식물근권세균을 처리한 감귤 잎에서 주사전자현미경을 통한 감귤 더뎅이병균의 생장 억제 관찰 (Observation of Growth Inhibition of Elsinoe fawcettii on Satsuma Mandarin Leaves Pre-treated with Rhizobacterial Strains by a Scanning Electron Microscope)

  • 감귤 더뎅이병을 일으키는 E. fawcettii에 항균효과가 있는 식물근권세균 B. gladioli MRL408-3, TRH423-3, P. fluorescens THJ609-3, TRH415-2에 의해 감귤 더뎅이병이 감소하였다. 이들 식물근권세균을 전 처리한 Satsuma mandarin 감귤 잎에 감귤 더뎅이병균을 접종한 후 주사전자현미경을 이용하여 관찰하였다. 식물근권세균을 전 처리한 잎에서 무처리한 잎에 비해 감귤 더뎅이병의 병반수가 감소되었다. 특히 B. gladioli MRL408-3 균주를 전 처리한 식물에서 감귤 더뎅이병의 병반수가 뚜렷하게 억제되었다. 주사전자현미경을 통해 관찰 결과 식물근권세균을 전 처리한 감귤 잎 표면에서 병원균의 발아율과 발아관의 길이가 감소되는 것을 확인하였다. 시판 농약인 imibenconazole을 처리한 잎에서 감귤 더뎅이병균의 성장이 가장 뚜렷하게 억제되었는데, 이는 병반수가 가장 적게 형성된 것과 상통한다. 이들 결과를 통하여 식물근권세균에 의해 감귤 잎 표면에서 감귤 더뎅이병균의 발아와 생장이 억제되고 그 결과 감귤 더뎅이병의 발생이 감소되는 것으로 판단된다.

한국산 가송이(가칭)의 균사생장 특성과 소나무에 외생균근 형성 (Mycelial Growth and in vitro Ectomycorrhizal Synthesis on Pinus densiflora Seedlings of Tricholoma bakamatsutake in Korea)

  • 가송이(Tricholoma bakamatsutake)는 송이(Tricholoma matsutake)의 근연종으로 식용 가능한 외생균근성 버섯 중의 하나이다. 본 연구는 우리나라 신갈나무림에서 수집한 가송이 균주들의 생리학적 특성에 관한 최초의 보고로, 자실체의 조직으로부터 순수 분리한 균주를 대상으로 다양한 조건(배지, 온도, 질소원)에서 이들의 배양 특성을 조사하였다. 대부분의 균주들은 PDA 배지, 20 또는 $25^{\circ}C$의 배양온도에서 균 생장력이 높았으며, 시험균 중 2균주는 그들의 균사생장에 질산염보다 암모늄 형태의 무기질소원을 더 선호하였다. 가송이는 근연종인 송이와 최적배양 조건이 유사하기는 하나, 균사생장 속도가 송이보다 더 느림을 알 수 있었다. 가송이의 모든 균주들은 carboxymethylcellulose 배지 상에서 cellulase 활성을 나타냈다. 또한 가송이 2균주는 소나무 실생묘에 이들 균을 접종한 지 3 또는 8개월 후 균근을 각각 형성하였으며, 균을 접종하지 않은 묘목보다 균을 접종한 묘목의 생체량이 더 큰 것으로 조사되었다.

위생캔으로 제조한 Bacillus Starter 발효대두의 품질 평가와 금속물질 함량에 관한 연구 (Studies on the Quality Evaluation and Metal Content of Sanitary Canned by Fermented Soybean of Bacillus starter)

  • In order to improve the quality and sensory evaluation of Fermented Soybean(Chungkugjang meju) were investigated. The samples were prepared and fermented by the inoculation of Bacillus strains (B. subtilis, number 1, B. natto, number 2) that the product made With sanitary canned food. 1. The water content of samples was 46.75~50.60%, pH 5.35~6.95 and total acidity 3.26~3.62. 2. The reducing sugar of content for sample was 9.49~10.05%, Amino-N, 67~396mg% and the activity of protease was 0.36~1.49unit/g. 3. The heavy metals analyzed from sample cans, iron, tin and lead content of sample were 5.32~5.84ppm, 27.31~29.04ppm and 0.019~0.021ppm. 4. Therefore, results that Chungkugjang-meju manufactured from B. natto starter induced better product Quality and sensory test than that of the B. subtilis strain.

