Comparison of Organic Acid Contents and Xanthine Oxidase Inhibitory Activities of Commercial Fruit Juices and Vinegars (시판 과일 주스와 식초의 유기산 함량과 Xanthin Oxidase 저해 활성 비교)

  • Xanthine oxidase (XO) inhibitors play an important role in the treatment of gout and many other diseases related to superoxide anion metabolism. In this study, four commercial fruit juices and three vinegars were evaluated for their inhibitory activity of XO (XOI), as well as contents of organic acids by HPLC with UV detection. Five different organic acids were detected in commercial samples: acetic acid and malic acid were the most prominent in vinegars and fruit juices, respectively. The vinegars showed high XOI activity (33.8~64.9%) related to the great concentration of acetic acid ($R^2=0.7192$). The presence of acetic acid in vinegar could be responsible for its XOI effect.

Antioxidant Activity and Agronomic Characteristics of Colored Waxy Corns (유색 찰옥수수의 항산화력 및 작물학적 특성)

  • This study was carried out to evaluate major agronomical characterization and phenol compound contents, Xanthine oxidase inhibitory activity (XO), Catalase activity, Superoxide dismutase activity (SOD) and 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) radical sacvenging activity were analyzed in colored waxy corns. The mean of stem height and ear length were 248.8 cm and 18.6 cm, respectively. The pericarp thickness in CNU108 $(30.3{\mu}m)$ was thinner than other hybrids. The period of tasseling days in CNU69 and CNU202 were very shorter than other hybrids (59 days). 100-kernel weight of CNU50 was 35.6 g and heavier than the others. The antioxidant activities such as xanthin oxidase (XO), catalase and superoxide dismutase (SOD) were higher in CNU70 and CNUI 38. Especially highest EDA (electron donating ability) in DPPH radical scavenging effect was 94.8% and 94.6% in CNU160 and CNU193, respectively. In the results, the antioxidant enzyme activity and antioxidant acticity were higher in CNU109 and CNU 34 hybrids. The hybrids, CNU34, CNU70, CNU108, CNU 138 and CNU193 may be considered higher functional color waxy corn.

Antioxidant and Oxygen Radical Scavenging Capacities of the Extracts of Pear Cactus, Mulberry and Korean Black Raspberry Fruits

  • In this study, we evaluated the phytochemical compositions of the fruits of the pear cactus, mulberry, and Korean black raspberry to determine how they related to antioxidant capacity. Specifically, the total soluble polyphenol, flavonoid and anthocyanin contents were determined, and the phenolic compounds were evaluated by HPLC. In addition, the antioxidant capacity was evaluated by electron paramagnetic resonance (EPR), SOD-like activity and nitrate scavenging activity. Korean black raspberry was found to have the highest level of phytochemicals, followed by mulberry and pear cactus (p<0.05). Furthermore, EPR revealed that Korean black raspberry extract (100 ${\mu}g$/mL) removed 78.6% of the superoxide radicals in a xanthin-xanthin oxidase. Additionally, when the removal of hydroxyl radicals was evaluated, the extract of pear cactus showed 50% scavenging activity at 1000 ${\mu}g$/mL. Korean black raspberry extract showed the highest SOD-like activity, which was 44% (p<0.05). Finally, the nitrate scavenging activity of both extracts of mulberry and Korean black raspberry were found to be 93%, while that of the pear cactus was 63% at pH 1.2 (p<0.05).

  • Biological activities and anticarcinogenicity of Korean Pear peel were investigated, Electron donating activity and superoxide dismutase (SOD)-like activity of fraction II, III were up to 80% and 50-60% at 50 ppm, respectively, Inhibitory effects on xanthine oxidase were about 80% at 50 ppm, breast adenocarcinoma was about 60% at 2,000 ppm, higher III than II Inhibitory effect on prostate adenocarcinoma was about 23% at 500 ppm. In conclusion, Korean pear peel was expected to use as a functional material.

