In previous paper, we described the induced callus of Corydalis remota contains a significant amount of alkaloids. This study describes an optimal condition to maximize alkaloid production. The suspension cultures maintained alkaloid production ability after fifth subculture and a small amount of alkaloid seemed to be released out of cells. The yields of alkaloid by cultured cells was varied depending on the concentrations of NAA, carbon sources and phosphate ion and depending on the vitamin combinations and concentrations. Biosynthetic precursor and an elicitor treatment also affected the total alkaloid yield of the cultures. The optimal conditions for alkaloid production were as follows: 1) MS basal salt containing 30 g/l of glucose, 1.0 mg/l of NAA, and vitamins of LS medium should be used. 2) The culture should be treated with tyrosine 20 mg/l, and yeast extract 1.5 ml/l after the culture reached a stationary phase of growth. Five alkaloids were isolated from the cultures and they were characterized. The spectral data unambiguously revealed that the isolated compounds were dihydrosanguinarine, protopine. tetrahydropalmatine, allocyptopine and ambinine, respectively.

In a preliminary screening of plant extracts for the antigastritic and antiulcer actions in rats, the extracts of head of Panax ginseng Radix showed positive activity in HCl ethanol-induced gastric lesion. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent butanol fraction reduced significantly HCl ethanol-induced gastric lesion at the oral dose of 500 mg/kg. In pylorus ligated rats, hexane and butanol fraction showed decreases in the volume of gastric secretion and acid output, of which effects were stronger in butanol fraction. Further assays with butanol fraction disclosed that it significantly suppressed the aspirin-induced and Shay ulcer. The butanol fraction at the intraduodenal dose of 500 mg/kg showed significant stimulation of mucus secretion.

Hairy roots induced from stems of Rubia cordifolia var. pratensis were cultured in the liquid medium under a variety of auxins to find the optimal condition for the growth and production of pigments. Culture of the hairy roots on NN liquid medium containing NAA 0.5 mg/l was best for growth of hairy roots. Production of yellow anthraquinone derivatives and purpurin in hairy roots was enhanced by the culture on NN liquid medium without auxins. Effects of L-phenylalanine, L-tyrosine and juglone, synthesized via the shikimic acid pathway, on growth and production of pigments in hairy roots were studied in the present study. Concentration of exogeneous L-phenylalanine. L-tyrosine and juglone in liquid culture system of hairy root containing NAA 0.1 mg/l was decreased quickly in its early stages of the culture period. Addition of juglone to NN liquid medium containing NAA 0.1 mg/l enhanced the productivity of pigments in hairy roots.

As part of our search for less toxic antimicrobial agents from natural resources, antimicrobial activity of rutin isolated from Sophora japonica was tested in vitro against four kinds of gram positive bacteria and four kinds of gram negative bacteria by serial broth dilution method. Among eight kinds of bacteria tested, the antimicrobial activity of rutin was the most potent against Mycobacterium smegmatis showing MIC of $375\;{\mu}g/ml$. The acute toxicity of rutin was examined in mice and rats. and $LD_{50}$ value administered intraperitoneally in mice was 650 mg/kg(Confidence limit: 894-1,473 mg/kg). There were no significant changes in serum biochemical values and histopathological changes as compared with those of control after intraperitoneal administration with rutin in rats.

Three medicinal species of the genus Polygala were examined by comparative morphology, anatomy and TLC analysis. They were classified into three species, and distinctly divided into two groups in this study. Group I is composed of P. japonica, P. sibirica and Group II P. tenuifolia. Considered on the relationships between two groups by the differences of leaf shape, adnated part of petal, flower colour, fruit shape etc., Group II may have been independently evolved from the common ancestor by having one raw of palisadelike chlorenchyma under the stem epidermis through the different pathway. It is also suggested that the taxa of Group II appear to be more advanced than those of Group I.

