Anti-fatigue activity of a mixture of seahorse (Hippocampus abdominalis) hydrolysate and red ginseng

  • Seahorse, a syngnathidae fish, is one of the important organisms used in Chinese traditional medicine. Hippocampus abdominalis, a seahorse species successfully cultured in Korea, was validated for use in food by the Ministry of Food and Drug Safety in February 2016; however. the validation was restricted to 50% of the entire composition. Therefore, to use H. abdominalis as a food ingredient, H. abdominalis has to be prepared as a mixture by adding other materials. In this study, the effect of H. abdominalis on muscles was investigated to scientifically verify its potential bioactivity. In addition, the anti-fatigue activity of a mixture comprising H. abdominalis and red ginseng (RG) was evaluated to commercially utilize H. abdominalis in food industry. H. abdominalis was hydrolyzed using Alcalase, a protease, and the effect of H. abdominalis hydrolysate (HH) on the muscles was assessed in C2C12 myoblasts by measuring cell proliferation and glycogen content. In addition, the mixtures comprising HH and RG were prepared at different percentages of RG to HH (20, 30, 40, 50, 60, 70, and 80% RG), and the anti-fatigue activity of these mixtures against oxidative stress was assessed in C2C12 myoblasts. In C2C12 myoblasts, $H_2O_2$-induced oxidative stress caused a decrease in viability and physical fatigue-related biomarkers such as glycogen and ATP contents. However, treatment with RG and HH mixtures increased cell viability and the content of fatigue-related biomarkers. In particular, the 80% RG mixture showed an optimum effect on cell viability and ATP synthesis activity. In this study, all results indicated that HH had anti-fatigue activity at concentrations approved for use in food by the law in Korea. Especially, an 80% RG to HH mixture can be used in food for ameliorating fatigue.

Temporal and Spatial Downregulation of Arabidopsis MET1 Activity Results in Global DNA Hypomethylation and Developmental Defects

  • DNA methylation is an epigenetic mechanism for gene silencing. In Arabidopsis, MET1 is the primary DNA methyltransferase that maintains CG DNA methylation. Plants having an overall reduction of MET1 activity, caused by a met1 mutation or a constitutively expressed MET1 antisense gene, display genome hypomethylation, inappropriate gene and transposon transcription, and developmental abnormalities. However, the effect of a transient reduction in MET1 activity caused by inhibiting MET1 expression in a restricted set of cells is not known. For this reason, we generated transgenic plants with a MET1 antisense gene fused to the DEMETER (DME) promoter (DME:MET1 a/s). Here we show that DME is expressed in leaf primordia, lateral root primoridia, in the region distal to the primary root apical meristem, which are regions that include proliferating cells. Endogenous MET1 expression was normal in organs where the DME:MET1 a/s was not expressed. Although DME promoter is active only in a small set of cells, these plants displayed global developmental abnormalities. Moreover, centromeric repeats were hypomethylated. The developmental defects were accumulated by the generations. Thus, not maintaining CG methylation in a small population of proliferating cells flanking the meristems causes global developmental and epigenetic abnormalities that cannot be rescued by restoring MET1 activity. These results suggest that during plant development there is little or no short-term molecular memory for reestablishing certain patterns of CG methylation that are maintained by MET1. Thus, continuous MET1 activity in dividing cells is essential for proper patterns of CG DNA methylation and development.

중국 연변 수학 교과서의 실천과 종합응용 영역에 나타난 학습내용 분석 (Analysis of the contents of Practice and Synthetic Application area in Yanbian Textbooks)

  • 본 연구에서는 중국의 수학과 '교육과정표준'에서 4개의 하위 영역 중 하나로 제시하고 있는 실천과 종합응용의 내용을 구현한 연변의 초등학교 수학 교과서 내용을 분석하였다. 이를 위해 연변교육출판사에서 출판한 12권의 초등학교 수학 교과서를 이용하여 실천과 종합응용 내용을 분석하였다. '교육과정표준'의 실천과 종합응용에서는 수학적 지식과 경험을 활용하고 응용하여 도전적이고 종합적인 문제를 해결하고, 주요 학습내용에 대한 이해와 연계를 체험하도록 제시하고 있다. 교과서 분석 결과, 실천활동에서는 상황적 배경이 대부분 교실 안에서 할 수 있는 활동으로 제한되어 있다는 것과 종합응용에서는 수학 지식의 상호연계성을 인식할 수 있는 활동이 적다는 한계를 보였다. 수학적 과정 면에서는 문제해결이 주를 이루고 있으며, 부분적으로 의사소통 활동이 제시되어 있었으며, 추론 활동이 적게 나타났다. 또 수학적 활동에서도 대부분 체험활동이 주를 이루고 있으며, 수학적 지식을 타 교과나 타 영역에 통합할 수 있는 통합적 활동이 부족한 것으로 나타났다.

