소리쟁이 뿌리 열수 추출물의 폴리페놀 함량 및 추출물의 항산화 효과 (Polyphenol Contents of Rumex crispus Root Extract with Hot Water and its Antioxidative Effect)

  • This study was conducted to investigate the extraction characteristics including total polyphenol compound content (TPC) and the antioxidative effect of Rumex crispus root extract on commercial corn oil. Extraction yield was increased with extraction temperature but decreased with extraction ratio. No significant differences in aromatics content were found among the extracts prepared with various extraction conditions; extraction ratio, extraction temperature, extraction pH and composition of extractant. Total flavonoid content of the extract was increased with extraction temperature and extraction pH, and highest value of it was found when extractant composition of ethanol in water was 75%. Total polyphenol compound content (TPC) of the extract with 10 fold extraction ratio showed the highest value, but no significant difference in TPC was found with extraction temperature. Reducing power and DPPH (${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging ability (RSA) of the extract in spectrophotometric absorbance were decreased with extraction ratio but increased with extraction temperature and showed 63.1%~98.4% and 67.6%~86.7% of those of ascorbic acid in reducing power and DPPH RSA, respectively. The antioxidative effects of the extract on corn oil were 84.8~93.0% of that of commercial butylated hydroxytoluene, the antioxidant index value was highest when the ethanol composition to water in extractant was equal ratio.

붉나무 껍질 에탄올 추출물의 추출특성과 항산화 활성에 관한 연구 (Extraction Characteristics and Antioxidant Activity of Ethanol Extract of Rhus javanica Bark)

  • 붉나무의 유용성에 대한 기초자료를 마련하고자 붉나무 수피를 에탄올로 추출하여 추출물의 몇 가지 이화학적 특성과 항산화 활성을 측정하였다. 추출물의 가용성 고형분의 함량은 73.5 mg/100g, 당도는 $17.8^{\circ}Brix$로 측정되었으며, 총방향족 화합물, 총플라보노이드 함량 및 총페놀성 화합물의 함량은 각각 흡광도 0.508, 49.88 mg/100g 및 296.6 mg/100g으로 측정되었다. 추출물의 환원력은 대조구로서 추출물의 가용성 고형분과 동량의 ascorbic acid의 27.5 %로 측정되었으나, 철환원력은 ascorbic acid와 유사하게 측정되었다. 추출물의 DPPH라디칼 소거능은 95.16 %로서 ascorbic acid와 거의 유사하였고, 금속이온 봉쇄능은 ascorbic acid의 74.73 %보다 높은 81.58 %로 측정되었다. 그리고 pH 2에서 추출물의 아질산염 소거능은 51.76 %로 측정되었으며, Rancimat test에 의한 대두유 산화 억제효과가 있음을 확인하였다.

비수리(Lespedeza cuneata G.Don) 추출물과 담금주의 이화학적 특성비교 (Comparison of Physicochemical Properties between Extracts and Soaked Liquor of Lespedeza cuneata G.Don)

  • 전국의 산야에 자생하는 야생 초본의 하나인 비수리(Lespedeza cuneata G.Don)의 천연물 자원으로서의 유용성에 대한 기초자료를 마련하기 위하여 비수리의 물과 에탄올 추출물의 몇 가지 이화학적 특성과 항산화 활성을 측정하고, 비수리 담금주의 결과와 비교하였다. 비수리 추출물의 유리 아미노산은 phosphoethanolamine을 비롯한 22종이 검출되었으며, 백분율 함량은 phosphoethanolamine 20.36>L-proline 18.02>ammonia 14.48>L-aspartic acid 12.96>${\gamma}$-amino-n-butyric acid(GABA) 10.67 % 등의 순서로 측정되었다. 총 플라보노이드와 총 페놀성 화합물의 함량은 담금주>증류수 추출물>에탄올 추출물의 순서로 비교되었다. 항산화 활성으로서 DPPH(1,1'-diphenyl-2-picrylhydrazyl) 라디칼 소거능에 기준한 전자 공여능은 담금주가 ascorbic acid의 81.4%로 가장 높았으며, 에탄올 추출물>증류수 추출물의 순서로 측정되었다. 철 환원력(ferric reducing antioxidant power)과 ABTS(2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) 라디칼 소거능은 담금주>증류수 추출물>에탄올 추출물의 순서로서 추출물 중의 플라보노이드와 페놀성 화합물 함량에 비례하였다. 그리고 아질산염 소거능은 전자 공여능의 순서와 같이 담금주>에탄올 추출물>물 추출물의 순서로 비교되었으며, 담금주의 경우 pH 1.2에서 96.6%로서 물 추출물의 2.6배에 해당되는 높은 소거활성으로 측정되었다.

