Use of Bacteriocinogenic Pediococcus acidilactici in Sausage Fermentation

  • The bacteriocin produced by Pediococcus acidilactici KFRI 168 exhibited a wide antimicrobial spectrum including many strains of lactic acid bacteria, Listeria monocytogenes, Staphylococcus aureus, and Enterococcus faecium by both disk and deferred assay methods. Inhibition of Lis. monocytogenes and Stph. aureus were observed only from deferred assay. Gram-negative bacteria were not inhibited. Bacteriocin production was observed at 10 h, and was maximized at 16 h in MRS broth incubated at $37^{\circ}C$. In a beaker sausage fermented with P. acidilactici KFRI 168, viable counts of Stph. aureus, Salmonella, Escherichia coli, Clostridium perfringens, and Lis. monocytogenes were reduced by 2.8, 2.3, 2.4, 0.7, and 0.5 log CFU/g, respectively. Inoculated P. acidilactici KFRI 168 maintained its viable count of more than $10^8$ CFU/g during the whole fermentation period, and it took less than 8 h to reduce sausage pH below 5.

키토산을 첨가한 개상용 레진의 세균부착에 관한 연구 (ADHERENCE OF ORAL BACTERIA ON VARIOUS DENTURE RELINING MATERIALS WITH CHITOSAN)

  • The purpose of this study was to evaluate the rate of adherence of bacteria on various denture relining materials and to find out the effects of chitosan, when it was added to denture relining materials. Denture relining materials such as Tokuso rebase normal $set^{(R)}$, Mild $rebaron^{(R)}$, $Kooliner^{TM}$, and New $truliner^{TM}$ were used in this study. The adherence of Streptococcus mutans was studied on the surfaces in the denture relining materials with chitosan and in those without chitosan. When chitosan was added to M17 broth and MRS broth, the viable cell count of Streptococcus mutans was reduced. The viable cell count of Streptococcus mutans on the specimens decreased in the following order : Mild rebaron, Tokuso rebase normal set, and Newtruliner and Kooliner. The deture relining materials with chitosan showed a lower rate of adherence of Streptococcus mutans than those without chitosan.

황색포도상구균에 대한 마우스의 지연성과민반응 발현 (Induction of Delayed-Type Hypersensitivity Reaction to Staphylococus aureus in Mice)

  • The development of delayed-type hypersensitivity(DTH) reaction to Staphylococcus aureus in mice was studied, Mice received 3 injections of $10^8$ viable S. aureus subcutaneously showed a marked footpad swelling when mice were challenged with $10\;{\mu}g$ staphylococcal protein antigen into footpad(The percent increase of footpad thickness at 24 h after challenge wsa 35% approximately). Histological observation of footpad of immunized mice showed a marked thickness of subcutaneous tissue due to edematous reaction and massive infiltration of lymphocytes and neutrophils which are characteristic cells in DTH reaction. Intensity of DTH reaction of mice immunized with viable bacteria was much higher than that of mice immunized with staphylococcal protein or heat-killed bacteria. The DTH reaction to S. aureus could be transferred to normal recipient mice by both spleen cells and lymph node cells.

알긴산 나트륨을 이용한 유산균 캡슐화의 상업화 공정 개발 (Development of a Commercial Process for Micro-Encapsulation of Lactic Acid Bacteria Using Sodium Alginate)

  • 바이오 고분자인 알긴산 나트륨(Sodium Alginate)을 이용하여 기존의 방법에 비해 생산성이 우수한 캡슐화의 상업화 공정을 개발하고자 하였다. 또한, 동일 공정으로 키토산을 알긴산과 함께 캡슐화하여 알긴산 나트륨으로 캡슐화 된 유산균과 비교하였다. 유산균 캡슐화의 상업화 공정의 주요 공정은 캡슐화 후 기존의 동결건조 대신에 본 연구진이 개발한 생산성이 우수한 유동화 건조 방법에 의하여 건조시간을 15~24이상 단축할 수 있었지만, 생균수는 동결건조와 유동층 건조의 비율이 1:0.75로 동결건조 방법이 좋았다. 하지만 건조에 드는 비용과 시간을 고려 해 볼 때 유동층 건조 방법으로 상업화 공정이 가능함을 확인할 수 있었다. Chitosan-alginate 캡슐은 알긴산 칼슘캡슐과 생균수를 비교하였을 때, 알긴산을 이용한 캡슐은 희석배수 $10^{-9}$, 즉 약 $1{\times}10^9$ 마리 이상의 균이 존재하고, 키토산을 이용한 캡슐은 희석배수 $10^{-3}$, 즉 약 $1{\times}10^3$ 마리의 균이 존재함을 확인 할 수 있었다. 본 연구의 기술로 제조된 유산균 캡슐은 pH 4.65, 6.01에서 96시간 이상 동안 안정하였지만, pH 7.07, 8.35에서는 1시간 이내에 모두 붕해되었다. 이는 유산균 캡슐이 위산에서 안정성을 보이고 pH 7이상을 띠는 소화기관인 소장과 대장에서는 쉽게 붕해가 일어날 수 있음을 알 수 있었다.

