Initial screening assay for depigmenting agents includes in vitro mushroom tyrosinase assay and antioxidant assay. Based on this screening result, melanin synthesis in melanocyte, in screened samples, is further measured. Measuring cellular melanin needs time, human resource, and skills. Therefore initial screening method should be reliable. We examined, 34 Chinese herbs, correlated the screening assay methods with cellular melanin. No reliable relationship was observed between factors, indicating the limitation in the use of these assays, probably due to the complexicity of melanogenesis.

In this study, the extracted oils and commercial oils (Clarisse) were analyzed after GC-MSD fragrance component analysis for each natural essential oil obtained through steam distillation extraction (cypress, seokchangpo, lavender) of a total of three plants grown and grown in Jangheung area. Easy, cinnamon, frankincense) was combined to confirm cytotoxicity, antioxidant, anti-inflammatory, and whitening effects, confirming its potential as a cosmetic material. As a result of the analysis of fragrance components, sabinene was identified as the main component of cypress oil, asarone in seokchangpo oil, and L-Linalool in lavender oil. At a concentration of 100 𝜇L/mL with no cytotoxicity of 6 essential combination oils, NO production was inhibited by 27.76%, DPPH radical scavenging activity was 99.69%, ABTS radical scavenging activity was 94.66%, and tyrosinase inhibitory activity was 55.9%. Possibility of being useful as a raw material for functional cosmetics was presented.
Total phenolic content was the highest in 60% ethanol extracts at 21.91 mg/g, and inhibitory activity against tyrosinase of 60% ethanol extracts was higher than ethanol extracts of other concentration. The inhibitory compounds against tyrosinase from Persimmon leaves were purified using Sephadex LH-20, MCI-gel CHP-20 column chromatography with gradient elution. Two purified compounds were isolated as a result. The chemical structures of each compound were determined and identified using $^1H$-NMR and $^{13}C$-NMR, FAB-Mass. The compounds were confirmed as (+)-gallocatechin and prodelphinidin B-3. The tyrosinase inhibitory activities of purified (+)-gallocatechin and prodelphinidin B-3 were 29.5 and 40.2%, respectively. The inhibitory activities of (+)-gallocatechin and prodelphinidin B-3 against melanin biosynthesis in melanoma cell were 32.5 and 46.7%. The safety of essence with tyrosinase inhibitory compounds from persimmon leaves was also assessed by various safety profiles. First, changes in pH (4.90~4.95) and viscosity (23,000~26,000 cP) was not detected for 60 days. Essence also showed stability against temperature and light for 60 days. All these findings suggest that extracts from persimmon leaves have a great potential as a cosmetical ingredient with a potent whitening effect.

This study is performed to investigate the effects of citrus essential oils on melanin production in B16 melanoma cells and reactive oxygen species (ROS) generation in RBL 2H3 cells. Five kinds of citrus essential oil (bergamot, grapefruit, lemmon, mandarin, petigrain) did not have any influence on DPPH radical scavenger activity, cell growth and cytotoxicity in B16 melanoma cells. In purified tyrosinase assay, both mandarin and petigrain essential oils dose-dependently inhibited its activity, but bergamot did not. In $1{\mu}M\;{\alpha}-MSH-stimulated$ B16 melanoma cells, all of 5 citrus essential oils inhibited melanin production in $\underline{a}$ dose dependent manner. On the other hand, four kinds of citrus essential oil dose-dependently increased ROS generation in RBL 2H3 mast cells, but mandarin did not. From the above results, it is possible that citrus essential oils nay be developed to be anti-melanogenic agent on the basis of their inhibitory effect on MSH-induced melanin production. Hut we can not rule out the possibility of the induction of allergy and inflammation since citrus essential oils caused ROS generation in RBL 2H3 mast cells.

To develop a new natural whitening agent for cosmetics, we investigated effects of Acanthopanax sessiliflorum Seeman on melanogenesis. We prepared phenolic acid-rich extract including two phenolic acids, chlorogenic acid and caffeic acid, as predominant constituents from Acanthopanax sessiliflorum Seeman. Phenolic acid-rich extract showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.43{\pm}0.35{\mu}g/mL$ and $158.91{\pm}1.57{\mu}g/mL$, respectively. Phenolic acid-rich fraction reduced melanin contents of B16 melanoma cells dose-dependantly and the decrease was $27.27{\pm}2.66%$ at a concentration of $100{\mu}g/mL$. And the phenolic acid-rich fraction reduced intracellular tyrosinase activity about $53.67{\pm}8.55%$ at a concentration of $100{\mu}g/mL$. Phenolic acid-rich extract inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that phenolic acid-rich fraction reduced melanin formation by the inhibitions of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that phenolic acid-rich extract could be used as a whitening ingredient in cosmetics.
The purpose of this study was to investigate the biological activities of an aqueous extract of Angelica gigas (Ag) fermented by Saccharomyces cerevisiae (Sc). First, the soluble solids of the F/3 group, in which the Ag was fermented by Sc for 3 days, decreased from $1^{\circ}Bx$ to $0.9^{\circ}Bx$. On the other hand, the pH increased with the number of days of fermentation. The result of a TLC experiment confirmed that it gradually decomposed into a low-molecular weight sugar form upon fermentation. The total phenolic compounds and flavonoid contents were higher in the fermented group than in the non-fermented group. K and Ca contents were increased by fermentation in the following order: F/3, NF, and F/0 groups. Decursin and decursinol angelate contents were highest in the F/3 group. The DPPH (${\alpha}$, ${\alpha}{\prime}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging activity of the NF, F/0, and F/3 groups were 41.89%, 39.51%, and 60.26%, respectively. The inhibition activities of tyrosinase and lipoxygenase were stronger in the F/3 group than in the NF group. This experiment showed that the fermentation of Ag Nakai can lead to an increase in its antioxidant ability, physiological activity, whitening and anti-inflammatory effects. Thus, this oriental herbal medicine can be developed into a functional material that can be utilized in the development of cosmetic products in future.

