  • The Ras superfamily of small G-proteins acts as a molecular switch on the intracellular signaling pathway. Upon ligand stimulation, inactive GTPases (Ras-GDP) are activated (Ras-GTP) using guanine nucleotide exchange factor (GEF) and transmit signals to their downstream effectors. Following signal transmission, active Ras-GTP become inactive Ras-GDP and cease signaling. However, the intrinsic GTPase activity of Ras proteins is weak, requiring Ras GTPase-activating protein (RasGAP) to efficiently convert RAS-GTP to Ras-GDP. Since deregulation of the Ras pathway is found in nearly 30% of all human cancers, it might be useful to clarify the structural and physiological roles of Ras GTPases. Recently, RasGAP has emerged as a new class of tumor-suppressor protein and a potential therapeutic target for cancer. Therefore, it is important to clarify the physiological roles of the individual GAPs in human diseases. The first RasGAP discovered was RASA1, also known as p120 RasGAP. RASA1 is widely expressed, independent of cell type and tissue distribution. Subsequently, neurofibromatosis type 1 (NF1) was discovered. The remaining GAPs are affiliated with the GAP1 and synaptic GAP (SynGAP) families. There are more than 170 Ras GTPases and 14 Ras GAP members in the human genome. This review focused on the current understanding of Ras GTPase and RasGAP in human diseases, including cancers.

  • Urbanization increases the amount of impervious surface and artificial heat emission, resulting in urban heat island (UHI) effect. Local climate zones (LCZ) are a classification scheme for urban areas considering urban land cover characteristics and the geometry and structure of buildings, which can be used for analyzing urban heat island effect in detail. This study aimed to examine the UHI effect by urban structure in Suwon and Daegu using the LCZ scheme. First, the LCZ maps were generated using Landsat 8 images and convolutional neural network (CNN) deep learning over the two cities. Then, Surface UHI (SUHI), which indicates the land surface temperature (LST) difference between urban and rural areas, was analyzed by LCZ class. The results showed that the overall accuracies of the CNN models for LCZ classification were relatively high 87.9% and 81.7% for Suwon and Daegu, respectively. In general, Daegu had higher LST for all LCZ classes than Suwon. For both cities, LST tended to increase with increasing building density with relatively low building height. For both cities, the intensity of SUHI was very high in summer regardless of LCZ classes and was also relatively high except for a few classes in spring and fall. In winter the SUHI intensity was low, resulting in negative values for many LCZ classes. This implies that UHI is very strong in summer, and some urban areas often are colder than rural areas in winter. The research findings demonstrated the applicability of the LCZ data for SUHI analysis and can provide a basis for establishing timely strategies to respond urban on-going climate change over urban areas.

  • Ultraviolet (UV) irradiation is a relevant environment factor to induce cellular senescence and photoaging. Both autophagy- and silent information regulator T1 (SIRT1)-dependent pathways are critical cellular processes of not only maintaining normal cellular functions, but also protecting cellular senescence in skin exposed to UV irradiation. In the present studies, we investigated whether modulation of autophagy induction using a novel synthetic SIRT1 activator, heptasodium hexacarboxymethyl dipeptide-12 (named as Aquatide), suppresses the UVB irradiation-induced skin aging. Treatment with Aquatide directly activates SIRT1 and stimulates autophagy induction in cultured human dermal fibroblasts. Next, we found that Aquatide-mediated activation of SIRT1 increases autophagy induction via deacetylation of forkhead box class O (FOXO) 1. Finally, UVB irradiation-induced cellular senescence measured by $SA-{\beta}-gal$ staining was significantly decreased in cells treated with Aquatide in parallel to occurring SIRT1 activation-dependent autophagy. Together, Aquatide modulates autophagy through SIRT1 activation, contributing to suppression of skin aging caused by UV irradiation.

  • This study aims to reduce the production cost of mushrooms and therefore boost the income of mushroom producers. The addition of a 10% concentration of herb medicine refuse was adequate for promoting mycelium growth in Flammulina velutipes. A moisture content of 60% in the culture medium was adequate for promoting hyphal growth. The optimum temperature and humidity were $16^{\circ}C$ and 70%-85%, respectively. The average yield of Flammulina velutipes fruting body per 1,100 ml bottle was 275-282 g, which is similar to that of the control. The outbreak of diseases such as mildew and bacterial blotch in mushroom was never observed in the bottle treated with herb medicine refuse. Fewer deformed and second-class quality mushrooms were observed than in the control group. Deviation reduced, and mushroom growth was observed to be significantly uniform. The winter mushroom had a white fruiting body with a hemispherical shape. The diameter of the mushroom cap was 9.2-9.3 mm, stipe length was 12.7-12.8 cm, and thickness was 3.3-3.4 mm; these were almost the same as those in the control group. The partial hardness of winter mushrooms in both the experimental and the control groups showed that the stipe ($54.6-57.3g/cm^2$) is a little harder than the mushroom cap ($46.8-47.6g/cm^2$). The calories and nutrients per 100 g of mushrooms in the control and samples were similar to each other.

  • Gibberllic acid ($GA_3$) is one of gibberellins (GAs) that are a class of plant growth hormones that exert profound and diverse effects on plant growth and development. $GA_3$ is essentially non-UV absorbing and is difficult to assay by UV-detector. For effective extraction of gibberellic acid from fruits by using liquid-liquid extraction, optimized pH and extraction solvent were established. The selective and sensitive derivative of $GA_3$ for HPLC/UV-vis was derivatized using phenacyl bromide, and the experimental factors, including reaction time, reaction temperature and amount of derivatizing reagent and base were investigated for the effective synthesis. The derivatized $GA_3$ with phenacyl bromide was effectively analyzed by HPLC/UV-vis. The structure of derivatized $GA_3$ was confirmed by HPLC/ESI-MS. For apple, LOD and LOQ were 0.008 mg/kg and 0.027 mg/kg, respectively. For pear, LOD and LOQ were 0.003 mg/kg, 0.012 mg/kg, respectively. The established method can be applied to more effective analysis of $GA_3$ from plant and food.
