Menstrual cycle 1) In disorder of menstrual cycle, persons who have Bate menstruation are more than persons who have premature menstruation. 2) Persons who have normal menstrual cycle are in 124 persons(28.51%), the others who have severe premature menstruation or late menstruation more than one time for 4-7 months are in 311 Persons.(71.49%) 2. Menstrual duration and amount 1) Persons who have normal menstrual amount we in 66-89%, hypermenorrhea is in 1-11%, hypomenorrhea is in 5-21%. 2) In the study of menstrual duration, persons more than one thirds are irregular in thier menstrual amount every menstruation. 3) In the study of MMQ, persons who are irregular in thier menstrual amount every menstruation are in 125persons.(29.76%) 3. Menstrual pain 1) Persons who have slight menstrual pain are in 289 persons(65.98%), the middle is in 86 persons(19.63%) the severe is in 34 persons(7.76%) by MMP. 2) Persons who are irregular in thier menstrual pain every menstruation are in 145 persons.(33.11%) Conclusion : In menstrual cycle, there are more persons who have irregular menstrual cycle than normal.(71.49%) In menstrual duration and amount, more persons have normal menstrual amount.(66-89%) In menstrual pain, persons who have slight menstrual pain are the most.(65.98%)
Purpose: Examine changes in the aspect and state of patient with infertility who admitted to Korean oriental hospital. Look at the changes in last 8 years and seek how oriental medicine can approach to infertility patients who admitted to Korean oriental hospital in last 8 years. Methods: Total of 7434 new patients with infertility who admitted to Gynecology of Oriental Hospital, $\bigcirc\bigcirc$ University Medical Center, from October 2002 to December 2009 was compared to infertility patients based on bottom collected data in this study, total of 1061 infertility patients were targeted for this study. Results: 1.There is no distinct increases in age between women who admitted to oriental medicine and patient's average age, it has shown constantly increasing trend in their age though. However, the average age of infertility patients have shown significant increasing range than average age of all infertility patients. 2. After analysis the result of infertility patient's rate of overall patients, the rate of infertility patients are remaining the same or slowing down in 2005, as the starting year(Table 2). The significant relationship was found in diagnosis and treatment of gynecology(Table 4~5) for infertility patients. In 2005 as the starting year, as patients who admitted to oriental hospital after diagnosed or treated at gynecology are increasing, the rate of infertility patients of overall patients admitted to hospital is slowing down, which is decreasing. 3. There is increasing number of patient diagnosed with "unknown cause" who admitted to Gynecology of Oriental Hospital after year of 2005. 4. The increasing number of patient diagnosed with "unknown cause" who admitted to oriental hospital has a tendency to be important factor in infertility treatment of oriental medicine. Conclusion: Unknown causes of infertility patients who admitted to oriental hospital will continuously increase as senses of society, economic, and social climate change.
Purpose: These studies were undertaken to evaluate the anti-oxidative and neuroprotective effects of Gamisoyo-san(GMSYS). Materials and Methods: We studied the antioxidant effects of GMSYS by assessing the DPPH free radical and the ABTS radical cation inhibition activities, the total polyphenolic contents(TPC). To evaluate the effects of GMSYS in the human neuroblastoma cells, we measured the cell viabilities in SH-SY5Y cells treated with GMSYS. Then we observed the protective effects of GMSYS against 6-OHDA induced neurotoxicity in SH-SY5Y cells. To confirm the neuroprotective effects of GMSYS in the primary culture of mesencephalic dopaminergic cells, we counted the TH-immunopositive cells and measured the NO and TNF-$\alpha$ after the treatment of GMSYS and 6-OHDA. Results: The DPPH free radical and the ABTS radical cation inhibition activities were increased in a dose dependent manner and the IC50 were $133.60{\mu}g/m{\ell}$ and $106.20{\mu}g/m{\ell}$, respectively. The TPC was 0.78%. There were no differences between the various concentrations of GMSYS and the control in the cell viability of SH-SY5Y cells. The neuroprotective effects of GMSYS were shown in the co-treatment group at the low concentrations of $25{\mu}g/m{\ell}$ and the post-treatment group at all concentrations. After the treatment of GMSYS and 6-OHDA in the primary culture of dopaminergic cells, the TH-immunopositive cells were significantly increased in $0.2{\mu}g/m{\ell}$ of GMSYS than the 6-OHDA group. The NO and TNF-$\alpha$ were significantly decreased in $0.2{\mu}g/m{\ell}$ of GMSYS than the 6-OHDA group. Conclusions: This study shows that GMSYS has the antioxidant and neuroprotective effects, especially in the mesencephalic dopaminergic cells. We suggest that GMSYS could be useful for the treatment of postmenopausal depression related with the degeneration of dopamine neuron.
