Modulatory Effect of Linoleic Acid During Brucella abortus 544 Infection in Murine Macrophage RAW264.7 Cells and Murine Model BALB/c Mice

  • In this study, we investigated the effects of linoleic acid (LA) treatment on Brucella abortus infection in professional phagocyte RAW264.7 cells, particularly during the pathogen's invasion and intracellular growth in these cells, as well as in murine model BALB/c mice focusing on bacterial splenic proliferation and immunoregulatory activities. LA inhibited the growth of Brucella in a dose- and time-dependent manner. The ability of the pathogen to enter the phagocytes was inhibited as was its survival within these cells. This was accompanied by increased nitrite accumulation in these cells at 24 h post-infection. The concentration of LA used in the present study did not affect the total body weight or liver function of the mice. During Brucella infection, the total splenic weight of these animals was not changed; rather, resistance to bacterial proliferation was enhanced in the spleen. Furthermore, mice treated with LA displayed elevated levels of IL-12 and IFN-γ but reduced levels of IL-10 during infection. The findings in this study showed the regulatory role of LA against B. abortus infection suggesting its potential use in designing intervention strategy for brucellosis.

Proliferation Effect of Conditioned Medium Produced by Lymph Node Stromal Cells

  • CS21 lymphoma cells that preferentially metastasize to lymph nodes after s.c. inoculation into BALB/c mice were grown in vitro in the presence of CA12 stromal cells isolated from lymph nodes. To obtain fundamental data on identification and characterization of the soluble factor(s) produced by CA12 stromal cells, we investigated the biological profile of the conditioned medium produced by CAl2 stromal cells. CAl2 conditioned medium has no affinity with Con A. CAl2 conditioned medium is associated with the proliferation of splenic T- and thymic T-cells with-out adding mitogen, although the conditioned medium cannot induce the differentiation of thymocytes. Additionally, we showed that H-7, not HA-1004 inhibits CS21 cell proliferation. These results suggest that CAl2 conditioned medium has a specific soluble factor(s) produced by lymph node stromal cells.

  • Mancozeb, a polymeric complex of zinc and manganese salts of ethylene bisthiocarbamate (EBDC), is used widely in agriculture as fungicides, insecticides, and herbicides. Mancozeb can be occupationally and environmentally exposed to human and has been reported to induce estrogenic activity, therein it is considered as an endocrine disrupter. After female ICR mice were treated Mancozeb orally at the doses of 250, 1,000 and 1,500 mg/kg/day for consecutive 30 day, we investigated the effects of Mancozeb on the immunopathological parameters (body-, thymus-, spleen-, liver- and kidny-weight, splenic cellularity, hematological parameters) and mitogen (Con A, LPS)-induced splenocyte proliferation (SP). Liver- and kidney- weight were increased, but body- and thymus-weight, number of splenocytes and WBC were decreased, when compared with control group. When splenocytes isolated from the mice exposed to Mancozeb for 30 days were cultured in presence of mitogens, the SP against Con A was significantly and dose-dependently decreased and the SP against LPS was also slightly decreased. Our present results indicate that subacute exposure of Mancozeb to mice might show immunotoxic effect.

Effects of Acanthopanacis cortex Extracts on the Cytokine-inducing and Immune response in Mice (생쥐에서 오가피에 의한 싸이토카인 유도와 면역반응에 관한 효과)

  • This experimental study was carried out to evaluate the effects of Acanthopanacis cortex on Cytokine-inducing and and immune response in Mice. In order to investigate the effect of Acanthopanacis cortex, the following was performed; Cytotoxicity, in vitro, the fraction of $CD4^+$, $CD8^+$, $B220^+$ in splenic cell, gene expression of IL-12(p35), IL-12(p40), IFN-${\gamma}$, and splenic cell proliferation by Acanthopanacis cortex. Analysis of cytokine gene expression was carried out by RT-PCR amplification. Amplified PCR products were electrophoresed on 1.2% agarose gel, and the analysis (Ht) was used to 1D-density program. The results were obtained as follows. Acanthpanacis cortex showed didn't have cell toxicity under $12{\mu}g/m{\ell}$ group on mouse lung fibroblast cells. In an in vitro model using mouse peripheral blood mononuclear cells (PBMCs), extract of Acanthpanacis cortex induced multiple cytokine, including interleukin-12 (p35), interleukin-12 (p40), interferon-gamma (IFN-${\gamma}$). The extract also enhanced the percentages of the $CD4^+$, and $CD8^+$ in the untreated control were $22.1{\pm}3.3$ to $38.4{\pm}2.1$, and $5.0{\pm}0.4$ to $10.7{\pm}0.3%$, respectively. From above findings, it is suggested that Acanthopanacis cortex is able to anti-cancer and activate immune response system.

The Effect of Chemical Sympathectomy on Moxibustion-Induced Immunomodulation in the Rat Spleen (백서의 비장에서 화학적 교감신경절제가 뜸(구(灸))자극으로 유도된 면역변조에 미치는 영향)

