Isolation and Characterization of Bacillus Species Possessing Antifungal Activity against Ginseng Root Rot Pathogens (인삼 뿌리썩음병에 길항력이 있는 Bacillus 균의 분리 동정 및 특성 조사)

  • Ginseng (Panax ginseng C. A. Meyer) is an economically important crop in Korea. While the consumption of the crop is gradually increasing, the yield is decreasing due to the injury of continuous cultivation or infection of soil-borne fungal pathogens such as Cylindrocarpon destructans, Fusarium solani, Rhizoctonia solani and Sclerotinia nivalis. In order to find promising biocontrol agents, we have isolated 439 soil bacteria from ginseng cultivated soil and tested their antifungal activities against ginseng rot pathogens. Among them, 3 strains were finally selected and tested for the elucidation of their genetic and biochemical properties. They were identified as Bacillus amyloliquefaciens using phylogenetic analysis based on 16S rRNA gene sequences. Moreover, all selected strains showed positive reaction for PCR detection targeting biosynthetic gene sequences of iturin A and surfactin. The results provided promising evidences that the bacterial strains isolated from ginseng cultivated soil can be novel biocontrol agents for ginseng cultivaion.

Bioequivalence Evaluation of the Tiropramide Formulation by GC/MS (티로프라미드 주사제의 생물학적 동등성 평가를 위한 GC/MS 방법)

  • The bioequivalence study of two tiropramide products was evaluated in 16 health male volunteers following intra-muscular injection. Test product was Tiram$^{(R)}$ injection (S Pharm. Co, Ltd.) and reference product was Tiropa$^{(R)}$ injection(D Pharm. Co., Ltd.). The drug concentration in plasma was determined by GC/MS for over a period of 8 hours after injection. Analysis of variance reveal that there are no differences in AUC (area under the plasma concentration-time curve from time zero to infinity), Cmax (maximum plasma concentration) and Tmax (time to reach Cmax). The differences of mean AUC, Cmax and Tmax between two products were 0.73, -1.385 and -12.994%, respectively. Minimum detectable differences (%) at ${\alpha}=0.05$ were all less than 20% given as a guideline (10.05, 17.90 and 19.01% for AUC, Cmax and Tmax, respectively). From these results, the two formulations of tiropramide are bioequivalent and thus, may be prescribed interchangeably.

Selection and Antagonistic Mechanism of Bacillus thuringiensis BK4 against Fusarium Wilt Disease of Tomato (토마토시들음병의 생물학적 방제를 위한 토착길항세균 Bacillus thuringiensis BK4의 선발과 길항기작)

  • In oder to select an antifungal substance-producing antagonistic bacterium against Fusarium oxysporum casuing fusarium wilt on tomato, strain BK4 was isolated from local soil of Gyeoungbuk and was identified as Bacillus thuringiensis by 16s rDNA analysis, biochemical test, and Mcirolog TM 3.0 System. The antibiotic of B. thuringiensis BK 4 was highly produced at $30^{\circ}C$ in nutrient broth (pH 9.0). The crude antibiotic was even stable at $121^{\circ}C$ and more stable at slight alkalic condition than acid condition. It was also remained $50{\%}$ activity at pH 3.0. B. thuringiensis BK4 showed the inhibition of spore germination and the biocontrol ability against F. oxysporum causing fusarium wilt of tomato in vivo test. According to these results, B. thuringiensis BK4 was enough to use with a microbial agent for biocontrol against fusarium wilt.

  • The purpose of this study was to confirm the antifungal activity, plant growth-promoting activity, and mineral solubilizing activity of 18 types of bacteria isolated purely from rhizosphere soil. The potential of isolates of the genus Bacillus and Pseudomonas as biocontrol agents was confirmed through the antifungal activity of these isolates. This activity has been determined to be due to various hydrolytic enzymes on the cell wall of plant pathogenic fungi and the production of siderophores in isolates. In addition, most of the isolates have been found to have aminocyclopropane-1-carboxylate deaminase production activity, indole-3-acetic acid production activity, and nitrogen fixation activity. These characteristics are believed to have a positive effect on root development, growth, and the productivity of crops via a reduction in the concentration of ethylene under conditions of environmental stress, to which plants are commonly exposed. In addition, on testing for the solubilizing activity of the isolates for phosphoric acid, silicon, calcium carbonate, and zinc, some isolates were found to have mineral solubilizing activities. Inoculation of these isolates during plant growth is expected to assist plant growth by converting nutrients necessary for growth into usable forms that can be absorbed by plants. The 18 isolated strains can be used as biocontrol agents due to their antifungal activity, plant growthpromoting activity, and mineral solubilizing activity.

