Studies on the Production of Acid Digestive Enzyme -Isolation and Characterization of a Fungal Strain Which Produces Acid Enzymes- (내산성(耐酸性) 소화효소제(消化酵素劑)의 생산(生産)에 관(關)한 연구(硏究) -내산성(耐酸性) 효소생산균(酵素生産菌)의 분리(分離)와 효소(酵素) 생산조건(生産條件)에 관(關)하여-)

  • A fungal strain which produced high levels of acid protease and amylase was isolated from the atmosphere for application to the manufacture of digestive enzme preparation. This study was carried out to elucidate its microbiological characteristics, environmental conditions for production of the enzymes, and relationships between the enzyme activity and acidity. 1. The isolate was identified as a fungal strain which belonged to Aspergillus niger by the manual of Rafer and Fennel, and was found to be a strain producing high levels of acid protease and amylase. 2. The optimal pH of tile enzymes produced by the strain were: protease, 2.0;, ${\alpha}-amylase$, 4 to 5; and glucoamylase, 3 to 5. 3. The optimal culture conditions for production of the enzymes were: protease (at pH 2.5), 2 to 3 days incubation on wheat bran at $30^{\circ}C$; ${\alpha}-amylase$ and glucoamylase(at pH 3.0), 3 days incubation at $30^{\circ}C$. 4. The production of acid protease and glucoamylase was increased approximately by 20 percent when 2 percent of corn starch was added to the wheat bran medium. 5. The addition of 0.3 percent ammonium sulfate to the wheat bran medium resulted in enhancing the enzyme production, especially of acid prctease.

Development of the Integrated Management System of the Control Points (기준점 통합관리시스템 개발)

  • Control stations managed by national and local governmes are associated with other survey work and constructing geography information and they are important assets in the national level as the positional standard of the country. Since these control points are managed as some type of register and the control points could not be easily updated due to the loss of control stations from construction work or urban development. Therefore, the users could not understand the present situation of the changed control stations. In this background, the aim of this study was to develop control station management system which the managers can use to efficiently maintain control points and to support the usage of the survey control points. For developing this system, we have designed input, update, network, analysis and statistic functions, and have constructed the system using Mapobject as main engine with other languages such as Visual C++ and Visual Basic. The graphic data used in this system are 1/5,000 digital map and digital cadastral map, and the attribute data of each control station are point name, map tile name, longitude and latitude coordinates, TM coordinates, surveying data with the format of year-month-day and control situation photos and so on. In the result of constructing this control station management system, we could achieve integrated management of graphic, attribute and positioning information of each control station.

Method development for quantitative analysis of naturally occurring radioactive nuclides in building materials (실내 건축자재 중 천연방사성핵종의 정량분석법 연구)

  • Naturally occurring radioactive materials (NORMs) increase radiation exposure to the public as these materials are concentrated through artificial manufacturing processes by human activities. This study focuses on the development of a method for the quantitative analysis of $^{232}Th$, $^{235}U$, and $^{238}U$ in building materials. The accuracy and precision of inductively coupled plasma mass spectrometry (ICP-MS) for determination of digestion processes was evaluated for certified reference materials (CRMs) digested using various mixed acid (e.g., aqua regia, hydrofluoric acid, and perchloric acid) digestions and a $LiBO_2$ fusion method. The method validation results reveal that a $LiBO_2$ fusion and $Fe(OH)_3$ co-precipitation should be applied as the optimal sample digestion process for the quantitative analysis of radionuclides in building materials. The radioactivity of $^{232}Th$, $^{235}U$, and $^{238}U$ in a total of 51 building material (e.g., board, brick, cement, paint, tile, and wall paper) samples was quantitatively analyzed using an established process. Finally, the values of $^{238}U$ and $^{232}Th$ radioactivity were comprehensively compared with those from the indirect method using ${\gamma}$-spectrometry.

  • The study area, Bonggil-ri, Gyeongju, SE Korea, is composed of Cretaceous sedimentary rocks, and Tertiary igneous rocks and dykes. A research on fracture developing history and density distribution was carried out on well exposed Tertiary granites. The fractures developed in this area have the following sequence; NW-SE trending duo-tile shear bands (set a), NNW-SSE trending extensional fractures (set d), WNW-ESE trending extensional or normal fractures (set b), NE-SW trending right-lateral fractures (set c), WNW-ESE trending reverse fault reactivated from normal faults (set e) and NW-SE trending left-lateral faults reactivated from shear bands (set a) under brittle condition. According to the result of fracture density analysis, the fracture density in this area depends on rock property rather than rock age, and also higher fracture density is observed around fault damage zones. However, this high fracture density may also be related to the cooling process associated with dyke intrusion as well as rock types and fault movement. Regardless of the reason of the high fracture density, high fracture density itself contributes to fluid flow and migration of chemical elements.