Purpose: The purpose of this study is to identify the real condition of urinary incontinence(UI), interstitial cystitis(IC), atrophic viginitis(AV) for elderly women and analyze the cognition of traditional korean medicine(TKM) for them. Methods: We utilized questionnaire from May to June, 2010. Questionnaires were taken from 125 women using senior welfare center and nursing home, aged over 65 up to 92. The data were analyzed by $X^2$-test using SPSS/PC ver 18.0 program. Results: The prevalences of UI, IC and AV symptoms were 50.4%, 40.8%, 56%, respectively. The average I-QoL score for UI was $82.62{\pm}21.16$, and the average ICSI score for IC was $8.16{\pm}2.50$. After adjustment for each of the variables considered in this study, alcohol was associated with UI and age, BMI(body mass index) were associated with IC. Most of respondents have no experience(94.4%) or don't know (79.2%) about TKM for UI, IC and AV. 44 women(37.3%) indicated that they weren't willing to use TKM for UI, IC and AV. Reasons for not taking TKM were because of 'no knowledge of TKM(34.1%)' and 'more accustomed to western treatment(34.1%)'. Conclusion: Although the prevalence of UI, IC, AV for elderly women was high, the actual percentage of treatment for these diseases was low, furthermore, patients were not aware of TKM and had very few experiences of TKM for these diseases. The development and increased promotion about TKM program for elderly women's urogenital diseases is needed.
Purpose: To provide the information of efficacy for herbal formulas of high frequency, it was evaluated the anti-inflammatory effect. In many studies, plantderived anti-inflammatory efficacies have been investigated for their potential inhibitory effects on lipopolysaccharide (LPS)-stimulated macrophages. This study was performed to examine the anti-inflammatory effects of herbal formulas of high frequency on LPS-stimulated RAW 264.7 cells. Methods: Anti-inflammatory activity was investigated in 25 herbal formula extracts in vitro and in vivo. To investigate the anti-inflammatory effect in vitro model, using LPS-stimulated macrophages, RAW 264.7 cell line. The productions of nitric oxide(NO), prostaglandin(PG)$E_2$, interleukin(IL)-6 and tumor necrosis factor(TNF)-$\alpha$ were examined in RAW 264.7 cells, in the presence of the herbal formulas. RAW 264.7 cells were incubated with LPS $1\;{\mu}g/mL$ and herbal formulas for 18 hours. As an in vivo, using a rat model of carrageenin-induced paw edema. The paw volume was measured at 2 and 4 hours following carrageenin-induced paw edema in rats. Results: 8 kinds of herbal formula inhibited NO production by LPS-stimulated in some concentration, but the effect of NO inhibition is weak. 12 kinds of herbal formula inhibited $PGE_2$ production by LPS-stimulated over the 30%. Among them Gumiganghwal-tang, Sagunja-tang, Samchulkunbi-tang, Insampaedok-san and Hwangryunhaedok-tang inhibited IL-6 production by LPS-stimulated but TNF-$\alpha$ was not inhibited. 12 kinds of herbal formula reduced the carrageenin-induced paw edema in rats. Particularly, 3 kinds of herbal formula(Gumiganghwal-tang, Ssanghwa-tang and Soshiho-tang) were better than indomethacin. Conclusion: These results suggest that Gumiganghwal-tang, Sangunja-tang, Samchulkunbi-tang, Insampaedok-san and Hwangryunhaedok-tang have antiinflammatory activity.
