Enzymes are important class of catalyst for biotransformation. Stability and reusability of enzymes during the catalysis process is a key issue. Activity of enzyme can be enhanced by its immobilization on a suitable substrate by creation of specific microenvironment. A variety of membranes has been used as substrate due to the biocompatibility and simpler method to tune hydrophilicity/hydrophobicity property of the membrane surface. In this review, polymer membranes including cellulose, polyacrylonitrile (PAN), polydimethylsiloxane (PDMS), polyvinylidene fluoride (PVDF), polyethersulfone (PES) are introduced and discussed in detail. Biodegradation of organic contaminants by immobilized enzyme is an environmental friendly process to reduce the contamination of environment in pharmaceutical company and textile industries. The controlled hydrolysis of oil can be performed in enzyme immobilized membrane bioreactor (EMBR), resulting in reducing carbon emission and reduced environmental pollution. Bioethanol and biodiesel are considered alternative fossil fuels that can be prepared in EMBR.

억새(Miscanthus spp.)는 우리나라 등 동아시아가 원산이며 바이오매스량이 많아 바이오에너지 원료작물로서 잠재성이 크기 때문에 2세대 바이오에탄올 생산 원료로 주목을 받고 있고, 축산깔개 및 토양개량제 등으로도 이용되고 있다. 미국과 유럽 등에서는 생태계 교란 방지를 위해 4배체 물억새(M. sacchariflorus)와 2배체 참억새(M. sinensis)의 종간 교잡 이질 3배체인 불임성 억새(M. x giganteous)을 주로 재배하고 있으나, 단일 유전형의 품종으로 병해충과 자연재해에 취약하여 다양한 억새 품종의 개발이 시급한 실정이다. 본 연구는 억새를 재료로 하여 반수체 및 배수체 확보를 통한 배수성 별 특성 평가와 함께 기내배양을 통해 탈분화 및 재분화 시스템을 구축하여 억새의 육종 효율을 높이기 위해서 실시하였다. 억새 종자로부터 캘러스의 유도는 MS배지와 N6배지에 1mg/L 2.4-D를 첨가하였을 때 비배발생 캘러스(nonembryogenic callus)가 유도되었고, N6배지에 3~5 mg/L 2,4-D를 첨가하였을 때 배발생 캘러스(embryogenic callus)가 발생하였다. MS배지보다는 N6배지에서 캘러스 유도율이 높았으며, 식물생장조절제로 2,4-D와 BA 조합처리 보다 2,4-D 단용 처리하였을 때 캘러스 유도율이 더 높았다. 억새 종에 따른 캘러스 유도율은 물억새가 25.2~49.3%, 참억새는 30.3~52.0%였고 거대억새는 62.6~81.1%로 나타났다. 억새 신초 및 줄기로부터의 캘러스 유도율은 물억새가 4.4~17.2%, 참억새는 1.8~7.7%, 거대억새는 15.3~19.9%로 나타나 종자에 비해 매우 낮았다. 미성숙화기로부터의 캘러스 유도는 억새 종에 따른 차이가 없었으며, 3mg/L 2,4-D 첨가 배지에서 캘러스 유도율이 비교적 낮았고(60~80%), 1mg/L와 5mg/L의 2,4-D가 첨가된 배지에서 캘러스 유도율이 높게(90~95%) 나타났다.

This study was carried out to evaluate the antioxidant, anti-inflammation, and tyrosinase inhibitory activity of Melissa officinalis extracts and their fractions. The total polyphenol contents of the extracts were 33.02-302.76 mg GAE/g and total flavonoid contents were 9.98-325.07 mg CE/g. In 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, DPPH radical scavenging activity was observed in the extracts and water fractions of M. officinalis and similar to that of ascorbic acid ($30{\mu}M$). In addition, the treatment of chloroform fraction significantly inhibited the production of nitric oxide (NO) in RAW 264.7 cells, indicating that they have anti-inflammatory activity. Tyrosinase inhibitory activity of $200{\mu}g/mL$ of 100% ethanol reflux extract showed better inhibitory activity than arbutin treatment at statistically significant level. As a result, it is considered that M. officinalis can be used as an effective cosmetic ingredient having antioxidant, anti-inflammation, and whitening activity.

