Monitoring of Commercial Cephalopod Products Sold on the South Korea Market using DNA Barcode Information (DNA 바코드를 이용한 국내 유통 두족류 제품의 원재료 모니터링 연구)

  • Cephalopods are one of the most important fishery resources in the world because of their desirable taste and nutritional value. In south Korea, one of the countries in which a large amount of seafood is consumed, cephalopods (e.g., octopus, squid, and cuttlefish) have an annual consumption rate of over 400,000 metric tons. In this study, octopus and squid products (n=28) sold on the market were monitored by analyzing sequences of DNA barcode markers (cytochrome c oxidase subunit I and 16S ribosomal RNA genes). For species identification, the NCBI BLAST database was screened with the sequences and analyzed as a query. In this BLAST search, twelve squid products showed 99-100% sequence identity to Dosidicus gigas (n=3) and Todarodes pacificus (n=9). In the case of the other 16 products that were declared using octopus as raw materials on the labels, six products were identified as Cistopus taiwanicus (n=1), Amphioctopus marginatus (n=1), Scaeurgus unicirrhus (n=1), and Dosidicus gigas (n=3). Monitoring results indicated that a significant percentage (37.5%) of mislabeling was present in octopus products sold on the South Korean market.

Food Component Characterization of Muscle around Pectoral Fin, Salmon Fillet Processing By-products (연어 Fillet 부산물인 가슴지느러미 부근 근육의 식품성분 특성)

  • For the effective use of salmon processing by-products, the food components of muscle around pectoral fin (MAPF) were investigated and compared with those of salmon fillet muscle (SFM). The proximate composition of MAPF was 64.4% for the moisture, 17.2% for the crude protein, 16.2% for the crude lipid and 1.4% for the ash. The pH, volatile basic nitrogen (VBN) content and yield of MAPF were 6.73, 7.8 mg/100 g and 7.4 (g/100 g SFM), respectively. The trichloroacetic acid (TCA) soluble-N content of MAPF was 479 mg/100 g, which was lower than that (612 mg/100 g) of SFM. The Hunter value of MAPF was 54.86 for L value, 22.08 for a value, 22.41 for b value and 52.35 for ${\Delta}$E value, which were similar to those of SFM. The total content in amino acids of MAPF was 17.1 g/100 g, its major amino acids were aspartic acid, glutamic acid, leucine and lysine. The major fatty acids of MAPF were 16:0, 18:1n-9 and 18:2n-6, 20:5n-3 and 22:6n-3. No differences were found in the major fatty acids and the major amino acids between MAPF and SFM. The calcium and phosphorus contents of MAPF were 19.1 mg/100 g and 211.7 g/100 g, respectively. These results suggested that MAPF could be used as seafood resources.

Solubility Improvement of Cuttle Bone Powder Using Organic Acids (유기산처리에 의한 갑오징어갑 분말의 가용성 개선)

  • As a pan of a study on effective use of seafood processing by-products, such as cuttle bone as a calcium source, we examined on the kind of organic acid (acetic acid and lactic acid), reaction concentration (mole ratio of calcium to mole of organic acid), reaction temperature $(20\~60^{\circ}C)$ and reaction time (6$\~$24 hours) as reaction conditions for the solubility improvement of cuttle bone powder. The high soluble cuttle bone powder was also prepared from the optimal reaction conditions and partially characterized. From the results on examination of reaction conditions, the high soluble cuttle bone powder was prepared with 0.4 in mole ratio of a calcium to mole of a acetic acid at room temperature for 12 hours, Judging from the patterns of IR and X-ray diffraction, the main component of the high soluble cuttle bone powder was presented as a form of calcium acetate, and a scanning electron micrograph showed an irregular form. The soluble calcium content in the high soluble cuttle bone powder was $5.3\%$ and it was improved about 1,380 times compared to a raw cuttle bone powder. For the effective use of the high soluble cuttle bone powder as a material for a functional improvement in processing, it should be used after the calcium treatment at room temperature for about 1 hour in tap water or distilled water. from these results, we concluded that it is possible to use the high soluble cut시e bone powder as a material for a functional improvement in processing.

