Isolation of Bacillus amyloliquefaciens H41 Producing Growth Inhibition Factor against Vibrio anguillarum (어병균 Vibrio anguillarum 생육 저해 인자를 생산하는 Bacillus amyloliquefaciens H41의 분리)

  • To investigate the possible use of probiont in fish farming industry, a bacterium with inhibitory effect against Vibrio anguillarum was isolated from gastrointestinal tract of the marine fish of yellow tail. It was identified to be Bacillus amyloliquefaciens H41 based on biochemical and physiological characterization. The optimal growth conditions of the isolated strain were 1% peptone, 1.5% yeast extract, 1% sucrose, 0.5% NaCl, 0.05% $MgSO_4{\cdot}7H_20$, pH 7.0-8.0, and 20 hr of incubation between $28-35^{\circ}C$ under aeration. The culture supernatant of the isolated strain showed inhibition activity against V. anguillarum. Inhibition activity was cleared by forming a clear zone by a paper-disk method. The maximal production of growth inhibition factor was induced by cultivation under 1% peptone, 1.5% yeast extract, 1% sucrose, 1% NaCl, 0.05% $MgSO_4{\cdot}7H_20$, pH 7.5 and at $35^{\circ}C$. The highest growth inhibition factor production was observed after 16-24 hr cultivation under aeration. The culture supernatant of the isolated strain showed inhibition activity whereas no inhibition activity was shown from the standard B. amyloliquefaciens KCTC 1724 strain. The growth inhibition affected only against V. anguillarum among other pathogenic Vibrios tested here.

Isolation and Characterization of Six Microorganisms from the Digestive Tract of the Cricket Gryllus bimaculatus (쌍별귀뚜라미(Gryllus bimaculatus) 소화기관에서 분리한 6종류의 특성규명)

  • We describe the isolation and characterization of six different intestinal microorganisms from the digestive tract of the cricket Gryllus bimaculatus. Based on 16S rRNA gene sequences, we obtained six isolates belonging to four different genera: Staphylococcus, Bacillus, Citrobacter, and Proteus. All the isolates were resistant to ampicillin. Ampicillin is an irreversible inhibitor of the enzymeetranspeptidase, which is needed to make bacterial cell walls. None of the isolates were resistant to kanamycin, which binds to the 30S subunit of the bacterial ribosome and then inhibits total protein synthesis. Gram staining was conducted, in addition to morphological classification under a microscope. Four grampositive isolates and two gram-negative isolates were detected. The gram-positive isolates were GL1 (round shaped, 2 am in diameter), GL2 (rod shaped, $2.5{\mu}m$ in length), GL3 (rod shaped, $2{\mu}m$ in length), and GL4 (round shaped, $1.5{\mu}m$ in diameter). The gram-negative isolates were GL5 (rod shaped, $2{\mu}m$ in length) and GL6 (rod-shaped, $2.5{\mu}m$ in length). Notably, two of the isolates, GL2 and GL4, secreted specific extracellular proteins. These were determined by MALDI-TOF-MS spectral analysis to be a 87 kDa collagenase, 56 kDa hypothetical protein, and 200 kDa hypothetical protein. The six isolates in this study could be used for various biotechnological applications and pest management, both in the field and in greenhouse systems. In addition, it would be interesting to determine the relationship between these isolates and their host.

Soil Microbial Community Analysis in Large Patch (Rhizoctonia solani AG2-2 IV) (갈색퍼짐병 발병토양의 미생물 군집 분석)

  • Large patch, caused by Rhizoctonia solani AG2-2 IV, is a soil-born disease that is the most important of warm season turfgrass such as zoysia and Bermuda grass. This study was conducted to analysis of the soil microbial community structure on large patch. Center of the large patch (CLC), edge (CLE) and healthy (CLH) part of microbial communities were examined using metagenomics in Phylum level. Distribution trends of the rhizosphere microorganisms were similar to the order Proteobacteria, Acidobacteria, Chloroflexi, Firmicutes, Planctomycetes, Gemmatimonadetes, Nitrospira, Cyanobactria and Verrucomicrobia in soil collections. Contrastively Actinobacteria was more 56% abundant in healthy part soil (16%) than in the center (9.28%) or edge (10.84%) parts. Taxonomic distributions were compared among the CLC, CLE and CLH, total 6,948 OTUs were detected in the CLC, 6,505 OTUs for the CLE and 5,537 OTUs were detected in the CLE. Distributions of Actinobacteria OTUs were appeared 615 OTUs in the CLC, 709 OTUs in the CLE and 891 OTUs in the CLH. Among Actinobacteria, 382 OTUs were overlapped in the all soils. Not matched OTUs of CLH (286 OTUs) was detected 23 times higher than CLC (91 OTUs) and CLE (126 OTUs).

