhydrine-negative reaction. The active components were heat-liable, acid-unstable non-proteinous macromolecules (>30 KD). In vivo analysis using ICR male mouse showed that the buckwheat extract was superior than the aspirin in pulmonary thrombosis, KCN-induced coma and death. Our results suggest that the buckwheat is a potential as an antithrombosis agent and medicinal food.

Successful Laparoscopic-Assisted Ovariohysterectomy in a Cat with Pyometra (고양이의 자궁축농증에서 복강경을 이용한 난소자궁절제술)

  • Laparoscopic-assisted ovariohysterectomy was performed in a cat (2.7 kg) with pyometra. A 10-mm operative laparoscope was inserted into the abdomen through an umbilical port. A transabdominal suspension suture was placed to maintain the exposure of the ovarian vascular pedicle. The ovarian vascular pedicle and suspensory ligament were progressively cauterized and transected with 5-mm multifunction bipolar grasping forceps. Both ovaries and the uterus were exteriorized via 5-mm caudal port enlarged to 2 cm. The uterine body and associated arteries were ligated, transfixed, and transected under direct vision. No peri- or post-operative complications were encountered. To the authors' knowledge, this is the first report of the use of laparoscopy for the treatment of pyometra in the cat.

An Etiologic Study of Rabbit Dermatitis at Large Rabbit Farms in South Korea (집단 사육 되는 토끼에서 호발하는 피부병에 관한 병인론적 연구)

  • This study was carried out between August and September 2007 to determine the causative agents and epidemiologic features of rabbit dermatitis in Korea. Rabbits were shipped to the laboratory in the College of Veterinary Medicine from 10 rabbit farms. A total of 520 hair, blood, and skin specimens collected from skin lesions of 40 rabbits with suspected dermatopathy were examined mycologically, bacteriologically, and parasitologically. The positive rates of dermatophytosis, bacterial skin dermatitis, and ectoparasite dermatitis were 95, 92.5, and 7.5%, respectively. The etiologic agents of dermatophytosis were identified as Trichophyton mentagrophyte (95%), non-dermatophytic filamentous fungi such as Aspergillus s(5%), and Cryptococcus humilocus (2.5%). With respect to bacteria-related skin dermatitis, Staphylococcus coagulase negative was the most common etiological agent. Staphylococcus aureus was the second most frequent causative agent. Most of the pathogenic isolates were resistant to tetracycline, and aminoglycosides such as amikacin and gentamicin were the most effective drugs against the pathologic bacteria isolated. Ectoparasites were rarely detected in this study. Only Psoroptes cuniculis was detected in 3 (7.5%) out of the 40 tested rabbits. The role of ectoparasites as a causative agent of dermatitis in rabbits in this study was minimal. Our results provide important information related to rabbit dermatitis treatments and researches.

Isolation and Identification of α-Glucosidase Inhibitory Compounds, Hyperoside, and Isoquercetin from Eleutherococcus senticosus Leaves (가시오갈피(Eleutherococcus senticosus) 잎으로부터 α-Glucosidase의 저해 활성 물질, Hyperoside와 Isoquercetin의 분리 및 구조·동정)

  • In the present investigation, the anti-diabetic potential of 80% ethanol extract of Eleutherococcus senticosus leaves (EEES) was examined based on ${\alpha}$-glucosidase inhibitory activities. EEES was sequentially fractionated with n-hexane, chloroform, ethyl acetate (EtAOc), n-butanol, and $H_2O$. Of the various fractions, EtAOc fraction effectively inhibited ${\alpha}$-glucosidase activity by 68.05%. Therefore, EtAOc fraction was selected for further isolation and identification studies. EtAOc fraction was separated by medium pressure liquid chromatography with silica and ODS gel to yield eight fractions (EAA~EAH). Based on the results of ${\alpha}$-glucosidase inhibitory activity, EAH fraction was re-chromatographed to yielded four more fractions (EAHA~EAHD). Of these, EAHC fraction showed higher ${\alpha}$-glucosidase inhibitory activity of 93.60%. EAHC fraction was re-chromatographed and yielded EAHCA and EAHCB fractions. Further, identification and chemical structures of these two fractions were analyzed using $^1H$-NMR, $^{13}C$-NMR, and mass spectra data. Based on the results of the spectral data, the isolated compounds were identified as hyperoside and isoquercetin. Results of the present study indicate that the isolated compounds, hyperoside, and isoquercetin from leaves of E. senticosus could be used for the development of new anti-diabetic drugs.