Purpose: This study was designed to find out the anti-cancer effects of Samultang-Gami which was composed of Rehmanniae Radix(RR), Angelicae Gigantis Radix(AGR), Cnidii Rhizoma(CR), Paeoniae Radix(PR), Cortex Moutan Radicis(CMR), Hedyotis Diffusa(HD) and Caesalpinia Sappan on HeLa, HepG2 and AGS cells. Methods: Various cancer cell lines including HeLa, HepG2 and AGS cells, were used. In vitro anti-cancer effects were measured by MTT assay using cancer cell lines treated with various concentrations of 70% ethanol extract of Samultang-Gami. Expression of cell cycle arrest mediators including Bax, Bcl-2, p53 and DARP-1 proteins were measured by Western blot analysis. Results: 1. Samultang-Gami decreased the viability of HeLa and HepG cells in a dosedependent manner. 2. AGR, CMR, PR and HD decreased the viability of HeLa, HepG2 and AGS cells. 3. We could observe that the decreased Bax and Bcl-2 expression level and the increased PARP-1 expression level by Samultang-Gami extracts treated in HeLa cells. 4. We could observe that the decreased Bcl-2 expression level and the increased Bax, p53 and PARP-1 expression level by RR extracts treated in HeLa cells. and also could observe that the reduction of the protein level of Bcl-2, p53 and PARP-1 and the increase of the protein level of Bax by PR in HeLa cells. 5. We could observe that the increased p53 expression level, the decreased PARP-1's that and the unchanged Bax and Bcl-2's that by Samultang-Gami extracts treated in HepG2 cells. 6. We could observe that the reduced Bcl-2 expression level by each of RR extracts and PR extracts in HepG2 cells. 7. The treatment of Samultang-Gami in AGS cells didn't have any effect on the expression level of Bax, Bcl-2, p53 and PARP-1. 8. We could observe that the increased p53 and PARP-1 expression level by each of CR, RR and PR extracts in AGS cells. Conclusion: Taken together, we suggest that Samultang-Gami exhibits cytotoxic effects on HeLa, HepG2 and AGS cells, causing apoptosis. The results showed that Samultang-Gami may do so by regulating the expression of specific target molecules that promote efficient apoptotic cell death in a dose-dependent manner.
Purpose : This study was designed to investigate the effects of Jeongkyeong-Tang(JKT) on the polycystic ovary(PCO) induced by estradiol valerate(EV) in rats. Methods : PCO was induced by single intramuscular injection with EV(4mg) in female rats. Normal group(n=8) were injected with sesame oil and orally administrated distilled water for sixty days. PCO control group(n=8) were injected with EV and orally administrated distilled water for sixty days. JKT treated group(n=8) were injected with EV and orally administrated JKT for sixty days. Then we measured weights of body, ovaries and adrenal glands, and measured content of serum estrogen. The histomorphometrical changes of ovaries were also evaluated. The expressions of nerve growth factor(NGF) were analyzed in the central nervous system, adrenal glands and ovaries by immunohistochemistry. Results : - The weights(mg) of ovaries in JKT treated group (69.7${\pm}$6.7) were significantly increased( p<0.001) compared with PCO control group(46.7${\pm}$12.2). - The numbers of secondary follicles in JKT treated group(4.00${\pm}$l.31) were significantly increased(p <0.05) compared with PCO control group(2.25${\pm}$1.39). - The numbers of mature follicles in JKT treated group(5.50${\pm}$1.51) were significantly increased(p<0.01) compared with PCO control group(2.88${\pm}$1.13). - The numbers of atretic follicles in JKT treated group(2.75${\pm}$l.16) were significantly decreased(p<0.001) compared with PCO control group(6.88${\pm}$2.03). - The numbers of corpora lutea in JKT treated group(4.13${\pm}$1.46) were significantly increased(p<0.01) compared with PCO control group(2.13${\pm}$1.46). - The contents(pg/ml) of serum estrogen in JKT treated group(115.18${\pm}$18.29) were significantly decreased(P<0.01) compared with PCO control group(153.06${\pm}$29.47). - The expressions of NGF-immunoreactive cells in the ovarian granulosa cells in JKT treated group were lesser observed than PCO control group. Conclusion : From the above results, we concluded that Jeongkyeong-Tang has inhibitory effect on the development of EV-induced polycystic ovary. And it's effect may be related with decreased NGF activities in the ovary.
Purpose: This study was undertaken to evaluate the effects of the different administration duration of Cynomorii Herba extract solution on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis and the activities of sperm hyaluronidase. Methods : We used the 8-week-old ICR mice and administered 0.3mg/g extract solution of Cynomorii Herba once a day for 30, 60, 90 and 120 days. The control group was administered the normal saline in the same way and duration. We examined the number of total, motile and normal sperm from the cauda epididymis. And we compared the testicular tissue especially seminiferous tubules between control and treated groups by histochemical methods. At the end we observed the difference of sperm hyaluronidase activities between control and treated groups. Results : The significant differences were observed in the concentration of total sperm, the motility and normality of spermatozoa of the Cynomorii Herba extract solution administered groups compared to the control group in 60 and 90 days groups. In the histological analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the Cynomorii Herba extract solution administered groups compared to the control group, respectively. Also, the activity of hyaluronidase was significantly increased in the Cynomorii Herba extract solution administered groups compared to the control group. Conclusion : This study shows that Cynomorii Herba has the beneficial effect on the concentration, morphology and motility of sperm especially in 60 and 90 days administration group. We can suggest that Cynomorii Herba extract solution be useful for the treatment of male sexual dysfunctions and infertility.

