Monitoring of Patulin Content in Domestic Apples of Processing and Market Products (국내산 가공용 사과 및 시판제품의 Patulin 함량 모니터링)

  • This study analyzed 102 samples from 24 local areas to monitor patulin contamination of apples for processing by the areas. According to the analysis, patulin was detected in 47 samples among totally 92 ones and those of 15 samples from 12 areas were found to be 50 or more ppb so measures to deal with the patulin contamination were necessary. As ways to reduce residues of patulin, when 200 ppm ascorbic acid were treated with they was decreased by 93.4% to 12.89 ppb, and when 100 ppm activated carbon was added they were declined by 95.8% to 2.68 ppb. Treatment with pectinase did not show any significant difference and effects of temperature was not considerable under established sterilizing conditions(90, 105, 120, 135 and $150^{\circ}C$/25 sec). In conclusion, treatment with ascorbic acid and activated carbon on apple juice was found to decrease residues of patulin but more systemic researches were needed to determine it in the future.

A Study on the Development of Regional Low-flow runoff Model at the Youngsan River Basin (영산강 유역의 지역갈수 유출 모형 개발에 관한 연구)

  • The present day, interest in low flow statistics has been increased, primarily because of the increase in water demands and the contamination of water quality and the limitation of water resource development and the annual water supply stability. The model for estimating low flow statistics is points of this study chosen catchments in the Youngsan basin. For the purpose of this study. the low-flow discharge at 23 points, where absent gauging station is unmeasured and the data is used for the model. Finally, the regional formula to estimate the low flow statistics at the unmeasured point is proposed.

Adsorption and Leaching Characteristics of Ionic Pesticides in the Soils of Jeju Island, Korea (제주도 토양 중 이온계 농약의 흡착 및 용탈 특성)

  • The adsorption and leaching characteristics of five ionic pesticides including four acidic pesticides (2,4-D, dicamba, MCPA, and MCPP) and one amphoteric pesticide (imazaquin) in agricultural soils were investigated. Soils around spring waters that were heavily affected by pesticide run-off and soils around wells considering the regional characteristics in Jeju Island were collected at 24 stations. The Freundlich constant, $K_F$ value, which is a measure of the adsorption capacity, decreased in the order of 2,4-D > MCPA > MCPP > dicamba > imazaquin. The adsorption capacity of these ionic pesticides decreased with increasing pH owing to the effects of ionization of pesticides and different ionizable functional groups of soils. The leaching of ionic pesticides in the soil column showed a reverse relationship with their adsorption in soils, namely, the ionic pesticides were leached more quickly for the pesticides with lower adsorption capacity. The groundwater contamination potential of the ionic pesticides was evaluated in the order of imazaquin > MCPA > MCPP > dicamba > 2.4-D according to the groundwater ubiquity score based on soil Koc and the half-life of the pesticide.

Isolation of Major foodborne Pathogenic Bacteria from Ready-to-Eat Seafoods and Its Reduction Strategy (해산물식품 중 식중독원인균의 오염패턴 및 저감화 방안)

  • The contamination frequency of major foodborne pathogenic bacteria was investigated from 213 seafood samples including sliced raw fish and shellfish in Busan and CyeongNam province area. Tested microorganisms were Salmonella spp. Staphyloroccus aureus, Vibrio parahaemolyticus, Escherichia coli O157:H7, Bncillus cereus, Listeria monocytogenes and Campylobacter jejuni. The frequency of isolated microorganisms was V. parahaemolyticus (30.5%), B. cereus (9.9%), S. aureus (3.8%) and other pathogenic bacteria (1.4%). from July to October, total isolation rates were greater than 50% and V. parahaemolyticus was dominant among the microorganisms isolated. The bacteria isolation rate (49.2%) in raw seafoods including shellfishes was higher than one (28.9%) in sliced raw fish. V. parahaemelyticus isolates were resistant to ampicillin (96.9%), amikacin (29.2%) and tetracycline (27.7%), and B. cereus isolates were resistant to ampicillin (100%), Penicillin G (100%), rifampicin (71.4%) and tetracycline (14.3%). The growth of V. parahaemolyticus and B. rereus was greatly inhibited below $10^{\circ}C$, but increased at ambient temperature. Washing seafood with tap water showed to reduce total count of remaining V. parahaemolyticus. Thus temperature control under $10^{\circ}C$, sufficient washing and prompt eating appeared to reduce the risk of food poisoning by these bacteria in seafoods.

