  • Rust disease was found on broad bean (Vuia faba) in several farmer's fields located in Changseon-myon, Nam-hae-gun, Gyeongnam province in Korea. The infection rate of the disease in the surveyed area was 84.3% in 2002. The typical symptoms of the disease appeared first as small, white spots on leaves, stems and pods. Spots, then turn brownish, their edges rised slightly, and powdery mass of yellowish brown spores revealed on them. Severely infected leaves were eventually defoliated. Uredospores were light brown in color, ellipsoid or ovoid in shape and 24~38$\times$21~26 ${\mu}{\textrm}{m}$ in size. Teliospores were dark brown in color, ellipsoid or ovoid in shape and 22~42$\times$17~27 ${\mu}{\textrm}{m}$ in size. Pedicels were sienna to luteous in color and 52~116 ${\mu}{\textrm}{m}$ in size. The causal fungus was identified as Uromyces viciae-fabae, based on morphological characteristics. This is the first report on the rust of broad bean caused by Uromyces viciae-faae in Korea.

  • In June 2011, walnuts (Juglans regia) from orchards in Kimcheon and Muan, in southern Korea, were found to exhibit downy mildew-like symptoms of a foliar disease. Whitish polygonal efflorescence was produced on the abaxial surface of affected leaves and discolored light green blotches on the corresponding adaxial surfaces. In the later stage of disease development, diseased tissues collapsed and became necrotic. Based on morphological and cultural characteristics, the causal fungus was identified as Microstroma juglandis. The sequence of ITS rDNA of the present isolate showed 100% similarity with those of M. juglandis obtained from GenBank databases, thus confirming its identity. Pathogenicity tests were conducted on leaves of walnut seedlings, fulfilling Koch's postulates. The disease has been previously reported in North America, Europe, Oceania and some western Asia. This is the first report of downy leaf spot on walnuts in East Asia.

  • Bacterial canker, caused by Clavibacter michiganensis subsp. michiganensis, is a very damaging disease to tomato (Lycopersicon esculentum) farm in Korea. It infects tomato, spreads through the xylem and causes bacterial wilt and canker. Selection of resistant cultivar is the best way to prevent or reduce the occurrence of the disease. Thirty-nine tomato cultivars, twenty-one cherry tomato cultivars and thirteen rootstock tomato cultivars were inoculated with Clavibacter michiganensis subsp. michiganensis, to evaluate tomato cultivarspecific resistance against bacterial canker. In the evaluation of 73 major commercial cultivars, 'Sunmyung', 'Sweet', 'Akiko', 'Dadaki', 'Match', 'Magnet', 'Friend', and 'Greenpower' were found to have a high level of resistance to bacterial canker of tomatoes.

  • The Sclerotium blight was found on Neofinetia falcata at Yong-in city, Gyenggi-do, Korea. The symptom occurred low leaves yellowish and wilt of a whole plant. Severely infected plants were blighted and dies eventually. White mycelial mats appeared on the surface of basal stem and bulbs and the sclerotia were formed on stems, roots, and sphagnum moss. The sclerotia were spherical in shape, 1~3 mm in size and white to brown in color. The optimum temperature for the growth and sclerotia formation was $25{\sim}30^{\circ}C$ on PDA. On the pathogenicity test, the first symptom was appeared 5 days after inoculation and development to severe stem rot and blight. The causal fungus was identified as Sclerotium rolfsii and we suggested to call that the new Sclerotium blight on Neofinetia falcata caused by Sclerotium rolfsii in Korea.

  • The mean and RMS velocity field of the respiratory gas flow in tile human airway was studied experimentally by particle image velocimetry(PIV). Some researchers investigated the airflow for the mouth breathing case both experimentally and numerically. But it is very rare to investigate the airflow of nose breathing in a whole airway due to its geometric complexity. We established the procedure to create a transparent rectangular box containing a model of the human airway for PIV measurement by combination of the RP and the curing of clear silicone. We extend this to make a whole airway including nasal cavities, larynx, trachea, and 2 generations of bronchi. The CBC algorithm with window offset (64*64 to 32*32) is used for vector searching in PIV analysis. The phase averaged mean and RMS velocity distributions in Sagittal and coronal planes are obtained for 7 phases in a respiratory period. Some physiologic conjectures are obtained. The main stream went through the backside of larynx and trachea in inspiration and the frontal side in expiration. There exist vortical motions in inspiration, but no prominent one in expiration.

