In vitro Evaluation of Biological Activities of Wa-song (Orostachys japonicus A. Berger) and Korean Traditional Plants Mixture (와송과 한약재 복합물의 in vitro 생리활성 평가)

  • This study was carried out to determine the biological activities of Wa-song (Orostachys japonicus) hot water extracts. Four types of extract samples were prepared, including Wa-song, traditional plants mixture [PM; mixture of Baekbokryung (Poria cocos), Changchul (Atractylodis rhizoma), and Sa-in (Amomum xanthoides)], and two different ratio composites of these (mixture of PM and Wa-song extract, 1:1 (v/v); PMO-1 and 1:3 (v/v); PMO-3). Their biological activities were measured using various in vitro assays. Total phenolic and flavonoid contents of PM were higher compared to those of Wa-song, and those of PMO-1 were higher than those of PMO-3. Further, PMO-1 contained higher ABTS and DPPH radical scavenging, reducing power, and nitrite scavenging activities than PMO-3. On the contrary, PMO-3 contained higher tyrosinase and inhibitory activities of MCF-7 and HT-29 cancer cells than PMO-1. According to the results, biological activities of PMOs were significantly higher than those of Wa-song extract and PM in in vitro assays. Therefore, we expect that PMOs could show higher biological activities than Wa-song extract alone in vivo.

Antioxidant Activity of Wa-song (Orostachys japonicus A. Berger) According to Drying Methods (건조 방법에 따른 와송의 항산화 효과)

  • Antioxidant activity of wa-song (Orostachys japonicus A. Berger) was analyzed to clarify the influence of extractive solvent and drying method such as sun, hot-air and freeze drying. The contents of total phenols and flavonoids were significantly higher in 95% ethanol extracts than water extracts. Ability of reducing power and DPPH radical, hydroxyl radical and nitrite scavenging ability were higher in the order of hot-air dried sample> freeze dried sample> sun dried sample and these abilities were also higher in 95% ethanol extracts than water extracts. In conclusion, antioxidant activities of wa-song extracts were in proportion to the contents of total phenols and flavonoids. Also, hot-air drying is the superior method for the enhancement of antioxidant activity of wa-song.

Antioxidant Effect of Wa-song (Orostachys japonicus A. Berger) Extracts on Edible Oil and Fat (유지 기질에 대한 와송 추출물의 항산화 효과)

  • The antioxidants activities in water and 95% ethanol extracts of wa-song (Orostachys japonicus A. Berger) dried by sun, hot air and lyophilization were measured in vitro reaction system. In reaction system containing linoleic acid, the antioxidant activities against lipid oxidation enhanced in proportion to storage time. The antioxidant activity of ethanol extracts was higher than that of water extracts. In the drying methods, wa-song extracts showed higher antioxidant activity in the other of hot air-dried, lyophilized and sun-dried. The lipid oxidation system containing the promoting factors, such as $Fe^{+2}$ and $Cu^{+2}$ions, also showed that the ethanol extract of hot air-dried wa-song possessed the highest antioxidant activity. Soybean oil and lard being added with various levels (0.1, 0.5 and 1.0 g/100 g) of the ethanol extract of hot air-dried wa-song were stored at $60^{\circ}C$ (for 28 days) or $180^{\circ}C$ (for 48 hr). Its extract appeared to lower the acid value of soybean oil, wheras it failed to lower the acid value for lard at early storage time, but it was lower than control and BHT after 28 days. Anicidine value and peroxide value were lower soybean and lard added wa-song extract than control and BHT for storage for 28 days at $60^{\circ}C$. In 28 days, its value was significantly decreased in proportion to sample concentration. TBA value was increased during storage time at $60^{\circ}C$, but it was significantly decreased by sample concentration after storage 36 hours at $180^{\circ}C$ storage. Therefore results suggest ethanol extract of hot air-dried wa-song could be potential candidates for natural antioxidants for materials containing lipid.

Effect of Wa-song(Orostachys japonicus A. Berger) Extract on the Oxidative Stability of Edible Oil During its Heating (식용유지의 가열시 와송 추출물이 산화안정성에 미치는 영향)

  • This experiment was conducted to investigate the effect of wa-song (Orostachys japonicus A. Berger) extracts on the oxidative stability of edible oil. Wa-song was dried from hot air (HWE) and freeze (FWE), and then they were extracted by hot water. The different levels (0.1, 0.5 and 1.0 g/100 mL) of HWE and FWE were added to soybean oil and lard. The chromaticity of edible oils was generally increased with prolonged heating time and HWE was higher than FWE. The anisidine value showed significant increase during heating for 48hrs. After heating for 48 hrs, it was lower than control, except for HWE added sample of 0.5 and 1.0 g/100 mL, FWE added sample of 1.0 g/100 mL. In lard, it was lower in sample added wa-song extract than control. The acid value was not significant by added amount of wa-song. Its value in HWE added sample was lower than FWE, after heating for 48 hrs. POV was lower HWE than FWE, also. After heating for 24 hrs, TBA values in soybean oil containing HWE and FWE added sample was lower than control. In lard, its value in HWE and FWE added sample was lower than control during heating for $12{sim}48\;hrs$. Therefore, those results suggested that HWE has higher antioxidant activity than FWE added sample, and then oxidative stability of HWE in edible oil was more potential for lard during its heating.

