A Study on Freezing Characteristics of Pavements Using Data of Test Road (시험도로 자료를 이용한 포장의 동결특성 연구)

  • To prevent the lowering of structural capability due to freezing and thawing in cold winter, numerous researches on frost heaving have been performed. As the result, the freezing index contour map of the Korea peninsula has been made for the design of the anti-freezing layer of pavements. However, the validity of the anti-freezing layer needs to be evaluated because systematic investigations on the variation of freezing depth with the thickness and material types of pavement layers and the configuration of the ground have been rarely performed. The freezing index of the Korea Highway Corporation test road site was calculated and the freezing depths of the concrete and asphalt pavements of the test road were investigated using the ambient and pavement temperature and water content. In addition, the investigated freezing depths were compared to the values estimated by existing freezing depth models. This is the preliminary study on the freezing-related data measured at the test road. The results with higher reliability will be produced by the long-term accumulation of the data and the analysis on it.

Effects of Cryopreservation of Sperm and Embryos on fertilization, Development and Pregnancy in Int Application (정자와 수정란의 동결이 ICSI 시술에서 수정, 발생 및 임신에 미치는 영향)

  • The cryopreservation of germ cells, sperm and embryos, has been largely used to increase the effect of artificial reproductive techniques for human infertility, but the efficiency of germ cell cryopreservation has been conkoversial till now. Thus, the effect of the cryopreservation of human sperm used for ICSI and the effect of the cryopreservation of embryos produced by ICSI on fertilizatiof development and pregnancy were investigated. Sperm freezing did not affect fertilizatiort development and pregnancy rates. Also, there was no significant difference between ejaculated and testicular sperm in ferclizatiort development and pregnancy. Embryo freezing methods, slow freezing and vitrificatior did not differ each other in viability and pregnncy rates. However, ICSI embryo freezing significantly decreased pregnancy rate compared to fresh embryos freezing (p<0.05). In conclusiof this result suggested that cryopreservation of sperm for ICSI did not affect on the resulted embryo development and pregnancy, but ICSI embryo cryopreservation would significantly inhibit pregnancy.

  • 돼지 난자의 유리화 동결 처리 방법 중 난자를 담는 용기(loading vessel)의 재료로 최근에 알려진 것으로 open-pulled straws(OPS)[Vajta등, Mol Reprod Dev, 51:53-58, (1998)], electron microscope grids(EMG) [Martino등,Biol Reprod, 54:1059-1069, (1996)〕, nylon loop system(NLS) [Lane등, Fertil Steril,72: 1073-1078, (1999)] 등이 보고되고 있다. OPS는 1/4cc straws를 열을 가하여 길게 뽑아 내벽을 얇게 함으로써 filing된 난자나 수정란이 액체 질소와 접촉했을 때 유리화가 신속하게 되도록 하는 방법으로 돼지에서는 별로 보고된 것이 없다. EMG는 열전도가 예민한 전자현미경용 copper grid를 이용한 방법으로 최근 국내 기술진의 연구성적을 포함한 몇몇 학자들에 의하여 보고되었고 NLS는 0.5mm직경의 nylon loop를 이용하여 급속 동결한 성적이 보고되었으나, 돼지 난자에 응용 된 것은 없다. 따라서 이와 같은 동결 재료는 사람과 반추류, mouse외에 돼지 난자에 대하여는 전혀 시도되지 않았지만 유리화 동결기술에서 가장 중요한 실험으로 생각된다. 성공적인 유리화 동결을 위해서는 수정란이 냉각의 전도성이 빠르고, 작은 용액을 수정란과 같이 filling해야 하며 모든 동작이 신속 간편해야 하며 융해 방법도 초급속도의 융해가 요구되므로 이에 부합되어야 한다. 연구 목적은 돼지 난자를 유리화 동결/융해 시 동결 재료-straw/glass, copper grid, nylon 3가지에 대한 제작 방법, 난자 loading, 동결 처리, 보관 방법, 융해 방법 등을 난자의 회수, 수정 후 생존율을 비교 조사하여 가장 우수한 방법을 선택할 목적이었다. 수행 내용은 3가지의 재료의 sample을 제작하고 소독한 다음 준비된 돼지 COCs을 40시간동안 IVM한 후 난자를 5~l5개 정도로 선정 하여 준비된 VS 용액에 평형처리 하였다. 각 재료의 용기에 loading 한 후 동결/보관하였고, 융해는 역순으로 평형하여 maturation 배지에 3~4시간 배양한 다음 경검하고 IVF한 후 NCSU-23 배지에 담아 IVC 배양하면서 cell cleavage상태를 확인하였다.

STUDIES ON THE FREEZING OF YELLOW SEA BREAM (옥돔의 동결에 관한 연구 2. 동결에 의한 옥돔조직의 변화)

  • Histological changes of yellow sea bream, Branchiostegus japonicus japonicus, were observed under microscope during freezing storage for 6 months at $65^{\circ}C$ and $-35^{\circ}C$, and the results were compared with the muscle structure of fresh muscle. The freezing storage of yellow sea bream at $-^{\circ}C$ showed more changes in muscle structure than that stored at $-35^{\circ}C$. In the view point of the changes of the muscle structure, the yellow sea bream can be stored up to 1 month at $65^{\circ}C$ and 3 months at $-35^{\circ}C$. The freezing of yellow sea bream at $-5^{\circ}C$ showed higher extracellular freezing than that at $-35^{\circ}C$.