Effect of Feeding Fermented Food Wastes on Consumer Acceptability of Pork Belly (남은 음식물 발효사료 급여가 돈육 삼겹살의 소비자 기호도에 미치는 영향)

  • The objective of this experiment was to compare tile effect of feeding the commercial feeds (control) or fermented food waste feeds (FEWF) on consumer's acceptability and preference of pork belly. The bellies from carcasses of gilts and barrows were used. The consumers evaluated raw meat for color, freshness, fat amount (5=too much fat; 1= not enough fat), fat acceptability and overall acceptability on 5-point stale (5=most desirable; 3=moderate; 1=least desirable) and grilled belly for flavor, taste, texture, juiciness and overall acceptability on 5-point scale (5=most desirable; 3=moderate: 1=least desirable). Raw belly of control had higher scores in color than belly fed FFWF. However, there were no differences between treatments for freshness, fat contents, acceptability of fat contents and overall acceptability (p>0.05). There were no differences in any sensory trait between control and FFWF belly after cooking (p>0.05). Mean scores for preference or overall-liking of raw and cooked belly also were not significantly different between two groups (p>0.05). This results mean that feeding FFWF to pork didn't change the sensory Properties of belly meat.

Antioxidant Activity and Chemical Characteristics of Orostachys malacophyllus and Fermented Orostachys malacophyllus (와송과 발효 와송 추출물의 이화학적 특징 및 항산화 활성)

  • Orostachys malacophyllus grow on the old roofing tile or on the rock of mountain and is belong to Crassulaceae family. After air drying for Orostachys malacophyllus (OM), using the mixture of lactic acid bacteria (Lactobacillus bulgaricus, Streptococcus thermophilus, Lactobacillus acidophilus) was fermented (FOM). OM and FOM extracted using water (W), ethanol (E) and methanol (M) and were measured extracts yield, pH and Brix. Extracted OM and FOM were tested by in vitro experimental models of α,α´-diphenyl-β-picrylhydrazyl (DPPH) radical scavenging activity, Fe/Cu reducing power, linoleic acid peroxidation using ferric thiocyanate and thiobarbituric acid (TBA) methods and peroxidation of rat liver homogenate. In addition, the bioactive materials (phenolic compounds, flavonoids and minerals) were measured. The highest phenolic compounds and flavonoids were OME 122.2 mg/100 g and OME 84.0 mg/100 g. OM and FOM′s major minerals were K, Ca and Mg. The highest free radical scavenging activity showed in FOMM (93.9%), OMM (93.4%), FOME (92.1%) and OME (91.9%) at 0.5% additional level. Fe reducing powers were strong in FOME and FOMM and Cu reducing powers were strong in OME and FOMM. Antioxidant activities on lipid peroxidation using rat homogenate as measured by TBARS method showed strong in FOME and on lipid peroxidation of linoleic acid as measured by ferric TBA method showed strong in OME and FOME and measured by ferric thiocyanate showed strong in FOME and FOMM.

The Effect of Germination of Perilla Seed on the Oxidative Stability of the Oil (들깨기름의 산화안정성에 미치는 들깨 종실 발아의 영향)

  • The fresh perilla seed and tile one-year stored perilla seed were solvent extracted for their oil. On the other hand, the fresh seed and the stored seed were germinated in the dark at $25{\sim}28^{\circ}C\;for\;2{sim}3$ days and then solvent extracted. The above four kinds of perilla oil, that is, the oil from the nongerminated and fresh seed(NFO), the oil from the nongerminated and one-year stored seed (NSO), the oil from the germinated and fresh seed(GFO), and the oil from the germinated and one-year stored seed(GSO) were analyzed with regards to the chemical composition, and the effects of germination of the seed on the oxidative stability of perilla oil were studied. The iodine value and the saponification value were similar in all the perilla oils, but the acid value was increased by germination of the seed. The contents of free fatty acid and diacylglycerol were increased by germination of the seed, while the content of triacylglycerol was decreased. Of the polar lipid components, the content of phosphatidyl ethanolamine was greatly increased by germination of the seed. The contents of total tocopherol of perilla oil from the fresh seed and the one-year stored seed were 494 ppm and 439 ppm, respectively, and by germination of the seed increased to 560 ppm in GFO and 515 ppm in GSO, respectively. Especially a great change in the content of ${\gamma}-tocopherol$ was observed. The oxidative stability of perilla oil was increased by germination of the seed and the increase was distinct in the case of the one-year stored seed compared with that in the case of the fresh seed.