Effects of Green Tea Catechin on the Superoxide Dismutase, Glutathione Peroxidase and Xanthine Oxidase Activites of Kidney in Diabetic Rats (녹차 Catechin이 당뇨쥐 신장조직의 Superoxide Dismutase, Glutathione Peroxidase 및 Xanthine Oxidase)

  • The purpose of this study was to investigate the effect of green tea catechins on the antioxidative defense enzyme activity of kidney in diabetic rats. Sprague-Dawley male rats weighting 100$\pm$10g were randomly assigned to one normal and three STZ-induced diabetic groups; catechin free diet(DM-0C group), 0.25% catechin diet(DM-0.25C group) and 0.5% catechin diet(DM-0.5C group). Diabetes was induced by intravenous of 55mg/Kg body weight of STZ in sodium citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Rats were sacrified at the 6th day of diabetic states. Superoxide dismutase(SOD) activity in kidney was decreased by 25% and 20% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. Glutathione peroxidase(GSHpx) activity in kidney was were no significant differences the diabetic groups compared to normal group. Xanthin oxidase(XOD) activity was increased by 110% and 63% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. The contents of superoxide radical(O$_2$)in kindney were 116% and 33%, respectively, higher in DM-0C and DM-0.25C groups than normal group. DM-0.5C group and normal groups were similar levels in their superoxide radical contents of kidneys. Levels of TBARS(thiobarbituric acid reactive substance) in kidney were increased by 62% in DM-0C group, when compared with normal group, but those of DM-0.5C group were similar to that of normal groups. These results indicate that free radical generation system was weakened and free radical scavenger system was enhance in kidney of STZ-induced diabetics rats by dietary catechin. Thereby it may reduce renal disorders such as oxidative damage and aging of tissue.

  • In this study, to evaluate the anti-obesity effects of mulberry leaf tea and its fermented product by Monascus pilosus, we investigated body and organ weight, blood and liver biomarkers in mice fed 1% tea infusions instead of water for 8 weeks. Mice were divided into three groups such as a normal control (NC), unfermented mulberry leaf tea infusion (UMI) and fermented mulberry leaf tea infusion (FMI). Although it is not significant, tea infusion groups showed reduction of body weight gains compared with NC group. Moreover, contents of LDL-cholesterol and lipid peroxide (LPO), altherogenic index, and xanthin oxidase (XO) activity were significantly decreased, and glutathione S-transferase (GST) activity was significantly elevated. The results from this study suggested that UMI and FMI may have an anti-obesity activity, upregulate antioxidant enzymes and reduce levels of oxidants related to liver damage.

Antioxidant Activities of Korean Perilla Leaves (Perilla frutescens) by Various Cultivars (잎들깨 품종에 따른 추출물의 항산화 활성 및 생리활성 평가)

  • This study aimed to investigate the biological activities, including polyphenolic content, flavonoid content, and antioxidant activities, of various cultivars of Korean perilla leaves. The results indicated that among nine cultivars (Namcheon dlggae, Saedora, Nulbora, Donggel 1, Donggell 2, Soim, Sangyeop, Somirang, and Saebom) of perilla leaves, the total polyphenolic content (gallic acid equivalent mg/g, GAE) was the highest in "Nulbora," while it was lowest in Namcheon dlggae. Moreover, flavonoid content in the extracts of nine cultivars leaves was in the range of 132.93~268.50 mg catechin equivalent/g sample. The antioxidant effects of the perilla leaves were determined using two different in vitro bioassays measuring DPPH and ABTS radical-scavenging activities. The results revealed that antioxidant activity was also higher in "Nulbora" compared with other cultivars. Xanthin-oxidase-inhibition activity ranged from 65.65% to 80.58%, with "Nulbora" exhibiting the highest activity, although the difference with other cultivars was not significant. "Nulbora" extracts reduced the expression of pro-inflammatory genes and several cytokines, including IL-6 activation induced by LPS in macrophages in the range of 100-50 ㎍/mL. These results suggest that extracts from perilla leaves can be used as bioactive and functional materials that could be important in industrial applications in the future.