강도가 점증하는 전신활동에 따른 반응시간의 변화와 정신작업의 정확도 (Variation of reaction time and accuracy of mental work with strength of whole-body activity gradually increasing)

  • 산업현장에서는 높은 강도의 육체적 작업에 병행하여 정신적 작업이 요구되는 경우가 있다. 육체적 작업 후에 피로의 회복을 위한 충분한 시간이 주어지지 않는 경우, 이 피로에 수반되는 정신적 피로에 의해 정신적 작업의 정확도는 저하된다는 보고가 있다. 그러나 본 연구에서는 높은 강도의 근육활동에 숙련된 그룹과 숙련되지 못한 그룹 모두에서 본 연구를 위한 실험조건 하에서는 오히려 육체적 활동 후의 정신적 작업의 정확도가 증가한다는 것이 밝혀졌다. 또한 물리적 자극에 대한 이들 그룹의 반응시간을 측정하여 정신작업의 정확도와의 관계를 해석하였다. 그 결과 반응시간은 높은 강도의 근육활동에 숙련된 그룹에서만 운동강도에 따라 안정적으로 점증하였고, 모든 피실험자 그룹에 대해서 정신작업의 정확도와 반응시간은 밀접한 관계에 있지 않음을 알 수 있었다.

Conflicts between Protected Franchise Territory and Unrestricted Delivery Trading Area

  • Propose: The expansion of the delivery market in franchise business is a positive means of advertising and promotion for franchisees and contributes to substantial sales growth for stores. However, unrestricted and uncontrolled delivery sales and business activity of franchisee directly lead to encroachment of business territory between franchisees, resulting in increased operating costs of franchisees and conflicts between franchisees over rights and interests on franchise business. Moreover, in relation to the restrictions on the opening of competitors, it is incapacitating the law intent of prohibiting unfair encroachment of business territory, in the issue of the Fair Franchise Act in regard to guarantee of exclusive sales and business activity rights of franchisee. This study aims to point out major problems arising out of franchisee's infinite competition on delivery sales and business activity that are not restricted or controlled and then suggest legal supplements, policy tasks, and practical implications for improvement on the issues. Literature Review: In franchising business transactions, vertical restraints are associated with the exclusive territory establishment, control of transaction area, restriction of intangible sales and business activity of franchisee. Therefore, in the franchise business, it is necessary to take positive interpretation and application on e-commerce, the area of e-commerce, and delivery trading area and find proper and practical ways, by virtue of constructive attitude of each actor, to reduce the encroachment of business territory and various conflicts caused by unrestricted delivery trading area. Conclusion and suggestion: The finding shows that unrestricted or uncontrolled delivery sales and business activity of franchisees cause encroachment of business territory and many conflicts among franchisees. And this matters also weaken the legislation of the law on the protection of the business territory and can be negative factors that disrupt the protection of identity and sound development of the franchise industry in the long run. Therefore, in the franchise business, it is necessary to take positive interpretation and application on e-commerce, the area of e-commerce, and delivery trading area and find proper and practical ways, by virtue of constructive attitude of each actor, to reduce the encroachment of business territory and various conflicts caused by unrestricted delivery trading area.

Heterogeneity in liver histopathology is associated with GSK-3β activity and mitochondrial dysfunction in end-stage diabetic rats on differential diets

  • While liver histopathology is heterogeneous in diabetes, the underlying mechanisms remain unclear. We investigated whether glycemic variation resulting from differential diets can induce heterogeneity in diabetic liver and the underlying molecular mechanisms. We generated end-stage non-obese diabetic model rats by subtotal-pancreatectomy in male Sprague-Dawley rats and ad libitum diet for 7 weeks (n = 33). The rats were then divided into three groups, and fed a standard- or a low-protein diet (18 or 6 kcal%, respectively), for another 7 weeks: to maintain hyperglycemia, 11 rats were fed ad libitum (18AL group); to achieve euglycemia, 11 were calorie-restricted (18R group), and 11 were both calorie- and protein-restricted with the low-protein diet (6R group). Overnight-fasted liver samples were collected after the differential diets together with sham-control (18S group), and histology and molecular changes were compared. Hyperglycemic-18AL showed glycogenic hepatopathy (GH) without steatosis, with the highest GSK-3β inactivation because of Akt activation during hyperglycemia; mitochondrial function was not impaired, compared to the 18S group. Euglycemic-18R showed neither GH nor steatosis, with intermediate GSK-3β activation and mitochondrial dysfunction. However, euglycemic-6R showed both GH and steatosis despite the highest GSK-3β activity and no molecular evidence of increased lipogenesis or decreased ApoB expression, where mitochondrial dysfunction was highest among the groups. In conclusion, heterogeneous liver histopathology developed in end-stage non-obese diabetic rats as the glycemic levels varied with differential diets, in which protein content in the diets as well as glycemic levels differentially influenced GSK-3β activity and mitochondrial function in insulin-deficient state.

