Experimental Effects of Acanthopanax sessiliflorus $S_{EEM}$ Extracts Following Gamma-ray Irradiation on Immuno-stimulating and anti-tumor activity in mice (방사선(放射線)이 조사(照射)된 오갈피 나무의 추출물(抽出物)이 면역기능(免役機能) 및 항암(抗癌) 기능(機能)에 미치는 실험적(實驗的) 효과(效果))

  • Objectives : This experimental study was designed to investigate the effects of Acanthopanax, sessiliflorus SEEM extracts following gamma-ray irradiation on immuno-stimulating and anti-tumor activity in terms of proliferation of tumor cells, thymocytes, splenocytes, and NO production from peritoneal macrophages in mice. Methods: 10AS and 100 AS were the bark powders of Acanthopanax sessiliflorus $S_{EEM}$ stems which were exposed in 10 kGy or 100 kGy of electron beam respectively. Results : Treatment with either 10AS or 100AS increased proliferation rates of thymocytes and splenocytes significantly, and treatment with l0AS also decreased proliferation rates of tumor cells significantly. Treatment with either l0AS or l00AS promoted NO production from peritoneal macrophages significantly. Conclusion : These results suggested that AS has direct inhibition effect of tumor growth and immuno-stimulating activity. In conclusion, we demonstrate that AS could be used to treat cancer patient as complementary or alternative medicine to typical anti-cancer medication.

  • Disulfiram (DSF) and diethyldithiocarbamate (DDC), a reduced form of DSF, protect the liver against toxicant-induced injury through inhibition of cytochrome P450 2E1. The effect of DSF and DDC on the levels of major hepatic microsomal epoxide hydrolase (mEH) and glutathione S-transferase (GST) expression was comparatively studied, given the view that these enzymes are involved in terminal detoxification events for high energy intermediates of xenobiotics. Treatment of rats with a single dose of DSF (20-200 mg/kg, po) resulted in 2- to 15-fold increases in the mEH mRNA level at 24 hr with the ED$_{50}$ value being noted as 60 mg/kg. The mEH mRNA level was elevated ~15-fold at 24 hr after treatment at the dose of 100 mg/kg, whereas the hepatic mRNA level was rather decreased from the maximum at the dose of 200 mg/kg, indicating that DSF might cause cytotoxicity at the dose. In contrast to the effect of DSF, DDC only minimally elevated the mEH mRNA level at the doses employed. DSF moderately increased the major GST mRNA levels in the liver as a function of dose, resulting in rGSTA2, rGSTA3/5 or rGSTM1 mRNA levels being elevated 3- to 4-fold at 24 hr post-treatment, whereas the rGSTM2 mRNA level was not altered. DDC, however, failed to stimulate the mRNA levels for major GST subunits, indicating that the reduced form of DSF was ineffective in stimulating the GST the expression. The effect of other organosulfides including aldrithiol, 2, 2'-dithiobis(benzothiazole) (DTB), tetramethylthiouram disulfide (TMTD) and allyl disulfide (ADS) on the hepatic mEH and GST mRNA expression was assessed in rats in order to further confirm the increase in the gene expression by other disulfides. Treatment of rats with aldrithiol (100 mg/kg, po) resulted in a 16-fold increase in the mEH mRNA level at 24 hr post-treatment. DTB, TMTD and ADS also caused 5-, 9- and 12-fold increases in the rnRNA level, respectively, as compared to control. Thus, all of the disulfides examined were active in stimulating the mEH gene in the liver. The organosulfides significantly increased the rGSTA2, rGSTA3, rGSTA5 and rGSTM1 mRNA levels at 24 hr after administration. In particular, aldrithiol was very efficient in stimulating the rGSTA and rGSTM genes among the disulfides examined. These results provide evidence that DSF and other sulfides effectively stimulate the mEH and major GST gene expression at early times in the liver and that DDC, a reduced form of DSF, was ineffective in stimulating the expression of the genes, supporting the conclusion that reduced form(s) of organosulfur compound(s) might be less effective in inducing the mEH and GST genes through the antioxidant responsive element(s).

