Antioxidant and Anti-inflammatory Activity of Stem Bark Extracts from Ulmus davidiana var. japonica

  • Ulmus davidiana var. japonica is a deciduous tree used in traditional medicine. In this study, we examined the antioxidant and anti-inflammatory activity of extracts and compounds isolated from U. davidiana var. japonica stem barks for development of cosmetic phyto-materials. Phytochemical examination of the stem bark led to the isolation and characterization of three known compounds, (+)-catechin (1), (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2), and procyanidin B3 ((+)-catechin-($4{\alpha}{\to}8$)-(+)-catechin) (3). Their bioactivities including antioxidant (DPPH ree radical scavenging assay) activity, anti-inflammatory (nitric oxide nhibition assay) were evaluated. Most of the crude extracts and isolates indicated significant antioxidant potential compared with BHT and ${\alpha}$-tocopherol as controls. Futhermore, all compounds showed higher inhibitory activities for NO production in Raw 264.7 cells than the L-NMMA using the positive control. Eespecially, (+)-catechin (1) and (+)-catechin-7-O-${\beta}$-D-apiofuranoside (2) which could inhibit more than 90% of the NO production at a concentration of 100 ${\mu}g/m{\ell}$, implying excellent anti-inflammatory activity.

Anti-Inflammatory Activity of Constituents Isolated from Ulmus davidiana var. japonica

  • Twenty six compounds (1-26) were isolated from the root barks of Ulmus davidiana var. japonica. The anti-inflammatory activity of the isolated compounds were evaluated agai nst the generation of inflammatory chemical mediators in bone marrow-derived mast cells. Among them, compounds 10, 11, 13, 15 and 19 inhibited not only cyclooxygenase-2 dependent prostaglandin $D_2$ generation but also 5-lipoxygenase dependent leukotrien $C_4$ generation in a concentration-dependent manner. In addition, compounds 11, 12, 13, 15 and 19 also inhibited $\beta$-hexosaminidase release, a marker of mast cell degranulation reaction, from bone marrow-derived mast cell. These results suggest that the anti-inflammatory activity of U. davidiana might in part occur by both the inhibition of eicosanoid generations and the degranulation reaction of mast cells.

Manufacture and Physiological Functionalities of Some Natural Plant Fermentation Broths and Liquor (몇 가지 천연식물 발효액의 제조 및 생리기능성)

  • Three kinds of plant fermentation broths and one liquor were prepared using persimmon, Ulmus davidiana var. japonica and barley, and investigated its physiological functionality. Anti-aging antioxidants activity was the highest of 99% in Ulmus dacidiana bar. japonica liquor and also was high in the persimmon vinegar(68.3 %). SOD-like activity show also 80.6% in the Ulmus davidiana var japonica liquor however the other fermentation broths were less 25%. Antihypertensive angiotensin I-converting enzyme inhibitory activity and fibrinolytic activity were weak or not detected in all the fermentation broths. Anti-dementia acetylcholinesterase inhibitory activity were showed 54.7% in the Ulmus davidiana var. japonica liquor. Our results showed that the Ulmus davidana var. japonica liquor has the potential to become a new functional liquor because of high antioxidant activity and anti-dementia activity.

Anti-Obesity and Lipid Metabolism Effects of Ulmus davidiana var. japonica in Mice Fed a High-Fat Diet

  • The root bark of Ulmus davidiana var. japonica (Japanese elm) is used in Korea and other East Asian countries as a traditional herbal remedy to treat a variety of inflammatory diseases and ailments such as edema, gastric cancer and mastitis. For this study, we investigated the lipid metabolism and anti-obesity efficacy of ethyl alcohol extract of Ulmus davidiana var. japonica root bark (UDE). First, HPLC was performed to quantify the level of (+)-catechin, the active ingredient of UDE. In the following experiments, cultured 3T3-L1 pre-adipocytes and high-fat diet (HFD)-fed murine model were studied for anti-obesity efficacy by testing the lipid metabolism effects of UDE and (+)-catechin. In the test using 3T3-L1 pre-adipocytes, treatment with UDE inhibited adipocyte differentiation and significantly reduced the production of adipogenic genes and transcription factors PPARγ, C/EBPα and SREBP-1c. HFD-fed, obese mice were administered with UDE (200 mg/kg per day) and (+)-catechin (30 mg/kg per day) by oral gavage for 4 weeks. Weight gain, epididymal and abdominal adipose tissue mass were significantly reduced, and a change in adipocyte size was observed in the UDE and (+)-catechin treatment groups compared to the untreated control group (***p < 0.001). Significantly lower total cholesterol and triglyceride levels were detected in UDE-treated HFD mice compared to the control, revealing the efficacy of UDE. In addition, it was found that lipid accumulation in hepatocytes was also significantly reduced after administration of UDE. These results suggest that UDE has significant anti-obesity and lipid metabolism effects through inhibition of adipocyte differentiation and adipogenesis.

