Effects of Yupoongyangyeong-tang(YYT) on the Changes of Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats (유풍양영탕(愈風養營湯)이 정상 흰쥐의 국소 뇌혈류량 및 평균혈압에 미치는 영향(影響))

  • Objectives: The present study was carried out to investigate the effects of Yupoongyangyeong-tang(YYT) on changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in rats. In addition, the author also investigated action mechanisms of YYT on changes in rCBF and MABP. Methods and Results : In this study, treatment with YYT elevated rCBF in a dose-dependent manner, but MABP levels were elevated only in the 10 mg/ml treatment group. Pre-treatment with indomethacin. an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. Pre-treatment with methylene blue, an inhibitor of guanylate cyclase, inhibited increase of rCBF induced by YYT, too. In addition. pre-treatment with indomethacin also inhibited increase of MABP. However, pre-treatment with methylene blue did not affect MABP levels. Conclusion : These results suggest that YYT is useful to treat patients with diseases related to cerebral ischemia, because YYT can increase rCBF. In addition, the mechanisms are thought to be related to both cyclooxygenase and guanylate cyclase.

Effect of Nodus Nelumbinis Rhizomatis Extract on the Regulation of Regional Cerebral Blood Flow in Rats (흰쥐에서 우절(藕節) 추출물의 국소 뇌혈류량 조절 효과)

  • Objectives : In brain disorders such as ischemic stroke, the final outcome depends largely on the duration and the degree of the ischemia as well as the susceptibility of various cell types in the affected brain region. In the present study, the effects of Nodus Nelumbinis Rhizomatis Extract(NNRe) were tested for the anti-oxidative action of rCBF. Methods : Regional cerebral blood flow(rCBF) were determined by LDF methods. LDF allows for real time, noninvasive, continuous recordings of local CBF. The LDF method has been widely used to trace hemodynamic changes in the superficial or the deep brain structures in experimental stroke research. Results : NNRe treatment showed no change on rCBF in methylene blue, ODQ and L-NNA pretreated rats. 120 minutes of MCAO and followed reperfusion, 0.1% concentration of NNR treatment improved the altered cerebral hemodynamics of cerebral ischemic by increasing rCBF. Conclusions : The ischemia/reperfusion induced oxidative stress may have contributed to cerebral damage in rats, and the present study provides clear evidences for the beneficial effect of NNR on ischemia/reperfusion induced brain injury.

  • This Study was designed to investigate the effects of Patholobi Caulis on the change of regional cerebral blood flow (rCBF) and blood Pressure (MABP) in normal and Cerebral ischemic rats. And, this Study was designed to investigate the inhibition of lactate dehydrogenase (LDH) activity in neuronal cells. The results were as follows : In normal rats, Patholobi Caulis significantly increased rCBF in a dose-dependent manner, and MABP was somewhat increased. In ischemia rats, rCBF was significantly and stably increased by Patholobi Caulis (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. Patholobi Caulis significantly inhibited LDH activity in neuronal cells. It was suggested that Patholobi Caulis had an anti-ischemic effect through the improvement of cerebral hemodynamics and inhibitive effect on the brain damage.

  • Oriental medicine prescription has been used for the treatment of various clinical symptoms associated with cerebral apoplexy. However, single herb drug does not used cerebral apoplexy and there mechanisms of action have not been defined, and it is not yet known what effects they have on the hemodynamics of cerebral circulation. The study was aimed to investigate the effect batryticatus bombycis(BB) on the vascular systems including changes in blood pressure (BP), and regional cerebral blood flow (rCBF) and of male Sprague-Dawely rats. The changes in rCBF were determinated by laser-Doppler flowmetry. 1. Blood pressure was not affected by BB in rats. 2. rCBF was increased by BB in a dose-dependent manner. 3. Pretreatment with methylene blue(1mg/kg), and propranolol(1mg/kg) did not inhibited BB induced increased in rCBF. 4. Pretreatment with propranolol(1mg/kg) was increased by BB in a dose-dependent manner in blood perssure. 5. Pretreatment with ODQ($10{\mu}g/kg$) and L-NNA(1mg/kg) were inhibited BB induced increased in rCBF. 6. Pretreatment with L-NNA(1mg/kg) was increased rCBF in a dose-dependent manner. These results suggest that BB causes a diverse response of blood pressure and regional cerebral blood flow(rCBF). The increased in rCBF is also mediated by nitric oxide synthease and guanylate cyclase.

  • Yongyanggaktang has been used in oriental medicine as a therapeutic agent for cerebral disease and eclampsia for many years. The purpose of this study was to determine the effect of Yongyanggaktang and its exception of Cornu Saigae Tataricae(羚羊角) in YYG about regional cerebral blood flow(rCBF) in rats. 1. rCBF was increased by Yongyanggaktang in dose dependently. 2. rCBF was increased by exception of Cornu Saigae Tataricae(羚羊角) in YYG at high dosages. 3. rCBF was decreased by Yongyanggaktang pretreated propranolol. 4. L-NNA did not affect rCBF. 5.ODQ did not affect rCBF. Above the results. Yongyanggaktang relates to the symphathetic action.

