  • A mutant strain of Lactobacillus paracasei (ML-7) was derived by ultraviolet irradiation of the parent L. paracasei LS-2 strain, followed by colony selection on pH 3.5 MRS agar plates, on which L. paracasei LS-2 cannot grow. The L. paracasei ML-7 mutant strain produced an increased amount of organic acids compared to the L. paracasei LS-2 parent strain. Broiler chickens were randomly fed with one of three experimental diets: a basal diet as a control, and basal diets supplemented with 0.3% of the L. paracasei LS-2 and L. paracasei ML-7 strains. When concluding the study, the final body weights of the broilers fed with the supplemented basal diets were found to be significantly higher (1.9% for LS-2 and 6.2% for ML-7) than the control broiler group. Broilers fed with basal diet supplemented with the mutant L. paracasei ML-7 strain showed increased body weight and food intake, compared to broilers fed with the basal diet with or without the parent L. paracasei LS-2 strain.

  • Transgenic plants from hypocotyl segments of buckwheat were produced with the Agrobacterium strain LBA4404 harboring the binary vector pBI121 containing chimeric genes of neomycin phosphotransferase II (npt II) and $\beta$-glucuronidase (gus). Two weeks after co-cultivation with Agrobacterium, most of the hypocotyl segments gradually became brown and died on the selection medium containing 100mg/$\ell$ of kanamycin. Plants regenerated from the hypocotyl explants grown on selection medium were GUS-positive in the leaf, stem and vascular tissues by histochemical assay, and varied in gus activity (440-2568 pmol, 4-MU/mg protein) by fluorimetry. The plants showing GUS activity were confirmed of containing GUS and NPT-II genes by polymerase chain reaction (PCR). Within 3 months, transgenic buckwheat plants were able to obtained from the hypocotyl segments.

  • In order to cloning of the nif genes of Enterobacter agglomerans NFB-264, the digested total DNA of the strain was ligated to pBR 322 and transformed into E. coli. Through the negative selection and colony hybridization, the transformants were obtained. The recombinant plasmids, pNEL 10 and pNES 20 were extracted from these transformants. It was known from Southern hybridization that pNEL 10 contained the 12 Mdal foreign DNA fragment hybridized with nif Q-X probe and pNES 20 included the 5 Mdal foreign DNA fragment hybridized with nif NE and nif YK probe.

  • A sisomicin-producing strain, Micromonospora inyoensis, was treated with various mutagens. The optimal death rates to obtain the high-producers of sisomicin were 90% for ultraviolet light, 99% for nitrosoguanidine, and 99.3% for nitrous acid, respectively. In place of the method of liquid culture, an agar plug method, which is easy and convenient, was used to measure the antibiotic-producing abilities of the strains isolated from mutagen-treated cells. On the other hand, as the selection method of overproducing mutants after mutagenesis, gradient agar plates which included various antibiotics and salts were used. Among the antibiotics and salts tested, gentamicin and kanamycin as antibiotics, and CuCl$_2$ and HgCl$_2$ as salts, were effective to select the high-producers of sisomicin.

  • Several researchers have developed a number of theoretical time factors to determine the coefficient of consolidation by biezocone excess pore water dissipation test in soft clay deposits. However, depending on the assumptions and analytical techniques, the estimated coefficient of consolidation could be in a considerably wide range even for a specific degree of consolidation. These solutions are obtained from an initial excess porewater pressure distribution which can be determined from. either the cavity expansion theory or the strain path method. The 야ssipation of the initial excess porelvater pressure has been usally simulated by means of linear-uncoupled consolidation analysis and then the dissipation curve is normalized by the initial excess porewater pressure for easy use. However. since there is no guidelines or rules on which method gives the best solution for obtaining the coefficient of consolidation from the dissipation curve, the final selection was only based on engineer's extrience and Judgements. Thus, such an arbitrary selection might be inappropriate for a specific site to characterize the consolidation behavior. In this paper, we reviewed various theoretical time factors and, based on this consideration, we mentioned needs for researches in selecting a specific solution that is compatible for Korean clays. Also we listed some source of errors that can be encountered in the procedure of dissipation analysis.