The purpose of this study is to provide evidence for discovering functional materials through the anti-inflammatory efficacy screening of randomly selected medicinal herbs. We prepared 70% ethanol extracts from 10 herbs and evaluated for the inhibitory effect of NO production on LPS-stimulated mouse macrophage cell line Raw 264.7. As a result, it was confirmed that the Chrysanthemi Zawadskii Herba (CZ) extract had the highest effect of inhibiting NO production induced by LPS. We therefore measured and compared NO inhibitory effects at different concentrations (10, 50, 250 ㎍/mL) of 70% ethanol and water extract of CZ. It was observed that both ethanol and water treatment groups inhibited NO production in a concentration-dependent manner in both ethanol and water treatment groups. In particular, it was confirmed that the CZ 70% ethanol extract (99.97%) had a higher NO inhibitory effect than the water extract (93.32%) in the high concentration (250 ㎍/mL) treatment group. There was no effect of CZ extract on cell viability at all concentrations used in the experiment. Moreover, it was shown that CZ ethanol extract remarkably inhibited the expression of VCAM-1 induced by TNF-𝛼, and it was slightly decreased even by treatment with water extract. This study suggests that Chrysanthemi Zawadskii Herba has potential as a functional substance that regulates vascular inflammation.

In this study, we used extracts obtained from five different solvents (water, ethanol, hexane, ethyl acetate, butanol) of Achyranthes japonica (AJ) and also AJ fermented with Lactobacillus plantarum (LP) to confirm effects on the anti-inflammatory activity in RAW264.7 cells. Experiments of measuring nitric oxide (NO) and cytokine production were performed in lipopolysaccharide (LPS)-induced RAW264.7 cells, and the expression of both cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) was observed by a western blot method. The cytotoxicity of RAW264.7 was confirmed by the cell counting kit (CCK) assay at a concentration of $100{\mu}g/mL$, which has no toxicity. As a result of the inhibition of NO production, the inhibition rate of AJ-LP extracted with ethanol samples was about 74% higher than that of using the control group. Interleukin-6 (IL-6), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and Interleukin-$1{\beta}$ (IL-$1{\beta}$), which are inflammatory cytokines, also showed an excellent efficacy with inhibition rates of about 57, 70, and 74%, respectively. Comparing to the results of COX-2 and iNOS expression in the AJ group, the inhibition rate of 20-hydroxyecdysone was the highest than others. On the other hand, the COX-2 expression level of AJ-LP group decreased about 16% compared to that of the control group, and the iNOS expression level was also decreased about 7%. These results suggest that the extract of AJ fermented from L. plantarum can be used as an anti-inflammatory natural material.

Arthrospira platensis is one of the oldest algae in the world and has been reported to have anti-aging properties, including phycocyanin, tocopherol and beta-carotene. In this study, we tried to search protective activities against UVB-induced reactive oxygen species(ROS) of Arthrospira platensis under indoor cultivation ethanol extracts(ICAE) and outdoor cultivation ethanol extracts(OCAE). The anti-oxidative capacities were evaluated by DPPH radical scavenging activity and SOD-like activities at various concentrations(0.1, 0.5, $1mg/m{\ell}$) of ICAE and OCAE. Zebrafish embryos and HaCaT cells were exposed to UVB radiation and treated with various concentrations(0, 0.01, 0.05, 0.1, 0.5, $1mg/m{\ell}$) of ICAE and OCAE. ROS levels of zebrafish and HaCaT cells were generated by UVB radiation. ROS levels were detected using a fluorescent microscope after DCFH-DA staining. The DPPH radical scavenging activity of ascorbic acid was 73% and SOD-like activity was 86% in the positive control group. ICAE and OCAE at $1mg/m{\ell}$ concentration showed 43, 57% DPPH radical scavenging activity and 20, 19% SOD-like activity. Anti-oxidative of ICAE and OCAE had lower effects than the positive control ascorbic acid but significant results. ROS of UVB-induced zebrafish embryos and HaCaT cells were higher than negative control. ICAE and OCAE treated group decreased ROS concentration dependently than UVB-induced positive control group. These results suggest that Arthrospira platensis ethanol extract may have usability value as a cosmetic material for skin protection.