  • The study was aimed to investigate the effect cuscutae semen(CS) on the vascular systems including changes in blood pressure (BP), regional cerebral blood flow(rCBF) and pial arteriolar diameter of male Sprague-Dawely rats. The changes in rCBF were determinated by laser-Doppler flowmetry, and the changes in diameter of pial arteriole were measured through a closed crainal window. 1. Blood pressure was not affected by CS in rats. 2. rCBF was increased by CS in a dose-dependent manner. 3. Pretreatment with methylene blue(Img/kg), and propranolol(1mg/kg) significantly inhibited CS induced increased in rCBF. 4. Pretreatment with indomethacin(1mg/kg) did not inhibited CS induced increased in rCBF. 5. Pial arterial diameter was increased by CS in a dose-dependent manner. These results suggest that CS causes a diverse response of blood pressure, regional cerebral blood flow(rCBF), and pial arteral diameter. The increased in rCBF is also mediated by adrenergic ${\beta}-receptor $ and guanylate cyclase.

Study on the Effects of Chukdamtanggamibang on blood pressure, regional cerebral blood flow(rCBF) and smooth muscle (척담탕가미방이 혈압, 뇌혈류량 및 평활근에 미치는 효능에 관한 연구)

  • This study was aimed to investigate the effect Chukdamtanggamibang on the vascular systems including changes in blood pressure and regional cerebral blood flow(rCBF) of male Sprague-Dawely rats, contractile force of guinea pig`s tracheal smooth muscle and abdominal aorta and femoral artery in rabbits. Blood pressure was not affected by Chukdamtanggamibang in rats. rCBF was significantly increased by Chukdamtanggamibang in a dose-dependent manner. Contractile force of isolated guinea pig`s tracheal smooth muscle evoked by His ({TEX}$ED_{50}${/TEX}) were inhibited significantly by Chukdamtanggamibang. Propranolol, indomethacin and methylene blue did not significantly alter the inhibitory effect of Chukdamtanggamibang. Contractile force of isolated rabbit`s abdominal aorta and femoral artery evoked by NE ({TEX}$ED_{50}${/TEX}) were inhibited significantly by Chukdamtanggamibang. ODQ and L-NNA significantly attenuated the inhibitory effects of Chukdamtanggamibang in abdominal aorta, whereas propranolol did not significantly alter the inhibitory effect of Chukdamtanggamibang. These results indicate that Chukdamtanggamibang can relax hitamine-induced contraction of guinea pig`s tracheal smooth muscle and that this inhibition involves, in part, the relation to the soluble guanylyl cyclase synthesis and nitric oxide (NO) synthesis.

Quantitative Analysis of Regional Cerebral Blood Flow using $^{99m}Tc-HMPAO$ SPECT in Parkinson's Disease (파킨슨병에서 $^{99m}Tc-HMPAO$ SPECT를 이용한 국소뇌혈류의 정량적 분석)

  • Regional cerebral blood flow were measured in 10 patients with Parkinson's disease and 12 normal persons using $^{99m}Tc-HMPAO$ SPECT. Reconstructed images were interpreted qualitatively and were compared with those findings of CT. For the quantitative analysis, six pairs of region of interest matched with the perfusion territories of large cerebral arteries and cerebellar hemisphere were determined. From the count values, indices showing the degree of asymmetry between right and left cerebral or cerebellar hemisphere, cerebral asymmetry index (ASI) and percent index of cerebellar asymmetry (PIA), and an index showing change of each region, region to cerebellum ratio (RCR) were obtained. ASI of normal persons and patients were $0.082{\pm}0.033$ and $0.108{\pm}0.062$, respectively and PIA were $-0.4{\pm}0.7%$ and $-0.7{\pm}1.0%$, respectively, which showed no statistically significant difference between normal persons and patients. Among 10 RCR's, those of both regions of basal ganglia and both regions of anterior cerebral artery were significantly reduced. We concluded that the most significant reduction of regional cerebral blood flow in patients with Parkinson's disease was observed in the regions of basal ganglia and in the regions of anterior cerebral artery, and the degree of change in hemispheric blood flow was similar in both hemisphere.

  • SPECT of regional cerebral blood flow was performed using $^{99m}Tc-HMPAO$ in 28 patients with cerebrovascular disease and quantitative analysis was done. Cerebral asymmetry index and percent index of asymmetry of cerebellar hemisphere of patients with cerebral infarction or ischemia were $0.764{\pm}0.576$ and $-5.6{\pm}7.1%$ and those of intracranial hemorrhage was $0.416{\pm}0.251$ and $-2.5{\pm}4.1%$ respectively. According to PIA of cerebellar hemisphere, 12 patients showed crossed cerebellar diaschisis. $^{99m}Tc-HMPAO$ SPECT seemed to be a useful tool for the evaluation of regional cerebral blood flow.

  • The study was designed to investigate the effects of Palmul-Tang(PMT) on the change of cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD) and mean arterial blood pressure(MABP)] in normal and cerebral ischemic rats. The change of rCBF and MABP were determinated by laser-doppler flowmetry(LDF), and the change of PAD was determinated by video-microscopy. The results in normal rats were as follows ; PMT significantly increased rCBF and PAD in a dose-dependent, and PMT increased MABP in a dose-dependent. This results were suggested that PMT significantly increased rCBF by dilating PAD. The results in cerebral ischemic rats were as follows ; Both rCBF and PAD were significantly and stably increased by PMT(10㎎/㎏, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. The present authors thought that PMT had an anti-ischemic effect through the improvement of cerebral hemodynamics.
