A Comparision of Disinfection Efficacy for RGP Lens by Using Multi-purpose Solution and Cleaner (다목적 용액 및 세척제를 이용한 RGP 렌즈 소독방법의 비교)

  • The aim of this study is to establish RGP lens care system using RGP lens multi-purpose solution(MPS) and cleaner. Each RGP lens was incubated in Pseudomonas aeruginosa standard strain, and the numbers of P. aeruginosa colony remaining after treatment by using MPS and/or cleaner including surface activating components were estimated. Soaking only in MPS for 30 min reduced the number of P. aeruginosa up to 50%. 95% of P. aeruginosa was eliminated at RGP lenses soaked in MPS for 4 hr, but P. aeruginosa was not detected at RGP lenses soaked for 6 hr. When only cleaner was used for RGP lens care, 70% of P. aeruginosa was eliminated. This result showed that using cleaner had more effective at removing P. aeruginosa than soaking in MPS for 30 min. When soaking in MPS for 30 min after using cleaner reduced the number of P. aeruginosa up to 36% of that which soaking only in MPS for 30 min. This result suggested that using cleaner could be helpful to improve the disinfection efficacy of short time soaking in MPS.

Upregulaton of Bradykinin Receptor Mediated by Nucleoside Diphosphate Kinase and Flagellin from Pseudomonas aeruginosa (Bradykinin Receptor의 발현에 미치는 녹농균유래 Nucleoside Diphosphate Kinase 및 Flagellin의 효과)

  • Immune defense responses against Pseudomonas aeruginosa infection play an important role in maintaining homeostasis in the human body. Previously, we reported that expression of the bradykinin receptor (BR) is induced in response to P. aeruginosa infection. However, the factors responsible for the induction was uncertain. Here, we found that the type III secretion system (T3SS) is responsible for the induction of BR expression, and nucleoside diphosphate kinase (Ndk), as a novel T3SS effector, mediates the upregulation. The Ndk-mediated expression of BR was not induced by fliC mutant treatment, indicating the involvement of flagellin, one of the well-known pathogen-associated molecular patterns (PAMPs). Taken together, this study demonstrated that Ndk cooperates with flagella in the development of defense responses against P. aeruginosa infection.

Improving Detection Method of Pseudomonas aeruginosa, an Important Index Organism of Bottled Water Quality (먹는샘물의 녹농균 분포 및 검사방법 개선에 관한 연구)

  • Since the official allowance of bottled water at Korean domestic market in 1995, Pseudomo~zas aemginosu has been detected from 2.3% and 1.2% of source and products of bottled water sa~nple tested, respectively. according to the nation-wide dala froin May 1995 to December 1996. Therefore, P aeivginosa was the second most important parameter, next to colifoi~ns, anlong the bacieriological parameters regulated for bonled water. The official standard method initially adopted the Japanese officlal method and Standard Methods of the US, w~hich is using aspai-agiii-acetamid mnedia(A-A method). how eve^; the method showed low specificity regardless of the high sensitivity. Analysis of Quorum Sensing-Related Phenotypes of Pseudomonas aeruginosa Clinical Isolates (녹농균 임상균주의 쿼럼 센싱 관련 표현형 분석)

  • Pseudomonas aeruginosa is a Gram (-) opportunistic human pathogen causing a wide variety of infections on lung, urinary tract, eyes, and burn wound sites and quorum sensing (QS), a cell density-sensing mechanism plays an essential role in Pseudomonas pathogenesis. In order to investigate the importance of QS in the Pseudomonas infections of Korean patients, we isolated 189 clinical strains of P. aeruginosa from the patients in Pusan Paik Hospital, Busan, South Korea. The QS signal production of these clinical isolates was measured by signal diffusion assay on solid media using reporter strains. While most clinical strains (79.4%) produced the QS signals as similar level as a wild type strain, PAO1 did, where LasR, the initial QS signal sensor-regulator was fully activated, a minority of them (4.2%) produced much less QS signals at the level to which LasR failed to respond. Similarly, while 72.5% of the clinical isolates produced QS signals enough to activate QscR, an another QS signal sensor-regulator, some few of them (9%) produced the QS signals at much lower level where QscR was not activated. For further analysis, we selected 74 clinical strains that were obtained from the patients under suspicion of Pseudomonas infection and investigated the total protease activity that is considered important for virulence. Interestingly, significant portion of them showed very low protease activity (44.6%) or no detectable protease activity (12.2%). When the biofilm-forming ability that is considered very important in chronic infection was examined, most isolates showed lower biofilm-forming activity than PAO1. Similarly, significant portion of clinical isolates showed reduced motility (reduced swarming activity in 51.4% and reduced twitching activity in 41.9%), or non-detectable motility (swarming-negative in 28.4% and twitching-negative in 28.4%). Our result showed that the clinical isolates that produced QS signals at the similar level to wild type could have significantly reduced activities in the protease production, biofilm formation, and motility, and some clinical isolates had unique patterns of motility, biofilm formation, and protease production that are not correlated to their QS activity.