This study was to investigate the Taraxinic acid from Taraxacum coreanum on tyrosinase activity and melanogenesis. In B16BL6 cell, Taraxinic acid did not show cytotoxicity even at concentrations of up to 100 ㎍/mL. In addition, tyrosinase inhibitory activity and melanogenesis inhibitory activity were confirmed by stimulation with α-melanocyte stimulating hormone (α-MSH) in the presence of taraxinic acid. Taraxinic acid was added to cells at concentrations of 10, 50 and 100 ㎍/mL and treated for 48 hours to confirm tyrosinase inhibitory activity and melanin production. The tyrosinase inhibitory activity increased in proportion to the amount of the sample, and showed an inhibitory activity of about 54.5% at a concentration of 50 ㎍/mL. Melanin production decreased in proportion to the sample amount, and it was about 62.2% at the concentration of 10 ㎍/mL. From the above results, it was found that Taraxinic acid had higher tyrosinase activity and melanin synthesis inhibitory activity in melanocyte than arbutin. The results suggest that Taraxinic acid can be utilized in natural whitening cosmetics.

In this study, we analyzed the cosmetic applicability of extract from snow tea, native to Lijiang, Yunnan-province, China. After confirming the species as N. grossedentata through DNA analysis of Lijiang snow tea, experiments were conducted using representative tea, green tea, and a representative control group for each efficacy analysis. Both teas were extracted using 70% (v/v) ethanol aqueous solution. The polyphenol content in the Lijiang snow tea extract (gallic acid equivalent, 23.9 ± 3.2 mg/mL) was higher than that in green tea extract (16.4 ± 2.3 mg/mL). In contrast, the antioxidant (Radical scavenging, IC50 104 ㎍/mL), tyrosinase enzyme inhibitory (whitening agent, IC50 40.7 ㎍/mL), and Escherichia coli growth inhibitory (preservative) activities (IC50 2.85 mg/mL) were analyzed based on the solid content in the extract, and it was confirmed that the activities of Lijiang snow tea extract were superior to those of green tea extract (radical scavenging, IC50 234 ㎍/mL. It also showed similar efficacy to previously used active substances such as antioxidants (vitamin C, IC50 108 ㎍/mL), whitening agents (vitamin C, IC50 80㎍/mL), and preservatives (methylparaben, IC50 4.35 mg/mL). However, green tea was found to be better in collagenase inhibition activity (anti-wrinkle). Through this study, the cosmetic application potential of Lijiang snow tea is high.
This study aimed to confirm the possibility of being used as a cosmetic material material through the verification of the whitening and anti-inflammatory activities of an ethyl acetate fraction from Glechoma hederacea var. longituba (GHEA). The observed electron donating and ABTS+ radical scavenging abilities of GHEA were 89.6% and 88.7%, respectively, at 1,000 ㎍/ml concentration, with a tyrosinase inhibitory effect of 22.3% at the same concentration. For cell viability, a rate of 80% or more was observed in all concentrations that treated GHEA on melanoma and macrophage cells. Protein and mRNA expression inhibition was measured by Western blot and RT-PCR for 25, 50, and 100 ㎍/ ml concentrations, and it was confirmed that expression decreases in a concentration-dependent manner as GHEA concentration increases. The inhibition of the whitening-related factors MITF and TRP-2 were superior following GHEA treatment than those of the control group treated with kojic acid of 100 ㎍/ml concentration. For tyrosinase, the lowest mRNA expression rate was 29.1% at 100 ㎍/ml which confirmed excellent inhibition. In analyzing the effects of pro-inflammatory cytokines IL-1β, IL-6, and TNF-α on protein and mRNA expression, IL-6 and TNF-α showed high protein and mRNA inhibition compared to a vitamin C control group. Based on these experimental results, GHEA could be applied as a natural cosmetic material.

This study was performed to investigate the enhancement of UV-protection activities and skin-whitening effects from Angelica gigas Nakai extracts on ultra high pressure extraction process. Extraction at $60^{\circ}C$ treated by ultra high pressure for 15 minute and associated with ultrasofication (HPE15) was showed more than double yield, compare conventional extraction, as 12.24% (w/w) from A. gigas. Extracts of HPE15 reduced expression of MMP-1 on UV-irradiated CCD-986sk cells as 122.2% and revealed high inhibitory potency on tyrosinase as 69.4% by adding samples. Extracts of HPE15 from A. gigas showed strong inhibition effect on melanin production test by Clone M-3 cells as 82.4% by adding extracts. From the preliminary observations, we considered that the extracts from A. gigas could be potent natural materials for skin-whitening agent, and could be used as a potential anti-aging agent for the photo-damaged skin.