  • This study is aimed to investigate the structure of world pollack markets and the position/competitiveness or Korea for WTO/NAMA Negotiation. First or all, it is clearly pointed out that many limitations and problems are inherent in FAO statistics that is widely utilized to investigate the structure of international seafood markets. Especially, it is impossible to find not only the data for Russia that is the top production and export country of pollack, but also the data for importing/exporting countries for pollack. In order to make up for these problems, the data for export and import of major countries are collected and analyzed. The results of analysis show the followings. First, it is clearly investigated that classification of fish products are different for countries. Second, it is understood the structure of international pollack market in actuality. The pollack market is segmented by frozen, fresh, dried, fillet, roe, surimi, etc. In addition, the pollack market has grown as much as 600,000 tons in amount and $1.2billion in value. Third, competitiveness of Korea in international pollack markets is measured quantitatively. It shows that Korea has low RAC index and TSI index, but high RMI index. Thus, it is identified that Korea becomes the largest pollack importing country. Fourth, the partial equilibrium analysis on pollack import market of Korea indicates that the frozen pollack has both price elasticity and substitution elasticity, while the fresh pollack has income elasticity.

Listeriosis and Listeria monocytogenes (리스테리아증과 Listeria monocytogenes)

  • Listeria monocytogenes, one of five species in the genus Listeria and the only one currently believed to be pathogenic for humans, is a small gram-positive, nonsporeforming, aerobic, motile and hemolytic rod-shaped bacterium. The bacterium is widespread in the environment, having been isolated from soil, dust, animal feed, water, sewage, almost every type of animal that has been cultured, and asymptomatic humans. L. monocytogenes causes listeriosis, a disease which most often affects humans with a compromised immune system. Included are pregnant woman, infants and adults suffering from such diseases as cancer, cirrhosis of liver or AIDS or are being treated with drugs such as corticosteroids. Listeriosis is manifested by such syndromes as pregnancy infections, granulomatosis infantiseptica, sepsis, meningoencephalitis, and focal infections. Infections, can be treated successfully with penicillin, ampicillin, or erythromycin. However, a mortality rate of about 30% has occurred in outbreaks of listeriosis. Food-associated outbreaks of listeriosis have been attributed to coleslaw (Canada, 1981), pasteurized milk (U.S., 1983), and soft cheese (U.S., 1985). Presence of L. monocytogenes in various dairy foods has prompted recall of such products from the U.S. market-place. L. monocytogenes also has been found in raw meats and seafood.

Diet and Colorectal Cancer Risk in Asia - a Systematic Review

  • Diet is one of the major factors that can exert a majorly influence on colorectal cancer risk. This systematic review aimed to find correlations between various diet types, food or nutrients and colorectal cancer risk among Asian populations. Search limitations include dAsian populations residing in Asia, being published from the year 2008 till present, and written in the English language. A total of 16 articles were included in this systematic review. We found that red meats, processed meats, preserved foods, saturated/animal fats, cholesterol, high sugar foods, spicy foods, tubers or refined carbohydrates have been found by most studies to have a positive association with colorectal cancer risk. Inversely, calcium/dairy foods, vitamin D, general vegetable/fruit/fiber consumption, cruciferous vegetables, soy bean/soy products, selenium, vitamins C,E and B12, lycophene, alpha-carotene, beta-carotene, folic acid and many other vitamins and minerals play a protective role against colorectal cancer risk. Associations of fish and seafood consumption with colorectal cancer risk are still inconclusive due to many varying findings, and require further more detailed studies to pinpoint the actual correlation. There is either a positive or no association for total meat consumption or white meats, however their influence is not as strong as with red and processed meats.

Formulation of Surimi and Surimi-based Products with Acceptable Gelling Ability from Squid Muscle (가열 젤 형성능을 가진 오징어 Surimi와 Surimi-based 제품을 위한 첨가물의 최적화)

  • We investigated the optimum formulation to improve the gelling ability of squid, Dosidicus gigas, surimi. The solubility of minced squid muscle was highest at pH 10.7, and lowest at pH 5.0. The yields of conventional surimi and protein recovery after alkaline pH-shift processing were $68.1{\pm}2.4%$ and $65.3{\pm}2.6%$, respectively, whereas the protein recovery with acidic pH-shift processing was only $21.2{\pm}1.6%$. The addition of 5% starch decreased the breaking force regardless of the kind of starch, while the mixture of corn, potato, and wheat starch (total 15%) increased the breaking force by up to 1.9 fold. The addition of 5% egg white, 5% porcine plasma protein, 0.3% $CaCl_2$, and 0.3% Polymix GA significantly increased the breakingforce (P<0.05). None of the ingredients examined in this study significantly affected the deformation value (P<0.05). The optimum concentrations of egg white and $CaCl_2$ to obtain a breaking force of 55 g and a whiteness of 70 were 2.69% and 0.22%, respectively.