Isolation of Higher Alcohol-Producing Yeast as the Flavor Components and Determination of Optimal Culture Conditions

  • Ten yeast strains affecting doenjang flavor were isolated from soybean fermented foods (traditional meju and doenjang), among which Zygosaccharomyces sp. Y-2-5, showing excellent growth, glucose consumption, pH, and flavor production, was selected. Higher alcohols produced by Zygosaccharomyces sp. Y-2-5 related to flavor were 2-propanol, 1-propanol, 2-methyl-1-propanol, 1-butanol, and 3.3-dimethyl-2-butanol. Optimal culture conditions for Zygosaccharomyces sp. Y-2-5 were 10% (w/v) NaCl, pH 4.0, 3.0% (w/v) glucose concentration, and inoculation time day 0 or 15 doenjang fermentation.

Agrobacterium으로 형질전환시킨 갈퀴꼭두선이의 세포배양에 의한 천연염료생산 (Production of Anthraquinone Derivatives by Rubia cordifolia var. pratensis Transformed by Agrobacterium spp)

  • The cells of Rubia cordifolia var. pratensis were transformed by Agrobactrium tumefaciens strain 11157. Surface-sterilized young leaves and stems of the plants were cocultivated with bacterial suspensions. Crown galls induced from stems were cultured with variation of culturing conditions and compared with untransformed cells. The growth rates and production of anthraquinone pigments of cells were remarkably improved by transformation. Furthermore, hairy roots were induced by inoculation or cocultivation with Agrobacterium rhizogenes strains.

오리에서 발생한 바이러스성 간염과 살모넬라균증의 혼합감염 (Concurrent Occurrence of Duck Viral Hepatitis and Salmonellosis in Ducklings)

  • Ducklings collected from three farms, having history of rapid onset and spread of nerve signs including kick spasmodically with legs and opisthotonos, were pathologically, barteriologically, virologically examined. Grossly, multiple petechial to ecchymotic hemorrhages were detected in the swollen liver. Histopathologically, diffuse coagulative necrosis of hepatocytes was characteristic in acute cases. Chronic cases revealed marked bile duct hyperplasia rather than hepatocyte necrosis. Some of these cases exhibited multiple granulomas consisting of macrophages, heterophil, fibrin and necrotic cell debri. Filtered homogenate of livers sampled from ducklings caused embryo death with marked hemorrhage and swollen of liver after inoculation into chorioallantoic membrane. Three strains of Salmonella app.,S montevideo, S hadar, and S give, which were biochemically and serologically identified, weir isolated from ducklings of three farms, respectively. From these results, these ducklings were concurrently infected with duck hepatitis virus and Salmonella spp.

Protection Against Salmonella Typhimurium, Salmonella Gallinarum, and Salmonella Enteritidis Infection in Layer Chickens Conferred by a Live Attenuated Salmonella Typhimurium Strain

  • In the present study, we investigated the protection conferred by a live attenuated Salmonella enterica serovar Typhimurium (ST) strain against Salmonella Typhimurium, Salmonella Gallinarum (SG), and Salmonella Enteritidis (SE) infection in layer chickens. Birds were orally primed with the attenuated ST strain at 7 days of age and then boosted at 4 weeks post prime immunization (PPI). Sequential monitoring of plasma IgG and mucosal secretory IgA (sIgA) levels revealed that inoculation with ST induced a significant antibody response to antigens against ST, SE, and SG. Moreover, significant lymphoproliferative responses to the 3 Salmonella serovars were observed in the immunized group. We also investigated protection against virulent ST, SE, and SG strain challenge. Upon virulent SG challenge, the immunized group showed significantly reduced mortality compared to the non-immunized group. The reduced persistence of the virulent ST and SE challenge strains in the liver, spleen, and cecal tissues of the immunized group suggests that immunization with the attenuated ST strain may not only protect against ST infection but can also confer cross protection against SE and SG infection.

