Experimental Studies on Hair Growth Effect of Extracts of Cortex Mori Radicis in Hair Removed C57BL/6N Mice (상백피(桑白皮) 추출물(抽出物)의 발모효과(發毛效果)에 관한 실험적 연구)

  • Cortex Mori radicis has been used as a components of antidiabetics, antiasthma and diuresis in Oriental Medicine. This experiment examined the effect of an extracts, obtained from the acetone(CM-A) and aqueous(CM-W) extracts of Cortex Mori radicis, on hair growing activity of the C57BL/6N mice after topical application to skin. We investigated the number of hair follicle and mast cells, and changes of subpopulation of splenocytes and thymocytes in skin for 16 day. The results were as follows : The Hair growing effect in experimental groups was more increased in 85%(CM-A) and 90%(CM-W) than control group(10%) in hair depilated area. The number of hair follicle in experimental groups(CM-A and CM-W) was more increased than control group. Splenic B/T lymphocytes of CM-A group were decreased compare to control group. CD4/CD8 positive TH cells in splenic T lymphocytes of CM-W group were increased compare to control group. These results suggest that CM-A and CM-W may be used in treatment of alopecia areata.

Immunomodulating Effect of Edible Mushrooms in Mice (식용버섯의 면역조절에 미치는 영향)

  • This study was carried out to evaluate the immunomodulatory capacity of edible mushrooms, including Lepista nuda, Corprinis comatus, Letinus edodes, and Pleurotus eryngii, in mice. BALB/c mice were administered 1, 50, and 500 mg/kg body weight of various mushrooms five times a week over 4 weeks through gastric intubation. The control mice were administered distilled water. No significant changes in body weight were observed. IL-4 and IFN${\gamma}$ production was evaluated with splenic T lymphocytes stimulated in vitro with phytohemagglutinins for 48 hr. The mice group administered L. edodes showed significantly higher ratio of IFN${\gamma}$ versus IL-4 than the other groups. In addition, the ratio of plasma IgG2a versus IgG1 was also significantly elevated in mice treated with L. edodes. However, no significant change was observed in ratio of IgG2a versus IgG1 in splenic B lymphocytes stimulated in vitro with lipopolysaccharides for 7 days. These results indicate that L. edodes can enhance type-1 helper T cell-mediated cellular immunity.

Immunotoxicological Evaluation of Pollen Intake Using Mice Model (실험동물을 이용한 화분섭취의 면역안전성 평가)

  • Pollen has been used for Prevention or treatment of certain diseases such as diabetes, arthritis, or cancer in traditional medicine. In addition, pollen is under investigation as a host cell for a gene expression. This study was undertaken to evaluate the immunologic safety of pollen intake. BALB/c mice were administered with 500, 50,5, or 0.5 mg/kg bw of lily pollen for five times a week for four weeks through gastric intubation. Comparing the control mice administered with distilled water, no significant changes were observed in body weight gain, weight of liver, spleen, lung, and his-topathological findings of liver and kidney of the mice groups administered with the pollen. Plasma level of IgG1, IgG2a, and IgE was not different among the groups. When splenic B lymphocytes were stimulated in vitro with lipopolysaccharides for 7 days, level of IgGl and IgGwa produced in the culture supernatants was not significantly different among the groups. Furthermore, no significant alteration was observed in IL-4 and $IFN{\gamma}$ producing ability with splenic T lymphocytes stimulated in vitro with phytohemagglutinins for 48 hours between the pollen-administered and the control mice. Overall, this study suggests that the lily pollen intake is Inducing no significant modulation of humoral and cell-mediated immunity in mice.

Effect of Kami-Kwiryong-Tang on immune cells in BALB/c Mice (가미귀용탕(加味歸茸湯)이 생쥐의 면역세포(免疫細胞)에 미치는 영향(影響))

  • The purpose of this research was to investigate the effects of Kami Kwiryong Tang (KKT) on the immune cells in BALB/c mice. KKT (500mg/kg) was administerd p.o. once a day for 7 days. KKT decreased the proliferation of thymocytes, but did not affect the proliferation of splenocytes. KKT enhanced the subpopulation of Th (CD4+CD8- single positive cells) cells in splenic T-lymphocytes, but decreased the subpopulation of Th cells in thymocytes. KKT enhanced the production of ${\gamma}$-interferon and interleukin-2, but did not affect the production of interleukin-4 in mice serum. KKT did not affect the production of nitric oxide, but enhanced the phagocytic activity in peritoneal macrophages. These results suggest that KKT is a potent prescription on immune response via the production of cytokines from splenic Th1 cells and the increase of phagocytic activity in vivo.

Comparative Study of 3 kinds of Black Soybean on Murine Immune Cells (생쥐의 면역세포에 대한 검은콩 3종의 비교 연구)

  • The purpose of this research was the comparative study of 3 kinds of black soybean on murine immune cells. The 3 kinds of black soybean are Glycine max Merr. with inner color-yellow (GY), Glycine max Merr. with inner color-greenish (GG) and Rhynchosia volubilis Lour. (RV). All of the black soybean increased the viability of murine thymocytes in vitro. The combined treatment of GY or GG and mitogen did not affect the viability of splenic T- and B-lymphocytes compared with mitogen-treated group, but the combined treatment of RV and mitogen increased their action compared with mitogen-treated group. Also, the 3 kinds of black soybean were given p.o. once a day for 7 days, respectively. RV increased the population of thymic-$CD8^+$, splenic-$CD8^+$ and $B220^+$ cells in vivo. Furthermore, GY and GG did not affect the phagocytic activity and the production of nitric oxide in peritoneal macrophages in vitro, but RV enhanced their action. These results suggest that immunopotentiative action of Rhynchosia volubilis Lour. is more potent than their of Glycine max Merr.

