Aloe-Emodin-Mediated Photodynamic Therapy Attenuates Sepsis-Associated Toxins in Selected Gram-Positive Bacteria In Vitro

  • Sepsis is an acute inflammatory response that leads to life-threatening complications if not quickly and adequately treated. Cytolysin, hemolysin, and pneumolysin are toxins produced by gram-positive bacteria and are responsible for resistance to antimicrobial drugs, cause virulence and lead to sepsis. This work assessed the effects of aloe-emodin (AE) and photodynamic therapy (PDT) on sepsis-associated gram-positive bacterial toxins. Standard and antibiotic-resistant Enterococcus faecalis, Staphylococcus aureus, and Streptococcus pneumonia bacterial strains were cultured in the dark with varying AE concentrations and later irradiated with 72 J/cm-2 light. Colony and biofilm formation was determined. CCK-8, Griess reagent reaction, and ELISA assays were done on bacteria-infected RAW264.7 cells to determine the cell viability, NO, and IL-1β and IL-6 pro-inflammatory cytokines responses, respectively. Hemolysis and western blot assays were done to determine the effect of treatment on hemolysis activity and sepsis-associated toxins expressions. AE-mediated PDT reduced bacterial survival in a dose-dependent manner with 32 ㎍/ml of AE almost eliminating their survival. Cell proliferation, NO, IL-1β, and IL-6 cytokines production were also significantly downregulated. Further, the hemolytic activities and expressions of cytolysin, hemolysin, and pneumolysin were significantly reduced following AE-mediated PDT. In conclusion, combined use of AE and light (435 ± 10 nm) inactivates MRSA, S. aureus (ATCC 29213), S. pneumoniae (ATCC 49619), MDR-S. pneumoniae, E. faecalis (ATCC 29212), and VRE (ATCC 51299) in an AE-dose dependent manner. AE and light are also effective in reducing biofilm formations, suppressing pro-inflammatory cytokines, hemolytic activities, and inhibiting the expressions of toxins that cause sepsis.

EVALUATION OF MICROBIAL RISK IN SOIL AMENDED WITH ORGANIC FERTILIZERS FROM STABILIZED SWINE MANURE WASTE

  • This study evaluated microbial risk that could develop within soil microbial communities after amended with organic fertilizers from stabilized swine manure waste. For this purpose, we assessed the occurrences and competitiveness of antibiotic resistance and pathogenicity in soil microbial communities that were amended with swine manure wastes stabilized by a traditional lagoon fermentation process and an autothermal thermophilic aerobic digestion process, respectively. According to laboratory cultivation detection analysis, soil applications of the stabilized organic fertilizers resulted in increases in absolute abundances of antibiotic resistant bacteria and of two tested pathogenic bacteria indicators. The increase in occurrences might be due to the overall growth of microbial communities by the supplement of nutrients from the fertilizers. Meanwhile, the soil applications were found to reduce competitiveness for various types of antibiotic resistant bacteria in the soil microbial communities, as indicated by the decrease in relative abundances (of total viable heterotrophic bacteria). However, competitiveness of pathogens in response to the fertilization was pathogens-specific, since the relative abundance of Staphylococcus was decreased by the soil applications, while the relative abundance of Salmonella was increased. Further testes revealed that no MAR (multiple antibiotic resistance) occurrence was detected among cultivated pathogen colonies. These findings suggest that microbial risk in the soil amended with the fertilizers may not be critical to public health. However, because of the increased occurrences of antibiotic resistance and pathogenicity resulted from the overall microbial growth by the nutrient supply from the fertilizers, potential microbial risk could not be completely ruled out in the organic-fertilized soil samples.

해산물식품 중 식중독원인균의 오염패턴 및 저감화 방안 (Isolation of Major foodborne Pathogenic Bacteria from Ready-to-Eat Seafoods and Its Reduction Strategy)

