Effect of Making a Hole in Zona Pellucida by Laser on Hatching of Frozen-thawed ICR Mouse Embryos (레이저를 통한 투명대내의 천공이 동결융해 ICR 마우스 수정란의 부화에 미치는 영향)

  • This study was performed to investigate the effect of laser-assisted hole in the zona pellucida (ZP) of frozen-thawed ICR mouse embryos on the process of hatching that is critical for expanded blastocysts to implant into endometrium, Vitrification medium, composed of ethylene glycol and sucrose supplemented with 7.5% (w/v) PVP, was used to freeze $2{\sim}4$ cell stage embryos recovered from oviducts of superovulated and mated female mice before storing them in $LN_2$. Right after thawing them, a laser beam was shot to make a hole in ZP followed by culturing in KSOM for $96{\sim}120\;hr$ and examining development to blastocyst and hatching every 12 hr. Laser-treated embryos showed significantly higher hatching rate compared to control (92.9% vs. 22.1%, p<0.05). From around Day 4, blastocysts developed from laser-treated embryos started hatching while the blastocysts of control group failed to hatch showing a lot of shrinkage. This study shows that a laser-assisted hole in ZP improves the hatching rate of blastocysts developed from frozen-thawed, in vitro cultured ICR mouse embryos.

Programming Characteristics of the Multi-bit Devices Based on SONOS Structure (SONOS 구조를 갖는 멀티 비트 소자의 프로그래밍 특성)

  • In this paper, the programming characteristics of the multi-bit devices based on SONOS structure are investigated. Our devices have been fabricated by 0.35 $\mu\textrm{m}$ complementary metal-oxide-semiconductor (CMOS) process with LOCOS isolation. In order to achieve the multi-bit operation per cell, charges must be locally frapped in the nitride layer above the channel near the source-drain junction. Programming method is selected by Channel Hot Electron (CUE) injection which is available for localized trap in nitride film. To demonstrate CHE injection, substrate current (Isub) and one-shot programming curve are investigated. The multi-bit operation which stores two-bit per cell is investigated. Also, Hot Hole(HH) injection for fast erasing is used. The fabricated SONOS devices have ultra-thinner gate dielectrics and then have lower programming voltage, simpler process and better scalability compared to any other multi-bit storage Flash memory. Our programming characteristics are shown to be the most promising for the multi-bit flash memory.

Identification, Characterization, and Efficacy Evaluation of Bacillus velezensis for Shot-Hole Disease Biocontrol in Flowering Cherry

  • Though information exists regarding the pathogenesis of the shot-hole disease (SH) in flowering cherry (FC), there has been a lack of research focusing on SH management. Therefore, here, we investigated the inhibitory activities of antagonistic bacteria against SH pathogens both in vitro and in vivo as well as their biochemical characteristics and bioactive compounds. Two biosurfactant-producing bacterial antagonists, identified as Bacillus velezensis strains JCK-1618 and JCK-1696, exhibited the best effects against the growth of both bacterial and fungal SH pathogens in vitro through their cell-free culture filtrates (CFCFs). These two strains also strongly inhibited the growth of the pathogens via the action of their antimicrobial diffusible compounds and antimicrobial volatile organic compounds (VOCs). Crude enzymes, solvent extracts, and biosurfactants of the two strains exhibited antimicrobial activities. Liquid chromatography/electrospray ionization time-of-flight mass spectrometric analysis of the partially purified active fractions revealed that the two antagonists produced three cyclic lipopeptides, including iturin A, fengycin A, and surfactin, and a polyketide, oxydifficidin. In a detached leaf assay, pre-treatment and co-treatment of FC leaves with the CFCFs led to a large reduction in the severity of the leaf spots caused by Epicoccum tobaicum and Bukholderia contaminans, respectively. In addition, the two antagonists produced indole-3-acetic acid, siderophore, and a series of hydrolytic enzymes, along with the formation of a substantial biofilm. To our knowledge, this is the first report of the antimicrobial activities of the diffusible compounds and VOCs of B. velezensis against the SH pathogens and their efficiency in the biocontrol of SH.

  • Recently, with the rapid development in the industries such as mechanical plants, automobiles, ships and marine structures, it is enlarged by the use of the SS 41 steel. This mechanical plants and marine structures are exposed m corrosion because of Cl-under marine environments. To protect their accidents, mainly applied anti-corrosion epoxy coating and various protective its structures. In this study, corrosion control characteristics on the epoxy coating were investigated by the galvanic corrosion of impressed voltage tester under marine environments The main results obtained are as follows; 1. Corrosion current density of amine-epoxy coating becomes more increased than that of other epoxy coating and the time area rate of pin hole and pit until 5% becomes most rapid. 2. The potential of SUS 304 stainless steel(cathode) for Al-epoxy coating is nearly zero potential. 3. Corrosion current density of Amine-epoxy by shot blast becomes more decreased than that of not shot blast and cathodic potential becomes more noble. 4. As distance of anode and cathode is more decreased, corrosion current density of epoxy coating is more increased and cathodic potential becomes less noble.