Purpose: This study was performed to investigate the effects of Keukhachukeo-Tang (KCT) on the development of experimentally-induced endometriosis in rats. Methods : Endometriosis was induced in rats by autotransplanting uterine tissue to the peritoneum and devided them into three groups: (1) sham-operated group (n=8), (2) surgically induced endometriosis and untreated control group (n =8), (3) surgically induced endometriosis and KCT treated group. KCT (1,200 mg/head) was orally administrated for 15 days after operation. Then we measured the body weight, the volumes of endometriotic implants. The weight (body, left uterus and ovaries) and concentrations of cytokines (MCP-1, TNF-${\alpha}$, IL-l${\beta}$) in serum and peritoneal fluid were also measured. Histopathology, immunohistochemistry for COX-2, and histochemistry for mast cells in transplanted uterine tissue were performed. Results : - The volumes(mm$^3$) of endometriotic implants in KCT-treated group (107${\pm}$66) were significantly decreased (p<0.05) compared with control group (405${\pm}$318). - The contents(pg/ml) of MCP-1 in peritoneal fluid in KCT-treated group (6,940${\pm}$893) were significantly decreased (p<0.01) compared with control group (8,632${\pm}$1,245). - The contents(pg/ml) of TNF-${\alpha}$ in peritoneal fluid in KCT-treated group (847${\pm}$330) were significantly decreased (p<0.05) compared with control group (1,245${\pm}$362). - The percentages(%) of positive epithelial layers for COX-2 in KCT-treated group (31${\pm}$10) were significantly decreased (p<0.01) compared with control group (50${\pm}$8). - The numbers of mast cells in adjacent tissue of transplanted uterine tissue in KCT-treated group (69${\pm}$18) were significantly decreased (p<0.01) compared with control group (109${\pm}$30). - The numbers of mast cells in stroma of transplanted uterine tissue in KCT-treated group(16${\pm}$5) were significantly decreased (p<0.01) compared with control group (26${\pm}$8). - Histopathologically, proliferation of endometriotic epithelia and stroma, and infiltration of inflammatory cells in transplanted uterine tissue of KCT-treated group were weakly observed than those of control group. Conclusion : From the above results, Keukhachukeo-Tang (KCT) have inhibiting effects on the development of transplanted uterine tissue. And these effects are related to the decreased concentration of MCP-1 and TNF-${\alpha}$, decreased expression of COX-2, and decreased infiltration of mast cells by administration of Keukhachukeo-Tang.
Objectives: The purpose of this study is to investigate the effect of Melandrii Herba (MH), Akebia Quinata Decaisne (AQ), and Tetrapanax Papyriferus (TP) on milk secretion and aquaporin (AQP) expression in lactating mice. Methods: For the experiment, the mice were divided into three groups, which were orally administered MH (2,720 mg/kg), TP (400 mg/kg) and AQ (2,800 mg/kg) extracts respectively for 3 weeks from Day 1 after the birth, compared with the control group (C group), which was administered distilled water. A group consisted of six infantile mice per postpartum mouse. For comparison with the C group, non-pregnant SKH-1 mice were used as the virgin group. Results: 1. When it comes to the immunohistochemical staining for prolactin receptors in the mammary glands, the AQ and MH groups showed a strong immune response to the secretory epithelial cells constituting the mammary alveoli, while the TP group represented a weaker immune response. 2. In the immunohistochemical staining for AQP in the mammary glands, AQP1 showed a strong immune response in the walls of capillaries and venules around the mammary alveoli, and AQP3 in the epithelial cells constituting the mammary alveoli, and AQP5 in some tissues between the mammary alveoli. AQP1 was expressed in the order of TP group>AQ group=C group>MH group, and AQP3 was MH group and AQ group>TP group=C group, and AQP5 was MH group>C group>AQ group and TP group. 3. In the Western blot, AQP1 was expressed in the order of TP group>AQ group>C group>MH group, and AQP3 was MH group>AQ group>C group>TP roup, and AQP5 was MH group>TP Group>C group>AQ group. All of AQP1, 3, 5 expression were significantly higher in the C group than in the Virgin group. Conclusions: The administration of Akebia Quinata Decaisne, Tetrapanax Papyriferus and Melandrii Herba have the effect of improving prolactin levels in postpartum mice and increasing the expression of prolactin receptor and AQPs in the mammary glands, suggesting that lactation might be enhanced by the development of the mammary glands.