Inhibitory effect of bacteriocin-producing lactic acid bacteria against histamine-forming bacteria isolated from Myeolchi-jeot

  • The objectives of this study were to identify the histamine-forming bacteria and bacteriocin- producing lactic acid bacteria (LAB) isolated from Myeolchi-jeot according to sequence analysis of the 16S rRNA gene, to evaluate the inhibitory effects of the bacteriocin on the growth and histamine accumulation of histamine-forming bacteria, and to assess the physico-chemical properties of the bacteriocin. Based on 16S rRNA gene sequences, histamine-forming bacteria were identified as Bacillus licheniformis MCH01, Serratia marcescens MCH02, Staphylococcus xylosus MCH03, Aeromonas hydrophila MCH04, and Morganella morganii MCH05. The five LAB strains identified as Pediococcus acidilactici MCL11, Leuconostoc mesenteroides MCL12, Enterococcus faecium MCL13, Lactobacillus sakei MCL14, and Lactobacillus acidophilus MCL15 were found to produce an antibacterial compound with inhibitory activity against the tested histamine-producing bacteria. The inhibitory activity of these bacteriocins obtained from the five LAB remained stable after incubation at pH 4.0-8.0 and heating for 10 min at $80^{\circ}C$; however, the bacteriocin activity was destroyed after treatment with papain, pepsin, proteinase K, ${\alpha}$-chymotrypsin, or trypsin. Meanwhile, these bacteriocins produced by the tested LAB strains also exhibited histamine-degradation ability. Therefore, these antimicrobial substances may play a role in inhibiting histamine formation in the fermented fish products and preventing seafood-related food-borne disease caused by bacterially generated histamine.

A study on the market-based fisheries resource management for the sustainable fishery (지속적 어업을 위한 시장기반 수산자원관리 연구)

  • The market-based fisheries resources management system was reviewed and the improvement scheme was studied for seeking the sustainability of marine ecosystems and their fisheries resources. A demand-side policy is currently emerging to the management of fisheries resources by reducing over-exploitation, based on the consumer's selection. The role of consumers in the sustainable fishery was studied by comparing the social responsibility and consumer's activity between Korea and foreign countries and international NGO groups, based on the FAO guideline. The adoption of the Marine Stewardship Council (MSC)'s eco-labelling certificate was suggested by expanding the existing environment-friendly certification system in Korea. This new system will contribute to the protection of young immature fish and spawners in coastal and offshore fisheries as well as to the international trade of seafood in terms of fish products from well-managed fisheries. It is noted that the consumer's activities in the markets as regulators will contribute to the ecosystem health and sustainability.

Characterization of Acid-soluble Collagen from Alaska Pollock Surimi Processing By-products (Refiner Discharge)

  • The study was carried out to examine on the refiner discharge from Alaska pollock as a collagen resource by characterizing biochemical and functional properties of collagen. The refiner discharge from Alaska pollock surimi manufacturing was a good resource for collagen extraction according to the results of total protein, heavy metal, volatile basic nitrogen, collagen content, amino acid composition, and thermal denaturation temperature (TDT). TDT of acid soluble collagen from refiner discharge showed $20.7^{\circ}C$, which was similar to that of collagen from Alaska pollock muscle and was higher than that of collagen from Alaska pollock skin. TDT of acid-soluble collagen from refiner discharge was, however, lower than those of skin collagens from warm fish and land animal. Acid-soluble collagen from refiner discharge of Alaska pollock could be used as a functional ingredient for food and industrial applications according to the results of water and oil absorption capacities, and emulsion properties. In addition, if the thermal stability of the acid-soluble collagens is improved, collagen from refiner discharge from Alaska pollock could be more effectively used.
