Study on Skin Permeation of Tocopheryl Acetate Using Swollen Micelle (Swollen Micelle을 이용한 Tocopheryl Acetate 의 피부흡수 연구)

  • A study was performed to increase skin permeation of tocopheryl acetate, an oil-soluble component among tocopheryl acetate, salicylic acid, and niacinamide, which are the active ingredients of the anti-hair loss toner. As a method of transparently solubilizing tocopheryl acetate while containing salicyl acid and niacinamide, we used a swollen micelle structure that increased the size of the micelle formed by the aggregation of surfactants. The prepared swollen micelle solution contains three kinds of active ingredients, and poloxamer 407 and octyldodeceth-16 were used as surfactants to increase transparent properties and stability. In addition, isostearic acid was used as a co-surfactant to increase the size of micelles. To evaluate the physical properties of the prepared swollen micelles, turbidity at room temperature (25 ℃) was measured. The Franz diffusion cell method was used to evaluate the skin permeation rate of tocopheryl acetate among the hair-loss prevention components contained in swollen micelles. After 24 h, tocopheryl acetate showed a 6-fold improvement in skin permeation compared to the control group. Therefore, it can be seen that the swollen micelles developed in this study can be applied to hair-loss prevention products or solubilized formulations of various functional cosmetics.

  • When the surfactant is dissolved in an aqueous solution, it forms aggregate called micelles (<20 nm) in the solution, and micelles can form the solubilized formulation by supporting the active ingredient therein. Swollen micelles are formulations capable of carrying larger amounts of active ingredient than conventional solubilized formulations at 50~100 nm. Unlike liposomes or nanoemulsions, which require a separate process such as high pressure emulsification, Swollen micelle is a more efficient method of solubilization and particle formation from a productive point of view. In this study, stabilization experiments on swollen micelle formulations were carried out using poloxamer 407, and then optimized formulation experiments for tocopheryl acetate components were performed using Response Surface Methodology (RSM). Tocopheryl acetate, a surfactant that affects solubilization and an active substance, were set as a factor and the correlation between them was confirmed. As the evaluation method, stability and particle size distribution and size were confirmed by temperature and time, and the structure and shape of the swollen micelle carrying the active ingredient were confirmed by FIB. These results show that poloxamer 407 0.500%, octyldodeceth-16 0.387% and tocopheryl acetate 0.945% are the most optimized prescriptions for swollen micelle stabilized with tocopheryl acetate.

Effect of alpha-tocopheryl acetate, retinyl palmitate, and phytantriol on hair protection

  • A hair essence containing α-tocopheryl acetate, retinyl palmitate, and phytantriol (TRP-hair essence) was prepared. TRP-hair essence was excellent in thermal stability to the extent that it did not harden at all even at 210 ℃. TRP-hair essence potently protected the hair from heat stress, significantly reducing the protein leakage in heat-treated hair (p <0.001). Treatment of TRP-hair essence to dyed human hair significantly protected hair against heat stress (p <0.05) as well as improved hair cuticle and color persistence (p <0.05). In addition, as a result of directly treating human hair with TRP-hair essence, the cuticle and tensile strength of human hair were significantly improved (p <0.05). These results suggest that TRP-hair essence can be effective for hair protection and hair quality improvement.

  • The Maillard reaction is a non-enzymatic reaction between amino and carbonyl groups. During milk processing, lactose reacts with milk protein through this reaction. Infant formulas (IFs) are milk-based products processed with heat-treatments, including spray-drying and sterilization. Because IFs contain higher Maillard reaction products (MRPs) than breast milk, formula-fed infants are subject to higher MRP exposure than breast milk-fed ones. In this study, we investigated the optimization of conditions for minimal MRP formation with the addition of $\small{L}$-carnitine ($\small{L}$-car), pyridoxine hydrochloride (PH), and $\small{DL}$-${\alpha}$-tocopheryl acetate (${\alpha}$-T) in an IF model system. MRP formation was monitored by response surface methodology using fluorescence intensity (FI) and 5-hydroxymethylfurfural (HMF) content. The optimal condition for minimizing the formation of MRPs was with $2.3{\mu}M$ $\small{L}$-car, $15.8{\mu}M$ PH, and $20.6{\mu}M$ ${\alpha}$-T. Under this condition, the predicted values were 77.4% FI and 248.7 ppb HMF.

A Study on the Solubilizing and Emulsifying Action of Tocopheryl Acetate using Plant Surfactant (식물성계면활성제를 사용한 토코페릴아세테이트의 가용화와 유화력에 관한 연구)

  • This study is a study on solubilization and emulsifying power of tocopheryl acetate using vegetable surfactants. High purity polyglyceryl-10 isostearate and polyglyceryl-10 oleate were mixed to synthesize a vegetable surfactant with excellent solubilizing power and emulsifying power. The mixed raw material was named Solubil EWG-1100. The appearance of this raw material was a pale yellowish paste with a specific smell, specific gravity of 1.12, and acid value of 0.085. The HLB value of this surfactant was calculated by the Griffin's equation with an average value of 15.17. The behavior of this surfactant to solubilize tocopheryl acetate was mechanically verified. The performance of solubilization was evaluated by a method of visual evaluation and was measured by a transmittance rate at 650 nm using a UV spectrophotometer. As a result, in the formulation using 3% ethanol as a co-solvent, the concentration of surfactant was required to solubilize tocopheryl acetate was required about 5 times of natural surfactant. In the formulation without ethanol as a co-solvent, the concentration of surfactant was required to solubilize tocopheryl acetate required about 7 times of natural surfactant. In addition, the concentration of surfactant required to make an emulsifivation 10 % of tocopheryl acetate was 1 wt% of Solubil EWG-1100, and the emulsified particle size was 3.5 mm in cream formula. In order to obtain stable and fine emulsified particles, it was found that as the concentration of tocopheryl acetate increased, the concentration of Solubil EWG-1100 also was to increase. As a result of testing the solubilizing power of the surfactant according to the pH various change, it showed stable solubilizing power in the acidic region of pH=3.2, the neutral region of pH=7.0, and the alkaline region of pH=11.8. As application, based on these results, it is expected that it can be widely applied to the cosmetics field that develops skin care prescriptions, sensitive skin products, and heavy dry skin products.