Antibacterial Effect of Various Fermentation Products and Identification of Differentially Expressed Genes of E.coli (다양한 발효액의 항균효과와 대장균의 유전적 변화에 미치는 영향)

  • Pseudomonas aeruginosa (P. aeruginosa), Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) are typical opportunistic pathogens. Moreover, these bacteria are known to possess multidrug-resistant (MDR) properties. This study investigates the antimicrobial activity of six fermented products, which have varying efficacies against P. aeruginosa, E. coli, and S. aureus. To identify novel candidate genes, differential expression analysis was performed using an annealing control primer. In the disk diffusion method, Fig vinegar (FV) and Diospyros kaki Thunb vinegar (DTV) showed the greatest increase in inhibition compared to other fermented products, whereas fermented Korean traditional nature herb (FKTNH) had no antibacterial effect. This study identified down-regulation of Escherichia coli O157:H7 ompW gene for outer membrane protein W, whereas gene for synthetic construct Lao1 gene for L-amino acid oxidase were up-regulated in E. coli treated with 5% FV. Consuming fermented vinegar helps prevent bacterial infections. Especially, FV and DTV are potentially useful alternative natural products for multidrug resistance. Furthermore, both are expected to be used as effective natural antimicrobial agents, such as disinfectants.

Inhibition of Quorum Sensing and Biofilm Formation by Synthetic Quorum Signal Analogues in Pseudomonas aeruginosa (합성된 쿼럼 신호 유사 물질에 의한 녹농균 쿼럼 센싱 및 생물막 형성의 제어)

  • Pseudomonas aeruginosa is an opportunistic pathogen that causes various infections on urinary track, cornea, respiratory track, and burn wound site, and mainly relies on quorum sensing (QS) for its virulence. To control the infectivity of P. aeruginosa, we previously synthesized the structural analogues of a major QS signal, N-3-oxododecanoyl homoserine lactone (3OC12-HSL) to use as a QS inhibitor. Two of them (5b and 5f) had been confirmed to have an inhibitory effect on LasR, a major QS signal receptor of P. aeruginosa in the screening by the recombinant Escherichia coli reporter. To further evaluate these compounds, we tested their efficacy to control the QS and virulence of P. aeruginosa. Unlike the result from E. coli reporter, both 5b and 5f failed to affect the LasR activity in P. aeruginosa, but instead they selectively affected the activity of QscR, another 3OC12-HSL receptor of P. aeruginosa. Interestingly, their effect on QscR was complex and opposite to what we obtained with E. coli system. Both 5b and 5f enhanced the QscR activity at the low concentration range (< 10 ${\mu}m$), but high concentration of 5f (${\approx}$1 mM) strongly inhibited QscR. While 5b and 5f didn't affect the production of proteases, the key virulence factor, they significantly reduced the biofilm formation that is important in mediating chronic infections. Especially, 5f inhibited the initial attachment of P. aeruginosa, rather than the biofilm maturation. Based on our results, we suggest that 5f can be applied for an anti-biofilm agent without increasing virulence of P. aeruginosa.

Purification and Characterization of the S-type Pyocin of Pseudomonas aeruginwa 90-2-2205 Isolated from Korean Patients (한국환자유래의 녹농균 90-2-2205로부터 S형 Pyocin의 정제 및 특성)

  • The s-type pyocin was purified from the lysate of the mitomycin C-induced Pseudomonas aeruginosa 90-2-2205 cells by the other of ammonium sulfate precipitation, DEAE-Sephadex A-50 chromatography and Sephadex G-200 gel filtration. The purity was confirmed by the polyacry-lamide gel electrophoresis. The molecular weight of the purified pyocin was estimated 180, 000 by gel filtration. The pyocin was analyzed to be a complex of some polypeptides by the SDS-PAGE. The pyocin was stable by heat treatment and at pH 6-7.5 by adding 10-3% gelatin and 0.2M NaCl to the 10mM Tris-HCl buffer (pH7.5). Its killing action against the sensitive cells was assumably a single hit process.

  • New quinolone derivatives of 7-(cis- or trans-3-amino-4-mehtylthiomethylpyrrolidinyl) quinolone-3-carboxylic acids were synthesized by condensation of 7-halo substituted quinolone-3-carboxylic acids with cis- or trans-3-amino-4-mehtylthiomethylpyrrolidine. Some of these compounds showed broad spectra of antibacterial activities against Gram-positive and Gram-negative organisms except Pseudomonas aeruginosa, and exhibited much stronger activity against MRSA.