Induction of Apoptosis by Immature Prunus salicina Lindl. cv. Soldam (피자두(Prunus salicina Lindl. cv. Soldam) 미숙과의 apoptosis 유도 효과)

  • Apoptosis of Prunus salicina Lindl. cv. Soldam, which possesses hematopoiesis, osteoporosis prevention, and antimutagenic effects, at different growth stages was evaluated. Cytotoxic effect of acetone extracts of immature fruits against various tumor cell lines was higher than that of mature fruits, particularly in hormone-independent human breast cancer, MDA-MB-231 cell line. Immature fruit extract increased expression level of pro-apoptotic protein Bax and reduced that of anti-apoptotic protein Bcl-2, and stimulated caspase-3 activity in MDA-MB-231 cells. Results suggest immature fruit of P. salicina Lindl. cv. soldam to be natural source for development of functional food and medical agents to prevent human breast cancer.

Lipolysis Effect of Daucosterol Isolated from Mulberry (Morus alba) Leaves (뽕잎으로부터 순수분리한 daucosterol의 lipolysis 효과)

  • Plants are reservoirs of naturally occurring chemical compounds and of structurally diverse bioactive molecules. The aim of this investigation was to screen for the presence of phytochemicals responsible for the lipolysis activity in mulberry (Morus alba) leaves, which are important in traditional Asian medicinal plants. Powdered mulberry leaves were extracted with hexane, ethyl acetate, and methanol. Daucosterol was isolated from the EtOAc extract of mulberry leaves, and its structure was elucidated by NMR spectral analyses. The NMR assignments for the compound were determined using $^1H$, $^{13}C$, DEPT, COSY, HSQC, and HMBC NMR spectral data. Daucosterol showed a concentration-dependent lipolysis activity that may impart medicinal properties that can be exploited by medical practitioners for the treatment of various diseases. However, further studies should be conducted to elucidate additional mechanisms of daucosterol.

Relationship Between Sleep Quality and Depression Symptoms in Psychiatric Patients: Mediating Effect Interpretation Bias for Ambiguity and Anxiety Symptoms (정신건강의학과 환자의 수면의 질과 우울 증상의 관계: 모호함에 대한 해석 편향과 불안 증상의 매개효과)

  • Objectives : This study was designed to investigate the effect of sleep quality on depression symptoms and the mediating effect of interpretation bias and anxiety symptoms in psychiatric patients. Methods : Data accumulated for outpatients and inpatients in the Department of Mental Health Medicine at Hanyang University Guri Hospital were used. The measurement tools were Pittsburgh Sleep Quality Index (PSQI), Ambiguous/Unambiguous Situations Diary-Extended Version (AUSD-EX), Beck Anxiety Inventory (BAI), Beck Depression Inventory-II (BDI-II). Correlation analysis and bootstrapping analysis were conducted using SPSS 25.0 and SPSS Macro based on 162 patient data. Results : As a result of the study, the double mediating effect of interpretation bias for Ambiguity and anxiety symptoms was significant in the relationship between sleep quality and depression symptoms. Conclusions : In this study, it was confirmed that low sleep quality sequentially affects anxiety and depression symptoms through interpretation bias for ambiguity. Based on this, it is expected that the development of other psychiatric symptoms can be prevented by preferentially performing therapeutic intervention on preceding symptoms.
