In Vitro Selection of RNA Aptamer Specific to Salmonella Typhimurium

  • Salmonella is a major foodborne pathogen that causes a variety of human diseases. Development of ligands directly and specifically binding to the Salmonella will be crucial for the rapid detection of, and thus for efficient protection from, the virulent bacteria. In this study, we identified a RNA aptamer-based ligand that can specifically recognize Salmonella Typhimurium through SELEX technology. To this end, we isolated and characterized an RNase-resistant RNA aptamer that bound to the OmpC protein of Salmonella Typhimurium with high specificity and affinity ($K_d$ ~ 20 nM). Of note, the selected aptamer was found to specifically bind to Salmonella Typhimurium, but neither to Gram-positive bacteria (Staphylococcus aureus) nor to other Gram-negative bacteria (Escherichia coli O157:H7). This was evinced by aptamer-immobilized ELISA and aptamer-linked precipitation experiments. This Salmonella species-specific aptamer could be useful as a diagnostic ligand against pathogen-caused foodborne sickness.

도계장과 돈육가공장에서 분리된 살모넬라 속의 특성 연구 (Characteristics of Salmonella Species Isolates from Poultry Slaughterhouses and Pork Meat Processing Plants)

  • This study was carried out to isolate and identify Salmonella species from poultry slaughterhouses and pork meat processing plants during the period from January 1997 to August 1998, and analyze resistance of antimicrobial agents and plasmid profiles of isolated Salmonella strains. A total of 15 Salmonella strains was isolated from poultry carcasses, swine carcasses, pork meats and cutting boards. Identified Salmonella strains were S. typhimurium, S. heidelberg, S. hilingdon, S. mbandaka, and S. virginia. Ten (66.7%) of 15 Salmonella strains showed resistance to antimicrobial agents and five strains (33.3%) of them were resistant to two or more antimirobial agents. Plasmids were isolated from three Salmonella isolates which had two or more plasmids.

Effects of Bacillus subtilis on Growth Performance and Resistance to Salmonella Infection in Broiler Chickens

  • The experiment was undertaken to see the effects of Bacillus sp. on the growth performance and disease resistance to Salmonella sp. infections. The use of probiotic microbes in poultry is commonly practiced. In this study, Bacillus subtilis was tested using a total of 120 chicks of age of 1 day after hatching. The growth traits examined were body weight gain and feed conversion rate. And also, the Salmonella resistance of Bacillus subtilis was tested after the chicks were orally administered with Salmonella pullorum by gavage force injections. The result showed that Bacillus subtilis yielded a high feed efficiency, consequently increased growth rate. For the effect of Bacillus subtilis on Salmonella infection, Bacillus subtilis significantly improved the resistance to Salmonella pullorum infection. Various clinical symptoms of Salmonella infection were highly decreased by addition of Bacillus sp.

양계 일반농장과 동물복지농장에서의 환경 샘플링을 통한 살모넬라 검출율 비교 (Comparison of Detection Rate of Salmonella spp. in Environment Sampling of Conventional and Welfare Chicken Farms)

  • 최근 산업동물에서도 동물복지의 관심이 높아짐에 따라 복지 사육방식을 채택한 농가의 수가 점점 늘어나고 있다. 본 연구는 복지농장과 일반농장 간의 살모넬라 검출율을 확인하였다. 각 농가에서 얻어진 샘플들을 이용하여 비교한 결과, 산란계와 육계농장 모두 복지농장이 일반농장에 비해 높은 검출율을 나타냈다. 산란계 농장의 경우 통계학적 유의차는 없었으나(P value, 1.00), 일반 농장 5개 농가에서는 살모넬라가 검출되지 않았으며 복지 농장 5개 농가에서 채취된 130개 샘플 중에서 1개 샘플에서 Salmonella enterica subsp. Enteritidis(SE)가 검출되었다. 육계 농장의 경우, 일반농장 10개 농가의 200개 샘플 중에 5개 농가의 7개 샘플에서 살모넬라가 검출되었으며, 복지농장 5개 농가의 200개 샘플 중 4개 농가의 21개 샘플에서 검출되었으며, 일반농장과 복지농장의 살모넬라 검출율간 통계학적 유의차를 보였다(P value, 0.0057). 일반농장의 2개 분리주는 Salmonella enterica subsp. Senftenberg, SE로 혈청형이 확인되었고 복지농장의 2개 분리주는 Salmonella enterica subsp. Grampian, Salmonella enterica subsp. Virchow로 혈청형이 확인되었다. 앞으로 국내에서 사육방식의 차이에 따른 지속적인 연구와 복지농장에서의 미생물 모니터링이 필요하며, 계사내 적절한 방역대책도 필요하다고 사료된다.