The ether soluble fraction of the roots of Angelicae gigantis Radix caused a significant prolongation of hexobarbital(HB) induced sleeping time in mice. Through systematic fractionation of the ether fraction monitored by bioassays, two pyranocoumarins, decursinol angelate and decursin were isolated as active principles. Decursin, as a main component, exhibited significant prolongation of HB-induced hypnosis as well as significant inhibition of hepatic microsomal drug metabolizing enzyme(DME) activities at relatively high dose which indicated that it is a weak DME inhibitor.

Effects of hydrogen peroxide$(H_2O_2)$ on polyphenol oxidase (PPO) in Perillae Folium were investigated. The inactivation of this enzyme was dependent on $H_2O_2$ concentration. and the initial lag period was not shown. Preincubation of Perillae Folium PPO with $H_2O_2$ in the absence of a substrate resulted in rapid loss of enzymatic activity. The inactivation of PPO by $H_2O_2$ dependents temperature and pH. OH radical scavengers such as mannitol and sodium formate did not protect the enzyme against inactivation by $H_2O_2$. Substrate analogue such as phenylalanine protected the enzyme against inactivation by $H_2O_2$. and copper chelator such as sodium azide also protected the enzyme.

The roots of Korean Astragalus membranaceus Bunge. (Leguminosae), Astragali Radix, were extracted following general methods. During these extractions, first of all, 1,2-benzendicarboxylic acid diisononylester(compound A), ${\beta}-sitosterol(compound\;B)$, $3-0-{\beta}-D-galactopyranosyl-{\beta}-sitosterol(compound\;C)$ and $stigmast-4-en-6{\beta}-ol-3-one(compound\;D)$ were isolated. Three of these compounds were identified the first time isolation from the genus of Astragalus.

한국의 신분류군 식물 1신종,2품종을 다음과 같이 발표한다. Asiasarum koreanum J. Kim et C. Yook (국명신칭: 자주족도리풀)-충북 청원, 음성, 괴산일대 그리고 속리산 근교에 자라고 잎은 어릴 때에 뚜렷한 자주색을 띠고 꽃은 대형(大型)으로 열편(裂片)은 삼각형(三角形)에 접근되며 흔히 끝은 점첨두이고 흔히 꼬이며 꽃받침은 반구형으로 안쪽의 주름이 명확하며 열편에 반점이 있거나 또는 없다. 특히 다른 Asiasarum속에 비해서 화경 $7{\sim}9cm$로 매우 길다. 구환(口環)은 흑자색을 띠며 종자는 바깥면이 흑색으로 폭 1.86 mm, 길이 3.78 mm로 Asiasarum속에서 가장 크다. 화학성분에 있어서도 다른 분류군(족도리풀, 만주족도리풀 등)과는 달리 지상부에 ${\alpha}-copaene$, nerolidol등이 있고 뿌리에는 4-carene. 4-terpineol, ${\alpha-terpineol$이 있어 Asiasarum 속의 신종으로 하였다. Asiasarum sieboldii Miquel for. chungbu ensis C. Yook et J. Kim (국명신칭: 무늬족도리풀)-강원도 내설악 일대와 광덕산 일대에 자라며 개체의 크기는 족도리풀과 유사하나 잎이 얇고 잎의 상면에 유백색의 무늬가 있다. 꽃은 작고 열편은 자주색으로 삼각형이며 구환은 검은색을 띤다. 종자는 폭 1.58mm, 길이 3.2mm이고 뿌리는 빈약하여 약용으로는 곤란하다. 족도리풀과 유사하나 잎에 무늬가 있으므로 족도리풀의 신품종으로 하였다. Asiasarum heterotropoides var. mandschuricum Maximowicz for. glabrata C. Yook. J. Kim et J. Nam (국명신칭: 반들족도리풀)-경기도 광릉일대와 경남 가야산일대에 자라고 잎의 하면은 광택이 있고 상면은 무모(無毛)이다. 꽃은 작고 열편은 삼각형 모양이며 구환은 백색이며 각각의 세 열편은 뒷쪽으로 젖혀져 화판통(perianth tube)와 밀착하며 수평으로 퍼지지 않는다. 뿌리는 매운맛이 뚜렷하다. 형태학적으로 지상부는 만주족도리풀(Asiasarum heterotropoides var. mandschuricum)과 유사하나 잎에 전혀 털이 없고 열편의 모양이 다 르므로 만주족도리풀의 신품종으로 하였다.