Antimicrobial Activities of (-)Epicatechin from Ulmus davidiana var. japonica Cortex

  • The extract of Ulmus davidiana var. japonica cortex has known as natural anti-inflammatory substance in East Asia. For the identification of antimicrobial substance, it was extracted by using methanol and fractionated by using different organic solvents. The fraction of butanol was represented the highest antimicrobial activities. Therefore, the butanol fraction was purified and identified the chemical structure by $^1$H and $^{13}$ C-NMR spectra, FT-IR and EI/MS spectroscopies. The isolated antimicrobial substance was identified as cis-2-[3,4-dihydroxy phenyl]-3,4-dihydro-2H-1-benzopyran-3,5,7-triol, which has commonly known as (-)epicatechin. Its minimum inhibitory concentrations (MICs) against Staphylococcus aureus and Listeria monocytogenes were shown as 100 $\mu\textrm{g}$/mL, respectively.

Anti-Wrinkle Effect of Ulmus davidiana Extracts (유근피 추출물의 피부개선효과)

  • The bark of the root and stem of Ulmus davidiana var. japonica has been used as a traditional Korean medicine to treat inflammatory disorders. This plant reportedly shows antioxidant, anticancer, and anti-inflammatory effects. In this study, we investigated the protective effects of Ulmus davidiana var. japonica ethanolic extract (UDE) on UVB irradiation-induced wrinkle in hairless mice. We evaluated for their free radical-scavenging activities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the anti-elastase activities, and for their anti-matrix metalloproteinase-1 (MMP-1) activity in human skin fibroblast cells. In the wrinkle measurement and image analysis of skin replicas, the results showed that UDE significantly inhibited wrinkle formation caused by chronic UVB irradiation. These results suggest that UDE has anti-wrinkle activity.

Germination Behaviors and Seed Longevities of Three Ulmus Species in Korea (한국산(韓國産) 느릅나무속 3수종(樹種)의 종자저장성(種子貯藏性) 및 발아특성(發芽特性))

  • This study was conducted to investigate temperature effect on seed germination behaviors and seed viability changes by drying periods of three Ulmus species (Ulmus davidiana var. japonica (Rehder) Nakai, U. pumila L., and U. parvifolia Jacq.) distributed in Korea. Statistical analysis showed that temperature had a significant effect on mean germination time in three Ulmus species, but its effect on germination percentage was only shown in U. parvifolia (p<0.01). U. davidiana var. japonica and U. pumila germinated relatively consistent in all temperature conditions, especially the latter showed high germination behaviors in low temperatures (10-15$^{\circ}C$). U. parvifolia germinated well in high temperature (25$^{\circ}C$), while this species rarely germinated in low temperature conditions (10-20$^{\circ}C$). The results confirm that seeds of U. pumila and U. parvifolia are orthodox seeds which can be stored under 4-8% moisture condition, and U. davidiana var. japonica seed is recalcitrant seed which is difficult to store for a long time. In conclusion, these differences of the germination behaviors for three Ulmus species seem to be adaptive regimes of their natural habitats, respectively.

Antioxidative and Protective Effects of Ulmus davidiana var. japonica Extracts on Glutamate-Induced Cytotoxicity in PC 12 Cells (느릅나무 추출물의 항산화 효과 및 L-glutamate 유래 PC12 세포독성 보호효과)

  • Antioxidative and protective effects of Ulmus davidiana var. japonica against oxidative damages induced by glutamate in PC 12 cells were investigated. Inhibitory activity against $FeSO_{4}-H_{2}O_{2}$-induced oxidative stress and DPPH radical-scavenging activity were detected in ethyl acetate and butanol fractions of ethanol extracts from stems and roots. Ethyl acetate and butanol fractions of ethanol extracts from roots significantly inhibited glutamate-induced cytotoxicity and reactive oxygen species in PC 12 cells. These results demonstrate ethyl acetate and butanol fractions of ethanol extracts of U. davidiana var. japonica have potent protective effect against glutamate-induced oxidative stress.

Lignan and Neolignan Glycosides from Uimus davidiana var. Japonica

  • Four lignan xylosides and two neolignan glycosides were isolated from the stem and root barks of Ulmus davidiana var. japonica. Their structures were identified as lyoniside, nudiposide, 5'-methoxyisolarciresinol-9'-O-$\beta$- D-xylopyranoside, isolariciresiol-9'-O-$\beta$-D-xylopyranoside, rel-trans-dihydrodehydroconiferyl alcohol 4'-O-$\alpha$-L-rhamnopyranoside and icariside E3 by comparison of their spectral data with those reported in the literatures, respectively.