LAMP-3 (Lysosome-Associated Membrane Protein 3) Promotes the Intracellular Proliferation of Salmonella typhimurium

  • Lysosomes are cellular organelles containing diverse classes of catabolic enzymes that are implicated in diverse cellular processes including phagocytosis, autophagy, lipid transport, and aging. Lysosome-associated membrane proteins (LAMP-1 and LAMP-2) are major glycoproteins important for maintaining lysosomal integrity, pH, and catabolism. LAMP-1 and LAMP-2 are constitutively expressed in Salmonella-infected cells and are recruited to Salmonella-containing vacuoles (SCVs) as well as Salmonella- induced filaments (Sifs) that promote the survival and proliferation of the Salmonella. LAMP-3, also known as DC-LAMP/CD208, is a member of the LAMP family of proteins, but its role during Salmonella infection remains unclear. DNA microarray analysis identified LAMP-3 as one of the genes responding to LPS stimulation in THP-1 macrophage cells. Subsequent analyses reveal that LPS and Salmonella induced the expression of LAMP-3 at both the transcriptional and translational levels. Confocal Super resolution N-SIM imaging revealed that LAMP-3, like LAMP-2, shifts its localization from the cell surface to alongside Salmonella. Knockdown of LAMP-3 by specific siRNAs decreased the number of Salmonella recovered from the infected cells. Therefore, we conclude that LAMP-3 is induced by Salmonella infection and recruited to the Salmonella pathogen for intracellular proliferation.

Characterization of Salmonella species from poultry slaughterhouses in South Korea: carry-over transmission of Salmonella Thompson ST292 in slaughtering process

  • Importance: Salmonella outbreaks linked to poultry meat have been reported continuously worldwide. Therefore, Salmonella contamination of poultry meats in slaughterhouses is one of the critical control points for reducing disease outbreaks in humans. Objective: This study examined the carry-over contamination of Salmonella species through the entire slaughtering process in South Korea. Methods: From 2018 to 2019, 1,097 samples were collected from the nine slaughterhouses distributed nationwide. One hundred and seventeen isolates of Salmonella species were identified using the invA gene-specific polymerase chain reaction, as described previously. The serotype, phylogeny, and antimicrobial resistance of isolates were examined. Results: Among the 117 isolates, 93 were serotyped into Salmonella Mbandaka (n = 36 isolates, 30.8%), Salmonella Thompson (n = 33, 28.2%), and Salmonella Infantis (n = 24, 20.5%). Interestingly, allelic profiling showed that all S. Mbandaka isolates belonged to the lineage of the sequence type (ST) 413, whereas all S. Thompson isolates were ST292. Moreover, almost all S. Thompson isolates (97.0%, 32/33 isolates) belonging to ST292 were multidrug-resistant and possessed the major virulence genes whose products are required for full virulence. Both serotypes were distributed widely throughout the slaughtering process. Pulsed-field gel electrophoretic analysis demonstrated that seven S. Infantis showed 100% identities in their phylogenetic relatedness, indicating that they were sequentially transmitted along the slaughtering processes. Conclusions and Relevance: This study provides more evidence of the carry-over transmission of Salmonella species during the slaughtering processes. ST292 S. Thompson is a potential pathogenic clone of Salmonella species possibly associated with foodborne outbreaks in South Korea.

부산지역의 설사환자로부터 분리한 Salmonella sp. 및 Shigella sp.의 약제내성 유형에 관한 연구 (Drug Resistance Profiles of Salmonella sp. and Shigella sp. Isolated from Diarrheal Patients in Pusan, Korea)

  • 병원을 찾은 설사환자의 분변으로 부터 Salmonella sp.와 Shigella sp.을 분리하고 혈청형, 월별, 연령별분포 및 약제내성 양상을 조사하였다. 311명의 설사환자로 부터 34주(10.9%)의 Salmonella sp.와 25주(8%)의 Shigella sp.을 분리하였다. 분리된 Salmonella sp.의 혈청형은 group D(50%)가 가장 많았고, Shigella sp.은 대부분(96%)이 group D에 속하였다. 분리된 Salmonella sp.의 월별 및 연령별 분포는 계절과 연령에 크게 관계없이 분리되는 경향을 보였고, 이와는 달리 Shigella sp.은 7월(64%)과 10대(76%)에서 높은 분리율을 나타내었다. 분리균주에 대하여 약제내성을 조사한 결과 Shlmonella sp.는 streptomycin에 공시한 전 분리균이 내성을 나타내었고, erythromycin과 novobiocin(90.6%), penicillin G(65.6%) 및 cephalexin(46.9%)에, Shigella sp.는 erythromycin(95.8%), novobiocin(91.7%) 및 streptomycin(87.5%)에 각각 높은 내성을 나타내었다. 또한 Salmonella sp.와 Shigella sp.의 내성양상은 각각 18종류 및 4종류의 내성형으로 분포하고 있었다.