Acute Toxicities of Emulsifiable Concentrates and Granules of Valeriana fauriei Briquet and Alpinia galangal Swartz Essential Oils against Cyprinus carpio (길초근(Valeriana fauriei Briquet) 및 양강근(Alpinia galangal Swartz) 정유의 유제 및 입제제형의 잉어(Cyprinus carpio)에 대한 급성독성)

  • In this study, two essential oils (EOs) extracted from Aleriana fauriei and Alpinia galangal were formulated as an emulsifiable concentrate (EC) and a granule. In the evaluation of their acute toxicity on fishes, Cyprinus carpio adults were used and the toxicities were determined in a static condition. The formulations were prepared from the essential oil extracted by three different methods namely steam distillation (SD), solvent extraction (SE) and supercritical fluid extraction (SFE). The acute toxicities were calculated using $LC_{50}$ values. Among EOs, only the EO extracted by solvent showed acute toxicities on carps. Some of the EC, EOs of Aleriana fauriei did not exhibit toxicity, while EOs from Alpinia galangal showed potent acute toxicities on carps. Among the granules, granules formulated with Aleriana fauriei EO extracted by SD method and Alpinia galangal EO extracted by SFE method showed acute toxicities on fishes. Nevertheless, $LC_{50}$ of ECs and granules formulated with all types of EOs in this study was higher than the fish toxicity level III for pesticides suggested by Korea Rural Development Administration. Furthermore, cytochrome P450 1A and glutathione S-transferase were confirmed as biomarkers in carps in response to the exposure to Alpinia galangal EO extracted by SD and SFE method, tracking Alpinia galangal EO in the aquatic environment.

  • Volatiles exist ubiquitously in nature. Volatile compounds produced by plants and microorganisms confer inter-kingdom and intra-kingdom communications. Autoinducer signaling molecules from contact-based chemical communication, such as bacterial quorum sensing, are relayed through short distances. By contrast, biogenic volatiles derived from plant-microbe interactions generate long-distance (>20 cm) alarm signals for sensing harmful microorganisms. In this review, we discuss prior work on volatile compound-mediated diagnosis of plant diseases, and the use of volatile packaging and dispensing approaches for the biological control of fungi, bacteria, and viruses. In this regard, recent developments on technologies to analyze and detect microbial volatile compounds are introduced. Furthermore, we survey the chemical encapsulation, slow-release, and bio-nano techniques for volatile formulation and delivery that are expected to overcome limitations in the application of biogenic volatiles to modern agriculture. Collectively, technological advances in volatile compound detection, packaging, and delivery provide great potential for the implementation of ecologically-sound plant disease management strategies. We hope that this review will help farmers and young scientists understand the nature of microbial volatile compounds, and shift paradigms on disease diagnosis and management to aromatic (volatile-based) agriculture.

Sulfasalazine Induces Apoptosis and Cell Cycle Arrest in RAW 264.7 Macrophages (마우스 대식세포에서 설파살라진의 세포사멸 및 세포주기 정체에 미치는 영향 연구)

  • Sulfasalazine is a disease-modifying antirheumatic abiotic agent. It is a derivative of aminosalicylic acid and has been used for the treatment of various inflammatory diseases, such as rheumatoid arthritis, ulcerative colitis, and Crohn's disease, since it was first synthesized in 1941 and approved as a medicine in the United States in 1950. However, its mechanism of action has not yet been clearly identified. In this study, the effects of sulfasalazine on cell survival, apoptosis, and cell cycle progression in macrophages, which are major immune cells that regulate inflammatory responses, were investigated using mouse macrophage RAW 264.7 cells. Sulfasalazine inhibited the viability of RAW 264.7 cells in a dose-dependent manner, starting at a concentration of 0.25 mM. Annexin-V staining was used to confirm that the decrease in cell viability was due to apoptosis, and the number of Annexin-V-positive cells increased significantly at a concentration of 0.25 mM or higher. The effect of sulfasalazine on the expression of key proteins that regulate the G0/G1 phase of the cell cycle was also investigated. Sulfasalazine treatment significantly increased the expression of the cyclin-dependent kinase inhibitors p21 and p27 in RAW 264.7 cells. Although sulfasalazine is frequently used as a control drug in studies on inflammatory diseases, such as inflammatory colitis and rheumatoid arthritis, studies on its effect on macrophages are very limited. Therefore, the results of this study are expected to provide vital information on the use of sulfasalazine as a disease treatment.

Determination of Lethal Concentrations and Lethal Times of Extracts from Tanacetum cineariaiaefolium, Derris elliptica, and Sophora flavescens, to Control Green Peach Aphid, Myzus persicae (복숭아혹진딧물, Myzus persicae, 방제를 위한 제충국, 데리스, 고삼 추출물의 살충농도와 살충시간 결정)

  • Botanical extracts are employed in management of aphids. Extracts from Tanacetum cineariaiaefolium, Derris elliptica, and Sophora flavescens are widely used to control various insects. In this study, we determined concentrations of insecticidal active ingredients in commercial botanical extracts of these plants, and we investigated the time and concentration for lethal results with the green peach aphid, Myzus persicae. The concentrations of active ingredients, pyrethrins from T. cineariaiaefolium, rotenone from D. elliptica, and matrine and oxymatrine from S. flavescens, were determined after their fractionation by liquid chromatography followed by mass analysis and comparison with standard compounds. The extracts were tested for lethality in a bioassay with green peach aphids. Sprays at defined doses were applied to tobacco leaves infested with aphid nymphs. The lethal concentrations (LC50) were 20.4 ppm for pyrethrins, 34.1 ppm for rotenone, and 29.6 ppm for matrine at 48 h after treatments. At 100 ppm application levels, the lethal time LT50 was 13.4 h for pyrethrin, 15.1 h for rotenone, and 14.4 h for matrine. Kaplan-Meier analysis indicated the lethal times for the three botanical extracts at 100 ppm were significantly faster than application of a chemical insecticide, Sulfoxaflor, applied at the recommended level. These results provide baselines to develop and formulate single or mixed preparations containing botanical extracts to control green peach aphids on commercial crops.