  • To ensure the safety of the tunnelling without the loss of economy, the tunnel seismic profiling(TSP) method for the prediction ahead of tunnel face, begins to be used routinely in these days. TSP method does not interfere the tunnelling works while the horizontal drilling does, and its prediction length is longer than that of the drilling. Yet the most frequently adopted technique of TSP in Korea is the multi-shot and 2 receiver array using in-hole receivers, even though this array requires as many as 26 drill-holes for receiver installation and ballasting, which results in 3-6 hours of suspension in excavation work. In this paper, multi-receiver and lesser shot array using side-wall attached 3 component geophones is to be described to prove the efficiency in terms of the survey time as well as the reliability of the method by comparison of the predicted weak points and the face mapping results. The predictions mostly agreed with the real fractures or joint developed zones which have been confirmed during the excavation. It also has been found that TSP method can be effectively applied to perform draining ground water ahead of tunnel face by imaging the geologic discontinuities.

  • A numerical model of the igniter for the interior ballistics has been developed combining the lumped parameter model with the theoretical equation of the orifice. With the developed model of the igniter, the numerical study on characteristics of the interior ballistics according to the igniter configuration in terms of the igniter length, the side hole diameter, and the distribution of side holes has been conducted. As results of the calculation of the pressure difference between the breech and shot base, the low frequency oscillations have been influenced by the igniter length, while the high frequency oscillations have been affected by the side hole diameter and the distribution of side holes.

  • Carbon Fiber Reinforced Plastic (CFRP) and Titanium Alloy (Ti6Al4V) stack, extensively used in aerospace structural components are assembled by fasteners and the holes are made using drilling process. Drilling of stack in one shot is a complicated process due to dissimilarity in the material properties. It is vital to have optimal machining condition and tool geometry for better hole quality and tool life. In this study the tool wear and hole quality were analysed by experimental analysis using three modified twist drills and online tool condition monitoring using Acoustics Emission (AE) sensor. Helix angle and point angle influence tool performance and cutting force. It was found that a tool geometry (TG1) with high helix angle of $35^{\circ}$ with low point angle $130^{\circ}$ results in reduction in thrust force of 150-500 N range but the TG2 also perform almost similar to TG1, but when compared with the AErms voltage generated during drilling it was found that progressive rise in voltage in TG1 is less with respect to TG2 which can be attributed to tool life. In process wear monitoring was done using crest factor as monitoring index. AErms voltage were measured and correlated with the performance of the drills.

Channel hot electron (CHE) injection for programming can operate in multi-bit using localized trap in nitride film. CHE injection in our devices is achieved with the single power supply of 5 V. To demonstrate CHE injection, substrate current (Isub) and one-shot programming curve were investigated. The multi-bit operation which stores two-bit per cell is investigated with a reverse read scheme. Also, hot hole injection for fast erasing is used. Due to the ultra-thin gate dielectrics, our results show many advantages which are simpler process, better scalability and lower programming voltage compared to any other two-bit storage flash memory. This fabricated structure and programming characteristics are shown to be the most promising for the multi-bit flash memory.

  • Out of plant-associated bacteria, certain plant growth-promoting bacteria (PGPB) have been reported to increase plant growth and productivity and to elicit induced resistance against plant pathogens. In this study, our objective was to broaden the range of applications of leaf-colonizing PGPB for foliar parts of road tress and pepper. Total 1,056 isolates of endospore-forming bacteria from tree phylloplanes were collected and evaluated for the enzymatic activities including protease, lipase, and chitinase and antifungal capacities against two fungal pathogens, Colletotrichum graminicola and Botrytis cinerea. Fourteen isolates classified as members of the bacilli group displayed the capacity to colonize pepper leaves after spraying inoculation. Three strains, 5B6, 8D4, and 8G12, and the mixtures were employed to evaluate growth promotion, yield increase and defence responses under field condition. Additionally, foliar application of bacterial preparation was applied to the road tress in Yuseong, Daejeon, South Korea, resulted in increase of chlorophyll contents and leaf thickness, compared with non-treated control. The foliar application of microbial preparation reduced brown shot-hole disease of Prunus serrulata L. and advanced leaf abscission in Ginkgo biloba L. Collectively, our results suggest that leaf-colonizing bacteria provide potential microbial agents to increase the performance of woody plants such as tree and pepper through spray application.