From the EtOAc soluble portion of leaves of Fraxinus rhynchophylla, four coumarin glycosides have been isolated and identified as aesculin, cichoriin, scopolin and fraxin.

The effects of acid hydrolysis product of Panax ginseng and MeOH extract of Euphorbia humifusa on the growth of human brain tumor cells were evaluated using U-373 MG human astrocytoma and SK-N-MC human neuroblastoma cells as model cellular systems. These plant extracts induced cytotoxicity in both cells in a dose-dependent manner. These cytotoxic effects were significantly inhibited by GSH, an antioxidant, in both cells. BAPTA/AM, an intracellular $Ca^{2+}$ chelator, significantly blocked the cytotoxic effects of these extracts in U-373 cells, but enhanced these effects in SK-N-MC cells. These results suggest that the plant extracts may be a valuable choice for the studies on the treatment of human brain tumors.

This experiment was conducted to find out the effects of L-phenylalanine on the saikosaponin content of callus induced from Bupleurum falcatum leaf segments. In the fresh and dry weight of callus, the addition of 2,4-D than L-phenylalanine was significantly effective. However, the L-phenylalanine treatment rather than 2,4-D was effective for high saikosaponin accumulation in the callus of Bupleurum falcatum.

In order to product the saikosaponin which is one of the secondary product from Bupleurum falcatum efficiently through the tissue culture, several levels of agar and gellan gum as the gelling agent, 2,4-D as the growth regulator, and L-phenylalanine as the precursor were used with single or combination treatment on MS basal medium. Gellan gum was more effective than agar as the gelling agent in fresh and dry weight increase of callus induced from Bupleurum falcatum leaf segment. Gellan gum medium supplemented with L-phenylalanine produced 1.6 times of fresh weight more than that of agar. The fresh weight was remarkably high in gellan gum when the calli was treated with the combination of 2,4-D and L-phenylalanine similar to the single treatment of 2,4-D or L-phenylalanine. However, the saikosaponin content in callus was high in gellan gum with the single treatment of L-phenylalanine. Especially, the saikosaponin content in gellan gum supplemented with 1.0mg/L L-phenylalanine was 2 times(2.4 mg/g) higher than that in agar medium supplemented with 0.1 mg/L 2,4-D(0.9 mg/g).

Fruits of Rubus coreanum (Korean name: Bog-bun-ja) have been used in oriental traditional medicine as the remedies for impotence, pollution, premature ejaculation and frequency of urination etc. It is known to have phenolic compounds as an astringent. By means of chromatographic methods, four hydrolyzable tannins were isolated from the fruits of R. coreanum. The structures of these compounds were established as gallic acid, 2,3(S)-HHDP-D-glucopyranose, sanguiin H-4 and sanguiin H-6 on the basis of physicochemical and spectroscopic evidences.

In previous studies we reported that the levels of RBC, hemoglobulin (HGB) and hematocrit (HCT) in SAM P6 were increased significantly from 7 day after oral administration of the pilose antler extract, 5 g/kg/day, and were lasted during the study. Thus, this study was performed to elucidate mechanism of erythropoietic action by the extract administration. SAM R1 and SAM P6 were chosen as experimental animals. At age of 12 weeks, pilose antler extract were given 0.3 and 5 g/kg/day (p.o.) each for 0, 7 and 14 days in both animals. Complete blood cells such as RBC, HGB, and HCT were counted. And plasma concentration of erythropoietin (EPO) which is the major regulator of erythropoiesis was measured using $^{125}I-antierythropoietin$ IgG. Total iron concentration in plasma was also analyzed. The levels of RBC and HCT were increased significantly after administration at both doses of 0.3 and 5 g/kg/day in SAM P6, however, these were increased only at dose of 5 g/kg/day in SAM R1. The plasma EPO concentration was increased significantly after administration in SAM P6. The plasma concentrations of total iron were significantly decreased after administration of the extract in SAM P6. These results suggest that the changes in erythropoietic effects after the administration of pilose antler extract may be mediated, at least in part, through the change in the plasma EPO.