  • This thesis is to analyze characteristic of sustainable development in the Nak-dong River. The results are summarized in the followings. 1) The principles which did not consider the equilibrium between development and environment, ecosystem, security. precautionary measure, equity could devise a plan, changes of economic land area, sand & gravel extractions from the riverbed, the river contamination cleanup, dam build 2) The rules which disregard polluter pay and opening participation in public affairs could input of too much province budget and manpower, project team. The measures to solve these problems could be proposed expansion of environmental infrastructure, sustainable sand & gravel extractions from the riverbed, fortifying the assistance of the residents who live in Dam construction site and the peripheral area of Dam, lower local fund rates in national projects, and the creation of management organization.

Development and Application of a Method for Rapid and Simultaneous Determination of Three β-agonists (Clenbuterol, Ractopamine, and Zilpaterol) using Liquid Chromatography-tandem Mass Spectrometry

  • ${\beta}$-agonists are anabolic compounds that promote fat loss and muscle gain, and their administration to livestock may provide economic benefits by increasing growth rate and feed efficiency. For these reasons, ${\beta}$-agonists are also commonly added to livestock feed as growth promoters. This can introduce a significant risk of secondary human poisoning through intake of contaminated meat. A new method for the simultaneous determination of three ${\beta}$-agonists (clenbuterol, ractopamine, and zilpaterol) was developed in this study and applied to various meat samples. The limits of quantification, derived through a validation test following Codex guidelines, were $0.2{\mu}g/kg$ for clenbuterol and zilpaterol, and $0.4{\mu}g/kg$ for ractopamine. The average recoveries for clenbuterol, ractopamine, and zilpaterol ranged from 109.1% to 118.3%, 95.3% to 109.0%, and 94.1% to 120.0%, respectively. The recovery and coefficient of variation (CV) values fell within the acceptable range according to the Codex guidelines. This method reduced the analysis time without decreasing detection efficiency by modifying the pretreatment steps. This method could be utilized to manage the safety of imported meat products from countries where zilpaterol use is still permitted, thereby improving public health and preventing ${\beta}$-agonist poisoning due to secondary contamination.

  • As Standards and Specifications of the Saengsik-classes has been established since 2005 by KFDA. The microbial Standards and Specifications of the Saengsik-classes is as follows; no detection in Escherichia coli, colony forming unit less then 1,000/g in Bacillus cereus, colony forming unit less then 100/g in Clostridium perfringens respectively. Contamination levels of Total aerobic bacteria, Escherichia coli, Bacillus cereus and Clostridium perfringens in Saengsik-classes were monitored. Total aerobic bacteria counts in Saengsik-classes was $1{\times}10^1{\sim}5.3{\times}10^7cfu/g$, for Bacillus cereus $1{\times}10^2{\sim}9{\times}10^2cfu/g$, for Clostridium perfringens $1{\times}10^1cfu/g$. Escherichia coli, was not isolated from all Saengsik-classes. Thess results will provide information for introduction of HACCP system to ensure microbial safety of Saengsik-classes.

