Characterization of a Low Molecular Weight Heat-Shock Protein cDNA Clone from Nicotiana tabacum

  • We characterized a cDNA clone for a low molecular weight heat-shock protein (LMW HSP) from tobacco named TLHS-l. Nucleotide sequence determination of TLHS-1 identified an open reading frame for 159 amino acids. To the upstream of the open reading frame, a sequence of 124 nucleotides was determined. To the 3' downstream of the open reading frame, 212 nucleotides were identified which carried poly(A)-tail. Comparison of the open reading frame and hydropathy plot of TLHS-1 with the previously reported class I LMW HSPs showed high identity which classified TLHS-1 as a class I LMW HSP cDNA clone. We proposed that there are six consensus regions in class I LMW HSPs. RNA blot hybridization for TLHS-1 showed a typical expression pattern of heat-shock-inducible gene from three common tobacco cultivars. The open reading frame of TLHS-1 was overexpressed in Escherichia coli. TLHS-1 protein confers thermal protection of other proteins in vitro and in vivo. Thermal induced aggregation of citrate synthase was reduced by purified TLHS-1 protein, and thermal death rate at $50^{\circ}C$ was reduced in E. coli expressing TLHS-l. From these data, we can expect that TLHS-1 acts as a molecular chaperone.perone.

Assessment of Coarse Woody Debris in Gallery Forest in the Bombo-Lumene Reserve (Democratic Republic of Congo)

  • The objective of this research was to assess the amount of carbon stock of coarse woody debris (CWD) in Bombo-Lumene Reserve. Data on lying CWD was collected on 35 circular sampling plots using Line Intersect Sampling (LIS) method. A total of 230 samples CWD (${\geq}10cm$ diameter) were inventoried. The mean carbon stocks of CWD was $29.48Mg\;C\;ha^{-1}$, ranging from 4.32 to $73.54Mg\;C\;ha^{-1}$. The CWD carbon stocks displayed a wide range of variation in decay states. The allocation of CWD among the decay class of all the CWD samples reveals that the most important classes were class 1 and class 3 with 323.66 and $321.96Mg\;C\;ha^{-1}$, followed by class 4 with 264.56 and the last one was class 2 with $121.72Mg\;C\;ha^{-1}$. The results suggested that the dead wood component is important in carbon sequestration and should be taken into consideration for quantification of carbon stocks not only in Bombo-Lumene Reserve, but in all forest ecosystems in the Democratic Republic of Congo.

A Study on the Relationship between Learners' Online Class Satisfaction and LMS Satisfaction (학습자의 원격수업 만족도와 LMS만족도와의 관계연구)

  • This study aims to investigate the degree of influence of LMS on online classes conducted after COVID-19 at C University in Gyeongsangnam-do, and to examine the relationship between overall class satisfaction and LMS satisfaction variables. C University started the project to construct 'Next-generation Smart LMS' and operated it from the first semester of 2021. As a result of conducting a survey about overall class satisfaction and LMS satisfaction consisting of five variables with 140 learners who have experienced online classes at C University. The learners showed high satisfaction not only in overall classes but also LMS. As a result of regression analysis of LMS satisfaction and overall class satisfaction, it was found that the functional convenience of LMS and interaction with instructors had a significant effect on overall class satisfaction. This study has limitations in that it was conducted only at one college and a limited number of variables were measured.

Nucleotide Sequence and Homology Analysis of phnC Gene Encoding Glutathione S-transferase from Pseudomonas sp.DJ77 (Pseudomonas sp. DJ77에서 Glutathione S-transferase를 암호하는 phnC 유전자의 염기서열과 상동성 분석)

  • Pseudomonas sp. DJ77로부터 클로닝된 glutathione S-transferase 유전자(phnC)의 염기서열을 결정하였다. 603bp의 open reading frame(ORF)이 존재하였고 개시코돈 앞에서 Shine-Dalgarno sequence를, 종결코돈 뒤에서는 terminator sequence를 발견하였다. phnC 유전자에서 만들어지는 phnC 단백질은 21,416 Da으로 SDS-polyacrylamide gel 전기영동 결과와 일치하였다. PhnC는 Bulkholderia cepacia LB400, Cycloclasticus oligotrophus RB1의 GST와 각각 53.7%, 49%의 높은 상동성을 나타냈다. 아미노산 서열의 상동성과 필수잔기들의 존재유무로 판단할 때 PhnC GST는 theta class GSTs와 진화적으로 유연관계가 높았지만 alpha, mu, pi, sigma class GSTs에서 구조적, 기능적으로 중요하다고 알려진 아미노산 잔기들이 PhnC GST에도 보존되어 있었다. 또한, phnC 유전자의 위치가 C. oligotrophus RB1, B. cepacia LB400 등의 GST 유전자 위치와 유사하다는 점에서 PhnC 효소는 난분해성 방향족 탄화수소의 분해에 관여하는 것으로 생각된다.