Acaricidal Efficacy of Herbal Extracts against Tetranychus urticae (Acarina: Tetranychidae) (점박이응애, Tetranychus urticae (Acarina: Tetranychidae)에 대한 약용식물 추출물의 살비 효과)

  • Methanol and hexane extracts from 35 species in 27 families of herbal plants were evaluated for their acaricidal activities against two-spotted spider mite, Tetranychus urticae by leaf-dipping and spraying methods in laboratory, pot and field, respectively. Acaricidal activities were different depending on herbal plants. When T. urticae was released on leaves of Phaseolus vulgaris var. humilis that were dipped in the 1,000 ppm hexane extracts from fruits of Torreya nucifer and Daphan genkwa, seeds of Xanthium strumarium and Pharbitis nil at one minute they experienced 56.8, 47.8, 47.7, and 47.7% mortalities, respectively. Dipping time influenced acaricidal activity, that is, one minute dipping was more effective than 30 second dipping of Phaseolus vulgaris var. humilis leaves. However, extracts of X. strumarium and P. nil seeds had comparatively high activities. Concentration of hexane extract of X. strumarium and P. nil seeds revealing LC50 were 1,824 and 1,899 ppm, respectively. Extracts of both plants from hot and cold water were not effective representing <20% mortality at 1,000 ppm. However, hexane extracts of X. strumarium and P. nil seeds were effective against T. urticae on P. vulgaris var. humilis representing 76.3 and 71.3% mortalities in pot, respectively. Control effects of hexane extract of P. nil seed were 50.8 and 35.1% at 2,000 and 1,000 ppm against T. urticae on Chrysanthemum morifolium in greenhouse, respectively.

Limitation in Attraction Efficacy of Aggregation Pheromone or Plant Volatile Lures to Attract the Western Flower Thrips, Frankliniella occidentalis Infesting the Hot Pepper, Capsicum annuum, in Greenhouses (시설 고추재배지에서 꽃노랑총채벌레 집합페로몬과 식물 휘발성 유인제 효능의 한계성)

  • Mass trapping of the western flower thrips, Frankliniella occidentalis, has been considered as an option to control this pest. This study applied the commercial lures to the hot pepper-cultivating greenhouses and assessed the enhancement of the attracting efficiency by adding to sticky traps. There was no color difference in the attracting efficiency between blue and yellow sticky traps. However, the installation position of the traps was crucial in the greenhouses. The more thrips were captured within host cropping area than outside areas of the crop. In vertical trap position, it was the most optimal to install the traps at the crop crown. Using these installation parameters, the yellow sticky traps captured approximately 1% population of the thrips. To enhance the trapping efficiency, the commercial lures containing aggregation pheromone or 4-methoxybenzaldehyde were added to the yellow sticky traps. However, these commercial lures did not significantly enhance the trapping efficiency compared to the yellow sticky trap alone. In contrast, Y-tube olfactometry assays confirmed the high efficiency of the aggregation pheromone or another plant volatile (methyl isonicotinate) to attract the thrips. Interestingly, these lure components had lower attracting efficiencies compared to the hot pepper flowers. The high attractive efficiency of the flowers was supported by the observation that the commercial lure was effective to enhance the trapping efficiency of the yellow sticky trap against F. occidentalis in Welsh onion (Allium fistulosum) field without any flowers. This study indicates the limitation of the commercial lures in application to hot pepper fields for the mass trapping of F. occidentalis. It also suggests active volatile component(s) from hot pepper flowers to attract F. occidentalis.

Characteristics of agronomy to vitamin A strengthening rice at large scale GMO field (대규모 GMO 포장에서 비타민 A 강화 벼의 농업특성 검정)

  • This study was carried out to develop of macro-protocol and the biosafety guide Golden Rice, modified vitamin A at large scale GMO field. Commercialization of genetically modified (GM) plants will be required the assessment of risks associated with the release of GM plants that should include a detailed risk assessment of their impacts in the environment and human health. Prior to GM plant release, applicants should provide the information on GM crops for approval. A total of $4,700m^2$ GM field, each of Golden Rice and 'Nakdong' were used for further analysis on agronomic traits and pathogenesis. Generally plant growth and panicle number in the transgenic lines were lower than in the wild type under field condition. The major agronomic traits such as plant growth, grain quality in Golden Rice were similar than in 'Nakdong'. The yield difference was due to reduced ripened rate and the 1,000 grain weight. The grain shape was similar donor plant, however, color of brown rice was different from segregation. Also the difference of pathogenesis reaction was not significant in the leaf blast and sheath blight between Golden Rice and 'Nakdong'. These results provided the agronomic data and pathogenesis for risk assessment analysis of Golden Rice and suggested that the macro-protocol could be useful to detect GM plants.