LIFE CYCLE AND CHEMOTHERAPEUTIC CONTROL OF A FILARIAN WORM, PHILOMETROIDES CARASSII PARASITIC IN CARASSIUS AURATUS (붕어에 기생하는 철사충 Philometroides carassii의 생활사와 치료에 관하여)

  • Philometroides carassii (ISHII, 1931) is a parasitic nematode of Carassius auratus and gold fish. Recently a large number of parasitized fishes by this nematode have been reported by fish farmers in Korea. The present investigation was aimed to find out the life cycles of the parasites and effects of chemotherapeutic medicines on the emerged larvae from the adult parasites. Results of the study are summarized as follows : 1. Intermediate host of p. cararsii is a freshwater copepod, Cyclops sp. and the invaded lavae in the body cavity of copepod are capable for parasitizing the fish host after 3 hours of invasion. 2. The lavae eaten directly by the fish host are digested or excreted. 3. The larvae invaded in the fish intestine through the intermediate host pass into the body cavity in 1 to 10 days, and become subadult stage. After 18 to 20 months the subadults parasitic in the body cavity of the Carassius auratus, penetrate through muscle fissile into tile caudal fin where they become adults. Whereas those parasitic in the body cavity of the gold fish, 7 to 8 months the sutadults penetrate through muscle tissue into the caudal fin where they become adults. 1. Male subadults found in the body cavity of the Carassius auratus and gold fish, were larger than females, This is the first record of male subadults occurring in the fish host. 5. Among four different :drugs tested, most of all larvae died in 4 ppm of dipterex in 5 days. But no adults were harmed even in 6 ppm dipterex in 11 days.

  • Plasteins were synthesized from a peptic filefish protein hydrolysate by papain, $\alpha-chymotrypsin$ and protease(from Streptomyces griceus) under the optimum conditions of previous paper. L-glutamic acid diethylester and L-leucine ethylester were incorporated into plastein during the plastein reaction by papain. The structural changes of freeze-dried filefish meat, peptic hydrolysate, FPC and plasteins were observed by Scanning Electron Microscopy(SEM). The functional properties of plasteins also were measured. The solubility of plasteins was higher than that of FPC and the Glu-plastein had $95\%$ solubility in the range of pH 3-10. The dispersibility of Glu-plastein and protease plastein was similar to that of egg albumin, but those of the other plasteins were lower. The water holding capacity of plasteins was lower than that of egg albumin and C. Lipid absorption of Leu-plastein was tile highest, holding 1.80 ml/g, and that of the other plasteins was similar to that of egg albumin. The emulsifying activity of Leu-plastein was the highest, holding $61.2\%$, and that of Glu-plastein was the lowest, holding $50.7\%$. The emulsifying stability of plasteins was similar to that of the emulsifying activity. The emulsifying capacity of Leu-plastein was 384 ml/g(the highest), but that of Glu-plastein and $\alpha-chymotrypsin$ plastein was 248 ml/g(the lowest). The Leu-plastein shelved the highest foaming capacity, $373\%$. The foaming capacity of other plasteins was higher than that of egg albumin. The foaming stability of plasteins was superior to that of egg albumin. The viscosity of plasteins was lower than that of egg albumin. The microstructure of $\alpha-chymotrypsin$ plastein by SEM wassimilar to that of papain plastein, but other plasteins showed differences in their microstructure. The microstructure of Glu-plastein had a smooth shape.

Analysis of Importance by Defect Type in Apartment Construction (공동주택 건축공사 하자유형별 중요도 분석)

  • While numbers of apartment housing are continually rising in the domestic housing construction industry, apartment contractors are currently developing plane models, upgrading facilities, and relevant technology, and investing much efforts to meet the higher demands of consumers. However, the construction process of apartment housing involves the intricate properties of the construction industry such as materials, workforce, equipment, weather, and unpredictable situations. If any of these factors becomes discordant and results in interference and interruption of the construction process, then defects, both functional and aesthetic, are likely to occur due to errors in the plan of industry organizers and constructors. Therefore, this research identifies the types of defects in an apartment construction project and analyzes their relative importance. Firstly, this research reviews the previous research trends and will reduce the needs of this research. Afterward, defect repair costs corresponding to the different defect types are calculated by applying results of the research and performing frequency analysis on defect types included in 'Tenant preliminary research' on apartments constructed by Company A. As a result of analyzing the importance of defect type, the top six activities, including tile, floor, paper hanging, PL window, cabinetry, and kitchen cabinet, are found to be of high importance, and the top six activities in question need of repair and management of defects first. The results of this study will help establish a plan to initially respond to such problems as refusal to move in and filing a defect suit against delay in repairing defects.