  • Background: To investigate the role of sympathetic nervous system (SNS) in moxibustion-induced immunomodulation, the effects of chemical sympathectomy on moxibustion-induced changes in splenic NK cell cytotoxicity, T and B cell proliferation were studied in Sprague-Dawley male rats. Methods: Chemical sympathectomy was achieved with intraperitoneal injection of 6-hydroxydopamine 50 mg/kg/day for 3 successive days. Direct moxibustion (6-minute interval, 9 moxa ball, each of which weighing 0.007 g and burning for 40 seconds) was applied on unilateral anterior tibial muscle region where Zusanli (ST36) acupoint is located, once a day for 7 successive days. NK cell cytotoxicity was measured by $4hr-^{51}Cr$ release assay. Mitogen-induced lymphocyte proliferation was analyzed by [$^3H$]-thymidine incorporation assay. Results: NK cell cytotoxicity was suppressed by moxibustion, more in sympathectomized rats than in vehicle-treated rats. T cell proliferation induced by concanavalin A was not affected by moxibustion. B cell proliferation induced by lipopolysaccharide showed no significant change in vehicle-treated rats, but an increase in sympathectomized rats by moxibustion. Sympathectomy alone induced augmentation of NK cell cytotoxicity and suppression of T cell proliferation. Conclusion: These results suggest that SNS has no direct relation with moxibution-induced immunomodulation but has an important role in the mechanism to keep the homeostasis of immune system by tonically inhibiting excessive changes of various immune components.

Immuno-regulatory Property of Fruit-Extracts of Cornus kousa Burg. (산딸나무열매 추출물의 면역조절기능)

  • This study was conducted to investigate the immuno-regulatory effect and apoptosis of L1210 and HL60 leukemia cells of methanol-extracts of Cornus kousa Burg(CKB). The proliferation of mouse splenocytes and thymocytes enhanced by the addition of $10\;{\mu}g/ml$ of CKB. CKB were administered p.o. once a day for 7 days in adult male BALB/c mice. CKB increased the splenic and thymic T lymphocytes, especially the number of $T_H$ cells markedly increased by the treatment of CKB. CKB treatment induced the apoptotic cell death in L1210 mouse leukemia and HL60 human leukemia cells. In addition, CKB also accelerated the phagocytic activity in peritoneal macrophages and increased the production of plaque forming cells. These results suggest that CKB have an various immuno-regulatory property.

Effects of Kamichihyo-san on Anti-CD40 and Recombinant Interleukin-4 Induced Cytokine Production and Immunoglobulin E in Highly Purified Mouse B Cells (생쥐의 B 세포에서 anti-CD40과 rIL-4로 유도된 사이토카인 생산과 면역글로블린 E에 대한 가미치효산의 효과)

  • In order to evaluate the antiallergic effects of Kamiohihyosan(KCHS), studies were done. We measured the cytotoxic activity for lung fibroblast cell, cytokines transcript expression, production of IL-4, IL-10, IFN-γ, proliferation of B cell in anti-CD40mAb plus rIL-4 stimulated murine splenic B cells. The results were obtained as follows: KCHS was not showed cytotoxicity in the fibroblast lung cell, KCHS increased the gene synthesis of INF-γ, TNF-α, IL1-β, IL-6, IL-10(m-RNA), KCHS decreased the gene synthesis of IL-4, IL-5, TGF-β(m-RNA), KCHS decreased the appearance of IL-4, IgE significantly, KCHS increased the appearance of IL-10, IFN-γ significantly, KCHS decreased the proliferation of B cell significantly, The facts above prove that KCHS is effective against the allergy. Thus, I think that we should study on this continuously.

  • The purpose of this research was to investigate the effect of Sabaek-San(SBS) on the activity of immune cell and leukemia cell. The addition of SBS(1 ㎍/㎖) enhanced the proliferation of cultured-splenocytes and thymocytes. And also, administration of SBS(250, 500 mg/kg) accelerated subpopulation of splenic T lymphocytes in BALB/c mice. Administration of SBS eminently enhanced the production of IFN-γ, and IL-4. The treatment of high dose of SBS inhibit the proliferation of Jurkat cells and dose-dependently increased the apoptosis of cultured-Jurkat leukemia cells. These results suggest that SBS have a cell mediated immuno-regulatory effect.

KYGJT decreased the appearance of TNF-α significantly. 5. KYGJT decreased the appearance of IgE significantly. 6. KYGJT decreased the proliferation of B cell significantly. 7. KYGJT decreased the appearance of Histamin Release Production significantly. The facts above prove that KYGJT is effective against the allergy. Thus. I think that we should study on this continuously

Lipid A of Salmonella typhimurium Suppressed T-cell Mitogen-Induced Proliferation of Murine spleen Cells in the Presence of Macrophage (Salmonella typhimurium lipid A를 처리한 식세포 존재 조건에서 mitogen에 유도되는 이자 세포의 증식억제)

  • Infection with virulent or attenuated Salmonella typhimuriumhas known to induce reduction in proliferative responses of spleen cells. We investigated a role of lipid A from S. typhimurium, a B cell mitogen, on proliferation of spleen cells by T cell mitogens such as concanavaline A and phytohemagglutinin under in vitro and ex vivo conditions. Lipid A alone induced proliferation of spleen cells in vitroin a dose-dependent manner. However, subsequent treatment of concanavaline A or phytohemagglutin in after lipid A treatment induced proliferation suppression of murine spleen cells in vitro and ex vivo. Removal of macrophages from spleen cells, which were obtained from a lipid A-injected mouse, restored proliferation by concanavaline A and phytohemagglutinin, indicating that macrophages appeared to play a role in lipid A-induced suppression. Secreted molecules from macrophages did not accounted for the suppression because suppressive effect was not achieved when the supernatant from macrophage-containing spleen cell culture was conditoned to macrophage-depleted spleen cell culture. Co-culture of spleen cells from lipid A-treated and - untreated mice showed proliferation suppression as increasing cell numbers of lipid A-treated mouse. These data suggested that the cell-to-cell contact of macrophage with splenic lymphocyte cells is responsible for immune responses against lipid A, which is applicable to the case of human S. typhi infection.