Molecular Cloning of Chicken Major Histocompatibility Complex Class II Molecules

  • The chicken major histocompatibility complex (MHC), the B complex, is beginning to be analyzed at the DNA level. Inbred lines of chickens have been reported to possess 3~5 MHC class II genes. To further analyzed the molecular structure of the chicken MHC class II genes, cDNA clones coding for chicken MHC class II (B-L) ${\beta}$ chain molecules were isolated from chicken spleen and liver. Tissue-specific transcription of B-L ${\beta}$genes was studied by Northern blot analysis. A high level of expression was detected for spleen poly(A)$^+$ RNA whereas a faint signal was detected for liver poly(A)$^+$ RNA. Twenty-nine cDNA clones were isolated from the spleen and eight cDNA clones were isolated from the liver. Based on restriction maps, most clones could be clustered into one family of genes. Four cDNA clones were sequenced (S7, S10 and S19 from the spleen and L1, which was identical to S19, from the liver). Complete amino acid sequences of B-L ${\beta}$ chain molecules were predicated from the nucleotide sequences of the cDNA clones. Although both the nature and the location of the conserved residues were similar in chicken and mammalian sequences, some species-specific differences were found, suggesting that the structures of the B-L molecules are similar, but not identical to their mammalian counterparts.

  • Adenylyl cyclase (AC) catalyzes the formation of cyclic AMP (cAMP) from ATP. The cAMP produced by AC serves as a secondary messenger in a variety of signal transduction pathways, and controls various cellular functions in many organisms. ACs can be grouped into six classes based on their primary amino acid sequences. Eukaryotes and mycobacteria contain only members of class III AC. The catalytic cyclase domains of class III AC are active as dimers: mammalian ACs, which are composed of a single polypeptide with two catalytic cyclase domains, form the active site as a result of intramolecular dimerization of the catalytic cyclase domains. In contrast, mycobacterial ACs function as homodimers, since their polypeptides contain a single catalytic cyclase domain. Six amino acids are required for the catalytic activity of class III AC - two aspartate residues, a lysine-aspartate pair and an arginine-asparagine pair. 16 ACs belonging to the class III were identified in Mycobacterium tuberculosis H37Rv, and their characteristics are reviewed.

Assessment of Biological Water Quality Using Epilithic Diatoms in the Upper Region of Nakdong River (낙동강 상류 수계에서 부착돌말류를 이용한 생물학적 수질 평가)

  • This study assessed biological water quality using epilithic diatoms in the Yeong river, Naeseong stream and Wi stream in the upper region of the Nakdong river from May to October 2016. Epilithic diatoms were not mobile, so they could reflect long-term water quality. The sampled epilithic diatoms were identified a total 158 taxa which were composed to 2 orders, 3 suborders, 8 families, 34 genera, 143 species and 15 varieties. Dominant species were Achnanthes convergens and Achnanthes minutissima at Yeong river, Nitzschia inconspicua at Naeseong stream, and Achnanthes minutissima, Cocconeis placentula var. lineata and Navicula minima at Wi stream. As a result of the CCA, Electrical conductivity, total nitrogen and total phosphorus were important factors determining the diatom species composition in the upper region of the Nakdong river. The correlation between diatom indices (DAIpo & TDI) measured to be high in the correlation coefficient (0.87) from the result of correlation analysis. In the result of the assessment of biological water quality using DAIpo and TDI, Yeong river was rated as class A at most sites. Naeseong stream was rated as class C to D at all sites except for N1 which was rated as Class A. Wi stream was rated as class B to C for DAIpo of W1, and TDI was rated as class D. The assessment of biological water quality at this site showed inferior TDI result compared to that of DAIpo. DAIpo and TDI of W2 were rated as class A to D, and the water quality has changed a lot. W3 and W4 were mostly rated as class B and C respectively.

Macromodels for Efficient Analysis of VLSI Interconnects (VLSI 회로연결선의 효율적 해석을 위한 거시 모형)

  • This paper presents a metric that can guide to optimal circuit models for interconnects among various models, given interconnect parameters and operating environment. To get this goal, we categorize interconnects into RC~c1ass and RLC-c1ass model domains based on the quantitative modeling error analysis using total resistance, inductance and capacitance of interconnects as well as operating frequency. RC~c1ass circuit models, which include most on~chip interconnects, can be efficiently analyzed by using the model~order reduction techniques. RLC-c1ass circuit models are constructed using one of three candidates, ILC(Iterative Ladder Circuit) macromodels, MC(Method of Characteristics) macromodels, and state-based convolution method, the selection process of which is based upon the allowable modeling error and electrical parameters of interconnects. We propose the model domain diagram leading to optimal circuit models and the division of model domains has been achieved considering the simulation cost of macromodels under the environmental assumption of the general purpose circuit simulator such as SPICE. The macromodeling method presented in this paper keeps the passivity of the original interconnects and accordingly guarantees the unconditional stability of circuit models.