The Antioxidants Activities of Hot-Water Extracts of Wa-song (Orostachys japonicus A. Berger) on Edible oil and Fat (식용유지에 대한 와송 열수추출물의 항산화효과)

  • This study was conducted to investigate the antioxidant activity of hot water extracts of wa-song (Orostachys japonicus A. Berger) dried using hot air (HWE) and frozen (FWE). Varying levels (0.1, 0.5 and 1.0 g/100 g) of HWE and FWE were added to soybean oil and lard. Chromaticity, anisidine value, acid value, peroxide value (POV) and thiobarbituric acid (TBA) value in oils were measured periodically during their storage for 28 days at $60^{\circ}C$. The chromaticity of edible oils showed a general increas with prolonged storage as well increasing levels of extracts. The anisidine value was not significantly increased during storage for 14 days, but was significantly increased after storage for 21 days in soybean oil and lard. The anisidine value of HWE on soybean oil was $12.60{\pm}0.92{\sim}13.82{\pm}0.68$ after storage for 28 days its value was lower than that of the control and buthylated hydroxy anisol (BHT). The antioxidant activity of HWE was found to be more effective than that of FWE. The acid value of HWE was significantly increased during storage from 14 to 21 days in soybean oil and from 7 to 14 days in lard. The antioxidant activity of FWE was particulary effective at the primary stage of the reaction system of lard storage. POV was highly increased during the storage periods between 7 and 14 days in soybean oil. The antioxidant activity in all the samples tested did not significantly increase after storage for 14 days, except when 0.1 g/ 100 g of FWE added to lard. TBA values of all the samples were lower than that of control and 0.02% BHT during their storage. The antioxidant activities of wa-song within the reaction system of oils were more effective in soybean oil than in lard.

Design of Short Indolicidin Analogs with Enhanced Prokaryotic Selectivity (증가된 원핵세포선택성을 가진 짧은 인돌리시딘 유사체의 설계)

  • Indolicidin (ID) is a 13-residue Trp-rich antimicrobial peptide (AMP) isolated from bovine neutrophils. In addition to having a high antimicrobial potency, it is also toxic to mammalian cells. To develop novel ID-derived AMPs with shorter lengths and enhanced prokaryotic selectivities (meaning potent antimicrobial activity against bacterial cells without toxicity against mammalian cells) over the parental ID, several ID analogs were designed and synthesized. Finally, 10-residue ID analogs (SI, SI-PA, SI-WF and SI-WL) with much higher prokaryotic selectivity than the parental ID were developed. Our results suggest that the hydrophobic and aromatic amino acids at the central position of the analog SI with the highest prokaryotic selectivity are important for potent antimicrobial activity, but two Pro residues do not affect antimicrobial activity. The order of prokaryotic selectivity for ID and its designed analogs was SI > SI-PA > SI-WF > SI-WL > ID > SI-WA. Taken together, our designed short ID analogs could be developed as therapeutic agents for treating bacterial infections.

Flora and Life-form of the Uninhabited Islets, in Jeju-do (제주도 무인도서의 식물상과 생활형)

  • This study was survey a flora and life form spectrum of 11 uninhabited islets, in jeju-do. The vascular plants of each uninhabited islets were summarized as Beomseom 159 taxa, Chagwi-do 136 taxa, Daryeo-do 81 taxa, Hyeongje-do 61 taxa, Jigwi-do 39 taxa, Munseom 147 taxa, Ran-do 58 taxa, Saeseom 65 taxa, Seogeondo 60 taxa, Seopseom 183 taxa and Wa-do 46 taxa. Floristic richness was in order analyzed to be Seogeon-do 28.62, Ran-do 19.24, Daryeo-do 18.53, Wa-do 16.42, Munseom 16.14, Hyeongje-do 14.76, Seopseom 13.32, Beomseom 13.24, Chagwi-do 6.15, Saeseom 5.86 and Jigwi-do 2.51. Life form spectrum was surveyed to H-$D_5$-$R_4$-e; hemieryptophyte(H) 26.8%, not production seeds($D_5$) 54.4%, clonal growth by stolons and struck roots($R_4$) 46.9% and erect form(e) 30.3%. According to the life form spectrum hemieryptophyte dominated high portions, which suggests islet in jeju-do has hemieryptophytic climate and similar to middle parts in Korean Peninsular. However, were noticed by that it has more therophytes. As a result, Partially shows the characteristics change from the temperate climate to the warm temperate climate or a semitropical climate by a climatic change.

티벳버섯(Tibetan mushroom)에서 분리한 EPS(Exopolysaccharide)가 로타바이러스의 MA-104 세포 감염에 미치는 영향

  • 본 연구는 유산균(Lactobacillus fermentum, Lab. acidophilus, Lab. brevis)과 효모(Candida kefyr, Cryptococcus albidus, Pichia ohmeri)등이 주요 균총인 티벳버섯 배양액에서 crude EPS를 분리하여, 로타바이러스의 MA-104 세포감염 억제효과를 측정하였다. 분리된 crude EPS는 0.0026${\sim}$0.363%의 농도에서 MA-104 세포에 대해 독성효과가 없었다. 로타바이러스 Wa, S2, YO의 MA-104 세포 감염에 대해 EPS 0.0026%의 농도에서는 9.11${\sim}$20.06%의 억제율을 보였고, EPS 0.33%의 농도에서는 23.69${\sim}$38.09%의 억제율을 보였다.

  • The object of this research is showing the effect of additional hydration of BS with OPC and less amount of gypsum in WA for the binder of BS with RFA mortar on strength development. The test performed was using factors of 0, 10, 30, and 50 % of OPC replacement for effect on strength development with additional stimulation.
