조위승청탕의 마우스 경구 단회 투여독성 평가 (Single Oral Dose Toxicity Test of Choweseuncheng-tang, a Polyherbal Formula in ICR Mice)

  • The object of this study was to evaluate the single dose toxicity of Choweseuncheng-tang (CWS), a polyherbal formula have been traditionally used as prevention or treatment agent for various diseases as Tae-eumin prescription on Korean medicinal theory, Sasang-euihak, in male and female mice. Aqueous extracts of CWS (yield = 11.00%) was administered to female and male mice as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy; organ weight and histopathology of 14 principle organs were also examined. As results, we could not find any CWS treatment related mortality and clinical signs, changes in the body and organ weights, gross findings and changes in histopathology of principle organs, except for some dose-independent accidental findings. The results obtained in this study suggest that the 50% lethal dose and approximate lethal dose of CWS aqueous extracts in both female and male mice were considered as over 2,000 mg/kg, the limited highest dosage recommended by KFDA Guidelines, and can be safety used in clinics.

절식요법이 체성분 변화에 미치는 영향: 후향적 관찰연구 (The Changes of Body Compositions after Modified Fasting Therapy: A Retrospective Observational Study)

  • Objectives: The purpose of this study was to investigate the effects of modified fasting therapy on the changes of body compositions. Methods: We analyzed the medical records of 33 patients, who carried out modified fasting therapy at Dunsan Korean Medicine Hospital of Daejeon University from January 1st, 2011 to December 30th, 2015. They went through reducing food intakes period (7 days), fasting period (14 days) and refeeding period (14 days). Body compositions (weight, body mass index, skeletal muscle mass, percent body fat, basal metabolic rate, waist-hip ratio, visceral fat area) were reviewed at each state. And then the data was analyzed. Results: The body composition values (weight, body mass index [BMI], skeletal muscle mass, percent body fat, basal metabolic rate, waist-hip ratio, visceral fat area) decreased during the fasting therapy period, as a whole. The weight, BMI, percent body fat and visceral fat area decreased during the reducing food intakes period, the fasting period and the refeeding period. The skeletal muscle mass and basal metabolic rate significantly decreased during the reducing food intakes period and the fasting period, and insignificantly increased during the refeeding period. Conclusions: Results from this investigation showed that modified fasting therapy using fermented herbal medicine have positive effects on changes of body compositions.

A Repeated-dose Oral Toxicity Study of Orostachys japonicus Extract in Sprague-Dawley Rats

  • A 12-week repeated-dose oral toxicity study of water-soluble Orostachys japonicus extract (WOJ) was performed in Sprague-Dawley (SD) rats of both genders. Each group of ten rats was orally administered in doses of either 0 or 250 mg/day over a 12-week period. As a result, no WOJ-related changes were observed in terms of survival rate, clinical signs, body weight, or food intake. In addition, no difference in organ weight between the control and treated groups was detected. Furthermore, serum biochemistry parameters revealed some changes within normal ranges although significant decreases in total-bilirubin in the females. In spite of some alterations in serum biochemistry, the clinical signs, body weight changes from food intake, and autoptical remarks indicated that WOJ was not toxic. This study suggests that repeated treatment of O. japonicus very low toxicity and the NOAEL (no observed adverse effect dose) of WOJ exceeds 250 mg/kg in the SD rats.

  • Objectives: This study was to evaluate the single dose toxicity of Sobokchuko-tang (SBC) in male and female mice. Methods: Aqueous extract of SBC (yield=6.60%) was administered to female and male mice as an oral dose of 2,000, 1,000 and 500 mg/kg (body weight) according to the recommendation of Korea Food and Drug Administration (KFDA) Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy; organ weight and histopathology of 14 principle organs were also examined. Results: we could not find any SBC treatment related mortality and clinical signs, changes in the body and organ weights, gross findings and changes in histopathology of principle organs, except for pharmacological immunomodulatory effects related findings including significant increases of submandibular lymph node weights, hypertrophy and hyperplasia of lymphoid cells in the submandibular lymph nodes restrictly detected in 2,000 mg/kg treated female and male mice with some sporadic accidental findings. Conclusions: The results obtained in this study suggest that the 50% lethal dose and approximate lethal dose of SBC aqueous extracts in both female and male mice were considered as over 2,000 mg/kg, the limited highest dosage recommended by KFDA Guidelines, and can be safety used in clinics.

문헌 고찰을 통한 비만 치료 한약의 장내미생물 변화 연구 (A Study on Changes in the Gut Microbiome of Herbal Medicines for Treating Obesity Using Literature Review)

  • Objective This study identified useful herbal medicines and prescriptions for obesity treatment by analyzing herbal medicines and prescriptions that showed meaningful results in weight loss by changing the gut microbiota. Methods Using PubMed, we selected and studied 23 papers showing meaningful results on weight loss through changes in the gut microbiota using herbal medicines. Result Of the 23 selected papers, 17 were of studies using herbal medicines, and 6 of studies using prescriptions. Pomegranate peel, Flos Lonicera, Rehmannia glutinosa, Rhein, Coix seed, Platycodon grandiflorus, mulberry leaves, Poria cocos, asperuloside, Bupleuri radix, Astragalus polysaccharides, Ephedra sinica, Ganoderma lucidum, Hirsutella sinensis, Caulis spatholobi, aconite, and Bletilla striata were used as herbal medicine. Linggui zhugan-tang, Bofutsushosan, Shenling baizhu powder, Chowiseungcheng-tang, Daesiho-tang, and Yijin-tang were used as prescription. Conclusion Seventeen herbal medicines and six prescriptions associated with meaningful results in weight loss through changes in the gut microbiota, suggest the possibility of treatment and prevention of obesity through herbal medicine.