Mousse cake와 Tiramisu에 인위접종된 Salmonella Typhimurium의 식품공전 분리배지, Real-time PCR과 Loop-mediated isothermal amplification-bioluminescence의 검출 특성 비교 (Comparison of Isolation Agar Method, Real-Time PCR and Loop-Mediated Isothermal Amplification-Bioluminescence for the Detection of Salmonella Typhimurium in Mousse Cake and Tiramisu)

  • 최근 한국에서 발생한 Salmonella로 인한 식중독 사고는 2018년 9월 학교급식에서 제공된 초콜릿 무스 케이크가 원인이 되었다. 이 연구의 목적은 Salmonella Typhimurium이 인위적으로 접종된 무스케이크와 티라미수에서 3M Molecular Detection Assay 2 - Salmonella와 식품공전에 등재된 방법인 분리배지와 real-time PCR을 비교하는 것이었다. 무스케이크 2종과 티라미수 2종 25 g에 225 mL BPW를 넣고 $37^{\circ}C$에서 24시간 동안 증균 배양하였다. 배양 후, 3M Molecular Detection Assay 2 - Salmonella, 분리배지 그리고 real-time PCR로 분석하였다. 초콜릿 무스 케이크를 제외하고 3가지 방법은 유사한 결과를 보였다. 초콜릿 무스 케이크에서 분리배지와 3M Molecular Detection Assay 2 - Salmonella는 모든 접종수준에서 동일한 결과를 나타낸 반면 real-time PCR은 $10^4CFU/25g$ 수준에서 1번의 양성결과를 제외하고 모두 검출되지 않았다. 초콜릿 무스에 S. Typhimurium을 $10^2CFU/25g$ 수준으로 접종하였을때, real-time PCR를 이용한 검출은 15%에서는 부분적인 음성을 나타냈고, 20-100% 함량의 초콜릿 무스에서는 모두 음성이었다. Real-time PCR로는 chocolate이 15% 이상 함유된 식품에서의 Salmonella균 검출이 불가능하였지만, LMAP 기반의 3M Molecular Detection Assay 2으로는 chocolate 농도에 관계없이 검출이 가능하였다.

A Study on the Prevention of Salmonella Infection by Using the Aggregation Characteristics of Lactic Acid Bacteria

  • Salmonella is one of the major pathogenic bacteria that cause food poisoning. This study investigated whether heat-killed as well as live Lactobacillus protects host animal against Salmonella infection. Live and heat-killed Lactobacillusacidophilus was administered orally to Sprague-Dawley rats for 2 weeks before the rats were inoculated with Salmonella. Rise in body temperature was moderate in the group that was treated with heat-killed bacteria as compared to the Salmonella control group. The mean amount of feed intake and water consumption of each rat in the heat-killed bacteria group were nearly normal. The number of fecal Salmonellae was comparable between the live and the heat-killed L. acidophilus groups. This finding shows that L. acidophilus facilitates the excretion of Salmonella. Moreover, the levels of pro inflammatory cytokines, including tumor necrosis factor (TNF)-alpha and interleukin (IL)-1 beta, in the heat-killed L. acidophilus group were significantly lower when compared to the levels in the Salmonella control group. These results indicate that nonviable lactic acid bacteria also could play an important role in preventing infections by enteric pathogens such as Salmonella.

Evaluation of Salmonella cross contamination at retail chicken meat outlets in Kandy, Sri Lanka

  • This study was carried out to evaluate the Salmonella cross contamination at retail chicken meat outlets in Kandy, Sri Lanka and to identify the risk factors for Salmonella contamination at retail chicken meat outlets. Fifteen retail chicken meat outlets in Kandy area were randomly selected. Pre-tested questionnaires were used to collect data for identifying risk factors of Salmonella cross contamination at retail outlets and swab samples from meat contact surfaces/utensils were obtained. Out of 57 swab samples collected from meat contact surfaces/utensils, the overall prevalence of Salmonella cross contamination was 21%. Weighing scale (33%), meat containing trays/buckets (27%) and cutting board (25%) showed the highest percentage of Salmonella prevalence whereas knife and showcase showed relatively low percentages, 14% and 9%, respectively. Retail chicken outlets with slaughtering facilities had a significantly higher prevalence of Salmonella cross contamination than those without slaughtering facilities. This higher rate of Salmonella cross contamination at retail chicken meat outlets could be attributed to lack of proper cold chains and minimal facilities, and poor level of hygiene in those outlets.
