추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. Antioxidant Activities of Various Solvent Extracts from Ginseng (Panax ginseng C.A. Meyer) Leaves

  • Water, methanol and ethanol extracts of ginseng leaves were assayed for total phenolics and flavonoids, ascorbic acid, cupric and ferrous ion chelating activities, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power (FRAP) assay and ABTS radical cation decolourization (TEAC) assay for their antioxidant properties. The ethanol extract of ginseng leaves contained significantly (p<0.05) higher amounts of total phenolics and flavonoids (600.57 and 1701 mg/100 g) than methanol (374.43 and 1512.64 mg/100 g) and water extracts (248.30 and 680.05 mg/100 g). Among solvent extracts of ginseng leaves, the ethanol extract showed the most powerful antioxidant activities. However, the ferrous ion chelating activity of ginseng leaf extracts were lower than the cupric ion chelating ability. These differences in concentrations of key antioxidants among various solvent extracts seemed to be responsible for their differences in antioxidant activities. These results suggest that ethanol extract of ginseng leaves has the most effective antioxidant capacity compared to the methanol and water extracts tested in the present study. Thus, it can be applied for the effective extraction of functional material from ginseng leaves for the usage of pharmaceutical and/or food industries.

짚신나물, 삼백초의 항산화와 항암활성 효과 (Antioxidant and Anticancer Effects of Agrimony (Agrimonia pilosa L.) and Chinese Lizardtail (Saururus chinensis Baill))

  • The antioxidant activities of Agrimony (Agrimonia pilosa L.) and Chinese lizardtail (Saururus chinensis Baill) according to extraction methods were measured. SOD-like activity showed greater antioxidant effects with ethanol solvent than those with water. Ethanol extracts of Agrimony leaves showed the highest SOD-like activity of 94.4%. SOD-like activity differed according to the extraction solvents. The contents of polyphenolic compounds were higher in water extracts than those in ethanol extracts. The contents were 161.4 mg for Agrimony roots, 100.2 mg for Agrimony leaves, and 79.1 mg for Agrimony stalks in order. EDA in Agrimony leaves that were highest among medicinal plants were 83.4% in the water extract and 81.7% in the ethanol extract. The anticancer effects of the extracts by water and ethanol from Agrimony and Chinese lizardtail were experimented. The growth of stomach cancer cells, SNU-719 was inhibited 94.5% by the hexane fractions of Agrimony and also the growth of liver cancer cells, Hep3B was inhibited 83.2% by the hexane fractions of Agrimony, while the growth of normal cell, DC2.4 was not affected.

Loquat (Eriobotrya japonica) leaf extract inhibits the growth of MDA-MB-231 tumors in nude mouse xenografts and invasion of MDA-MB-231 cells

  • BACKGROUND/OBJECFTIVES: The present study was conducted to examine the inhibitory effect of loquat leaves on MDA-MB-231 cell proliferation and invasion. MATERIALS/METHODS: Female athymic nude mice were given a subcutaneous (s.c.) inoculation of MDA-MB-231 cells and randomly grouped to receive a s.c. injection of either 500 mg/kg ethanol, water extract or vehicle five times a week. Tumor growth, mitotic rate and necrosis were examined. MDA-MB-231 cells were cultured with DMSO or with various concentrations of loquat water or ethanol extract. Proliferation, adhesion, migration, invasion and matrix metalloproteinase (MMP) activity were examined. RESULTS: Tumor growth of xenograft nude mouse was significantly reduced by loquat extracts. The results of mitotic examination revealed that loquat extracts reduced tumor cell division. Both ethanol and water extracts significantly inhibited MDA-MB-231 cell proliferation. The protein expression of ErbB3 was significantly down-regulated by loquat leaf extracts. Loquat leaf extracts increased apoptosis of MDA-MB-231 cells following 24 hour incubation and the ethanol extract was more potent in inducing apoptosis than the water extract. Furthermore, loquat extracts inhibited adhesion, migration and invasion of MDA-MB-231 cells. MMP activity was significantly inhibited by loquat extracts. CONCLUSION: Our results show that extracts of loquat inhibit the growth of tumor in MDA-MB-231 xenograft nude mice and the invasion of human breast cancer cells, indicating the inhibition of tumor cell proliferation and invasion.

  • 추출조건에 따른 지골피의 몇가지 물리적 성질 및 화학성분조성을 조사하였다. 1. 고형분과 총당함량은 물추출이 에탄올 추출물보다 높았으나 반면에 탄닌산함량은 에탄올 추출물에서 높았다. 2. 지골피 물추출물이 탁도와 갈색도가 에탄올추출물보다 높았으며 색도 중 적도와 황색도는 유사하였다. 3. 용매별 분획추출결과 수율은 물 > n-부탄올 > 에틸에텔 > 초산에 틸 분획순으로 증가하였다.

사과 부위별 항산화 성분 및 항산화 활성 (Antioxidant Component and Activity of Different Part Extracts in Apple (Malus domestica cv. Fuji))

  • To improve the utility of apple, apple flesh (Malus domestica cv. Fuji), pomace and peels were dried and then extracted using solvents (water, 70% and 100% ethanol). The contents of polyphenols, flavonoids, ascorbic acid, ursolic acid and antioxidant activities were measured as extracted from the apple flesh, pomace and peel. The content of polyphenols was the highest in the apple peel when 100% ethanol was extracted. The content of flavonoids was about 3~8 times higher in apple peel than those of flesh and pomace, and 70% ethanol extract was the highest content. Ascorbic acid content was the highest in apple peel as well as flavonoid content. The content of ursolic acid in the apple flesh was not detected in water and ethanol extraction. In the case of apple pomace and peel, ursolic acid was not detected in water extract but it was detected when ethanol was extracted. The content of ursolic acid was highest in the apple peel. In the case of antioxidative activity, DPPH and ABTS radical scavenging ability showed the highest ability when apple peel was extracted with 70% ethanol.