Mechanical Constitutive Model for Frozen Soil (동토지반에 대한 역학적 구성모델)

  • Recently, growing interests in frozen ground have stimulated us to advance fundamental theories and systematic researches on soil behavior under freezing conditions. Unlike the well-established soil mechanics theory, temperature variation and phase change of pore-water cause water migration to cold side, ground heaving, sharp increase in earth pressure, etc., which bring about serious problems in frozen geotechnical structures. Elasto-plastic mechanical constitutive model for frozen/unfrozen soil subjected to fully coupled THM phenomena is formulated based on a new stress variable that is continuous in frozen-unfrozen transitional regions. Numerical simulations are conducted to discuss numerical reliability and applicability of the developed constitutive model: one-dimensional heaving pressure, tri-axial compression test, and one-side freezing tests. The numerical results show that developed model can efficiently describe complex THM phenomena of frozen soil, and they can be utilized to analyze and design the geotechnical structures under freezing conditions, and predict their long-term behavior.

  • Life can be kept in suspended animation either before fertilization at oocyte stage or after fertilization at different stages of embryonic development for a variety of reasons. It not only has potential applications in fertility preservation and management in human but also has important roles in the preservation and management of animal genetic resources, low-cost international movement of selected genetics, and rapid dissemination of germplasm through assisted reproductive technologies (ART) and genetic engineering. Currently, slow-freezing and vitrification are the two approaches by which oocytes and embryos can be cryopreserved for long-term storage. Both of these methods have their own advantages and disadvantages but allow the cryopreservation of oocytes and embryos with comparable efficiency. Vitrification of oocyte and embryos, although proven successful just 13 years after slow-freezing, is generally considered an emerging technology and appears to slow gain acceptance in both animal and human ART despite having controversial storage and contamination issues. In this manuscript, we discuss the basic techniques of oocyt/embryo cryopreservation and review the current status and recent developments in vitrification.

A Durability Assessment on Complex Deterioration of Concrete with Ground Granulated Blast-Furnace Slag Replacement (복합열화 환경하에서의 고로슬래그미분말 사용 콘크리트의 내구성능 평가)

  • This paper presents the experimental results of frost durability characteristics including freezing-thawing and de-icing salt scaling of the concrete for gutter of the road and marine structure. Mixtures were proportioned with the three level of water-binder ratio(W/B) and three binder compositions corresponding to Type I cement with 0%, 30% and 50% GGBS(Ground granulated blast furnace slag) replacement. Also, two different solutions of calcium chloride were used to evaluate their effect on the frost durability resistance. Specially, in case of complex of freezing and thawing with salt and carbonation, the deterioration of concrete surface is evaluated. Test results showed that the BFS30 and BFS50 mixture exhibited higher durability and lower mass loss values than those made with OPC mix and the use of GGBS can be used effectively in terms of economy and frost durability of the concrete to be in complex deterioration. Therefore, the resistance to complex deterioration with freezing-thawing was strongly influenced by the strength and the type of cement.

Evaluation of Freeze-Thaw Effect on the Modulus of Subgrade Soils from Impact Resonance Test (충격공진시험을 이용한 동결.융해에 따른 노상토의 탄성계수 평가)

  • Anti-freezing layer does not used in case of non frost heaving in subgrade soils. In this case, the modulus of subgrade soils were varied with freezing and thaw cycles under non frost heaving. That effect should be properly considered in pavement design. Impact resonance test that is nondestructive testing method was used for continuously determining the modulus of subgrade soils during freezing and thaw cycle. The modulus of subgrade soils was identical with freezing and thaw cycles under closed freezing and thaw system which is no water supplement into specimen during testing. There was also no difference in the modulus of subgrade soil between before and after freezing-thaw cycles for all specimens with different water content and density. That is thaw-weakening of subgrade soils do not occur under closed freezing and thaw system. The moduli at freezing conditions are varied with water content and density, but it can be ignored in practical design sense.

Evaluation of the Mechanical Characteristics of Frozen Sand, Considering Temperature and Confining Pressure Effects, in a Cryogenic Triaxial Compression Test (동결 삼축압축시험을 통한 동결 사질토의 온도 및 구속압력에 따른 역학적 특성 평가)

  • Most studies have conducted cryogenic triaxial compression tests with frozen specimens prepared in a separate mold by one-directional freezing. This method has the potential to generate residual stress in a frozen specimen and cannot be adopted to simulate the application of the artificial ground freezing method in the field. Therefore, in this study, novel equipment and procedure for the cryogenic triaxial compression test were proposed to overcome the limitations of existing test methods. Therefore, the mechanical characteristics of frozen sand, considering the effect of temperature and confining pressure, were evaluated. As the freezing temperature decreased, the brittleness of frozen sand increased, and the strength increased due to a decrease in the unfrozen water content and an increase in the ice strength. A higher confining pressure resulted in an increase in interparticle friction and the pressure melting phenomenon, which caused strength reduction. Thus, it was found that the mechanical behaviors of frozen sand were simultaneously affected by both temperature and confining pressure.

  • 본 연구는 개의 동결정액제조 시 동결보호 희석액 내에 첨가되는 당의 종류와 조합이 동결융해 후 정자의 생존율 및 운동성에 미치는 영향과 정자동결 시 straw size, 예비동결 시 액체질소표면으로부터 straw까지의 거리 및 융해온도가 생존율에 미치는 영향에 대하여 조사하고자 실시하였다. 포유류의 경우 부생식선의 정낭선에서 분비되는 정장내에는 정자의 체외사출 후 정자의 영양물질인 fructose 가 분비되는데, 동결정액제조 시 회석액내에 첨가되는 당 성분은 fructose 및 glucose를 일반적으로 사용하고 있다.