전통 막걸리의 저장중 효모와 세균의 변화 (Changes of Yeasts and Bacterial Flora during the Storage of Korean Traditional Makgeolli)

  • 한국 전통 생 막걸리의 최적 저장 조건을 확립하기 위하여 먼저 전통 생 막걸리를 제조한 후 $4^{\circ}C$$20^{\circ}C$에 저장하면서 효모와 총 세균, 젖산균 수의 변화를 조사하였다. 효모는 $20^{\circ}C$에서 30일 저장 후 89%가 감소 되었으나, $4^{\circ}C$저장에서는 큰 변화가 없었다. 총 세균수는 $20^{\circ}C$에서 30일 저장시 98.0%가 감소 되었고 젖산균 역시 99%가 감소되었다. 그러나 $4^{\circ}C$저장에서는 65%미만의 낮은 감소를 보였다. 위의 결과와 관능검사 결과 등을 종합하였을 때 본 연구에서 제조한 생 막걸리는 $4^{\circ}C$$20^{\circ}C$에서 15일까지도 큰 품질저하 없이 음용할 수 있는 상태로 저장이 가능한 것으로 판단된다.

유산균의 종류 및 귀리 첨가가 요구르트의 품질 및 항산화활성에 미치는 영향 (Effects of Oat Addition and Various Lactic Acid Bacteria on Quality Characteristics and Antioxidant Activity of Yogurt)

  • In this study, we investigated the effects of adding oat and lactic acid bacteria on the quality and functionality of yogurt. Yogurt was fermented with various lactic acid bacteria,; Lactobacillus acidophilus (LA), Lactobacillus delbrueckii sub. bulgaricus (LB), and Streptococcus thermophilussei (ST) and quality properties, β-glucan content, antioxidant activity were estimated. The quality of control and oat added yogurt (OY) showed significant differences depending on the type of strain and combination. The addition of oats significantly accelerated the lactic acid bacteria production, decreased the pH, and increased the titratable acidity and count of the viable cells compared to the control. Acid production was highest in ST, with the complex strains containing ST and LALBST showing high quality characteristics. The viscosity of oat yogurt was higher than that of the control group, and LALBST was also significantly higher than that of the control group. The β-glucan content of OY was 0.14-0.2%, and the organic acid content and antioxidant activity were also significantly increased by the addition of oats. As a result, it is thought that the addition of oats and a combination of lactic acid bacteria can be used for improving the quality and functionality of yogurt.

시중에 유통중인 오징어채볶음과 멸치볶음의 위생실태 (Sanitary Conditions of Sliced Squid Bokum and Anchovy Bokum Available in the Market)

  • Microbial distribution as well as content of salt and preservatives in side dishes was investigated by analyzing cell count of viable cells, coliform bacteria and food poisoning bacteria of sliced squid bokum and anchovy bokum, purchased at 17 different department stores and 2 different traditional market in Seoul, which are most preferred by many consumers to any other side dishes available in the market. 6.2$\times$103~1.2$\times$108 cells/g of viable cell was detected in 19 different samples of the sliced squid bokum, among which samples of the sliced squid bokum and 14 samples of the anchovy bokum contained 103~108 cells/g of coliform group. However food poisoning bacteria were not detected in all the samples tested. Salt content was 2.42~4.89 %w/w and 2.28~6.46 w/w for the sliced squid bokum and the anchovy bokum respectively. Analysis of preservatives by HPLC such as sorbic acid, benzoic acid, dehydroacetic acid and another 6 kinds of esters resulted that 1.0$\times$73.8 mg/100 g of sorbic acid was detected in the 19 samples of the sliced squid bokum, while only 6 samples of the 15 anchovy bokum samples contained sorbic acid.

Preparation of Yogurt Added with Angelica keiskei Juice and Its Quality Characteristics

  • Mixtures prepared from whole milk with added skin milk powder(2.5%, w/v) and Angelica keiskei juice (1.5%, w/v) were fermented with lactic acid bacteria (single and mixed culture of Lactobacillus bulgaricus and Strpetococcus thermophilus) for 24 hours. The fermented mixtures (curd yogurt) were evaluated for acid production (pH and titratable acidity), cell numbers, viscosity, sensory property and keeping quality. Results indicated that the addition of Angelica keiskei stimulated the acid production by lactic acid bacteria. The number of viable cells reached 4.5~7.3$\times$10$^{9}$ CFU/mL for Angelica keiskei-added yogurts, while 3.3~5.1$\times$10$^{9}$ CFU/mL for control yogurts. Viscosity of Angelica keiskei-added yogurts was higher (3,609~3,854 centipoises) than that of control yogurts(3,346~3,700 centipoises). Of the microorganism tested, mixed culture of Lactobacillus bulgaricus and Streptococcus thermophilus was most effective in acid production. The overall sensory score showed that Angelica keiskei yogurt fermented with Streptococcus thermophilus was evaluated as good as control yogurt. When yogurts were stored at 4$^{\circ}C$ for 12 days, pH, titratable acidity and viable cells of lactic acid bacteria were not significantly changed(p<0.05).