  • The objective of this research was to determine if leaf size (width and length) is correlated with seed size to the extent that leaf size can be used as a predictor of seed size in a population of soybean plants or lines. Twelve soybean strains, representing three distinct seed size groups, were analyzed. Data on seed size and leaf size of the 12 strains were obtained in 1994 and 1995 field experiments. Strain seed size was positively associated with leaf width (r=0.918) and leaf length (r=0.925). The results of our study indicate that there is a significant correlation between seed size and leaf size in soybean. It is possible that selection for greater seed size either leads to, or results from, greater leaf size.

  • Plant-based vaccines possess some advantages over other types of vaccine biotechnology such as safety, low cost of mass vaccination programs, and wider use of vaccines for medicine. This study was undertaken to develop the transgenic maize as edible vaccine candidates for humans. The immature embryos of HiII genotype were inoculated with A. tumefaciens strain C58C1 containing the binary vectors (V662 or V663). The vectors carrying nptII gene as selection marker and scEDIII (V662) or wCTB-scEDIII (V663) target gene, which code EIII proteins inhibite viral adsorption by cells. In total, 721 maize immature embryos were transformed and twenty-two putative transgenic plants were regenerated after 12 weeks selection regime. Of them, two- and six-plants were proved to be integrated with scEDIII and wCTB-scEDIII genes, respectively, by Southern blot analysis. However, only one plant (V662-29-3864) can express the gene of interest confirmed by Northern blot analysis. These results demonstrated that this plant could be used as a candidated source of the vaccine production.

  • Recombinant Saccharomyces cerevisiae expression systems were developed to produce a novel human anti-angiogenic protein called LK8, an 86 amino-acid kringle fragment protein with three disulfide linkages. Galactose-inducible LK8 expression plasmid was constructed, and LK8 production levels by four S. cerevisiae strains were compared in order to select an optimal host strain. S. cerevisiae 2805 was the most efficient among the strains tested. Elevating the LK8 gene copy number through multiple integration using 8-sequences as target sites resulted in more than a two-fold increase in the LK8 production level compared with the plasmidbased expression system. The maximum LK8 protein concentration of 25 mg/L was obtained from batch cultivation of the yeast transformant that harbors 16 copies of the LK8 gene. In conclusion, the strain integrated with the multiple LK8 gene secreted the protein with relatively high yield, although, the increased LK8 gene dosage over 11 copies did not lead to further enhancement in batch cultivations.

  • In the end milling operation the deflection of the cutter is an important factor affecting the accuracy of machining, with implications on the selection of cutting parameters and economics of the operation. Several studies were devoted to the end mill deflection and its effects, notably, providing a useful insight into the problem. Although the deflection affects adversely the accuracy, the flexibility of the cutter is beneficial in attenuating the overload in a sudden transient situation, as well as in attenuating chatter. The deflection of the end mill was studied both experimentally with strain gauge, tool dynamometer, laser measuring apparatus and on a finite element model of the cutting using ANSYS software. The deflection of machining tool with various helix angles was studied with FEM simulation and experiment. ANSYS analysis performed on the finite element model of the end mill provides deflection results which agree within 15.0% with the experimental ones.

  • We established a strategy for bioethanol production using the hydrolysate of rape stem, in which the inhibitor cocktail was added intentionally. The final goal of this study was to circumvent the detoxification process when the hydrolysate of lignocelluloisic biomass contained the toxic substances in high concentration. When six yeast strains were examined, Sacchromyces cerevisiae ATCC 96581 and Pichia stipitis CBS 7126 were relatively resistant to inhibitor cocktail. Then, using strains 96581 and 7126, we designed a process strategy for bioethanol production, assuming that the concentration of toxic substance in the hydrolysate of rape stem was remarkably high. When strains 96581 and 7126 were inoculated simultaneously, it was observed that strain 7126 produced bioethanol as well as strain 96581, although the concentration of inhibitor cocktail was 18.2% (v/v). Finally, throughout this co-cultivation of strains 96581 and 7126, bioethanol was produced about 6.0 (g/L), and bioethanol yield reached at 0.4 (g-bioethanol/g-reducing sugar) (78.4% of theoretical value).