  • Woodfordia fruticosa Kurz. (Lythraceae), commonly known as Dhayero, is used in the treatment of various ailments in Nepal. In this study, the antioxidant capacity of crude extract and different polarity fractions of Woodfordia fruticosa flowers were assayed for their scavenging abilities against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and nitric oxide radicals, competitive ${\beta}$-carotene bleaching, reducing power, metal chelating ability and total phenolic content. Crude extract and polar fractions showed stronger antioxidant capacity and contained very high level of total phenolics. They exhibited strong DPPH radical scavenging, nitric oxide scavenging and reducing power, medium ${\beta}$-carotene bleaching and poor metal chelating capacity. Positive correlation obtained between total phenolic content and antioxidant capacity assays, indicated the contribution of phenolics toards antioxidant capacity.

백연수(Leucaena leucocephala (Lam.) de Wit) 잎 에탄올 추출물의 항산화능 (Antioxidative ability of ethanol extract from the leaves of Leucaena leucocephala (Lam.) de Wit)

  • 백연수 잎으로부터 얻어진 50% 에탄올 추출물의 항산화능을 조사하였다. 백연수 잎 추출물의 수율은 $18.3{\pm}0.40%$, 총 폴리페놀 함량은 $121.3{\pm}11.04{\mu}g$ gallic acid equivalents/mg, 총 플라보노이드 함량은 $105.2{\pm}2.77{\mu}g$ quercetin equivalents/mg으로 나타났다. 백연수 잎 추출물은 유리라디칼 소거활성, 양이온라디칼 소거활성, 아질산염 소거활성, 환원력 및 지질과산화 억제 활성의 항산화능을 가지고 있었다. 백연수 잎 추출물의 모든 항산화 활성은 추출물의 농도에 비례하여 증가하였으며, 유리라디칼 소거활성, 양이온라디칼 소거활성, 아질산염 소거활성 및 환원력의 $EC_{50}$은 각각 133.51, 667.22, 2.58 및 $250.00{\mu}g/mL$이었다. 백연수 잎의 우수한 항산화 활성은 높은 폴리페놀과 플라보노이드 함량에 기인한 것으로 여겨진다.

톳 자숙액 에탄올 추출물의 항산화 활성 (Antioxidant Activity on Ethanol Extract from Boiled-water of Hizikia fusiformis)

  • Antioxidant activity of the ethanol extract from boiled-water of Hizikia fusiformis (EBH) were compared with those of BHA, L-ascorbic acid, gallic acid, caffeic acid and (-)-catechin. The free radical scavenging ability against DPPH (1,1-diphenyl-2-picrylhydrazyl), authentic peroxynitrite and reducing power were measured as indices of antioxidant activity. EBH showed the potent DPPH radical and peroxynitrite scavenging activities, showing 85.23 and 96.97% at final concentration of $1000{\mu}g/ml$, respectively. The reducing power increased with the increasing amount of EBH (final concentration of 1, 10, 100 and $1000{\mu}g/ml$). Total phenolic content of EBH was 588 mg (-)-catechin/g at the final concentration $1000{\mu}g/ml$. Total phenolic contents correlated with DPPH radical scavenging activity $(R^2=0.766)$ and reducing power $(R^2=0.944)$. These results suggested that EBH could be a natural antioxidative source containing antioxidative components.

오미자의 이화학적 특성 및 항산화 활성 (Physicochemical Properties and Antioxidative Activities of Omija(Schizandra chinensis Bailon))