Effects of the Administration of water extract of Juglandis Semen without Inner cortex and with Inner cortex on Activity of Splenocytes and Macrophages in Mice (호두 속껍질 없는 것과 있는 것의 물 추출물 투여가 생쥐의 비장세포 및 대식세포의 활성에 미치는 영향)

  • The purpose of this research was to investigate the effects of the administration of Juglandis Semen without inner cortex (JE) or with inner cortex (JEIC) on activity of splenocytes and peritoneal macrophages in BALB/C mice. JE (300 mg/kg, p.o.) did not affect the cell viability of T- and B-lymphocytes in murine splenocytes, but JEIC (300mg/kg, p.o.) increased the cell viability of T- and B-lymphocytes. Furthermore, JE decreased the population of B220$^+$ cells in splenocytes, but JEIC enhanced the population of Thyl$^+$ cells. Also, JEIC enhanced the population of splenic CD4$^+$ cells. JE decreased the production of nitric oxide and the phagocytic activity of peritoneal macrophages, but JEIC increased the production of nitric oxide and the phagocytic activity of peritoneal macrophages. These results suggest that JEIC is more potent than JE against the immune response induced by splenocytes and macrophages.

Effects of Boheo-tang and Boheo-tang plus Cervi Pantotrichum Cornu : on Immune Response in Postpartum mice (보허탕(補虛湯)과 보허탕가녹용(補虛湯加鹿茸)이 산후 생쥐의 면역반응에 미치는 영향)

  • Purpose: The purpose of this study is to investigate the effect of Boheo-tang (B) and Boheo-tang plus cervi pantotrichum cornu (B+CP) on immune response in postpartum C57BL/6N mice. Methods: Normal saline(control), B and B+CP (8${\mu}{\ell}$/g) were administerd p.o. twice a day for 20 days. Subpopulation of T and B lymphocyte were accessed by flow cytometric analysis. Results: Splenic T and B lymphocytes were increased by the treatment of B. Subpopulation of cytotoxic T lymphocytes in the spleen, were significantly increased by both treatment of B and B+CP. Subpopulation of cytotoxic T lymphcytes in the thymus, were significantly increased by both treatment of B and B+CP. IL-4 production was significantly increased by the treatment of B+CP. Conclusion: This study shows that treatment of Boheo-tang and Boheo-tang plus cervi pantotrichum cornu can improve postpartum immune response in C57BL/6N mice.

Effect of Yonggak-san on the Immuno-regulatory and Apoptosis of Leukemia cells (용각산의 면역조절 및 백혈병세포의 아폽토시스에 미치는 효과)

  • The purpose of this research was to investigate the effect of Yonggak-san (YGS) on the immune reaction and apoptosis of leukemia cells. Administration of YGS(500 mg/kg) enhanced proliferation of splenocytes, thymocytes and mesenteric lymph node cells, and also YGS accelerated subpopulation of splenic Band T, thymic T and mesenteric lymph node-T lymphocytes, especially significantly increased CD4+-TH cells in BALB/c mice. YGS accelerated phagocytic activity and production of nitric oxide in peritoneal macrophages. YGS induced apoptosis of transplanted-L1210 cells in vivo, increased apoptotic cell death of cultured-L1210 and/or Molt4 human leukemia cells, decreased of mitochondrial transmembrane potential of both cells in vitro. These results suggest that YGS have an immune-regulatory effect and anti-cancer property.

  • The purpose of this research was to investigate the immunoregulatory effect, apoptosis of L 1210 leukemia cells of Comus officinalis Sieb.et Zucc. The proliferation of cultured splenocytes and thymocytes were enhanced by the addition of SSY. Splenic, thymic and mesenteric lymph node-T lymphocytes, especially TH cells was significantly increased in SSY-administered (p.o. for 7 days) mice. SSY treatment induced the apoptosis of L1210 mouse leukemia cells. In addition, SSY accelerated the phagocytic activity and nitric oxide production in peritoneal macrophages. These results suggest that SSY have an immuno-regulatory property and anti-cancer effect.

Effect of Chungjokupye-tang on the Anti-cancer Property (청조구폐탕의 항암효과에 관한 연구)

  • The purpose of this research was to investigate the effects of Chungjokupye-tang(CJKPT) on the anti-carcinogenic action. The cell viability of mouse spienocytes and thymocytes were enhanced by the addition of CJKPT. CJKPT were increased of splenic and thymic T lymphocytes, such as T/sub H/ cells were markedly increased by the treatment of CJKPT in vivo. CJKPT treatment induced the apoptotic cell death of Jurkat and HL60 leukemia cells. CJKPT reduced mitochondrial transmembrane potential and increased the expression of ICE, c-myc and p53 gene in Molt-4cells dose dependant manner. These results suggest that CJKPT have an anti-carcinogenic action via immunoregulatory mechanism.