The Dffects of Retinoids on CRABPII cRNA Induction amd Collagen Synthesis on Human Dermal Fibroblast

  • Retinoids are essential regulators of spithelial cell growth and celluar differentiation. They are also known to be effective in photoaging. It was reported that topical application of retinoic acid improves facial wrinkle carsed by collagen synthesis reduction in photodamaged skin. Collagen synthesis by retinoic acid may contribute to the wrinkle effacement. Since celluar retinoic acid binding protein II is slsctively induced in human skin and dermal fibroblasts in vitro by retinoic acid, this response can be used to mesure retinoids potency and activity. In order to know the activity of retinoids and their relations with collagen synthesis, we treated dermal fibroblasts with retinoids for 48 hours at 10-6-10-7M and measured CRABPII mRNA level by quantitative Nortern blotting. We also measured the rate of collagen systhesis by retinoids using 3-dimensional dermal equivalent. CRABPII mRNA level was increased 3-fold by retinoic acid, 2.1-fold by retinol and 1.4-fold by retinaldehyde. Collagen systhesis was increased 34% by all-trans retinioc acid, 26% by retinol, 17% by retinaldehyde and 7% by retinyl palmitate. From the above results, retinoids were found to be a potent indecers of CRABPII mRNA and collagen synthesis. Though retinoic acid was the most effective, its use has been restricted because of the side effects. Instead, retinol can be a best candidate in cosmetics for the treatment of photodamaged skin in terms of efficacy and safety.

Different Phosphate Transport in the Duodenum and Jejunum of Chicken Response to Dietary Phosphate Adaptation

  • Intestinal phosphate (Pi) absorption across the apical membrane of small intestinal epithelial cells is mainly mediated by the type IIb Na-coupled phosphate co-transporter (NaPi-IIb), but its expression and regulation in the chicken remain unclear. In the present study, we investigated the mRNA and protein levels of NaPi-IIb in three regions of chicken small intestine, and related their expression levels to the rate of net phosphate absorption. Our results showed that maximal phosphate absorption occurs in the jejunum, however the highest expression levels of NaPi-IIb mRNA and protein occurs in the duodenum. In response to a low-Pi diet (TP 0.2%), there is an adaptive response restricted to the duodenum, with increased brush border membrane (BBM) Na-Pi transport activity and NaPi-IIb protein and mRNA abundance. However, when switched from a low-(TP 0.2%) to a normal diet (TP 0.6%) for 4 h, there is an increase in BBM NaPi-IIb protein abundance in the jejunum, but no changes in BBM NaPi-IIb mRNA. Therefore, our study indicates that Na-Pi transport activity and NaPi-IIb protein expression are differentially regulated in the duodenum vs the jejunum in the chicken.

탄산탈수효소 모사를 이용한 이산화탄소 고정화 및 탄산칼슘 합성 (Carbonic Anhydrase Mimicry for Carbon Dioxide Fixation and Calcium Carbonate Mineralization)

  • Copper (II) and Nickel (II) mimic complexes of enzyme carbonic anhydrase were evaluated under ambient condition for carbon dioxide capture and conversion process. The synthesized complexes were characterized by ATR-FTIR and UV-DR spectroscopy. It was found that all the complexes have biomimetic activity towards $CO_2$ using para-nitrophenyl acetate (p-NPA) hydrolysis as the model reaction. Interestingly, the proper geometry obtained by the restricted orientation of tripodal N atoms in Cu (II) complex of 2,6-bis(2-benzimidazolyl) pyridine showed the highest activity (1.14 au) compared to others. The $CO_2$ bio-mineralization to $CaCO_3$ was carried out via in-vitro crystallization approach. Results indicate that the biomimetic complexes have a role in determining $CaCO_3$ morphology. The present observations establish a qualitative insight for the design of improved small-molecule catalysts for carbon capture.

육용종계 산란기의 영양소 요구량과 사료급여 체계 (utrient Requirements and Feeding System of Broiler Breeder Hens)

  • Results of experiments on the nutrient requirements and feeding system of broiler breeder hens were reviewed, and daily requirements of energy and protein were calculated using the prediction equations reported by Scott(1977) and NRC(1981). The experimental reports on daily ME needs of broiler breeder hens were ranged from 400 to 450 kcal, however, the ME needs of caged hens were 92~93% to those of floor-housed hens due to the difference of ME need for activity. The ME needs of broiler breeders decreased with increasing environmental temperature corresponding to a drop of 25 kcal per day for each 5˚C rise. About 80~90% of the daily ME needs were used for body rnaintenance and activity of hens. Experimental results on daily protein needs of broiler breeder hens were ranged from 18 to 22 g, however, calculated protein needs decreased as the BW gain and eggmass output decreased after peak production, and about 60~65% of the daily protein needs were used for egg production. In the current practice, broiler breeder hens are restricted in feed, and consume their daily allowance in the first 2 to 6 h after dawn. The results suggest that eggshell quality can be significantly improved in hens fed during the afternoon when shell calcification is initiated, with no adverse effect on laying rate and fertility of eggs.