Circadian Rhythms of Melatonin, Thyroid-Stimulating Hormone and Body Temperature: Relationships among those Rhythms and Effect of Sleep-Wake Cycle

  • Plasma melatonin, thyroid-stimulating hormone (TSH) and body temperature were measured simultaneously and continuously before and after the sleep-wake cycle was shifted in 4 healthy males and changes in the circadian rhythm itself and in the phase relationship among these circadian rhythms were determined. Normal sleep-wake cycle (sleep hours: 2300-0700) was delayed by 10 h (sleep hours: 0900-1700) during the experiment. Even after this shift the typical melatonin rhythm was maintained: low during daytime and high during night. The melatonin rhythm was gradually delayed day by day. The TSH rhythm was also maintained fundamentally during 3 consecutive days of altered sleep-wake cycle. The phase was also delayed gradually but remarkably. The daily rhythm of body temperature was changed by the alteration of sleep-wake cycle. The body temperature began to decrease at the similar clock time as in the control but the decline during night awake period was less steep and the lowered body temperature persisted during sleep. The hormonal profiles during the days of shifted sleep/wake cycle suggest that plasma melatonin and TSH rhythms are basically regulated by an endogenous biological clock. The parallel phase shift of melatonin and TSH upon the change in sleep-wake cycle suggests that a common unitary pacemaker probably regulates these two rhythms. The reversal phase relationship between body temperature and melatonin suggests that melatonin may have a hypothermic effect on body temperature. The altered body temperature rhythm suggests that the awake status during night may inhibit the circadian decrease in body temperature and that sleep sustains the lowered body temperature. It is probable but uncertain that there ave causal relationships among sleep, melatonin, TSH, and body temperature.

The activation of CD99 inhibits cell-extracellular matrix adhesion by suppressing β1 integrin affinity

  • CD99 is known to be involved in the regulation of cell-cell adhesion. However, it remains unclear whether CD99 controls cell-extracellular matrix adhesion. In this study, the effects of CD99 activation on cell-extracellular matrix adhesion were investigated. It was found that engagement of CD99 with the stimulating antibody YG32 downregulated the adhesion of MCF-7 cells to fibronectin, laminin and collagen IV in a dose-dependent manner. The CD99 effect on cell-ECM adhesion was inhibited by overexpression of the dominant negative form of CD99 or CD99 siRNA transfection. Treatment of cells with $Mn^{2+}$ or by ${\beta}_1$ integrin-stimulating antibody restored the inhibitory effect of CD99 on cell-ECM adhesion. Cross-linking CD99 inactivated ${\beta}_1$ integrin through conformational change. CD99 activation caused dephosphorylation at Tyr-397 in FAK, which was restored by the ${\beta}_1$ stimulating antibody. Taken together, these results provide the first evidence that CD99 inhibits cell-extracellular matrix adhesion by suppressing ${\beta}_1$ integrin affinity.

Effect of Aqueous Extract from Asiasari Radix on ${\alpha}$-melanocyte Stimulating Hormone Induced Melanogenesis in B16F10 Melanoma Cells (세신의 열수추출물이 ${\alpha}$-melanocyte Stimulating Hormone에 의해 유도된 B16F10 세포의 멜라닌 생성에 미치는 영향)

  • The aqueous extract from Asiasari radix (AEAR) was used to investigate the effect of ${\alpha}$-melanocyte stimulating hormone induced melanogenesis in B16F10 mouse melnoma cells. The treatment with AEAR at the 1.0 and 2.0 mg/ml level significantly inhibited the biosynthesis of melanin without changes of cell growth and morphology compared with untreated control. The AEAR-treated cells at the 2.0 mg/ml level were more efficient than commercial arbutin at 0.1 mg/ml. The tyrosinase activity also significantly decreased in AEAR-treated cells at the 1.0 and 2.0 mg/ml level. The Western analyses confirmed the slightly decreased expression of tyrosinase by AEAR treatment. These results indicate that AEAR may contribute to the inhibition of melanin biosynthesis through regulating tyrosinase activity and expression and serve as a new candidate in the design of new skin-whitening or therapeutic agents.