Monitoring Bacillus cereus and Aerobic Bacteria in Raw Infant Formula and Microbial Quality Control during Manufacturing (영.유아용 식품원료의 Bacillus cereus와 일반세균 모니터링 및 제조공정 중 미생물 품질제어)

  • The purpose of this study was to examine the presence of Bacillus cereus, aerobic bacteria and coliforms in the raw material of infant formulas and investigate the manufacturing process in terms of microbial safety. Among ten kinds of raw infant formula material samples (n=20), Bacillus cereus appeared in two (n=4). Aerobic bacteria were not detected in raw infant formula material or maximum 4.15 log CFU/g. Eleven species of aerobic bacteria were isolated and 76% of them were Sphingomonas paucimobilis, Pseudomonas fluorescens, Rhizobium radiobactor, or Stenotrophomonas maltophilia. A Pearson's correlation analysis revealed that the most influential factors for detecting Bacillus cereus were aerobic bacteria and coliforms. In other words, when the measured values of aerobic bacteria and coliforms were higher, the possibility that Bacillus cereus would appear increased. In a regression model to predict Bacillus cereus, the rate of appearance was correlated with aerobic bacteria and coliforms, and its contribution rate for effectiveness was 86%. Improving microbial quality control by pasteurization, spray dry, popping and extrusion resulted in a decrease in the numbers of Bacillus cereus, aerobic bacteria and coliforms in the raw materials. The results suggest that a hazard analysis and critical control point system might be effective for reducing microbiological contamination.

Determination of Microbial Contamination in the Process of Rice Rolled in Dried Laver and Improvement of Shelf-life by Gamma Irradiation (김밥 제조공정에서의 미생물 오염도 평가 및 감마선 조사를 이용한 김밥의 보존안정성 향상)

  • Determination of regional microbial contamination in the process of rice rolled in dried laver (Kimbab) and effects of gamma irradiation on the improvement of hygienic quality and shelf stability were investigated. Total aerobic bacterial distribution of raw materials of Kimbab were; 10$^{6}$ ∼10$^{7}$ CFU/g in dried laver, 10$^3$ CFU/g in cucumber and below 10 CFU/g in steamed rice, ham, fried egg, and salted radish. Total coliform bacteria were 10$^3$ CFU/g in dried laver and detected below detection limit (10 CFU/g) in other raw materials. And it was arithmetically calculated that the levels of total aerobic bacteria and coliform bacteria in Kimbab does not exceed 10$^{5}$ CFU/g and 10$^1$ CFU/g under the aseptic process, respectively. However, microbial contamination levels in just prepared Kimbab in a market were about 10$^{6}$ CFU/g of total aerobic and coliform bacteria. Therefore, it was considered that microbial contamination of Kimbab is mainly originated from environmental uptake during the preparation. The representative media for putrefying bacterial growth were steamed rice. Coliform bacteria were mainly increased in ham and fried egg during storage. The bacteria in dried laver were radio-resistant and survived at 3 kGy of gamma irradiation. Coliform bacteria on EMB agar plate were eliminated at the dose of 2 kGy. The sensory acceptability of 2 kGy irradiated Kimbab was stable and the Kimbab can be preserved for 24 hour at 15$^{\circ}C$. Therefore, it was considered that optimal irradiation dose for radicidation of Kimbab was 2 kGy.

Development of a PCR Diagnostic System for Iris yellow spot tospovirus in Quarantine

  • Iris yellow spot virus (IYSV) is a plant pathogenic virus which has been reported to continuously occur in onion bulbs, allium field crops, seed crops, lisianthus, and irises. In South Korea, IYSV is a "controlled" virus that has not been reported, and inspection is performed when crops of the genus Iris are imported into South Korea. In this study, reverse-transcription polymerase chain reaction (RT-PCR) and nested PCR inspection methods, which can detect IYSV, from imported crops of the genus Iris at quarantine sites, were developed. In addition, a modified positive plasmid, which can be used as a positive control during inspection, was developed. This modified plasmid can facilitate a more accurate inspection by enabling the examination of a laboratory contamination in an inspection system. The inspection methods that were developed in this study are expected to contribute, through the prompt and accurate inspection of IYSV at quarantine sites to the plant quarantine in South Korea.