  • The present study was performed to develop a functional raw food material from hydrolyzed whey protein powder (23%-GNANA) medication containing sialic acid as a marker compound that is naturally occurring at 7% concentration in GMP (glycomacropeptide). GMP is used worldwide in foodstuffs for babies and infants and is obtained from the milk protein as safe food. While the purpose of our detailed evaluation was aimed to assess preliminary NOAEL values for and above 2,000 mg/kg/day, a clinical dose allowance for 23%-GNANA (as per characteristic of a functional health product, a highly refined test substance of 23% (v/v) sialic acid combined in GMP), at the same time we also wanted to assess the safety of GMP hydrolyzate lacking sialic acid but with identical properties as GMP. Animal safety evaluation was conducted using 23%-GNANA as the test substance, produced from hydrolyzed whey protein powder (product name: HELICOBACTROL-23; provided by Medinutrol Inc. [Korea]; composed of 23% sialic acid and GMP protein) after isolating the sialic acid using enzymes approved as food additives, with GMP as a raw material, and subsequently increasing the content of xx up to 23% through 80% (v/v) ethanol soaking and concentrating, in accordance with GLP Guideline. The animal safety evaluation mentioned above was made on the basis of toxicity in SPF Sprague-Dawley female and male rats dosed with 10 mL of the test substance diluted to 0, 1,250, 2,500, and 5,000 mg/kg directly into their stomachs for 90 d. This was determined in terms of the general symptoms and animal viability, weight and amount of feed intake, eye examination, uracrasia tests, hematological and blood biochemical disorder tests, blood coagulation test, abnormal intestine weight, abnormalities during postmortem and histopathological examinations. Statistical significance was set at P<0.05. Based on the toxicity determination, a certain minor effect associated with the test substance was observed in male rats with no major effects of the tested substance, in comparison with the control group dosed with sterilized water. Nevertheless, the NOAEL value, evaluated as per toxicity criteria, was verified as 5,000 mg/kg/day (P<0.05). Similarly, for female rats, a certain minor effect associated with the test substance was observed in 5,000 mg/kg/day dosed group, with no major effect, yet the NOAEL value (as assessed as per toxicity criteria) was determined to be 5,000 mg/kg/day (P<0.05), which was the same as for male rats. Accordingly, the NOAEL values of the test substances for all female and male rats were finally verified as 5,000 mg/kg/day (P<0.05). In conclusion, it was determined that the 23%-GNANA test substance exceeds 2,000 mg/kg/day, the clinical allowance characteristic for functional health food, and was finally evaluated to cause no safety concerns when used as a raw material in functional health food production, which was the ultimate goal of the present study.

Effect of Tremella fuciformis Berk on Anti-stress activities during Long-Term and Short-Term in Mice (마우스에서 흰목이버섯 (Tremella fuciformis Berk)의 장.단기적 항스트레스 효과)

  • The objective of this study was to evaluate the protective effect of aqueous extracts from Tremella fuciformis Berk(Tf AE) against stress during long-term and short-term in ICR mice. All the animals were randomly divided into two groups which had been bred for 5 months that were treated by immobilization stress for 8 weeks (total 7 months breeding, equivalent to human beings aged 20) with or without Tf AE, and one out of two groups was continuously bred until they become 18 months old (equivalent to human beings aged 60) without Tf AE. Afterwards, the changes of serum and hepatic metabolites were investigated on the basis of the index of stress-related in vivo oxidative damage. As a result, it was found that stress increases serum triglyceride (TG) and aspartate aminotransferase (AST) and decreases serum HDL-cholesterol in the long-term (total 18 months breeding) and short-term (total 7 months breeding). In addition, stress concerned the decrease of total antioxidant status (TAS) and superoxide dismutase (SOD) as well as the increase of malondialdehyde (MDA) in liver. On the other hand, Tf AE-fed groups reversed all these biochemical indices. These results suggest that stress in one's youth causes negative results in TG, HDL-cholesterol, LDL-cholesterol, AST, TAS, SOD and MDA measured in one's senescent. The administration of Tf AE in the stressed mice decreases serum TG and AST that are increased by stress, and exerts influence on the increase of serum HDL-cholesterol. Also Tf AE recovered the values of liver TAS, SOD and MDA in the stressed mice. In conclusion, Tf AE represented protective effect in the stressed mice to some degree.