The Cnidium monnieri (L.) Cusson is an annual plant distributed in China and Korea. The fruit of C. monnieri is used as a medicinal herb that is effective for the treatment of carbuncle and pain in female genitalia. However, the anti-cancer effects of CME have not yet been reported. In this study, we assessed the apoptotic effects and cell cycle arrest effects of ethanol extracts from C. monnieri on HCT116 colon cancer cells. The results of an MTT assay and LDH assay demonstrated a decrease in cell viability and the cytotoxic effects of CME. In addition, the number of apoptotic body and the apoptotic rate were increased in a dose-dependent manner through Hoechst 33342 staining and Annexin V-PI double staining. In addition, cell cycle arrest occurred at the G1 phase by CME. Protein kinase B (Akt) plays an important role in cancer cell survival, growth, and division. Akt down-regulates apoptosis-mediated proteins, such as mammalian target of rapamycin (mTOR), p53, and Glycogen Synthase kinase-3β (GSK-3β). CME could regulate the expression levels of p-Akt, p-mTOR, p-GSK-3β, Bcl-2 family members, caspase-3, and PARP. Furthermore, treatment with CME, LY294002 (PI3K/Akt inhibitor), BIO (GSK-3β inhibitor), and Rapamycin (mTOR inhibitor) showed that apoptotic effects occurred through the regulation of the AKT/mTOR/GSK-3β signaling pathway. Our results demonstrated CME could induce apoptosis and cell cycle arrest in HCT116 colon cancer cells.
In this study, physiological changes in a thermotolerant yeast Saccharomyces cerevisiae KNU5377 cell exposed to 48-hour alcohol fermentation at $40^{\circ}C$ were investigated. After 12 hours of alcohol fermentation at $40^{\circ}C$, the $C_{16:1}$ unsaturated acid of plasma membrane increased to 1.5 times more than the $C_{16:0}$ saturated fatty acid, and to about 2 times more for the $C_{18:1}$ unsaturated fatty acid. Fermentation at both $30^{\circ}C$ and $37^{\circ}C$ fermentation showed the same pattern as that done at $40^{\circ}C$. The pH of the alcohol-fermentation medium was reduced to pH 4.1 from a starting pH of 6.0 through the 12-hr fermentation and then maintained this level during the continuing fermentation. With the process of fermentation, the remaining glucose was reduced, but its amount remaining during the $40^{\circ}C$-fermentation was less reduced than those fermented at $30^{\circ}C$ and $37^{\circ}C$. In the study investigating the changing pattern of cellular proteins in the alcohol-fermenting cells, the SDS-PAGE and 2-D data indicated the most expressed dot was phosphoglycerate kinase, which is one enzyme involved in glycolysis. Why this enzyme was most expressed in the cells exposed to unfavorable conditions such as high temperature, increasing concentration of produced alcohol and long time exposure to other stress factors remains unsolved.

In this study, the feasibility of utilizing wood chips from pitch pine (Pinus rigida) was evaluated for bioethanol production by an organosolv pretreatment and enzymatic saccharification. When wood chips from pitch wood were pretreated with 75% (v/v) ethanol and 1.7% sulfuric acid as a catalyst at H-factor 2000, average pulp yield was 43.3%, which pretreated wood fibers showed higher glucan (55.8%) and lower lignin (12.2%) contents than untreated control (43.9% glucan and 27.8% lignin). After enzymatic saccharification, the organosolv pulps with 56.2% delignification rate reached above 97% conversion rate of cellulose to glucose. These results indicated that increasing the delignification rate causes micro pores on the surface of organosolv pulps resulting in improved the accessibility of enzyme onto the substrate. Moreover, it was in agreement with the SEM examination of wood fibers.
Emulsified transesterification of soybean oil into biodiesel was investigated using potassium hydroxide and sodium methoxide catalysts with methyl glucoside oleic polyester as a methanol-in-oil emulsifier. The transesterification reaction conditions were optimized to obtain high yields of fatty acid methyl esters of the quality defined by biodiesel standards. The developed process resulted in $95{\sim}96%$ of overall yield from soybean oil by alkali-catalyzed methanolysis at $45^{\circ}C$ of reaction temperature with 6:1 of methanol-to-oil molar ratio and l(v/v)% methyl glucoside oleic polyester in the presense of 0.8wt% KOH and 1.2wt% $NaOCH_3$.