  • 부산 및 경남일대에서 널리 유통되고 있는 생선회, 패류 등의 횟감류 213건을 대상으로 주요 식중독 원인균인 Salmonella spp., Staphylococcus aureus, Vibrio parahaemolyticus, Escherichia coli O157:H7, Bacillus cereus, Listeria monocytogenes 및 Campylobacter jejuni의 오염실태를 조사하였다. V. parahaermolyticus가 65건(30.5%), B. cerus가 21건(9.9%), S. aureus가 8건(3.8%) 및 기타 식중독균이 3건에서 검출되었다. 이를 월별 검출율로 비교해 보면, 7~10월에서 식중독균의 높은 검출율을 나타내었으며, 이러한 높은 검출율은 주로 V. parahaemolyticus에 기인된 것으로 조사되었다. 아울러 생선회에서의 검출율(28.9%) 보다는 패류 등의 기타 횟감류에서 더욱 높은 검출율(49.2%)을 나타내었다. V. parahaemolyticus의 경우 Ampicillin (96.9%), Amikacin (29.2%) 및 Tetracycline (27.7%)에 대해 내성을 나타내었으며, B. rereus의 경우는 Ampicillin (100%), Penicillin G (100%) 및 Rifampicin (71.4%)에 대해 높은 내성을 나타내었다. V. parahaemolyticus와 B. cereus의 생육은 보관온도 및 시간에 크게 영향을 받는 것으로 나타났으며, 수돗물 세척이 해산물에 잔존하는 V. parahaemolyticus의 균수를 크게 감소시키는 것으로 조사되어, 여름철 해산물 섭취로 인한 식중독 발생의 예방을 위해서 $10^{\circ}C$이하의 저온보관과 수돗물로써의 철저한 세척이 필수적인 것으로 판단되었다.

주요 임상 내성균에 대한 어성초 에탄올 추출물의 항균효과 (Antimicrobial Activity of Houttuynia cordata Ethanol Extract against Major Clinical Resistant Microorganisms)

  • 다제내성균의 증가는 사용할 항생제가 적거나 심지어 선택할 항생제가 없기 때문에 감염환자의 치료에 커다란 위협이 되고 있다. 최근 다양한 천연 추출물이 내성세균에서 항균효과 및 상승효과를 보인다는 보고가 많다. 어성초(Houttuynia cordata)는 항균, 항바이러스 및 항산화 효과를 보이는 전통적 약초이다. 이에 저자들은 임상의 주요 내성균에 대한 어성초의 항균효과를 알아보고, 이들의 적절한 항균효과 측정방법을 평가하고자 한다. 우선 현재 가장 많은 문제를 야기하는 주요 내성균인 methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Enterococcus faecium (VRE), carbepenem-resistant Acinetobacter baumannii(CRAB) 각각 10 균주를 선택하여, CLSI 기준으로 디스크 확산법 및 미량액체배지희석법으로 감수성 시험을 하였다. CLSI 기준으로 시행한 디스크 확산법에서 $4,096{\mu}g/mL$에서 $30,000{\mu}g/mL$ 까지의 농도에서 모든 균의 억제대는 관찰되지 않았다. 그러나 액체배지 희석법에서는 MRSA, VRE, CRAB의 $MIC_{90}$이 각각 $4,096{\mu}g/mL$, $8,192{\mu}g/mL$, $4,096{\mu}g/mL$를 보였다. 결론적으로 어성초는 MRSA, VRE, CRAB 등 주요 임상 내성균에서 우수한 항균효과를 보이며 천연물의 항균효과를 평가에 디스크 확산법보다 액체배지 희석법이 유용하다고 할 수 있다. 만약 디스크 확산법으로 항균효과를 측정하려면 천연물에 대한 균 접종 농도의 변경 등 새로운 기준이 요구된다.

솔잎과 녹차 추출물이 식중독세균의 생존에 미치는 영향 (Effect of Pine Needle and Green Tea Extracts on the Survival of Pathogenic Bacteria)

  • 솔잎과 녹차의 물추출물과 에탄올추출물을 액체배지에 0~2%(w/v)첨가하여 5종류의 식중독세균(Listeria monocytogenes, Staphylococcus aureus 196E, Aeromonas hydrophila, Escherichia coli O157:H7, Salmonella typhimurium)을 $10^{5}$~$10^{6}$ cells/ml 되게 접종하여 35$^{\circ}C$에서 24시간 배양하였다. 솔잎과 녹차의 추출물 종류와 농도에 따른 각 식중독세균에 대한 항균활성은 배양액의 생균수 변화를 측정하였으며 이 결과로부터 각 추출물의 최소저해농도와 최소사멸농도를 구하였다. 아울러 솔잎과 녹차의 추출물과 보존료인 sodium benzoate로서 각 세균에 대한 생육저해환의크기를 비교하였다. 솔잎추출물은 Gram 양성균(L. monocytogenes, S. aureus)과 Gram 음성균중 A. hydrophila에 대하여 0.4~l.6% 범위내에서 증식을 억제시켰으며 E. coli와 S. typhimurium은 솔잎추출물에 대하여 강한 내성을 나타내었다. 솔잎 추출물은 시험한 5균주에 대하여 사멸작용은 전혀 나타내지 않았다. 반면에 녹차추출물은 시험한 5균주 모두에 대하여 0.2~l.0% 범위내에서 증식을 억제시켰으며 L. monocytogenes를 제외한 모든 균주를 0.5~2.0% 범위내에서 사멸시키는 강한 항균활성을 나타내었다. 솔잎과 녹차추출물은 특히 S. aureus와 A. hydrophila에 대하여 보존료(sodium benzoate)보다도 강한 항균활성을 나타내었다. 전체적인 항균력의 크기는 녹차 에탄올추출물 > 녹차 물추출물 > 솔잎 에탄올추출물 > 솔잎 물추출물의 순이었다. 이 결과로 미루어 녹차추출물은 다양한 종류의 세균에 대하여 우수한 항균작용을 나타내는 천연 보존료로서의 활용가치가 있을 것으로 판단된다.다.