Quality Characteristics and Content of Polysaccharides in Green Tea Fermented by Monascus pilosus

  • In this study, we designed a method to manufacture elevated fermented green tea by using Monascus pilosus, which is known as a functional microbe, and observe its antioxidant abilities and quality characteristics. The water-soluble substance (WSS) content of the fermented tea by M. pilosus (FTM) was lower than that of the non-fermented tea (NFT), although the alcohol-insoluble substance (AIS) content of the FTM was higher than that of NTM. On the other hand, the fractionated distilled water-soluble polysaccharide (DWSP), CDTA-soluble polysaccharides (CDSP), sodium carbonate-soluble polysaccharide (SCSP) and KOH soluble hemicellulose (HC) obtained from the AIS of the FTM was markedly higher than that of NFT. In the antioxidant parameters, the electron donating ability of all fractions, except HC, extracted from FTM was higher than that of NFT, and iron chelating ability of all fractions, except CDSP, extracted from FTM was higher than that of NFT. Whereas the DWSP and SCSP obtained from the FTM were higher than that of NFT, the activity of the HC fraction from both NFT and the FTM could not be detected. In addition, the xanthin oxidase (XO) inhibitory activities of the DWSP, CDSP and the SCSP obtained from the NFT were significantly higher than that of FTM, the aldehyde oxidase (AO) inhibitory activities of the DWSP and SCSP extracted from the FTM were markedly higher than that of the NFT. Meanwhile, the acceptance of NFT and FTM had no significant difference, while the quality of aroma, taste and mouthfeel of the FTM was higher than that of NFT. These results suggest that the post-fermented tea by Monascus microorganisms may be responsible for functional components as well as contribute to the improvement of the tea quality.

Antioxidant Activities of Rice Bran Extracts for Wellness Convergence (융복합적인 웰리스를 위한 미강추출물의 항산화 활성)

  • The aim of present study is to investigate antioxidative effect of the Rice Bran Extracts and Defatted Rice Bran Extracts. Rice Bran Extracts used Rice Bran Water Extract, Rice Bran Ethanol Extract, Defatted Rice Bran Water Extract, Defatted Rice Bran Ethanol Extract. This study was carried out to examine quenching effects of Rice bran extracts on DPPH-, Riboflavin-, and Xanthin oxidase- originated superoxide activities. In addition, in order to determine whether Rice Bran Extract can be safely applied to human skin, the cytotoxic effects of Rice Bran Extract in Human Dermal Fibroblast cells were determined using MTS Assay. These results demonstrated that RBE and DRBE had anti-oxidative properties and did not induce the cytotoxic effects in Human Dermal Fibroblast cells. Therefore, these findings suggest that anti-oxidative properties of RBE and DRBE may be considered convergence with skin care.

Nutritional Characteristics and Physiological Functionalities of Polyplant Extracts from Some Vegetables and Medicinal Plants (몇 가지 채소와 약용 식물을 이용하여 제조한 식물 복합 추출물의 영양학적 특성과 생리기능성)

  • The nutritional characteristics and functionalities of the polyplant extracts(PPE) from the extraction of various plants were assessed for the development of a new functional food. The PPE contained 42.95 g of glucose, 22.61 g of fructose, and 12.48 g of sucrose per 100 g of PPE as free sugar, and its reducing sugar contents was measured at 480.6 mg per g of PPE. Among free amino acids, proline contents was the highest at 0.58 g per 100 g of PPE, and it contained only 12.65 mg of malic acid and 6.92 mg of tartaric acid per g of PPE as an organic acid. The antioxidant activity of PPE was the highest at 91.6%, whereas SOD-like activity and xanthin oxidase inhibitory activity, which are associated with anti-aging effects, were 21.6% and 9.3%, respectively. HMG-CoA reductase inhibitory activity was 10.0% and anti-dementia acetylcholinesterase inhibitory activity was quite low(5.1%).