  • The recombinant glycoprotein, LBD-005 (Lucky R & D Center, Biotechnology)stimulates the growth of stem cells and activates the development of the hematopoietic cell in a similar manner to the naturally occurring GM-CSF. This test was conducted to investigate if LBD-005 had any effect on fertility in male and female rats. Sprague-Dawley rats(88 of each sex) bred at KRICT, were dosed subcutaneously, at a volume of 2ml/kg body weight with LBD-005 at 0, 250, 500 or 1, 000mg/kg body weight. The male rats were dosed, from 6 weeks of asge, for 60 days prior to mating.

  • Osteoporosis is one of the most important public health problems facing the aging population. Drug therapy for osteoporosis can be divided operationally into two main categories: drugs that inhibit bone resorption, and thus reduce bone turnover, and those that stimulate bone formation, exerting an anabolic effect. Antiresorptive agents such as estrogens, calcitonin, and bisphosphonates are most effective in the prevention of osteoporosis. Formation-stimulating agents such as sodium fluoride or monofluorophosphate, parathyroid hormone fragments, and anabolic steroids are of potential value in the treatment of established osteoporosis, where bone mass is already low and benefit from antiresorptive drugs is likely to be small Recently, raloxifene, a selective estrogen receptor modulator, has become available in various countries for clinical use in the treatment of involutional osteoporsis. This paper will review the use of these drugs in postmenopausal woman.

  • The purpose of this study was to identify important factors influencing fragrance sensibility responses as well as to explore fragrance sensibility factors affecting attitude toward and buying intention of perfume products. For data collection, smelling experiments were conducted with seven groups of 34 female consumers who are aged from the 20's to 40's. Data were analyzed by ANOVA, t-test, factor analysis, and linear regression analysis. In results, five factors (pleasant, romantic, stimulating, modem, familiar factors) were generated from the factor analysis of 24 fragrance sensibility factors. These fragrance sensibility factors significantly affected both attitude toward and buying intention of perfume products. Pleasant, romantic and familiar factors were considered as important factors affecting attitude and buying intention while the stimulating factor affected only buying intention. Fragrance sensibility factors influencing attitude and buying intention of perfume products differed in terms of age and purchase level of consumers.

Leucostim, A Human Granulocyte-Colony Stimulating Factor, Facilitates Granulopoiesis After Bone Marrow Transplantation In Mice

  • In vivo administration of Leucostim, a human recombinant granulocyte colony-stimulating factor (G-CSF), was evaluated for the effects on survival, hematologic recovery, and colony forming unit- spleen (CFU-5) in murine bone marrow transplantation (BMT) model. Sublethally irradiated (9 Gy) mice received bone marrow cells from untreated mice, and then were treated with G-CSF subcutaneously at doses of 2.5,5, or $10\mu\textrm{g}$/kg or vehicle solution (control) for 14 days from one day after BMT. There was no effect of irradiation and BMT on mortality. The repeated subcutaneous injections of Leucostim for 14 days post- BMT significantly facilitated hematologic recovery compared with vehicle control in a dose-dependent manner. Moreover, mice treated with Leucostim had significantly increased numbers of CFU-s colonies on day 10 post-BMT. These results suggest that Leucostim, a new G-CSF, has beneficial effects on hematologic reconstitution after BMT.

  • The effect of water extract of antler on serum level of female hormones was investigated in ovariectemized rats. Sprague-Dawley rats were ovariectomized(ox). After a further 40 days the animals were administrated with water extract of antler (625mg/kg daily) for 10, 20 and 30 days, respectively. Serum level of estradiol, progesterone, leutenizing hormone and follicle stimulating hormone were measured. Significant increase of serum estradiol level was elicited at 20 and 30 days after treatment with water extract of antler, respectively, Associated with the increase of serum estradiol level, there was a concomitant decrease in serum follicle stimulating hormone level. Serum progesterone level was also significantly increased at 30 days after treatment with water extract of antler. Although serum leutenizing hormone level of ox rats treated with water extract of anther was slightly lower than that of untreated ox rats, the decrease was not significant.