Activities of Essential Oils from Perilla frutescens var. acuta against Antibiotic-Susceptible and -Resistant Vibrio and Salmonella Species

  • We determined the inhibitory activity of the essential oil fraction obtained by steam distillation from the fresh and dried leaves of Perilla frutescens var. acuta against some pathogenic Salmonella and Vibrio spp. The activities of compounds isolated from the essential oils, apiol and myristicin, were also tested and the results were compared with those of the essential oil fraction. The Perilla essential oil fraction and its main components showed significant inhibition against antibiotic-susceptive and antibiotic-resistant strains of the tested Salmonella and Vibrio strains. Synergistic or additive effects were identified by combing the oils with ampicillin by checkerboard-titer tests. We conclude that essential oils from P. frutescens can be useful in the treatment of Salmonella and Vibrio infections and as safe additives to food materials for the prevention of contamination of food by these bacteria. This is especially important because of the rapid increase in antibiotic-resistant strains, which could cause severe symptoms in humans.

인천지역 식중독 환자에서 분리한 병원성 세균의 항생제 내성 및 다제 내성 양상 (Antimicrobial Resistance and Multi-Drug Resistance Patterns of Pathogenic Bacteria Isolated from Food Poisoning Patients in Incheon)

  • Antimicrobial resistance and multi-drug resistance patterns have been carried out on total of 210 isolated of Salmonella spp. and pathogenic E. coli isolated from food poisoning patients on January through December 2012 in Incheon, Korea. The highest percentage of antibiotics resistance was found to the following antimicrobial agents: tetracycline 43.8%, ampicillin 34.8%, nalidixic acid 23.8%, sulfamethoxazole/trimethoprim and chloramphenicol 12.4%, and ampicillin/sulbactam 11.4%. The highest percentage of resistance was 37.5% to ampicillin for Salmonella spp. and 59.0% to tetracycline for pathogenic E. coli. Overall the multidrug resistance rates of 1 drug was 26.2%, 2 drugs 9.0%, 3 drugs 9.5%, 4 drugs 7.1%, and 5 or more drugs 12.46%. The multi-drug (MDR) strains to four or more antimicrobial agents among the resistant organisms were quite high: 15.9% and 22.1% for Salmonella spp. and pathogenic E. coli, respectively. The study implies that limitation of unnecessary medication use is pertinent in order to maintaining the efficacy of drugs.

사료 첨가용 생균제를 위한 Probiotics 유산간균의 분리 및 동정 (Isolation and Identification of Probiotic Lactobacillus Isolates for Calf Meal Supplements)

  • 한우와 홀스타인의 분변으로부터 MRS 배지와 LAPT 배지를 이용하여 무작위 선발법으로 54균주의 유산균을 1차로 분리하였다. 1차로 분리된 54균주에 대해 내담즙성이 우수한 10균주를 분리한 다음 내산성을 조사한 결과 인공위액 pH 2.5에서 LS1, LS15 및 LL6 균주가 각각 66.5%, 82.6% 및 80.7%의 생존율을 나타내었다. Sal. typhimurium, Sta. aureus 및 Cl. perfringens의 병원균에 대해 가장 큰 항균력을 보인 균주는 LL6와 LL7이었다. API CHL kit로 동정한 결과 LS1, LS2 및 LMI 균주는 모두 L. fermentum, LL6와 LL7 균주는 L. acidophilus, LS3 균주는 L. plantarum으로 각각 동정되었고, 나머지 4균주는 Lactobacillus spp. 로 동정되어 분리된 10균주 모두 안전성 있는 유산간균임을 확인하였다. 10종류의 항생제에 대한 내성을 조사한 결과 ampicillin, amoxicillin과 erythromycin에 대해서는 감수성이 있으나 colistin과 ciprofloxacin에 대해 모두 내성을 나타내었다. LB1, LL6 및 LL7 균주는 gentamicin과 neomycin에 대해 내성을 보여 주었다. 분리 동정된 균주 중에 내산성, 내담즙성 및 병원성 균에 대한 특성을 기준으로 판단할 경우 probiotic 유산균으로 사용 가능성이 높은 것은 LL6인 L. acidophilus로 나타났다.