계측장치 실장 차량을 이용한 동적 하중 모니터링 연구 (Study on the Dynamic Load Monitoring Using the Instrumented Vehicle)

  • 주행 차량의 축하중은 저속 혹은 고속 축중기(WIM)에 의하여 측정 할 수 있으나, 주행 차량의 샤시, 축 구조 등과 같은 차량 고유 특성과 주행 속도, 도로의 평탄도 등과 같은 주행 환경 특성에 따라 동적으로 변화하며, 이러한 순간적인 동적 하중 변화에 의해 정적 상태에서 측정된 기준 중량과 오차가 발생하게 된다. 본 연구에서는 향후 무인 과적단속 체계 도입에 앞서, 주행 차량의 동적 하중 변화 특성을 파악하여 통제 불가능한 환경적 기본 오차의 범위에 대해 분석하고, 고속 축중기의 중량정확도 성능평가 기준에 대한 척도를 적절히 설정하기 위한 실차 시험을 수행하였으며, 주요 시험 결과는 다음과 같다. 첫째, 총중량의 경우 저속일 때 약 1%, 고속일 때 약 4%의 변화가 나타났고, 축하중의 경우 저속일 때 약 1-3%, 고속일 때 약 2-9%의 변화가 나타났으며, 이러한 현상은 단일축보다 그룹내 개별축에서 더 크게 나타났다. 둘째, 정상 평탄도 구간에 비해 충격 구간에서는 총중량의 경우 최대 약 8배, 축하중의 경우 최대 약 3~12배의 변화가 나타났으며, 이러한 동적 하중 변화의 진동 주파수는 2.4-5.8Hz로 나타났으며, 약 30m를 주행한 후에 정상 상태의 진폭으로 수렴하는 것으로 분석되었다.

인삼에탄올엑기스가 연독성에 미치는 영향 (The Effects of Ginseng Ethanol Extracts on the Toxicity of Lead acetate in Mice)

  • The effects of ginseng ethanol extract on the toxicity of lead acetate in mice were examined. Mice were given intraperitoneally daily doses of lead acetate 50 mg/kg with ginseng ethanol extract 50 mg/kg, 100 mg and 200 mg/kg for 3 weeks. The exposure of lead acetate showcd the toxicity at all experimental assay such as the gain of body weight, the ratio of some organs weight to body weight, serum transaminase activity and creatinine value, hematocrit and WBC counts. These toxicities were inhibited significantly by the ginseng ethanol extract administration. The 50 mg/kg and 100 mg/kg administration of ginseng ethanol extract inhibited histopathological changes on kidney by lead acetate, whereas the 200 mg/kg administration of the fraction enhanced histopathological changes.

유기게르마늄(Ge-132)의 랫드와 마우스에 대한 급성경구독성 (Acute toxicity of Organogermanium, Ge-132 in Rats and Mice)

  • The acute oral toxicity of organogermanium, Ge-132 was evaluated in rats andmice. The changes of body weight and clinical signs were observed for 14 days after the oral administration of Ge-132, from 0.31 g/kg up to 5 g/kg for SD rats and from 1.25 g/kg up to 5 g/kg for ICR mice. No death and toxic effects were observed for 14 days. The body weight of rats was significantly decreased 1 day after the administration in the maximum dosing group, but the decrease of body weight returned to control level 3 days after dosing. No significant changes in 132. Therefore, Ge-132 has no special toxic effects up to 5 g/kg, and LD* values of Ge-132 Ge-132 are above 5 g/kg in rats and mice.

Mouse Single Oral Dose Toxicity Study of DHU001, a Polyherbal Formula

  • This study was conducted to obtain acute information of the oral dose toxicity of DHU001, a polyherbal formula in male and female mice. In order to calculated 50% lethal dose ($LD_{50}$) and approximate lethal dose (LD), test material was once orally administered to male and female ICR mice at dose levels of 2000, 1000, 500, 250 and 0 (vehicle control) ml/kg (body weight). The mortality and changes on body weight, clinical signs, gross observation, organ weight and histopathology of principle organs were monitored 14 days after treatment with DHU001. We could not find any mortalities, DHU001 treatment-related clinical signs, changes on the body and organ weights, gross and histopathological findings. The results obtained in this study suggest that $LD_{50}$ and approximate LD in mice after single oral dose of DHU001 were considered over 2000 mg/kg in both female and male mice.

  • Water extracts of 21 traditional Korean herbal drugs were prepared, and a dose range of 100 mg/kg to 400 mg/kg was administered orally into mice once a day for five days. Changes of serum enzyme activities of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), blood urea nitrogen (BUN), alkaline phosphatase, body weight changes and histo pathological examination of various organs were investigated. Water extract of Ephedra Herba caused severe body weight loss at a dose of 100 mg/kg and death from a dose level of 200 mg/kg by oral administration. Angelica koreanae Radix and Anthrisci Radix showed a slight body weight loss and damages to liver and kidney.