산화적 손상에 대한 화피(樺皮)의 효능 - 왕벚나무와 자작나무 수피의 항산화능 비교 평가- (Comparative Study of Antioxidant abilities on Prunus yedoensis and Betula platyphylla var. japonica)

  • The aim of the present study was to investigate the antioxidant efficacies between extracts of Prunus yedoensis(PY) and Betulae platyphyllae var. japonica(BP). HPLC pattern was different between barks and extract solvents. Content of total phenolic compound was the highest in ethanol extract of BP(382.201 mg/g ext.) and its content was 1.9 times higher than that from the water extract of PY. Total antioxidant efficacy also was the highest in ethanol extract of BP (292 copper reducing equivalents). Nitric oxide scavenging activity was almost 70% in ethanol extract of BP treated 200 ug/ml and it was higher than positive control(ascorbic acid). DPPH radical scavenging ability was up by 80% in all samples. ABTS cation decolorization from each barks was activated over 85% in all samples at 100 ug/ml concentration, especially, the activity was the highest (94.4%) in ethanol extract of BP. Hydrogen peroxide scavenging activities were also highest (45%) in ethanol extract of BP at 200 ug/ml concentration and were as high as positive control. Stimulation of the macrophages RAW 264.7 cells with lipopolysaccharide (LPS) increased intracellular ROS levels and ethanol extract of BP at 200 ug/ml concentration reduced ROS levels up to 41 %. The results indicated that the barks of PY and BP has potent antioxidant activities and ethanol extract of BP of them has the highest antioxidant activities.

매생이 추출물의 생리 활성과 항산화 활성에 관한 연구 (A Study on Physiological Activity and Antioxidative Activity of Maesangi(Capsosiphon fulvescens) Extract)

  • Physiological activity and antioxidative activity of Maesangi(Capsosiphon fulvescens) extracts with distilled water or 95% ethanol were investigated. For the evaluation of physiological and antioxidative activities, some evaluation assay methods such as measurement of Hunter color value, chlorophyll a/b value, total phenolics, reducing power and thiobarbituric acid(TBA) value of soybean oil were used. Proximate composition and mineral contents of Masaengi were orders of crude protein>crude fiber>moisture>crude ash>crude lipid, and K>Ca>Mg>Na>P>Fe>Zn, respectively. In ethanol extract, the content of total phenolic compounds in Maesangi was determined to half times of that in Dasima(Laminaria). The reducing power of Maesangi-ehtanol extract was about 5 % of vitamin C and was lower than that of Dasima-ethanol extract. The TBA value of Maesangi-ethanol extract on soybean oil oxidation was about 47 % and 68.4 % to control in three and eight days oxidation, respectively. But TBA value difference was not observed significantly with the dosage below 5mL of ethanol extract.

뽕잎 추출물의 돌연변이 억제효과 (Antimutagenic Effect of Mulberry Leaf Extract)

  • Background: The present study was carried out to asses whether mulberry leaves (MLs) have the potential to inhibit the mutagenic effect of cigarette smoke condensates (CSCs). Methods and Results: ML powder was extracted with 70% ethanol, and a yield of 35.1% by weight was obtained. The 70% ethanol extract of ML was further extracted sequentially using diethyl ether, chloroform, butanol, dichloromethane and water. The crude 70% ethanol extract of MLs and its solvent fractions did not show any mutagenic effect when tested at concentrations up to 1 mg/plate against Salmonella typhimurium TA98. In contrast, the crude 70% ethanol extract showed an inhibitory activity against the mutagenicity of CSCs in the presence of S-9 mixture. Among the solvent fractions, the diethyl ether fraction showed the highest inhibitory activity, which increased in a dose-dependent manner, inhibiting mutagenesis by approximately 97.1% at a concentration of 1 mg/plate. Conclusions: In this study, we found that a crude 70% ethanol extract of MLs and the diethyl ether fraction themselves are potentially not mutagenic, but inhibit the mutagenic effect of CSCs.

진달래꽃(Rhododendron mucronulatum Turczaninow) 추출물의 항산화, 항암 효과 및 tyrosinase 저해활성 (Antioxidant, Anticancer and Tyrosinase Inhibition Activities of Extracts from Rhododendron mucronulatum T.)

  • Several biological activities of Rhododendron mucronulatum were investigated. The electron donating abilities of ethanol extracts of Rhododendron mucronulatum were more than 90% at 100 ppm, 500 ppm and 1,000 ppm. Xanthine oxidase was inhibition about 46% by the ethanol extracts of R. mucronulatum at 500 ppm 48% of tyrosinase activity relating to skin-whitening was shown at 1,000 ppm. Uniquely, the anti-microbial effects of water extract and ethanol extract were shown only on Staphylococcus aureus. The water extract 1 mg/disc showed the higher activity than ethanol extract. The growth inhibition effect of each sample on lung cancer (A549) and melanoma (B16F10) cell lines were over 70% at 1,000 ppm, while the effects on the melanoma (G361) and liver cancer (HepG2) were about 50% at the same concentration.