  • To develop physiological functionality of Omija extracted with water was evaluated on antioxidative activity. Omija, high acid material with pH 3.6, contain $57.5{\pm}1.03%$ of moisture, and $18.8{\pm}0.12%$ of crude fat. This material have $12.6{\pm}0.04%$ of carbohydrate and $11.1{\pm}0.07%$ of crude protein as well, but ash and crude protein contents were found less than 10%. 10 mineral contents were also found, too; K and Ca showed the highest level, then Al, Mg, Na and Mn were followed. In composition amino acid contents, glutamic acid took the largest portion, $131.7{\pm}1.3$ mg/100 g, aspartic acid $51.5{\pm}0.6%$, and other composition amino acid under 50%. In case of free sugar contents, 7 types were found. Most of them were glucose and fructose. Total phenolic compounds showed the highest level, $2,862.6{\pm}31.7$ mg/100 g. $197.8{\pm}14.6$ mg/100 g of flavonoid and $225.6{\pm}18.2$ mg/100 g were included. In terms of electron donating ability, radical scavenging ability activated as the amount of Omija extract increased. In particular, Omija extract in 1,000 ${\mu}g/m{\ell}$ demonstrated almost similar electron donating ability, $72.4{\pm}0.21%$, to BHT. It was also found that antioxidant activities of electron donating ability, SOD-like ability, hydroxyl radical scavenging ability and nitrite scavenging ability were highly promoted as Omija extract concentration increased. The nitrite scavenging ability was significant when the extract belonged to strong acid region and doping concentrations increased.

용매별 옻나무 추출물의 항산화 활성 및 아질산염 소거능 (Antioxidant and Nitrite Scavenging Activities of Solvent Extracts from Rhus verniciflua Stokes)

  • This study explores the antioxidant activities of solvent extracts from Rhus verniciflua, and the results were as follows. Total phenolic compound and flavonoid contents of Rhus verniciflua were $4.32{\pm}0.17$ mg/100 g and $0.16{\pm}0.02$ mg/100 g, respectively. Electron-donating abilities of solvent extracts from Rhus verniciflua proportionally increased with concentration, and ethyl acetate extract ($95.4{\pm}0.4%$) showed higher activity than that of BHT ($93.3{\pm}1.7%$) at a concentration of 1,000 ${\mu}L$/mL. Reducing power of the solvent extracts from Rhus verniciflua was the highest in ethyl acetate extract ($2.45{\pm}0.03$) at a concentration of 1,000 ${\mu}L$/mL. Nitrite scavenging ability of the solvent extracts from Rhus verniciflua (pH 1.2, 1,000 ${\mu}L$/mL) was the highest in ethyl acetate extract ($95.5{\pm}0.7%$). Therefore, ethyl acetate extract from Rhus verniciflua has similar antioxidant activity as that of BHT.

Antioxidant Activity and Protection from DNA Damage by Water Extract from Pine (Pinus densiflora) Bark

  • Water extract from Pinus densiflora (WPD) was investigated for its antioxidant activity and its ability to provide protection from DNA damage. A series of antioxidant assays, including a 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assay, a reducing power assay, a metal-chelating assay, a superoxide radical scavenging assay, and a nitrite scavenging ability, as well as a DNA damage protection assay were performed. Total phenolic content was found to be 211.32 mg Tan/g WPD. The extract scavenged 50% DPPH free radical at a concentration of 21.35 ${\mu}g/mL$. At that same concentration, the reducing power ability of WPD was higher than that of ${\alpha}$-tocopherol. The extract chelated 68.9% ferrous ion at the concentration of 4 mg/mL. WPD showed better nitrite scavenging effect at the lower pH. Meanwhile, WPD exhibited a strong capability for DNA damage protection at 1 mg/mL concentration. Taken together, these data suggest water extract from Pinus densiflora could be used as a suitable natural antioxidant.

머루와인의 항산화성 및 아질산염 소거능 (The Antioxidant and Nitrite Scavenging Activity of Wild Grape(Vitis coignetiea) Wine)

  • 머루와인의 pH는 $3.7{\pm}0.1$, 총 안토시아닌 함량은 $4.3{\pm}0.3$, 색도는 $10.2{\pm}0.8$이었으며, 총 페놀과 플라보노이드의 함량은 각각 $18.8{\pm}3.9$ mg/100 g, $0.5{\pm}0.2$ mg/100 g이었다. 무기질은 총 10종이 검출되었으며, 특히 K의 함량이 $5.3{\pm}0.2$ mg/100 g으로 가장 높은 함량을 보였다. 전자공여능을 측정한 결과, 1,000 ${\mu}L/mL$의 농도에서 $90.4{\pm}1.8%$로 가장 활성이 뛰어났으며, 환원력을 측정한 결과, 농도가 1,000 ${\mu}L/mL$일 때 환원력은 0.932를 나타내었다. 그리고 아질산염에 대한 소거능을 측정한 결과, pH 1.2의 조건일 때 1,000 ${\mu}L/mL$의 농도에서 $76.3{\pm}1.3%$로 가장 뛰어난 소거능을 보였다. 머루와인의 농도가 증가함에 따라 실험 결과는 유의적으로 증가하는 경향을 보였다.