Topological Study of the Behavior of Inorganic Fine Powers and a Nanovesicle Hybridized Coating

  • In this study, the surface of inorganic fine powders is hybridized with nanovesicles containing tocopheryl acetate prepared with hydrogenated lecithin via a coating process. From AFM and SEM analyses it is found that the surface of the nanovesicle-coated fine powders lost their traditional topology and improved in terms of their roughness, skewness, and kurtosis. In addition, TEM observations revealed the formation of a 5 nm thick coating layer on the surface of the fine powders. These hybridized powders, in which bioactive materials such as tocopheryl acetate can be embedded, can be employed as a part of a drug delivery system due to their special ability to control release rate and temperature selectivity. Physical properties of the powders, i.e., the different angle and friction coefficient, were excellent.

Oxidative Stress Induces Hypomethylation of LINE-1 and Hypermethylation of the RUNX3 Promoter in a Bladder Cancer Cell Line

  • Increased oxidative stress and changes in DNA methylation are frequently detected in bladder cancer patients. We previously demonstrated a relationship between increased oxidative stress and hypomethylation of the transposable long-interspersed nuclear element-1 (LINE-1). Promoter hypermethylation of a tumor suppressor gene, runt-related transcription factor 3 (RUNX3), may also be associated with bladder cancer genesis. In this study, we investigated changes of DNA methylation in LINE-1 and RUNX3 promoter in a bladder cancer cell (UM-UC-3) under oxidative stress conditions, stimulated by challenge with $H_2O_2$ for 72 h. Cells were pretreated with an antioxidant, tocopheryl acetate for 1 h to attenuate oxidative stress. Methylation levels of LINE-1 and RUNX3 promoter were measured by combined bisulfite restriction analysis PCR and methylation-specific PCR, respectively. Levels of LINE-1 methylation were significantly decreased in $H_2O_2$-treated cells, and reestablished after pretreated with tocopheryl acetate. Methylation of RUNX3 promoter was significantly increased in cells exposed to $H_2O_2$. In tocopheryl acetate pretreated cells, it was markedly decreased. In conclusion, hypomethylation of LINE-1 and hypermethylation of RUNX3 promoter in bladder cancer cell line was experimentally induced by reactive oxygen species (ROS). The present findings support the hypothesis that oxidative stress promotes urothelial cell carcinogenesis through modulation of DNA methylation. Our data also imply that mechanistic pathways of ROS-induced alteration of DNA methylation in a repetitive DNA element and a gene promoter might differ.

Effects of Dietary Lycopene and Vitamin E on Egg Production, Antioxidant Status and Cholesterol Levels in Japanese Quail

  • Japanese Quails were used to evaluate the effects of dietary supplementation with vitamin E (dl-a-tocopheryl-acetate), lycopene, and their combination on egg production, egg quality, concentrations of malondialdehyde (MDA), vitamin E, A and cholesterol in serum and egg yolk. Quails (n = 120; 55 d old) were divided into four groups (n = 30/group) and fed a basal diet or the basal diet supplemented with lycopene (100 mg/kg diet), vitamin E (250 mg dl-${\alpha}$-tocopheryl-acetate/kg diet) or a combination of lycopene and vitamin E (100 mg/kg lycopene plus 250 mg dl-${\alpha}$-tocopheryl-acetate/kg diet). Vitamin E and lycopene did not affect (p>0.05) body weight, feed intake or egg weight. Egg production and Haugh unit were greater (p<0.05) in each supplemental group compared with the control group (p<0.05). Serum and liver MDA levels were decreased in supplemented groups compared with the control group. Separately or as a combination, supplemental lycopene and vitamin E increased serum and egg yolk vitamin E and A but decreased cholesterol concentrations (p<0.05). In general, when a significant effect was found for a parameter, the magnitude of the responses to vitamin and lycopene supplements was greatest with the combination of the lycopene and vitamin E, rather than that observed with each supplement separately. Results of the present study indicate that supplementing with a combination of dietary lycopene and vitamin E reduced serum and yolk cholesterol concentrations and improved antioxidant status.

  • The chromatographic separation of MeOH extract of the aerial parts of Aster glehni (Compositae) led to the isolation of three sesquiterpenes, two sterols and four terpenes. Their structures were determined to be $\beta$-amyrin acetate (1), phytol (2), alismol (3), $\alpha$-tocopheryl quinone (4), $\alpha$-spinasterol (5), 10-O-methyl alismoxide (6), alismoxide (7), and 3-O-(6'-O-palmitoyl-$\beta$-D-glucosyl)-spinasta 7,22-diene (8) by physicochemical and spectroscopic methods . These compounds (1-8) were first isolated from the Aster glehni.