가금유래 주요병원성세균의 분리와 분리균주에 대한 약제감수성조사 (Isolation of the Pathogenic Bacteria from Chicken and Antimicrobial Drug Sensitivity of the Strain Isolated)

  • A total of 1503 specimens were submitted to the Poultry Disease Diagnostic Service Laboratory during the year 1966 and 1978. The most frequently diagnosed diseases in order of prevalence were avian mycoplasmosis, staphylococcosis, colibacillosis, salmonellosis and pullorum disease, the percentages of the conditions being 24.6%, 20.0%, 18.0%, 12.6% and 6.4%, respectively, The drug resistance of pathogenic mirnoorganisms isolated during the year 1978 from chicken with colicabacillosis, staphylococcosis or salmonellosis were investigated by the use of disc diffusion technique, the results being as follow. 1) Drug resistance of 63 strains of Escherichia coli More than 95% of the strains tested were sensitive to colistin and gentamicin. The percentages of strains sensitive to kanamycin, chloramphenicol, ampicillin and nitrofurantoin were 66.7%, 60.3%, 60.3% and 47.6%, respectively. Majority of the strains were highly resistant to streptomycin and tetracyline. All the strains were resisistant to bacitracin lincomycin, oleandomycin, penicillin and erythromycin. All the strains tested were resistant to more than two among 10 drugs in common use such as penicillin, erythromycin, streptomycin, tetracycline, neomycin, chloramphenicol, kanamycin, ampicillin and gentamicin, and 27 different resistance patterns were noted. The most frequent multiple resistance pattern was PC, EM, SM and TC (11.1%). 2) Drug resistance of 48 strains of Salmonella More than 95% of the strains tested were sensitive to colistin, gentamicin ana ampicillin. The percentages of st rains sensitive to kanamycin, tetracycline, neomycin and nitrofurantoin were 81,3%, 79%, 72.9%, and 68.0% respectively. None of them was sensitive to streptomycin, oleandomycin, erythromycin, lincomycin and bacitracin. All the strains were resistant to more than one among 7 drugs in common use such as streptomycin, erythromycin, neomycin, tetracycline, kanamycin, ampicillin and gentamicin. The most frequent resistance pattern was SM and EM(66.7%). 3) Drug resistance of 54 strains of Staphylococci All the strains tested were sensitive to gentmaicin, kanamycin and cephalothin. Majority of them were highly sensitive to bacitracin, methicillin, nitrofurantoin and chloramphenicol. The Percentages of strains sensitive to streptomycin, ampicillin, lincomycin and tetracycline were 66.7%, 55.6%, 44.4% and 27.8%, respectively. Among them, 51 strains were resistant to more than one among 11 drugs in common use such as tetracycline, lincomycin, ampicillin, penicillin, streptomycin, erythromycin, neomycin, oleandomycin, chloramphenicol, methicillin and bacitracin, and thirty one different resistance patterns were noted.

Biological Activities of Essential Oils from Angelica tenuissima Nakai

  • The current study was conducted to evaluate the antibacterial and antioxidant activities of the essential oil fraction from the roots of Angelica tenuissima Nakai and its main components. We extracted the essential oil fraction from the roots of A. tenuissima using steam distillation and isolated its main components. Their antibacterial activities were determined by broth dilution test against food-borne pathogenic bacteria. Antioxidant activities were evaluated by DPPH-scavenging assay and reducing-power test. Also tested was their ability to inhibit the growth of two gastrointestinal cancer cell lines, Caco-2 and MKN-45. The A. tenuissima oil fraction and its main components, ligustilide and butylidene phthalide exhibited marked inhibitory effects against most of the tested antibiotic-susceptible and antibiotic-resistant bacterial strains with minimum inhibiting concentrations (MICs) from $0.21{\pm}0.08$ to $3.60{\pm}0.89mg/ml$. They also showed growth-inhibiting activity against Caco-2 and MKN-45 cells. The oil fraction showed significant antioxidant activities in DPPH radical scavenging assay and reducing-power test. Taken together, A. tenuissima essential oil could be used as a safe additive for preventing food contamination by pathogenic bacteria. Additionally, its antioxidative activity and the ability to inhibit gastrointestinal carcinoma cell lines could increase its value for functional foods and prevention of cancer.