  • Since the leaf inoculation procedures are time-consuming and require considerable growth chamber space, a rapid dioassay method for screening of pathogenicity of Didymella bryoniae, a casual agent of gummy stem blight in watermelon, was established in this paper. The method produced reliable results within 8 days ( 5 days for growing seedlings and 3 days for rapid disease response in the seedlings). After contaminants in the root of 4~5 day-old seedlings had been washed using sterilized water, 5 seedlings were dipped into a vial containing 12 ml of conidial suspension (106 cells/ml). After the vials were placed in a growth chamber (22$^{\circ}C$, RH 50%, 14hr light/10hr darkness) for 3 days, susceptibility and resistance of cultivars were determined by the degree of disease response on cotyledon. The result of obtained by the dip-inoculation method was well coincided with the results by the leaf inoculation procedures and the result that had been observed for several years in the field. Screening of collected watermelon cultivars by the dip-inoculation method revealed that all the 21 domestic cultivars collected were susceptible and only 3 foreign cultivars (PI 189225, PI 482322 and IT 188207) were resistant among 18 cultivars A cucumber cultivar (Marketer) and bitter cucumber were proven to be resistant against the D. bryoniae among 8 other different cucurbits tested. The SDS-PAGE patterns of total proteins from a susceptible (Keumcheon) and a resistant (PI 189225) watermelon cultivars were compared 0, 12, 24 and 36 hrs after inoculation. The amounts of two distinct protein bands (24 kDa and 70 kDa) were gradually increased after inoculation in both cultivars.

  • Five antifungal substances were isolated from the long-term storaged extract of common purslane, and identified as isobutyric, butyric, isovaleric, valerie and caproic acids belonging to short-chain fatty acids (C4C6). Each of these fatty acids showed more or less antifungal potency against spore germination and mycelial growth of Alternaria alternata Japanese pear pathotype in vitro. Antifungal potency of each fatty acid against spore germination was greater than that against the mycelial growth. No one of these fatty acids completely inhibited the mycelial growth at concentration lower than 200 ppm, while 50 ppm of caproic acid and 200 ppm of valerie acid completely inhibited the spore germination. The results of bioassay also suggested that chain-length of the fatty acids might be related with the antifungal potency, since fatty acids with longer chain showed higher antifungal potency.

  • Anthracnose occurred in bitter gourd grown in Jeongup areas of Korea in 2011. Anthracnose of bitter gourd appeared as dark brown circular spots on naturally infected leaves and fruits. The symptoms of infected leaves and fruits were small brown to dark brown spots and gradually enlarged to larger cylindrical dark brown lesions. The causal fungus of anthracnose isolated from the diseased plants was identified as Colletotrichum gloeosporioides based on the morphological and cultural characteristics and ITS rDNA sequence analysis. All isolates of C. gloeosporioides produced symptoms on the host leaves by artificial inoculation. This is the first report of anthracnose on bitter gourd caused by C. gloeosporioides in Korea.

  • To identify genome sequences of Apple scar skin viroid (ASSVd) isolates in Korea, the field survey was performed from 'Hongro' apple orchards located in eight sites in South Korea (Bongwha, Cheongsong, Dangjin, Gimchoen, Muju, Mungyeong, Suwon, and Yeongwol). ASSVd was detected by RT-PCR and PCR fragments were cloned into cloning vector. Full-length viral genomes of eight ASSVd isolates were sequenced and compared with 21 isolates reported previously from Korea, India, China, Japan and Greece. Eight isolates in this study showed 92.2-99.7% nucleotide sequence identities with those reported previously. Phylogenetic analysis showed that seven isolates reported in this study belong to the same group distinct from other groups.