We previously reported the morphological, physiochemical characteristics of Alismae Rhizoma. In this study, we examined the antimicrobial and antifungal activities of 85% ethanol and water extracts of Alismae Rhizoma using disc method. Ethanol extracts of Korean Alismae Rhizoma showed antimicrobial activity on B. subtilis and S. aureus on the concentration dependent manner, whereas, Chinese Alismae Rhizoma revealed antimicrobioal activity on E. coli and N. gonorrhoeae. The water solution(500 mg/ml) of each lyophilized powder of aqueous extracts didn't show any antimicrobial activity on B. subtilis, S. aureus and E. coli, on the contrary, they stimulated cell growth slightly. Ethanol extracts of Korean samples showed antifungal activity on A. niger, but not on C. albicans. Neither A. niger nor C. albicans were affected by Chineses samples.

The objective of this reseach is to find new antitumoral substances from natural products. Several of natural products have been used as food that were isolated into hexane(Hex.) and/or ethylacetate(EtOAc) extracts. we have tested cytotoxicities of these plants against human solid tumor cells. The cytotoxic activity of these plants were tested using Sulforhodamin B(SRB) assay. Hexane extracts of Chrysanthemum sinense, Allium tubersum. Beta vulgaris, Ixeris dentata have revealed cytotoxicities against five human solid tumor cells, and its cytotoxicities of each cell line were $10-100\;{\mu}l/ml$ ED50 (Effective dose that cause 50% inhibition of cell growth in vitro)

Spiraea prunifolia var. simpliciflora (Rosaceae) is a deciduous. latifoliate shrub growing in most parts of Korea. The roots of this plant have been used for malaria, as antipyretics and emetics. From the roots of this plant, sterol glycoside and two triterpenoids were isolated and the structures were elucidated by chemical and spectroscopic methods. They were identified as $3{\beta}-hydroxyurs-12-ene-28-oic$ acid (ursolic acid.1), $2{\alpha}$, $3{\beta}$, $19{\alpha}-trihy-droxyurs-12-ene-28-oic$ acid (tormentic acid.2) and ${\beta}-sitoste-rol-3-O-{\beta}-D-glucopyranoside$ (3).

This study was attempted to investigate the effect of Composite Preparation (Sam Whang Sasim-Tang: SWST, Whang Ryun Haedok-Tang: WRHT) on the activities of GOT and GPT, the content of total lipids, triglyceride, total cholesterol. Phospholipid and ${\beta}-lipoprotein$ in the serum, and the change ratio of body and liver weight in the experimentally induced hyperlipemic rats, making use of the extract 150, 200, 300 and 500 mg/kg p.o. Significant test was performed by comparision with the values of corresponding experimentally hyperlipemic rats. The activities of S-GPT was significantly decreased in all dose of SWST and WRHT respectively. The activities of S-GPT was significantly decreased in dose of SWST 500 mg/kg and in all dose of WRHT respectively. The content of total lipids, triglyceride, total cholesterol and phospholipids were significantly decreased in all dose of two the extract. The rate of decrease on total lipids and triglyceride were remarkable in dose of SWST 300, 500 mg/kg and WRHT 200, 500 mg/kg, and then the content of total cholesterol was more remarkable in dose SWST 300, 500 mg/kg, and WRHT 150, 300 mg/kg, and the content of phospholipids was more remarkable in dose of the extract 150, 200 and 300 mg/kg. The content of ${\beta}-lipoprotein$ was significantly decreased in dose of WRHT 150, 200 and in all dose of SWST. Increase ratio of the body and liver weight were significantly decreased in dose of two the extract 300, 500 mg/kg respectively.