Isolation and Characterization of Indole-3-acetic acid- and 1-aminocylopropane-1-carboxylyic Acid Deaminase-producing Bacteria Related to Environmental Stress (환경스트레스와 관련된 indole-3-acetic acid 및 1-aminocylopropane-1-carboxylyic acid deaminase 활성을 갖는 박테리아의 분리와 특성 연구)

  • In this study, strains isolated from soil samples collected from Busan, Changwon, and Jeju Island were examined to verify their abilities of phosphate solubilization and nitrogen fixation, production of indole-3-acetic acid (IAA), siderophore, and 1-aminocylopropane-1-carboxylyic acid (ACC) deaminase in order to select strains that promote plant growth and play a role in biocontrol of pests or pathogens. According to the results of this study, most of the isolated strains were found to have ability of phosphate solubilization, nitrogen fixation, IAA production, siderophore production, and production of ACC deaminase. These isolated strains might help plant growth by directly improving absorption of nutrients essential for phosphate solubilization and nitrogen fixation. In addition, they can promote plant growth and control resistance to plant diseases through extracellular enzyme activity and antifungal activity. In addition, most of the selected strains were found to survive in various environmental conditions such as temperature, salinity, and pH. Therefore, Pseudomonas plecoglossicida ANG14, Pseudarthrobacter equi ANG28, Beijerinckia fluminensis ANG34, and Acinetobacter calcoaceticus ANG35 were finally selected through a comparative advantage analysis to suggest their potential as novel biological agents. Further studies are necessary in order to prove their efficacy as novel biological agents through formulation and optimization of effective microorganisms, their preservation period, and crop cultivation tests.

Status of Fusarium Wilt Incidence on Summer Radish and Etiological Characteristics of the Causal Fungus in Korea (고랭지 여름 무에서 시들음병 발생 현황과 병원균의 병원학적 특성 연구)

  • Incidence of Fusarium wilt was surveyed in fields of summer radish in Gangwon province in Korea in 2018 and 2019. The disease started in early July and spread rapidly in hot summer of late July and August and in severe case, reached up to 80% in a field in Gangneung area. Symptoms in the seedling stage include poor growth and browning of internal tissue of root. During mid-growth, the leaves of diseased plant turned yellow over time, the surface of the roots changed from white to blackish, and the vascular tissues turned brown. A total of 23 isolates was obtained from the diseased plants and identified as Fusarium oxysporum f. sp. raphani by elongation factor-1α and intergenic spacer sequence analysis. Pathogenicity of the isolates was tested by artificial inoculation to the radish and other plants. All the isolates tested were pathogenic to radish plant, although there were differences in virulence on radish 11 cultivars. However, the isolates were not virulent to other plants except some cruciferous vegetables including Brussels sprouts, rocket, stock, and turnip. The results of pathogenicity test showed that it is necessary to rotate with crops other than cruciferous vegetables in order to prevent Fusarium wilt from radish fields.

Comparative Responses of Sugar Beet Cyst Nematode, Heterodera schachtii and Clover Cyst Nematode, H. trifolii to Different Nematicides (클로버씨스트선충과 사탕무씨스트선충 알과 유충에 대한 약제 반응 비교)

  • This study aimed to investigate the differences in the effects of nematicidal materials (Burkholderia rinojensis, fluazaindolizine, fluopyram, fosthiazate, hydrogen peroxide, and imicyafos) on eggs and larvae of the sugar beet cyst nematode, Heterodera schachtii and clover cyst nematode, H. trifolii, which have recently become problematic in highland Chinese cabbage cultivation areas in Korea. Comparison of the hatching rates according to the test materials showed differences between treatments. In the treatment with fluopyram, fosthiazate, and imicyafos, a very high hatching inhibitory effect of 99% was observed in both nematodes, and there was no difference in the efficacy of treatments between nematode species except for that with hydrogen peroxide. Regarding the nematocidal activity of treatments against hatched larvae, only the fluazaindolizine treatment, despite showing a high hatching inhibitory effect, showed a low nematicidal effect of less than 30% against both nematodes, whereas a mortality rate of more than 90% was observed in the other treatments. There was no difference between nematode species in the reactions of the 2nd instar juveniles to all nematicidal test materials after 24 and 48 h of treatment. Although there are limitations to the laboratory experiment, the reaction to the nematicidal materials of the two cyst nematodes that damage the same host was similar, and simultaneous control is suggested in the areas where the two nematodes occur together.

A Study on the Flora and its Introduced Disturbing Plants in Damyang Area of Mudeungsan National Park, Korea (무등산국립공원 담양 지구의 식물상과 생태계교란 식물에 관한 연구)

  • This study was investigated to find out the distribution of flora and remarkable plants and factors and management measures for influx of wild disturbed plants in the Damyang area of Mudeungsan National Park. The field survey was carried out 8 times from May 2019 to October 2020. First of all, in the Damyang area of Mudeungsan National Park, a total of 603 taxa in 107 families, 349 genera, 531 species, 57 varieties, 5 subspecies and 10 forms. This result was found to be about 12.33% of the total 4,881 taxa of vascular plants in Korea. In addition, Rare plants were classified as 15 taxa. Floristic special and Korean endemic plants were identified as 85 taxa and 13 taxa, respectively. Lastly, the naturalized plants that appeared in the Damyang area of Mudeungsan National Park were observed in 45 taxa with 45 species of 16 families and 36 genera. Moreover the naturalization rate was 7.46% and the urbanization index was calculated to be 14.01%. The wild disturbed plants were found in a total of 4 taxa. Especially, a sheep sorrel(Rumex acetosella) was focused on managing for physica and biological control at Sinseondae Eoksaepyungjeon.