Diagnosis of Lily Plant Fasciation Caused by Rhodococcus fascians in Jeju Island

  • To diagnose lily fasciation, lily bulbs showing fasciation were collected from several greenhouses in Jeju Island, South Korea. Bacteria were isolated from the lily bulbs and amplified with both primers for fasA in plasmid and for putative glycosyltransferase epsH gene in chromosome of Rhodococcus fascians. Three bacterial isolates were detected with the P450 primer set and identified as R. fascians by NCBI blast analysis. Twelve bacterial isolates were identified as R. fascians using RS02785 primer set, including the three bacterial isolates identified as the same pathogen using the P450 primer set. Pathogenicity of these bacterial strains identified as R. fascians was demonstrated. Apparent symptoms were observed on wounded lily leaves after inoculation with each bacterial suspension whereas no symptom was found on lily leaves treated with H2O. Furthermore, bacteria re-isolated from wounded sites were identified as R. fascians. Based on the results, these two sets of primers are recommended for quarantine of R. fascians.

Biocontrol of Southern Blight Caused by Sclerotium rolfsii in Pepper Plants Using Bacillus subtilis GJ6-14

  • Southern blight, caused by the soil-borne fungus Sclerotium rolfsii, is a serious disease that affects many economically important crops. In this study, we selected Bacillus subtilis GJ6-14, from a total of 260 strains, to control Southern blight in pepper plants. In both seedling and plant tests, GJ6-14 significantly suppressed disease incidence and severity compared to control, furthermore, GJ6-14 demonstrated efficient colonization in the rhizosphere by maintaining the population from log 5.41 to log 3.92 in the pathogen-inoculated plants, indicating its potential as a biocontrol agent. Molecular analysis revealed up-regulation of defense-related genes, such as a 7.6-fold increase in LOX1 and 15.5-fold increase in PR1, at 72 hr after inoculation of S. rolfsii in GJ6-14-treated plants, suggesting activation of plant defense mechanisms. Overall, our findings highlight the promising role of B. subtilis GJ6-14 as a potential biocontrol agent in sustainable management of Southern blight in pepper plants.

한국 전통 발효식품에서 분리한 인산가용화 효모의 특성 (Characterization of Phosphate Solubilizing Yeasts from Korean Traditional Fermented Foods)

  • 한국 전통 발효식품에서 분리한 효모로부터 인산 가용화 활성이 우수한 5균주를 선발하였다. 선발한 균주 중 2균주는 Pichia anomala로 동정되었고, 3균주는 각각 Pichia farinosa, Candida versatilis, Pichia subpelliculosa로 동정되었다. 인산 가용화 효모는 $20{\sim}35^{\circ}C$의 온도에서 잘 자라는 중 온성 효모였으며 P. farinosa Y669는 $45^{\circ}C$의 고온에서도 생장하였다. C. versatilis Y907 균주는 pH 5~6의 좁은 pH 범위에서 생장하였고 15%의 NaCl 농도까지 내성을 나타내는 호염성 균주였다. 그 외 4균주는 pH 4.0~8.0에서 생장하였으며 NaCl 10% 농도에서 내성을 나타내었다. 인산 가용화 균주는 토양에서도 8주 동안 $10^7{\sim}10^8$ cfu/g의 밀도를 유지하며 생존하였다. 분리균주 중 인산 가용화 활성은 P. subpelliculosa Y1101가 가장 우수하였으며 배양 11일 후 697.2 ug/mL의 유리인산을 생성하였다.
